The crystal transitions of Nylon 11 annealed and drawn at different temperatures (T-d) with different drawing ratios (n) were investigated by wide-angle X-ray diffraction (WAXD). The alpha -form of Nylon Il could be transformed from the delta'-form by annealing at high temperature, The results showed that the crystal transitions of Nylon 11 strongly depended on the thermal history and the conditions of drawing. The delta'-form Nylon Il could he gradually transformed into the alpha -form when it was drawn at high temperature and the alpha -form was only partly transformed into the delta'-form when it was drawn at low temperature. This should be due to the effect of the competition between thermal inducement and drawing inducement. The thermal inducement was favorable to producing the alpha -form, while the drawing inducement was favorable to producing the delta'-form. (C) 2001 Elsevier Science Ltd. All rights reserved.

A novel inorganic-organic hybrid material incorporating graphite powder and Keggin-type alpha -germanomolybdic acid (GeMo12) in methyltrimethoxysilane-based gels has been produced by the sol-gel technique and used to fabricate a chemically bulk-modified electrode. GeMo12 acts as a catalyst, graphite powder ensures conductivity by percolation, the silicate provides a rigid porous backbone, and the methyl groups endow hydrophobicity and thus limit the wetting section of the modified electrode. The GeMo12-modified graphite organosilicate composite electrode was characterized by cyclic and square-wave voltammetry. The modified electrode shows a high electrocatalytic activity toward the reduction of bromate, nitrite and hydrogen peroxide in acidic aqueous solution. In addition, the chemically-modified electrode has some distinct advantages over the traditional polyoxometalate-modified electrodes, such as long-term stability and especially repeatability of surface-renewal by simple mechanical polishing.

A new complex K5H[Co-4(VW9O33)(2)]. 5H(2)O was prepared and its structure was determined by X - ray diffraction analysis. The anion has alpha - beta - Keggin structure. Two (VW9O33)(7-) moieties (alpha - B isomers) are linked via four Co(11)O-6 groups. The single crystal of the title complex is monoclinic P2(1/n) with a = 1.2307(3) nm, b = 2.1250(4) nm, c = 1.5817(3) nm, beta = 91.86(3)degrees, V = 4.1343 (14) nm(3), R = 0.0895, R-w = 0.2180.

The king cobra neuotoxin CM-11 is a small protein with 72 amino acid residues. After its complete assignments of H-1-NMR resonance's were obtained using various 2D-NMR technologies, including of DQF-COSY, clean-TOCSY AND NOESY, the secondary structure was analysed by studying the various NOEs extracted from the NOESY spectra and the distribution of chemical shifts. The secondary structure was finally determined by MCD as follows: a triple-strand antiparallel beta sheet with I20-W36, R37-A43 and V53--S59 as its beta strands, a short alpha helix formed by W30-G35 and four turns formed by P7-K10, C14-G17, K50-V53 and D61-N64.

The melting points(T-m), crystalline temperature(T-c) and crystallinity(chi(c)) of propylene/alpha-olefin (pentene-l, octene-1 and decene-1) copolymers have been investigated, The results show that the T-m, T-c and chi(c) of the copolymers are lower than those of propylene homopolymer, indicating that lower alpha-olefin incorporation in copolymer has strongly hampered the crystallization of propylene, From critical crystalline sequence length of several propylene/alpha-olefin copolymers, it can be seen that a long chain alpha-olefin has much stronger effect on crystallization of PP than a short alpha-olefin does.

The construction of the shuttle, expression vector of human tumor necrosis factor alpha (hTNF-alpha) gene and its expression in a cyanobacterium Anabaena sp. PCC 7120 was reported. The 700-bp hTNF cDNA fragments have been recovered from plasmid pRL-rhTNF, then inserted downstream of the promoter PpsbA in the plasmid pRL439. The resultant intermediary plasmid pRL-TC has further been combined with the shuttle vector pDC-8 to get the shuttle, expression vector pDC-TNF. The expression of the rhTNF gene in Escherichia coil has been analyzed by SDS-PAGE and thin-layer scanning, and the results show that the expressed TNF protein with these two vectors is 16.9 percent (pRL-TC) and 15.0 percent (pDC-TNF) of the total proteins in the cells, respectively, while the expression level of TNF gene in plasmid pRL-rhTNF is only 11.8 percent. Combined with the participation of the conjugal and helper plasmids, pDC-TNF has been introduced into Anabaena sg PCC 7120 by triparental conjugative transfer, and the stable transgenic strains have been obtained. The existence of the introduced plasmid pDC-TNF in recombinant cyanobacterial cells has been demonstrated by the results of the agarose electrophoresis with the extracted plasmid samples and Southern blotting with alpha-(32)p labeled hTNF cDNA probes, while the expression of the hTNF gene in Anabaena sp. PCC 7120 has been confirmed by the results of Western blotting with extracted protein samples and human TNF-alpha monoclonal antibodies. The cytotoxicity assays using the mouse cancer cell line L929 proved the cytotoxicity of the TNF in the crude extracts from the transgenic cyanobacterium Anabaena sp. PCC 7120.

This paper presents an algorithm for simplifying NDL deductions. An array of simplifying transformations are rigorously defined. They are shown to be terminating, and to respect the formal semantis of the language. We also show that the transformations never increase the size or complexity of a deduction---in the worst case, they produce deductions of the same size and complexity as the original. We present several examples of proofs containing various types of "detours", and explain how our procedure eliminates them, resulting in smaller and cleaner deductions. All of the given transformations are fully implemented in SML-NJ. The complete code listing is presented, along with explanatory comments. Finally, although the transformations given here are defined for NDL, we point out that they can be applied to any type-alpha DPL that satisfies a few simple conditions.

Cytokine-driven signalling shapes immune homeostasis and guides inflammatory responses mainly through induction of specific gene expression programmes both within and outside the immune cell compartment. These transcriptional outputs are often amplified via cytokine synergy, which sets a stimulatory threshold that safeguards from exacerbated inflammation and immunopathology. In this study, we investigated the molecular mechanisms underpinning synergy between two pivotal Th1 cytokines, IFN-γ and TNF-α, in human intestinal epithelial cells. These two proinflammatory mediators induce a unique state of signalling and transcriptional synergy implicated in processes such as antiviral and antitumour immunity, intestinal barrier and pancreatic β-cell dysfunction. Since its discovery more than 30 years ago, this biological phenomenon remains, however, only partially defined. Here, using a functional genomics approach including RNAi perturbation screens and small-molecule inhibitors, we identified two new regulators of IFN-γ/TNF-α-induced chemokine and antiviral gene and protein expression, a Bcl-2 protein BCL-G and a histone demethylase UTX. We also discovered that IFN-γ/TNF-α synergise to trigger a coordinated shutdown of major receptor tyrosine kinases expression in colon cancer cells. Together, these findings extend our current understanding of how IFN-γ/TNF-α synergy elicits qualitatively and quantitatively distinct outputs in the intestinal epithelium. Given the well-documented role of this synergistic state in immunopathology of various disorders, our results may help to inform the identification of high quality and biologically relevant druggable targets for diseases characterised by an IFN-γ/TNF-α high immune signature

Transgenic mice were generated by using the alpha-myosin heavy chain promoter coupled to the coding sequence of a constitutively active mutant alpha 1B-adrenergic receptor (AR). These transgenic animals demonstrated cardiac-specific expression of this alpha 1-AR with resultant activation of phospholipase C as shown by increased myocardial diacylglycerol content. A phenotype consistent with cardiac hypertrophy developed in adult transgenic mice with increased heart/body weight ratios, myocyte cross-sectional areas, and ventricular atrial natriuretic factor mRNA levels relative to nontransgenic controls. These transgenic animals may provide insight into the biochemical triggers that induce hypertrophy in cardiac disease and serve as a convenient experimental model for studies of this condition.

Alpha-synuclein is a major component of Lewy bodies in Parkinson's disease and is found associated with several other forms of dementia. As with other neurodegenerative diseases, the ability of alpha-synuclein to aggregate and form fibrillar deposits seems central to its pathology. We have defined a sequence within the NAC region of alpha-synuclein that is necessary for aggregation. Exploitation of chemically modified analogues of this peptide may produce inhibitors of aggregation.

Convergent biochemical and genetic evidence suggests that the formation of alpha-synuclein (alpha-syn) protein deposits is an important and, probably, seminal step in the development of Parkinson's disease (PD), dementia with Lewy bodies (DLB) and multiple system atrophy (MSA). It has been reported that transgenic animals overexpressing human alpha-syn develop lesions similar to those found in the brain in PD, together with a progressive loss of dopaminergic cells and associated abnormalities of motor function. Inhibiting and/or reversing alpha-syn self-aggregation could, therefore, provide a novel approach to treating the underlying cause of these diseases. We synthesized a library of overlapping 7-mer peptides spanning the entire alpha-syn sequence, and identified amino acid residues 64-100 of alpha-syn as the binding region responsible for its self-association. Modified short peptides containing alpha-syn amino acid sequences from part of this binding region (residues 69-72), named alpha-syn inhibitors (ASI), were found to interact with full-length alpha-syn and block its assembly into both early oligomers and mature amyloid-like fibrils. We also developed a cell-permeable inhibitor of alpha-syn aggregation (ASID), using the polyarginine peptide delivery system. This ASID peptide was able to inhibit the DNA damage induced by Fe(II) in neuronal cells transfected with alpha-syn(A53T), a familial PD-associated mutation. Results of the search of the periodic changes of the 530.3 nm line intensity emitted by selected structures of the solar corona in the frequency range 1-10 Hz are presented. A set of 12 728 images of the section of the solar corona extending from near the north pole to the south-west were taken simultaneously in the 530.3 nm ("green") line and white-light with the Solar Eclipse Coronal Imaging System (SECIS) during the 143-seconds- long totality of the 1999 August 11 solar eclipse observed in Shabla, Bulgaria. The time resolution of the collected data is better than 0.05 s and the pixel size is approximately 4 arcsec. Using classical Fourier spectral analysis tools, we investigated temporal changes of the local 530.3 nm coronal line brightness in the frequency range 1-10 Hz of thousands of points within the field of view. The various photometric and instrumental effects have been extensively considered. We did not find any indisputable, statistically significant evidence of periodicities in any of the investigated points (at significance level alpha = 0.05).
