提取DNA是所有分子生物学实验的第一步,但在昆虫分子生物学研究中,常规提取总DNA的方法不能保留昆虫所有的形态特征.显然,这不适用于对于体形较小的珍稀标本.以鞘翅目卷象科(Coleoptera: Attelabidae)昆虫为实验材料,提供了一种新的取材方法,取下整个腹部,消化其肌肉组织后收回骨化腹板.该方法可在提取昆虫总DNA的同时保存全部外形特征及其生殖器.

Hidden Markov model (HMM)-based speech synthesis systems possess several advantages over concatenative synthesis systems. One such advantage is the relative ease with which HMM-based systems are adapted to speakers not present in the training dataset. Speaker adaptation methods used in the field of HMM-based automatic speech recognition (ASR) are adopted for this task. In the case of unsupervised speaker adaptation, previous work has used a supplementary set of acoustic models to estimate the transcription of the adaptation data. This paper first presents an approach to the unsupervised speaker adaptation task for HMM-based speech synthesis models which avoids the need for such supplementary acoustic models. This is achieved by defining a mapping between HMM-based synthesis models and ASR-style models, via a two-pass decision tree construction process. Second, it is shown that this mapping also enables unsupervised adaptation of HMM-based speech synthesis models without the need to perform linguistic analysis of the estimated transcription of the adaptation data. Third, this paper demonstrates how this technique lends itself to the task of unsupervised cross-lingual adaptation of HMM-based speech synthesis models, and explains the advantages of such an approach. Finally, listener evaluations reveal that the proposed unsupervised adaptation methods deliver performance approaching that of supervised adaptation.

巨须裂腹鱼(Schizothorax macropogon)是雅鲁藏布江的重要经济鱼类。作者利用2004~2006年在雅鲁藏布江水系收集的293尾样本,以背鳍条磨片作为主要鉴定年龄的材料,对西藏雅鲁藏布江巨须裂腹鱼种群进行了年龄结构和生长特性的研究。结果表明,巨须裂腹鱼的雅鲁藏布江种群由2~14龄和16龄共14个龄组组成。其中优势龄组为4~6龄(占74.85%)。群体总性比为(♀∶♂)=1∶0.881。体长与体重的关系方程分别为:W♀=0.023L2.904(♀),W♂=0.018L2.962(♂),属于

通过截污、控制养鱼、控藻和科学种植等措施,2~3年内,在长江中游地区水质为劣Ⅴ类的重富营养湖泊武汉小莲花湖重建了以伊乐藻(Elodea nuttallii)为优势种的沉水植物先锋群落,2004和2005年春季的沉水植被盖度分别达到98%和95%,抑制了水华的发生,改善了水质。利用冬季水位低、透明度相对高的时机种植伊乐藻、菹草(Potamogeton crispu)等这些适合冬春季生长的沉水植物是建立沉水植物先锋群落的一个有效途径。由于以伊乐藻或菹草为优势种的先锋群落不稳定,必须配备丰富物种、优化结构、增加

在换水静置条件下(25℃),氯化高汞(以Hg~(++)计)对大型溞的48小时LC_(50)值及其可信限为13.5(±2.1)微克/升。试验个体饲以斜生栅藻单个培养,在汞含量为1—21微克/升浓度下,产仔总数经方差分析表明组间差异显著(F>F_(0.05))。各浓度组(X)与产仔总数(Y)的关系为:Y=-51.823X+4320.960(r=-0.904,p<0.01)。净增殖率(R_0)随浓度升高而逐渐下降。存活和生长用方差分析及D值检验表明对照组与各浓度组之间(差异数分别小于各自的D值20.89和0.1

Ptychobarbus dipogon is an endemic fish in the Yarlung Tsangpo River, but its biology is poorly known. We sampled 582 specimens (total length, TL, between 70.6 and 593.0 mm) from April 2004 to August 2006 in the Lhasa River, Tibet. We estimated ages based on the counts of alternating opaque and translucent zones (annuli) in thin transverse sections of lapilli otoliths. Ages ranged from 1(+) to 23(+) years for males and 1(+) to 44(+) for females. The observed 44(+) years was the oldest reported for schizothoracine fishes. Females attained a larger size than males. The TL weight relationship was W=7.12 x 10(-6)TL(3.006) for combined sexes. The growth parameters fitted von Bertalanffy growth functions were L-infinity = 598.66 mm, k=0.0898 year(-1), t(0)=-0.7261 year and W-infinity = 1585.38 g for females and L-infinity = 494.23mm, k=0.1197 year(-1), t(0)=-0.7296 year and W-infinity = 904.88g for males. The longevities of 32.7 year for females and 24.3 year for males were similar to the observed ages. Using an empirical model we estimated the instantaneous rate of total mortality (Z) at 0.28 per year in the lower reaches. Z in the upper and middle stocks was close to the M because of unexploited or lightly exploited stock. Protracted longevity, slow growth, low natural mortality and large body size were typical characteristics of P. dipogon. The current declining trend of P. dipogon could be prevented by altering fishing regulations.

Toll-like receptor 3 (TLR3) participates in the innate immune response by recognizing viral pathogens. To investigate grass carp immune system responding to GCRV (grass carp reovirus) infection, the full-length cDNA sequence and genomic organization of grass carp TLR3 (CiTLR3) was identified and characterized. The full-length genome sequence of CiTLR3 is composed of 5668 nucleotides, including five exons and four introns. The full-length of CiTLR3 cDNA is 3681 bp in length and encodes a polypeptide of 904 amino acids with an estimated molecular mass of 102,765 Da and a predicted isoelectric point of 8.35. Analysis of the deduced amino acid sequence indicated that CiTLR3 has four main structural domains, including a signal peptide sequence, 14 LRR (leucine-rich repeat) motifs, a transmembrane region and a TIR (Toll/interleukin-1 receptor) domain. It is most similar to the crucian carp (Carassius auratus) TLR3 amino acid sequence with an identity of 99%. Quantitative RT-PCR analysis showed that CiTLR3 transcripts were significantly up-regulated starting at day 1 and continued through day 7 following GCRV infection (P < 0.05). These data implied that CiTLR3 is involved in antiviral defense, provide molecular and functional information for grass carp TLR3, and implicate their role in mediating immune protection against grass carp viral diseases. The polyetherketone (PEK-c) guest-host system thin films doped with 3-(1,1-dicyanothenyl)-1-phenyl-4,5-dihydro-1H-pryazole (DCNP) were prepared. Their second-order nonlinear optical (NLO) coefficients chi(33)((2)) were measured by using Maker fringe method for the polymer films doped with different weight percents of DCNP. Experimental results indicate that the second-order NLO properties of the poled polymer films could decrease with the chromophore loading increasing when the chromophore loading reaches a fairly high level. In this paper, the relationship between the macroscopic second-order NLO coefficient and the chromophore number density was modified under considering the role of the electrostatic interactions of chromophores in the polymer film. According to the modified relationship, the macroscopic second-order NLO coefficient is no longer in direct proportion with the chromophore number density in the polymer film. The effect of the electrostatic interactions of chromophores on second-order NLO properties was discussed. The attenuation of the macroscopic second-order NLO activity can be demonstrated by the role of the chromophore electrostatic interactions at high loading of chromophore in the polymer systems.

利用两步合金法获得了能与p-GaN形成良好欧姆接触的高反射电极。电极的构成采用透明电极+高反射金属方案,厚的Ag层或Al层覆盖在合金后的Ni/Au上以提高电极的反射率,以Pt和Au作为覆盖层,可有效地防止Ag的氧化和团聚,短时间的热处理有助于Ag基电极反射率的提高。可靠性测试表明Ag基电极的稳定性较好,对于Al基电极,热处理后反射镜上出现暗斑,导致其反射率的下降。最终获得了波长在460nm处反射率为74%的Ag基高反射电极。

制备了3-(1,1-二氰基塞吩)-1-苯-4,5-二羟基-H-噻唑(DCNP)与聚醚醚酮(PEK-c)组成的主客掺杂聚合物薄膜,用Maker条纹法测量了不同掺杂浓度下薄膜的二阶非线性系数λ~((2))_(33)。实验结果表明,聚合物中生色团的含量高到一定程度,其宏观二阶非线性随生色团含量的增加反而下降。该文在考虑聚合物中生色团分子相互作用的情况下,修正了聚合物宏观二阶非线性与生色团的含量之间的关系,此时聚合物宏观二阶非线性与生色团的含量已非简单的正比关系。讨论了生色团分子间的相互作用对聚合物宏观二阶非线性的影响,从微观上解释了生色团分子在较高含量下其相互作用对聚合物宏观二阶非线性的衰减。
