In 1862, Glasgow Corporation initiated the first of a series of three legislative acts which would become known collectively as the City Improvements Acts. Despite having some influence on the nature of the built fabric on the expanding city as a whole, the most extensive consequences of these acts was reserved for one specific area of the city, the remnants of the medieval Old Town. As the city had expanded towards all points of the compass in a regular, grid-iron structure throughout the nineteenth century, the Old Town remained singularly as a densely wrought fabric of medieval wynds, vennels, oblique passageways and accelerated tenementalisation. Here, as the rest of the city began to assume the form of an ordered entity, visible and classifiable, one could still find and addresses such as ‘Bridgegate, No. 29, backland, stair first left, three up, right lobby, door facing’ (quoted in Pacione, 1995).

Unsurprisingly, this place, where proximity to the midden (dung-heap) was considered an enviable position, was seen by the authorities as a major health hazard and a source not only of cholera, but also of the more alarming typhoid epidemic of 1842. Accordingly, the demolitions which occurred in the backlands of the Old Town under the first of the acts, the Glasgow Police Act of 1862, were justified on health and medical grounds. But disease was not the only social problem thought to issue from this district. Reports from social reformers including Fredrick Engels suggested that the decay of the area’s physical fabric could be extended to the moral profile of its inhabitants. This was in such a state of degeneracy that there were calls for a nearby military barracks to be relocated to more salubrious climes because troops were routinely coming into contact ‘with the most dissolute and profligate portion of the population’ (Peter Clonston, Lord Provost, June 1861). Perhaps more worrying for the city fathers, however, was that the barracks’ arsenal was seen as a potential source of arms for the militant and often illegal cotton workers’ unions and organisations who inhabited the Old Town as well as the districts to the east. In fact, the Old Town and East End had been the site of numerous working class actions and riots since 1787, including a strike of 60,000 workers in 1820, 100,000 in 1838, and the so-called Bread Riots of 1848 where shouts of ‘Vive La Revolution’ were reported in the Gallowgate.

The events in Paris in 1848 precipitated Baron Hausmann’s interventions into that city. The boulevards were in turn visited by members of Glasgow Corporation and ultimately, it can be argued, provided an example for Old Town Glasgow. This paper suggests that the city improvement acts carried a similarly complex and pervasive agenda, one which embodied not only health, class conflict and sexual morality but also the more local condition of sectarianism. And, like in Paris, these were played out spatially in a extensive reconfiguration of the urban fabric of the Old Town which, through the creation of new streets and a railway yard, not only made it more amenable to large scale military manoeuvres but also, opened up the area to capitalist accumulation. By the end of the works, the medieval heritage of the Old Town had been almost completely razed, the working class and Catholic East End had, through the insertion of the railway yard, been isolated from the city centre and approximately 70,000 people had been made homeless.

Bacteria must control the progression of their cell cycle in response to nutrient availability. This regulation can be mediated by guanosine tetra- or pentaphosphate [(p)ppGpp], which are synthesized by enzymes of the RelA/SpoT homologue (Rsh) family, particularly under starvation conditions. Here, we study the effects of (p)ppGpp on the cell cycle of Caulobacter crescentus, an oligotrophic bacterium with a dimorphic life cycle. C. crescentus divides asymmetrically, producing a motile swarmer cell that cannot replicate its chromosome and a sessile stalked cell that is replication competent. The swarmer cell rapidly differentiates into a stalked cell in appropriate conditions. An artificial increase in the levels of (p)ppGpp in nonstarved C. crescentus cells was achieved by expressing a truncated relA gene from Escherichia coli, encoding a constitutively active (p)ppGpp synthetase. By combining single-cell microscopy, flow cytometry approaches, and swarming assays, we show that an increase in the intracellular concentration of (p)ppGpp is sufficient to slow down the swarmer-to-stalked cell differentiation process and to delay the initiation of chromosome replication. We also present evidence that the intracellular levels of two master regulators of the cell cycle of C. crescentus, DnaA and CtrA, are modulated in response to (p)ppGpp accumulation, even in the absence of actual starvation. CtrA proteolysis and DnaA synthesis seem indirectly inhibited by (p)ppGpp accumulation. By extending the life span of the motile nonreproductive swarmer cell and thus promoting dispersal and foraging functions over multiplication under starvation conditions, (p)ppGpp may play a central role in the ecological adaptation of C. crescentus to nutritional stresses.

Low height castor bean hybrids and highly productive possibly need larger quantity of macronutients during its development cycle. However, in Brazil there is little information on extraction and exportation of macronutrients by low height castor bean hybrids, as on season as out-of-season growing. This study aimed to evaluate the nutrients extraction and exportation by Savana hybrid castor bean on season and out-of-season growing. The experiments were conducted during the season of 2005/2006 and out-of-season of 2006 in an Oxisol. The experimental design was a randomized block with four replications. The plots were consisted by harvest times of plants, which were composed by six plant rows to 5 m, spaced 0.45 m between rows. Samples were collected at 17, 31, 45, 59, 73, 97 and 120 days after emergence (DAE) during the season and at 17, 31, 45, 59, 80; 100 and 120 DAE in the out-of-season. The nutrients accumulation by Savana hybrid was small until the flowering (30 DAE), but intensified after this period. The period of greatest nutrient absorption occurred at 65 and 90 DAE for N, 70 and 80 DAE for P, 60 DAE for K and Mg, 55 and 60 DAE for Ca and S. The amounts absorbed per hectare by hybrid Savana ranged 155 to 31 kg of N, 17 to 3 kg of P, 71 to 13 kg of K, 56 to 12 kg of Ca, 28 to 4 kg of Mg and 29,5 to 7 kg of S in season and out-of-season, respectively. The yield was higher in growing season, but the nutrients extraction and exportation per ton of grain were similar among the seasons. Among 40% and 50% of N and S and most part of Mg, K and Ca accumulated in the plants returned to soil with crop residues, but less than 35% of P remained in the residues.

Bothropstoxin-I (BthTx-I) is a Lys49-PLA(2) from the venom of Bothrops jararacussu that lacks detectable catalytic activity, yet causes rapid Ca2+-independent membrane damage. With the aim of understanding the interaction between BthTx-I and amphiphilic molecules, we have studied the interaction of sodium dodecyl sulphate (SDS) with the protein. Circular dichroism and attenuated total reflection Fourier-transform infrared spectra of BthTx-I reveal changes in the alpha-helical organization of the protein at an SDS/BthTx-I molar ratio of 20-25. At SDS/BthTx-I ratios of 40-45 the alpha-helices return to a native-like conformation, although fluorescence emission anisotropy measurements of 2-amino-N-hexadecyl-benzamide (AHBA) demonstrate that the total SDS is below the critical micelle concentration when this transition occurs. These results may be interpreted as the result of SDS accumulation by the BthTx-I homodimer and the formation of a pre-micelle SDS/BthTx-I complex, which may subsequently be released from the protein surface as a free micelle. Similar changes in the alpha-helical organization of BthTx-I were observed in the presence of dipalmitoylphosphatidylcholine liposomes, suggesting that protein structure transitions coupled to organization changes of bound amphiphiles may play a role in the Ca2+-independent membrane damage by Lys49-PLA(2)s. In order to quantify the growth, accumulation and exportation of macronutrients by carrot 'Forto', and obtain equations that best represent them, it was conducted an experiment in the São Gotardo (MG), from May to September 2004. Samples were taken at 40, 50, 60, 70, 80, 90, 100, 110 and 120 days after sowing (DAS) to determine dry matter and accumulation of macronutrients in leaves and roots. At 40 DAS, five days after thinning, the plants had 0.18 g and 0.04 g in dry leaves (LDM) and of roots (RDM), respectively. From then, until 88 DAS, the partitioning of photoassimilates and accumulation of matter in the shoots were more pronounced. The RDM was low until 80 DAS, which corresponded to two thirds of the crop cycle, from when the amount of material allocated in this part of the plant had strong growth, surpassing, at 88 DAS, the amount of LDM. The nutrient accumulation was small in the first 60 DAS, coinciding with the period of lower dry matter accumulation. The order of decreasing macronutrient accumulation by the crop were: K > N > Ca > P > S > Mg, in the amounts of 906.7, 438, 155.46, 87.4, 58 and 37.63 mg plant-1, respectively. The root participated with 60.5% of the accumulation of N, 86.1% of P, 58% of K, 25.5% of Ca, 55.6% of Mg and 65.5% of S. Considering a population of 590,000 plants per hectare, the total quantity of nutrients by the crop of carrots was 258.3, 51.6, 534.8, 91.7, 22.2 and 34.2 kg N ha-1, P, K, Ca, Mg and S, respectively.
