Twenty-four Actinobacillus suis isolates obtained from several species of non-porcine mammals were compared to the representative porcine strains, ATCC 15557 (serotype O1) and H89-1173 (serotype O2), by O serotyping, DNA fingerprinting, PCR amplification of apxICA, apxIICA and apxIIICA toxin genes and by rrs (16S rRNA) gene sequencing. Only two strains, both equine, reacted with O1 antiserum while two others, one canine and the other feline, reacted with O2 antiserum. One equine strain reacted weakly with both antisera. No amplification of apx genes was found with the non-porcine O1 or the "not O1/O2" strains but amplification of the apxICA and apxIICA genes was observed with the two O2 strains. In addition, these two O2 strains had both BamHI and BglII fingerprints that were very similar to the porcine O2 reference strain, H89-1173 and rrs gene sequences that were identical to the A. suis reference strain ATCC 15557. Taken together, these data suggest that although many non-porcine A. suis isolates are not A. suis (sensu stricto), some isolates are genotypically as well as phenotypically similar to A. suis.

The present study was undertaken to identify changes in some important proteins involved in CO2 fixation (Rubisco, Rubisco activase (RA), Rubisco binding protein (RBP)), NH4+ assimilation (glutamine synthetase (GS) and glutamate synthase (GOGAT)), using immunoblotting, and in the antioxidative defense as a result of Cu or Mn excess in barley leaves (Hordeum vulgare L. cv. Obzor). Activities and isoenzyme patterns of superoxide dismutase (SOD), ascorbate peroxidase (APX), guaiacol peroxidase (GPX) and catalase (CAT), as well as the levels of ascorbate (ASC), non-protein sulfhydryl groups, hydrogen peroxide and oxidative damage to proteins were determined. Data were correlated to the accumulation of Cu or Mn in the leaves after 5 days supply of heavy metal (HM) excess in the nutrient solution. In the highest Cu excess (1500 μM), Rubisco LS and SS were reduced considerably whereas under the highest Mn concentrations (18,300 μM) only minor changes in Rubisco subunits were detected. The RBP was diminished under the highest concentrations of both Cu or Mn. The bands of RA changed differently comparing Cu and Mn toxicity. GS decreased and GOGAT was absent under the highest concentration of Cu. At Mn excess Fd-GOGAT diminished whereas GS was not apparently changed. The development of toxicity symptoms corresponded to an accumulation of Cu or Mn in the leaves and to a gradual increase in protein carbonylation, a lower SOD activity and elevated CAT and GPX activities. APX activity was diminished under Mn toxicity and was not changed under Cu excess. Generally, changes in the isoenzyme profiles were similar under both toxicities. An accumulation of H2O2 was observed only at Mn excess. Contrasting changes in the low-molecular antioxidants were detected when comparing both toxicities. Cu excess affected mainly the non-protein SH groups, while Mn influenced the ASC content. Oxidative stress under Cu or Mn toxicity was most probably the consequence of depletion in low-molecular antioxidants as a result of their involvement in detoxification processes and disbalance in antioxidative enzymes. The link between heavy metal accumulation in leaves, leading to different display of oxidative stress, and changes in individual chloroplast proteins is discussed in the article.

Administration of gonadotropins or testosterone (T) will maintain qualitatively normal spermatogenesis and fertility in hypophysectomized (APX) rats. However, quantitative maintenance of the spermatogenic process in APX rats treated with T alone or in combination with follicle stimulating hormone (FSH) has not been demonstrated. Studies reported here were conducted to determine whether it would be possible to increase intratesticular testosterone (ITT) levels in APX rats to those found in normal animals by administration of appropriate amounts of testosterone propionate (TP) and if under these conditions spermatogenesis can be maintained quantitatively. Quantitative analysis of spermatogenesis was performed on stages VI and VII of the spermatogenic cycle utilizing criteria of Leblond and Clermont (1952) all cell types were enumerated. In a series of experiments designed to investigate the effects of T on spermatogenesis, TP was administered to 60 day old APX rats twice daily for 30 days in doses ranging from 0.6 to 15 mg/day or from 0.6 to 6.0 mg/day in combination with FSH. The results of this study demonstrate that the efficiency of transformation of type A to type B spermatogonia and the efficacy of the meiotic prophase are related to ITT levels, and that quantitatively normal completion of the reduction division requires normal ITT levels. The ratio of spermatids to spermatocytes in the vehicle-treated APX rats was 1:1.38; in the APX rats treated with 15 mg of TP it was 1:4.0 (the theoretically expected number). This study is probably the first to demonstrate: (1) the pharmacokinetics of TP, (2) the profile and quantity of T-immunoactivity in both serum and testicular tissue of APX and IC rats as well as APX rats treated with TP alone or in combination with FSH, (3) the direct correlation of serum T and ITT levels in treated APX rats (r = 0.9, p < 0.001) as well as in the IC rats (r = 0.9, p < 0.001), (4) the significant increase in the number of Type B spermatogonia, preleptotene and pachytene spermatocytes and round spermatids in TP-treated APX rats, (5) the correlation of the number of round spermatids formed in IC rats to ITT levels (r = 0.9, p < 0.001), and (6) the correlation of the quantitative maintenance of spermatogenesis with ITT levels (r = 0.7, p < 0.001) in the testes of TP-treated APX rats. These results provide direct experimental evidence for the key role of T in the spermatogenic process. ^

Reactive oxygen intermediates (ROI) play a critical role in the defense of plants against invading pathogens. Produced during the “oxidative burst,” they are thought to activate programmed cell death (PCD) and induce antimicrobial defenses such as pathogenesis-related proteins. It was shown recently that during the interaction of plants with pathogens, the expression of ROI-detoxifying enzymes such as ascorbate peroxidase (APX) and catalase (CAT) is suppressed. It was suggested that this suppression, occurring upon pathogen recognition and coinciding with an enhanced rate of ROI production, plays a key role in elevating cellular ROI levels, thereby potentiating the induction of PCD and other defenses. To examine the relationship between the suppression of antioxidative mechanisms and the induction of PCD and other defenses during pathogen attack, we studied the interaction between transgenic antisense tobacco plants with reduced APX or CAT and a bacterial pathogen that triggers the hypersensitive response. Transgenic plants with reduced capability to detoxify ROI (i.e., antisense APX or CAT) were found to be hyperresponsive to pathogen attack. They activated PCD in response to low amounts of pathogens that did not trigger the activation of PCD in control plants. Our findings support the hypothesis that suppression of ROI-scavenging enzymes during the hypersensitive response plays an important role in enhancing pathogen-induced PCD.

We investigated the relationship between H2O2 metabolism and the senescence process using soluble fractions, mitochondria, and peroxisomes from senescent pea (Pisum sativum L.) leaves. After 11 d of senescence the activities of Mn-superoxide dismutase, dehydroascorbate reductase (DHAR), and glutathione reductase (GR) present in the matrix, and ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) activities localized in the mitochondrial membrane, were all substantially decreased in mitochondria. The mitochondrial ascorbate and dehydroascorbate pools were reduced, whereas the oxidized glutathione levels were maintained. In senescent leaves the H2O2 content in isolated mitochondria and the NADH- and succinate-dependent production of superoxide (O2·−) radicals by submitochondrial particles increased significantly. However, in peroxisomes from senescent leaves both membrane-bound APX and MDHAR activities were reduced. In the matrix the DHAR activity was enhanced and the GR activity remained unchanged. As a result of senescence, the reduced and the oxidized glutathione pools were considerably increased in peroxisomes. A large increase in the glutathione pool and DHAR activity were also found in soluble fractions of senescent pea leaves, together with a decrease in GR, APX, and MDHAR activities. The differential response to senescence of the mitochondrial and peroxisomal ascorbate-glutathione cycle suggests that mitochondria could be affected by oxidative damage earlier than peroxisomes, which may participate in the cellular oxidative mechanism of leaf senescence longer than mitochondria.

In recent years, it has become apparent that salicylic acid (SA) plays an important role in plant defense responses to pathogen attack. Previous studies have suggested that one of SA's mechanisms of action is the inhibition of catalase, resulting in elevated levels of H2O2, which activate defense-related genes. Here we demonstrate that SA also inhibits ascorbate peroxoidase (APX), the other key enzyme for scavenging H2O2. The synthetic inducer of defense responses, 2,6-dichloroisonicotinic acid (INA), was also found to be an effective inhibitor of APX. In the presence of 750 microM ascorbic acid (AsA), substrate-dependent IC50 values of 78 microM and 95 microM were obtained for SA and INA, respectively. Furthermore, the ability of SA analogues to block APX activity correlated with their ability to induce defense-related genes in tobacco and enhance resistance to tobacco mosaic virus. Inhibition of APX by SA appears to be reversible, thus differing from the time-dependent, irreversible inactivation by suicide substrates such as p-aminophenol. In contrast to APX, the guaiacol-utilizing peroxidases, which participate in the synthesis and crosslinking of cell wall components as part of the defense response, are not inhibited by SA or INA. The inhibition of both catalase and APX, but not guaiacol peroxidases, supports the hypothesis that SA-induced defense responses are mediated, in part, through elevated H2O2 levels or coupled perturbations of the cellular redox state.

The use of fossil fuels has been considered one of reason for the increase of pollution in the atmosphere and it may be related to the climate changes. Then, the research of the new sources of fuels will be important. Considering this, the use of biodiesel has been considered not as bad as petrol. The castor bean (Ricinus communis L.) is an important oilseed, which belongs to Euphorbiaceae family, and the oil found in the seed has important characteristics for biodiesel. This plant is considered as “rustic” as it does not need so much water for its development and oil production. Due to this, this plant has been considered to be ideal in semi-arid regions, such as the Northeast of Brazil. The aim of his study is to better understand the responses to abiotic stresses (drought and salinity) from castor bean plants using morphological, physiological and molecular tools. In order to do this, the castor bean plants were subjected to salt stress (50, 100, 150 and 200 mM NaCl) in a controlled environment and drought stress (5, 10, 15 days and 10 days cyclic). After these treatments, these plants were subjected to different analyzes: a) the expansion and retention of water from leaves; b) anatomy using leaves and roots. Based on these results, we found that castor suffered decrease in leaf area with increase drought stress, however restricted water loss, probably by accumulation of compatible solutes in the leaves. The anatomy data showed modifications in the vascular system. These modifications observed suggested that castor bean plant may be resistant to stress as it was verified in 5 days of drought as well as in 100 mM NaCl. In both conditions, these plants were fine. Probably these plants keep some solutes in the cell and then maintain the cell tugor. The data obtained in this study gave a better idea how castor bean plant responds to abiotic stress conditions - drought and salt stress

World consumption of vegetable oils has increased in recent years because of its application in food, chemical, pharmaceutical and, more recently, energy industry. However, oilseeds, which these oils are extracted, have low viability, affecting the cultivation and productivity of these species. The aim of this study was to analyze the effect of aging on the coordination of catalase (CAT) and ascorbate peroxidase (APX) antioxidant systems in safflower and sunflower. . Therefore, seeds were subjected to accelerated aging for 3, 6 and 9 days and grown in moistened paper towel for 72 hours. Additionally, before accelerated aging, sunflower seeds were pretreated by osmopriming with 10 mM ascorbate (ASC) or 3 amino 1,2,4 triazol (3-AT), a specific inhibitor of CAT activitie. The method of artificial aging used was efficient in both species, because it caused a decrease in germination, seedling development and growth, especially in safflower. The aging caused inhibition of CAT activity for both species and to compensate for such inhibition , sunflower increased mRNA expression of this enzyme , while safflower mobilized over the activity of APX. Analysis of the expression of malate synthase and sugar content demonstrated that sunflower seeds consumes lipid reserves in quiescent state, while the safflower is more dependent on carbohydrate. Pretreatment with 3-AT inhibited CAT activity and stimulated the APX, though with ASC acted reverse on these systems. None of the treatments recovered the physiological decline aging. It is concluded that aging change the oilseeds antioxidant metabolism, despite interspecies variations in response to this process, the depletion of the CAT antioxidant system was common. Because of this we propose that the measurement of CAT activity can be used to identify aging seed lots.

The plants are often exposed to variations in environmental conditions that may trigger metabolic disturbances leading to a consequent loss in productivity of crops. These stressful conditions usually induce an accumulation of reactive oxygen species (ROS) in the cell, a condition known how oxidative stress. Among these species, hydrogen peroxide (H2O2) is an important molecule involved in numerous signaling mechanisms. The present study aimed to understand the relationship between the different enzymatic mechanisms of elimination of H2O2 by catalase (CAT) and ascorbate peroxidase (APX) in leaf tissues of seedlings of the species Vigna unguiculata L. Walp, under conditions of oxidative stress induced by application of CAT inhibitor, 3-amino-1,2,4-triazole (3-AT), and H2O2 itself on the roots. Three experiments were conducted. The first experiment was performed applying the compound 3-AT (5 mM) during the time (hours). In the second experiment, seedlings were exposed to different concentrations of H2O2 (2.5, 5.0, 7.5, 10 mM) for 48 h. The third strategy included the pre-treatment with H2O2 (2.5 mM) for 24 h, followed by subsequent treatment with the inhibitor 3-AT and recovery control condition. Treatment with 3-AT causes a strong inhibition of CAT activity in leaf tissues accompanied by an increase of activity of APX. However a decrease in oxidative damage to lipids is not observed as indicated by TBARS. It was observed that activity of APX is directly linked to the content of peroxide. Inductions in the activities of CAT and APX were observed mainly in the seedlings treated with 2.5 mM H2O2. This can be associated with a decrease in oxidative damage to lipids. In contrast, one same tendency was not observed in treatments with higher concentrations of this ROS. These results suggest that the concentration of 2.5 mM H2O2 can induce responses antioxidants later in seedling cowpea. This concentration when applied as pre-treatment for 24 h promoted an induction systems removers CAT and APX, both in activity and in terms of gene expression. However this increment was not observed in the recovered plants and the plants subsequently subjected to 3-AT. Additionally, the pretreatment was not sufficient to attenuate the inhibition of CAT activity and oxidative damage to lipids caused by the subsequent application of this inhibitor. The results showed that the application of 3-AT and H2O2 in the root systems of seedlings of cowpea promote changes in the parameters analyzed in leaf tissues that indicate a direct response to the presence of these factors or systemic signaling mecanisms. H2O2 appears to activate the responses of two antioxidant systems in this study thar does not promote greater protection in case of additional treatment with 3-AT. This demonstrates the importance of the CAT system. In this work, complete results indicate that there is a difference between the signaling and the effects caused by exposure to H2O2 and by treatment with 3-AT

Oilseeds are a high-value natural resource, due to its use as a substitute for petroleum. However, the storage time can reduce seed viability and oil quality. Therefore, scientific efforts have been made to provide a increment of storage time, germination rates and plant establishment of high-value oilseeds. The seedling establishment depends of the plant pass over the functional transition stage, characterized by a metabolic change from heterotrophic condition to autotrophic one. The storage oil mobilization is performed by β-oxidation process and the glyoxylate cycle. Also, the functional transition involves acclimation to photosynthetic condition, which generally includes the participation of antioxidant system and the reactive oxygen species, the latter are produced in various reactions of primary and secondary metabolism. In the present study, Catalase was inhibited during the functional transition of sunflower and safflower, after were performed many analyzes to elucidate the effects caused on the SOD and APX antioxidant systems. Also, were checked the changes in expression pattern of the glyoxylate cycle enzymes markers, ICL and MLS. It was observed that after CAT inhibition, the SOD and APX antioxidant systems allow the seedling establishment. Besides, was verified that both oilseeds can be accelerate the reverse mobilization and the photosynthetic establishment when Catalase activity has dramatically decreased

La biofortificación tiene como finalidad incrementar la concentración de elementos biodisponibles en las plantas de cultivo para elevar la calidad nutricional. El selenio es un elemento traza de gran impacto para el metabolismo antioxidante de las plantas y su acumulación es pobre en especies como el tomate, de igual manera este elemento es esencial para los humanos, por lo que adicionarlo en las plantas forma parte de los programas de biofortificación. El propósito de este trabajo fue analizar experimentalmente la capacidad del selenito de sodio para incrementar la concentración de Se y modificar la actividad antioxidante en plantas de tomate. Para ello se usaron plantas de la variedad híbrido Toro y se aplicaron tres tratamientos 0, 2 y 5 mg L-1 de selenito de sodio (Na2SeO3) utilizando como vehículo el agua de riego, los tratamientos fueron aplicados bajo un diseño experimental completamente al azar. Se llevaron a cabo tres muestreos 40, 80 y 120 días después del trasplante y la subsecuente cuantificación de la acumulación de selenio y macronutrientes en hojas, tallos y frutos así como su impacto en la producción de frutos bajo un diseño experimental completamente al azar. Se determinó la altura de la planta, los diámetros de tallos, firmeza, sólidos solubles de frutos y la materia seca total de los diferentes tejidos. Se obtuvo una cuantificación del potencial oxido reducción y de la actividad de antioxidante específicos como la catalasa, glutatión peroxidasa, superóxido dismutasa, el ácido ascórbico y licopeno, para cada variable se llevó a cabo un análisis de varianza y posteriormente una prueba de comparación de medias de Tukey. La expresión de los genes cat, gpx, sod, apx y lic en los frutos fue también analizada y en este caso se aplicó un análisis de varianza no paramétrico de Fisher, para encontrar las diferencias entre tratamientos. Los resultados mostraron un incremento en la acumulación de Se, encontrándose hasta un 53.1% de aumento en la concentración en los frutos bajo el tratamiento 5 mg L-1 en comparación con el testigo, sin embargo, este incremento no tuvo un impacto notorio en la producción y el rendimiento del tomate, a pesar de la existencia de una correlación de Spearman positiva (r =0.9637) entre la producción de fruto y la concentración de Se. Los valores del potencial oxido-reducción se redujeron en promedio desde -41.4 mV para el tratamiento testigo y hasta -68.0 mV con el mayor tratamiento. Mientras tanto, la concentración de Se si influyó positivamente en los parámetros de calidad incluyendo el ácido ascórbico (hasta un 50 % de aumento con el tratamiento 5 mgL-1 ) y el licopeno (hasta un 66.9% de aumento con el mayor tratamiento). La actividad de las enzimas antioxidantes aumentó notablemente en el fruto con el tratamiento 5 mg L-1 de selenito encontrándose 60.9% de aumento para CAT, 33.4% para GPX y 26.0% para SOD. En cuanto la expresión génica hubo mayor nivel de transcriptos de los genes gpx, sod y apx bajo el tratamiento 2 mg L-1

A remediação de locais contaminados com metais pesados usando plantas hiperacumuladoras aparenta ser uma alternativa bastante viável. Neste trabalho comparou-se a acumulação e tolerância ao cádmio (Cd), ambas baseadas nas respostas ao stress oxidativo em três espécies de plantas diferentes: Brassica juncea (L.) Czem., Nicotiana tabacum L. e Solanum nigrum L., descritas na literatura como plantas bastante tolerantes ou até com características híper acumuladoras. As plantas cresceram num solo contaminado com diferentes concentrações de Cd (O- 35 mg kg-1) durante um período de 90 dias. O factor de translocação (FT), utilizado para medir a translocação efectiva do Cd da raiz para a parte aérea, variou consideravelmente entre as espécies desenvolvidas. A N. tabacum foi a planta que apresentou os maiores valores de FT. Neste trabalho foi a única planta que preencheu todas as condições para ser considerada hiperacumuladora para todos os níveis de contaminação do solo. Por outro lado, a S. nigrum apresentou os maiores valores de concentração de Cd nos tecidos, com um FT > 1, na presença de 5 mg Cd kg·1 de solo. Apesar da B. juncea ter apresentado um resultado de FT inferior às restantes, foi a única planta com valores crescentes de FT com o aumento da contaminação de Cd. O stress oxidativo nas plantas desenvolvidas foi avaliado pela peroxidação lipídica e pelas actividades da catalase (CAT), ascorbato peroxidase (APX), guaiacol peroxidase (GPX) e superóxido dismutase (SOO), quer na raiz quer na parte aérea. Foi observado um aumento significativo (versus controlo) na peroxidação lipídica e actividade enzimática da CATe APX na parte aérea da B. juncea, N. tabacum e S. nigrum para os níveis de contaminação mais elevados, 15 e/ou 35 mg Cd kg-1 A B. juncea apresentou maior sensibilidade na resposta da GPX, para todas as concentrações de Cd no solo. A peroxidação lipídica e a actividade da CAT foram superiores na parte aérea em relação à raiz para todas as plantas em todas as contaminações de Cd presentes no solo. A actividade da SOO não apresentou respostas consistentes para nenhuma das plantas. ABSTRACT: Remediation of sites contaminated with heavy metals using hyper accumulators seems a promising alternative to engineering approaches. ln this work, we compared cadmium (Cd) accumulation and tolerance (based on responses to oxidative stress) in three different species, Brassica juncea (L) Czem., Nicotiana tabacum L. and Solanum nigrum L., described in the literature as very tolerant or even as hyper accumulators. The plants were grown in soil spiked with different Cd concentrations (O- 35 mg kg- 1) over a period of 90 days. The translocation factor (TF), used to measure the effectiveness of translocating Cd from roots to shoots, depended greatly on the species. N. tabacum was the plant which exhibited the highest TF values. lt was the only plant under study that fulfilled the conditions of a hyper accumulator for all levels of soil contamination. On the other hand, S. nigrum presented the highest Cd concentration in plant tissues, with TF > 1 in the presence of 5 mg Cd kg-1 of soil. Although B. juncea had presented the lowest TF and Cd concentrations, it was the only plant with TF values increasing with the level of cadmium. Oxidative stress in plants was evaluated by lipid peroxidation and activities of catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX) and superoxide dismutase (SOO), both in roots and shoots. A significant enhancement (versus control) on lipid peroxidation and enzymatic activity of CAT and APX in shoots of B. juncea, N. tabacum and S. nigrum was observed for the highest levels of Cd in soil, 15 and/or 35 mg Cd kg-1. B. juncea presented the most sensitive response of GPX, for all levels of Cd in soil. Lipid peroxidation and CAT activity were greater in shoots than in roots for all plants and soil Cd concentrations. SOO activity did not present consistent trends for any plant.
