In postnatal tissues, angiogenesis occurs in nontumoral conditions on appropriate stimuli. In the nervous tissue, hypoxia, neural graft, increased neural function, and synaptic activity are associated with neoangiogenesis. We have investigated the occurrence of neoangiogenesis in the superior cervical ganglia (scg) of newborn rats treated for 8–21 days with 6-hydroxy-dopamine (6-OHDA), nerve growth factor (NGF), or 6-OHDA + NGF. The two latter treatments induced a significant increase in scg size. However, the increase after combined treatment far exceeded that of NGF alone. Similarly, histological and histochemical analysis revealed neuronal hypertrophy and endothelial cell hyperplasia associated with stromal hypertrophy (as described by laminin immunostaining) and increased vascular bed (as revealed by platelet/endothelial cell adhesion molecule-1 immunostaining) in 6-OHDA + NGF-treated pups. NGF, either alone or associated with 6-OHDA, also induced a significant up-regulation of NADPH diaphorase, neuronal nitric oxide synthase, and vascular endothelial growth factor expression in scg neurons. The present investigation suggests that the increase of scg size induced by NGF and 6-OHDA + NGF is associated with neoangiogenesis, and that the induction of vasoactive and angiogenic factors in neurons represents a further and previously undisclosed effect of NGF.

Nerve growth factor (NGF) is a polypeptide which, in addition to its effect on nerve cells, is believed to play a role in inflammatory responses and in tissue repair. Because fibroblasts represent the main target and effector cells in these processes, to investigate whether NGF is involved in lung and skin tissue repair, we studied the effect of NGF on fibroblast migration, proliferation, collagen metabolism, modulation into myofibroblasts, and contraction of collagen gel. Both skin and lung fibroblasts were found to produce NGF and to express tyrosine kinase receptor (trkA) under basal conditions, whereas the low-affinity p75 receptor was expressed only after prolonged NGF exposure. NGF significantly induced skin and lung fibroblast migration in an in vitro model of wounded fibroblast and skin migration in Boyden chambers. Nevertheless NGF did not influence either skin or lung fibroblast proliferation, collagen production, or metalloproteinase production or activation. In contrast, culture of both lung and skin fibroblasts with NGF modulated their phenotype into myofibroblasts. Moreover, addition of NGF to both fibroblast types embedded in collagen gel increased their contraction. Fibrotic human lung or skin tissues displayed immunoreactivity for NGF, trkA, and p75. These data show a direct pro-fibrogenic effect of NGF on skin and lung fibroblasts and therefore indicate a role for NGF in tissue repair and fibrosis.

Nerve growth factor (NGF) serum levels were measured in 49 patients with asthma and/or rhinoconjunctivitis and/or urticaria-angioedema. Clinical and biochemical parameters, such as bronchial reactivity, total and specific serum IgE levels, and circulating eosinophil cationic protein levels, were evaluated in relation to NGF values in asthma patients. NGF was significantly increased in the 42 allergic (skin-test- or radioallergosorbent-test-positive) subjects (49.7 +/- 28.8 pg/ml) versus the 18 matched controls (3.8 +/- 1.7 pg/ml; P < 0.001). NGF levels in allergic patients with asthma, rhinoconjunctivitis, and urticaria-angioedema were 132.1 +/- 90.8, 17.6 +/- 6.1, and 7.6 +/- 1.8 pg/ml (P < 0.001, P < 0.002, and P < 0.05 versus controls), respectively. Patients with more than one allergic disease had higher NGF serum values than those with a single disease. When asthma patients were considered as a group, NGF serum values (87.6 +/- 59.8 pg/ml) were still significantly higher than those of control groups (P < 0.001), but allergic asthma patients had elevated NGF serum levels compared with nonallergic asthma patients (132.1 +/- 90.8 versus 4.9 +/- 2.9 pg/ml; P < 0.001). NGF serum levels correlate to total IgE serum values (rho = 0.43; P < 0.02). The highest NGF values were found in patients with severe allergic asthma, a high degree of bronchial hyperreactivity, and high total IgE and eosinophil cationic protein serum levels. This study represents the first observation (that we know of) that NGF is increased in human allergic inflammatory diseases and asthma.

We have studied the neuropathological characteristics of the brain of rats receiving daily intracerebroventricular administration of freshly dissolved human immunodeficiency virus type 1 recombinant protein gp120 (100 ng per rat per day) given for up to 14 days. Histological examination of serial brain sections revealed no apparent gross damage to the cortex or hippocampus, nor did cell counting yield significant neuronal cell loss. However, the viral protein caused after 7 and 14 days of treatment DNA fragmentation in 10% of brain cortical neurons. Interestingly, reduced neuronal nitric oxide synthase (NOS) expression along with significant increases in nerve growth factor (NGF) were observed in the hippocampus, where gp120 did not cause neuronal damage. No changes in NGF and NOS expression were seen in the cortex, where cell death is likely to be of the apoptotic type. The present data demonstrate that gp120-induced cortical cell death is associated with the lack of increase of NGF in the cerebral cortex and suggest that the latter may be important for the expression of neuropathology in the rat brain. By contrast, enhanced levels of NGF may prevent or delay neuronal death in the hippocampus, where reduced NOS expression may be a reflection of a subcellular insult inflicted by the viral protein.

The present-day condition of bipolar glaciation characterized by rapid and large climate fluctuations began at the end of the Pliocene with the intensification of the Northern Hemisphere continental glaciations. The global cooling steps of the late Pliocene have been documented in numerous studies of Ocean Drilling Program (ODP) sites from the Northern Hemisphere. However, the interactions between oceans and between land and ocean during these cooling steps are poorly known. In particular, data from the Southern Hemisphere are lacking. Therefore I investigated the pollen of ODP Site 1082 in the southeast Atlantic Ocean in order to obtain a high-resolution record of vegetation change in Namibia between 3.4 and 1.8 Ma. Four phases of vegetation development are inferred that are connected to global climate change. (1) Before 3 Ma, extensive, rather open grass-rich savannahs with mopane trees existed in Namibia, but the extension of desert and semidesert vegetation was still restricted. (2) Increase of winter rainfall dependent Renosterveld-like vegetation occurred between 3.1 and 2.2 Ma connected to strong advection of polar waters along the Namibian coast and a northward shift of the Polar Front Zone in the Southern Ocean. (3) Climatically induced fluctuations became stronger between 2.7 and 2.2 Ma and semiarid areas extended during glacial periods probably as the result of an increased pole-equator thermal gradient and consequently globally enhanced atmospheric circulation. (4) Aridification and climatic variability further increased after 2.2 Ma, when the Polar Front Zone migrated southward and the influence of Atlantic moisture brought by the westerlies to southern Africa declined. It is concluded that the positions of the frontal systems in the Southern Ocean which determine the locations of the high-pressure cells over the South Atlantic and the southern Indian Ocean have a strong influence on the climate of southern Africa in contrast to the climate of northwest and central Africa, which is dominated by the Saharan low-pressure cell.

The role of p75 neurotrophin receptor (p75(NTR)) in mediating cell death is now well charaterized, however, it is only recently that details of the death signaling pathway have become clearer. This review focuses on the importance of the juxtamembrane Chopper domain region of p75(NTR) in this process. Evidence supporting the involvement of K+ efflux, the apoptosome (caspase-9, apoptosis activating factor-1, APAF-1, and Bcl-(xL)), caspase-3, c-jun kinase, and p53 in the p75(NTR) cell death pathway is discussed and regulatory roles for the p75(NTR) ectodomain and death domain are proposed. The role of synaptic activity is also discussed, in particular the importance of neutrotransmitter-activated K+ channels acting as the gatekeepers of cell survival decisions during development and in neurodegenerative conditions.

Glacial-interglacial fluctuations in the vegetation of South Africa might elucidate the climate system at the edge of the tropics between the Indian and Atlantic Oceans. However, vegetation records covering a full glacial cycle have only been published from the eastern South Atlantic. We present a pollen record of the marine core MD96-2048 retrieved by the Marion Dufresne from the Indian Ocean ~120 km south of the Limpopo River mouth. The sedimentation at the site is slow and continuous. The upper 6 m (spanning the past 342 Ka) have been analysed for pollen and spores at millennial resolution. The terrestrial pollen assemblages indicate that during interglacials, the vegetation of eastern South Africa and southern Mozambique largely consisted of evergreen and deciduous forests. During glacials open mountainous scrubland dominated. Montane forest with Podocarpus extended during humid periods was favoured by strong local insolation. Correlation with the sea surface temperature record of the same core indicates that the extension of mountainous scrubland primarily depends on sea surface temperatures of the Agulhas Current. Our record corroborates terrestrial evidence of the extension of open mountainous scrubland (including fynbos-like species of the high-altitude Grassland biome) for the last glacial as well as for other glacial periods of the past 300 Ka.

The purpose of this systematic review was to compare the effectiveness of topical treatments to minimize post-radiotherapy xerostomia. PubMed, Cochrane Library (CENTRAL) and LILACS databases were searched without restriction on date or language until the 6thAugust, 2015. Key-wordsused for searching were radiotherapy, xerostomia and saliva. Two independent reviewers screened titles and abstracts, carried out data extraction and assessed risk of bias. The first search identified 429 articles. From these, 117 studies were selected for full-text reading, from which 18 were included in the qualitative synthesis. From the eighteen articles included, seven were non- controlled clinical trial, one article was controlled clinical trial and ten studies were randomized clinical trials (three clinical trials were placebo controlled and seven were crossover). By the assessment of the quality of the studies included, ten showed high risk of bias, four showed moderate risk of bias and four presented low risk of bias. All interventions were considered effective in treating xerostomia (mucin, polysaccharides, aloe vera, rape oil, linseed oil, carboxymethylcellulose, polyethylene oxide, pilocarpine and systems of care for xerostomia - gel, paste and mouthwash). Meta-analysis could not be performed due to heterogeneity between thestudiesand interventions. This systematic review showed that a single and general protocol for topical treatment of xerostomia post-radiotherapy does not exist and that follow-up visits should be performed to validate the individualized treatment plan.

The distribution of pollen in marine surface sediments offshore of the west coast of South Africa has been investigated to aid in the interpretation of marine pollen records of onshore vegetation changes. A transect of sediment surface pollen samples retrieved from the Namaqualand mudbelt from just south of the Orange River mouth (29°S) to St Helena Bay (33°S) indicates distinctive pollen spectra reflecting vegetation communities on the adjacent continent. Pollen concentration increases southwards, partly in relation to greater pollen productivity due to higher biomass and density of fynbos vegetation and of sedimentary processes and low pollen concentrations consequent to dilution with silt and clay from the Orange River. The distribution of specific pollen taxa suggests that the Orange River is a major contributor of pollen to the northern mudbelt declining southwards, while the pollen distribution in the central mudbelt is largely attributable to seasonal inputs of pollen from offshore berg winds and local ephemeral Namaqualand rivers. The typical fynbos elements dominate in the southern mudbelt indicating a pollen source mainly in the fynbos vegetation types. These conclusions support a companion analysis of fossil pollen records of two marine sediment cores from the northern and southern mudbelt respectively. This study demonstrates that pollen records from marine sediment cores in the Namaqualand mudbelt have the potential to be a tool to reconstruct palaeovegetation on the adjacent continent. However, to better reconstruct the palaeoclimate of South Africa and fully understand the relations between terrestrial and marine deposits, more marine surface sediments along the western coast of South Africa as well as more terrestrial surface sediments need to be studied.

To better understand Holocene vegetation and hydrological changes in South Africa, we analyzed pollen and microcharcoal records of two marine sites GeoB8331 and GeoB8323 from the Namaqualand mudbelt offshore the west coast of South Africa covering the last 9900 and 2200 years, respectively. Our data corroborate findings from literature that climate developments apparently contrast between the summer rainfall zone (SRZ) and winter rainfall zone (WRZ) over the last 9900 years, especially during the early and middle Holocene. During the early Holocene (9900-7800 cal.yr BP), a minimum of grass pollen suggests low summer rainfall in the SRZ, and the initial presence of Renosterveld vegetation indicates relatively wet conditions in the WRZ. Towards the middle Holocene (7800-2400 cal. yr BP), a rather moist savanna/grassland rich in grasses suggests higher summer rainfall in the SRZ resulting from increased austral summer insolation and a decline of fynbos vegetation accompanied by an increasing Succulent Karoo vegetation in the WRZ possibly suggests a southward shift of the Southern Hemisphere westerlies. During the last 2200 years, a trend towards higher aridity was observed for the SRZ, while the climate in the WRZ remained relatively stable. The Little Ice Age (ca. 700-200 cal. yr BP) was rather cool in both rainfall zones and drier in the SRZ while wetter in the WRZ.
