Hefen stellen einen großen und wichtigen Teil der Mikrobiota während der Weinbereitung dar, da ohne ihre alkoholische Fermentation die Umwandlung von Most und Wein nicht möglich wäre. Ferner ist es ihre Vielzahl an Stoffwechselprodukten, die dem Aroma des fertigen Weines eine zusätzliche Komplexität verleihen. Auf der anderen Seite steht durch den Metabolismus verschiedenster so genannter Wildhefen die Gefahr von Qualitätsabstufungen der Weine, was allgemein als „Weinfehler“ betrachtet wird. Ziel dieser Arbeit war zum einen die taxonomische Einordnung von Saccharomyces-Spezies, sowie die Quantifizierung und Hemmung von ausgewählten Wildhefen während der Weinbereitung.rnEin Teil dieser Arbeit umfasste die Identifizierung der nahverwandten Mitglieder der Saccharomyces sensu stricto-Gruppe. Durch den Einsatz des DNA-Fingerpinting-Systems SAPD-PCR konnten alle die Gruppe umfassenden Spezies anhand spezifischer Bandenmuster nachgewiesen werden, wodurch eine Einordnung dieser schwer zu differenzierenden Arten möglich war. Die Differenzierung zwischen den einzelnen Spezies war in jedem Fall deutlicher als dies die Sequenzierung der 5.8S rDNA und ihre flankierenden ITS-Regionen vermochte. Die SAPD-PCR zeichnete sich zudem durch eine geringe Muster-Varianz bei verschiedenen Stämmen einer Art aus und konnte zuverlässig unbekannte Stämme bestimmen und bereits hinterlegte Stämme neu klassifizieren. Zudem konnte mit Hilfe dieses Systems Hybride aus Saccharomyces cerevisiae und S. bayanus bzw. S. cerevisiae und S. kudriavzevii detektiert werden, wenn diese Hybride aus relativ gleichen genomischen Anteilen der Eltern bestanden. rnZusätzlich wurde ein quantitatives PCR-System entwickelt, um die Gattungen Saccharomyces, Hanseniaspora und Brettanomyces in Most und Wein detektieren und quantifizieren zu können. Die hierfür entwickelten Primer zeigten sich spezifisch für die untersuchten Arten. Durch die serielle Verdünnung definierter DNA-Mengen konnte für alle drei Systeme eine Kalibrierungskurve erstellt werden, mit Hilfe derer die tatsächlichen Quantifizierungen durchgeführt wurden. Die qPCR-Analyse lieferte ähnliche Zellzahlen wie Lebendzellzahl-Bestimmungen und wurde nicht von anderen Spezies und von Traubensaft gestört. Die maximal detektierbare Zellzahl betrug 2 x 107 Zellen/ml, während die minimale Detektionsgrenze je nach Art zwischen 1 x 102 Zellen/ml und 1 x 103 Zellen/ml lag. Allerdings konnte eine effektive DNA-Isolierung dieser geringen Zellzahlen nur erreicht werden, wenn die Zellzahl durch artfremde Hefen künstlich erhöht wurde. Die Analyse einer Most-Vergärung mit den drei Spezies zeigte schlussendlich, dass die quantitative PCR sicher und schnell Veränderungen und Sukzessionen detektiert und so ein geeignetes Mittel darstellt, um Populationsdynamiken während der Weinherstellung zu beobachten. rnDer letzte Teil dieser Arbeit befasste sich mit der Inhibierung von Schadhefen durch zellwand-hydrolysierende Enzyme. Es konnte hierbei eine endoglykosidisch wirkende β-1,3-Glucanase aus dem Bakterium Delftia tsuruhatensis isoliert werden. Diese besaß eine ungefähre Masse von 28 kDa, einen isolektrischen Punkt von ca. 4,3 und wirkte mit einer spezifischen Aktivität von 10 U/mg Protein gegen das Glucan Laminarin. Zudem zeigte das Enzym ein Temperaturoptimum von 50 °C und ein pH-Optimum bei pH 4,0. Weinparameter wie erhöhte Konzentrationen an Ethanol, Phenolen und Sulfit beeinflussten die Wirkung des Enzyms nicht oder nur wenig. Neben der allgemeinen Wirkung gegen β-1,3-Glucane konnte hier auch gezeigt werden, dass ebenso gut die β-1,3-Glucane in der Zellwand verschiedener Hefen hydrolysiert wurden. Fluoreszenz- und rasterelektronen-mikroskopische Aufnahmen von Hefezellen nach Inkubation mit der β-1,3-Glucanase zeigten zusätzlich die Zerstörung der Zelloberfläche der Hefen. Die lytische Wirkung des Enzyms wurde an verschiedenen weintypischen Hefen getestet. Hierbei zeigten sich stammspezifische Unterschiede in der Sensitivität gegenüber dem Enzym. Außerdem konnte festgestellt werden, dass sowohl Wachstumsphase als auch Medium der Hefen Einfluss auf deren Zellwand hat und somit auch auf die Wirkung des Enzyms.rn

Gamma-radiation exposure has both short- and long-term adverse health effects. The threat of modern terrorism places human populations at risk for radiological exposures, yet current medical countermeasures to radiation exposure are limited. Here we describe metabolomics for gamma-radiation biodosimetry in a mouse model. Mice were gamma-irradiated at doses of 0, 3 and 8 Gy (2.57 Gy/min), and urine samples collected over the first 24 h after exposure were analyzed by ultra-performance liquid chromatography-time-of-flight mass spectrometry (UPLC-TOFMS). Multivariate data were analyzed by orthogonal partial least squares (OPLS). Both 3- and 8-Gy exposures yielded distinct urine metabolomic phenotypes. The top 22 ions for 3 and 8 Gy were analyzed further, including tandem mass spectrometric comparison with authentic standards, revealing that N-hexanoylglycine and beta-thymidine are urinary biomarkers of exposure to 3 and 8 Gy, 3-hydroxy-2-methylbenzoic acid 3-O-sulfate is elevated in urine of mice exposed to 3 but not 8 Gy, and taurine is elevated after 8 but not 3 Gy. Gene Expression Dynamics Inspector (GEDI) self-organizing maps showed clear dose-response relationships for subsets of the urine metabolome. This approach is useful for identifying mice exposed to gamma radiation and for developing metabolomic strategies for noninvasive radiation biodosimetry in humans.

BACKGROUND Besides α1,3-galactosyltransferase gene (GGTA1) knockout, several transgene combinations to prevent pig-to-human xenograft rejection are currently being investigated. In this study, the potential of combined overexpression of human CD46 and HLA-E to prevent complement- and NK-cell-mediated xenograft rejection was tested in an ex vivo pig-to-human xenoperfusion model. METHODS α1,3-Galactosyltransferase knockout heterozygous, hCD46/HLA-E double transgenic (transgenic) as well as wild-type pig forelimbs were ex vivo perfused with whole, heparinized human and autologous pig blood, respectively. Blood samples were analyzed for the production of porcine and/or human inflammatory cytokines as well as complement activation products. Biopsy samples were examined for deposition of human and porcine C3b/c, C4b/c, and C6 as well as CD62E (E-selectin) and CD106 (VCAM-1) expression. Apoptosis was measured in the porcine muscle tissue using TUNEL assays. Finally, the formation of thrombin-antithrombin (TAT) complexes was measured in EDTA plasma samples. RESULTS No hyperacute rejection was seen in this model. Extremity perfusions lasted for up to 12 h without increase in vascular resistance and were terminated due to continuous small blood losses. Plasma levels of porcine cytokines IL1β, IL-6, IL-8, IL-10, TNF-α, and MCP-1 as well as human complement activation markers C3a (P = 0.0002), C5a (P = 0.004), and soluble C5b-9 (P = 0.03) were lower in blood perfused through transgenic as compared to wild-type limbs. Human C3b/c, C4b/c, and C6 as well as CD62E and CD106 were deposited in tissue of wild-type limbs, but significantly lower levels (P < 0.0001) of C3b/c, C4b/c, and C6 deposition as well as CD62E and CD106 expression were detected in transgenic limbs perfused with human blood. Transgenic porcine tissue was protected from xenoperfusion-induced apoptosis (P < 0.0001). Finally, TAT levels were significantly lower (P < 0.0001) in transgenic limb as compared to wild-type limb xenoperfusions. CONCLUSION Transgenic hCD46/HLA-E expression clearly reduced humoral xenoresponses since all, the terminal pathway of complement activation, endothelial cell activation, muscle cell apoptosis, inflammatory cytokine production, as well as coagulation activation, were all downregulated. Overall, this model represents a useful tool to study early immunological responses during pig-to-human vascularized xenotransplantation in the absence of hyperacute rejection.

Normal humans have one red and at least one green visual pigment genes. These genes are tightly linked as tandem repeats on the X chromosome and each of them has six exons. There is only one X-linked visual pigment gene in New World monkeys (NWMs) but the locus has three polymorphic alleles encoding red, yellow and green visual pigments, respectively. The spectral properties of the squirrel monkey and the marmoset (both NWMs) have been studied and partial sequences of the three alleles are available. To study the evolutionary history of these X-linked opsin genes in humans and NWMs, coding and intron sequences of the three squirrel monkey alleles and the three marmoset alleles were amplified by PCR followed by subcloning and sequencing. Introns 2 and 4 of the human red and green pigment genes were also sequenced. The results obtained are as follows: (1) The sequences of introns 2 and 4 of the human red and green opsin genes are significantly more similar between the two genes than are coding sequences, contrary to the usual situation where coding regions are better conserved in evolution than are introns. The high similarities in the two introns are probably due to recent gene conversion events during evolution of the human lineage. (2) Phylogenetic analysis of both intron and exon sequences indicates that the phylogenetic tree of the available primate opsin genes is the same as the species tree. The two human genes were derived from a gene duplication event after the divergence of the human and NWM lineages. The three alleles in each of the two NWM species diverged after the split of the two NWMs but have persisted in the population for at least 5 million years. (3) Allelic gene conversion might have occurred between the three squirrel monkey alleles. (4) A model of additive effect of hydroxyl-bearing amino acids on spectral tuning is proposed by treating some unknown variables as groups. Under the assumption that some residues have no effect, it is found that at least five amino acid residues, at positions 178 (3 nm), 180 (5 nm), 230 ($-$4 nm), 277 (9 nm) and 285 (13 nm), have linear spectral tuning effects. (5) Adaptive evolution of the opsin genes to different spectral peaks was observed at four residues that are important for spectral tuning. ^

A collection of 8,000 Arabidopsis thaliana plants carrying 48,000 insertions of the maize transposable element En-1 has been generated. This population was used for reverse genetic analyses to identify insertions in individual gene loci. By using a PCR-based screening protocol, insertions were found in 55 genes. En-1 showed no preference for transcribed or untranscribed regions nor for a particular orientation relative to the gene of interest. In several cases, En-1 was inserted within a few kilobases upstream or downstream of the gene. En-1 was mobilized from such positions into the respective gene to cause gene disruption. Knock-out alleles of genes involved in flavonoid biosynthesis were generated. One mutant line contained an En-1 insertion in the flavonol synthase gene (FLS) and showed drastically reduced levels of kaempferol. Allelism tests with other lines containing En-1 insertions in the flavanone 3-hydroxylase gene (F3H) demonstrated that TRANSPARENT TESTA 6 (TT6) encodes flavanone 3-hydroxylase. The f3h and fls null mutants complete the set of A. thaliana lines defective in early steps of the flavonoid pathway. These experiments demonstrate the efficiency of the screening method and gene disruption strategy used for assigning functions to genes defined only by sequence.

Semipermeable cell walls or apoplastic “membranes” have been hypothesized to be present in various plant tissues. Although often associated with suberized or lignified walls, the wall component that confers osmotic semipermeability is not known. In muskmelon (Cucumis melo L.) seeds, a thin, membranous endosperm completely encloses the embryo, creating a semipermeable apoplastic envelope. When dead muskmelon seeds are allowed to imbibe, solutes leaking from the embryo are retained within the envelope, resulting in osmotic water uptake and swelling called osmotic distention (OD). The endosperm envelope of muskmelon seeds stained with aniline blue, which is specific for callose (β-1,3-glucan). Outside of the aniline-blue-stained layer was a Sudan III- and IV-staining (lipid-containing) layer. In young developing seeds 25 d after anthesis (DAA) that did not exhibit OD, the lipid layer was already present but callose had not been deposited. At 35 DAA, callose was detected as distinct vesicles or globules in the endosperm envelope. A thick callose layer was evident at 40 DAA, coinciding with development of the capacity for OD. Removal of the outer lipid layer by brief chloroform treatment resulted in more rapid water uptake by both viable and nonviable (boiled) seeds, but did not affect semipermeability of the endosperm envelope. The aniline-blue-staining layer was digested by β-1,3-glucanase, and these envelopes lost OD. Thus, apoplastic semipermeability of the muskmelon endosperm envelope is dependent on the deposition of a thick callose-containing layer outside of the endosperm cell walls.
