The objective of this work was to perform the screening of soybean genotypes as to their ability to respond to the induction of hairy roots by Agrobacterium rhizogenes‑mediated transformation. Four Brazilian soybean cultivars (BRSMG 68 Vencedora, BRS 137, Embrapa 48, and MG/BR 46 Conquista) and two North American ones adapted to Brazilian cropping conditions (Bragg and IAS‑5) were screened for their capacity to respond to A. rhizogenes in protocols for in vitro hairy root culture and ex vitro composite plant production. Four‑day‑old seedlings with uniform size were injected with A. rhizogenes harboring the plasmid p35S‑GFP. Seedlings expressing green fluorescent protein (GFP) in at least one hairy root were used to determine the transformation frequency. Using an axenic in vitro protocol, excised cotyledons from four‑day‑old seedlings were infected with A. rhizogenes harboring the pCAMBIA1301 plasmid, containing the gusA reporter gene. The transformation frequency and the number of days for hairy root emergence after bacterial infection (DAI) were evaluated. The transformation frequency and DAI varied according to the genotype. Cultivars MG/BR 46 Conquista and BRSMG 68 Vencedora are more susceptible to A. rhizogenes and can be recommended for transformation experiments.

Abstract:The objective of this work was to develop a scale to assess the severity of postharvest physiological deterioration (PPD) of cassava roots, and to validate this scale for accuracy and reproducibility estimates. A diagrammatic scale (0 to 100%) for the damaged roots was analyzed according to precision, accuracy, and reproducibility. Seven evaluators (four with experience and three without it) quantified the PPD severity, with or without the scale, considering 150 roots with different levels of PPD. Without and with the use of the scale, respectively, the inexperienced evaluators obtained coefficients of determination (R2) from 0.76 to 0.86 and 0.87 to 0.92, and the experienced evaluators obtained R2 from 0.90 to 0.96 and 0.96 to 0.97. The values of the intercept (a) obtained by both the experienced and inexperienced evaluators who did not use the scale were all significant, while after using the scale, only two evaluators got values that were not significantly different from one. Evaluation reproducibility between the evaluators ranged from 0.61 to 0.91 for the inexperienced ones and from 0.83 to 0.95 for the experienced ones. The proposed diagrammatic scale was considered appropriate to estimate the severity of PPD in cassava roots, and can be used to identify sources of tolerance to postharvest deterioration.

In imaging diagnosis, redundant nerve roots of the cauda equina are characterized by the presence of elongated, enlarged and tortuous nerve roots in close relationship with a high-grade lumbar spinal canal stenosis. This is not an independent entity, but it is believed to be a consequence of the chronic compression at the level of the lumbar canal stenosis and thus may be part of the natural history of lumbar spinal stenosis. The present paper is aimed at reviewing the histopathological, electrophysiological and imaging findings, particularly at magnetic resonance imaging, as well as the clinical meaning of this entity. As the current assessment of canal stenosis and root compression is preferably performed by means of magnetic resonance imaging, this is the imaging method by which the condition is identified. The recognition of redundant nerve roots at magnetic resonance imaging is important, particularly to avoid misdiagnosing other conditions such as intradural arteriovenous malformations. The literature approaching the clinical relevance of the presence of redundant nerve roots is controversial. There are articles suggesting that the pathological changes of the nerve roots are irreversible at the moment of diagnosis and therefore neurological symptoms are less likely to improve with surgical decompression, but such concept is not a consensus.

This work presents the use of potentiometric measurements for kinetic studies of biosorption of Cd2+ ions from aqueous solutions on Eichhornia crassipes roots. The open circuit potential of the Cd/Cd2+ electrode of the first kind was measured during the bioadsorption process. The amount of Cd2+ ions accumulated was determined in real time. The data were fit to different models, with the pseudo-second-order model proving to be the best in describing the data. The advantages and limitations of the methodology proposed relative to the traditional method are discussed.

Organic extracts were obtained from roots of Canavalia ensiformis and evaluated for allelopathic potential on the germination of the weed seeds: Mimosa pudica, Cassia tora and Cassia occidentalis showing a strong allelopathic potential. After that, a systematic study of these crude extracts was made using specific protocols developed in capillary electrophoresis (CE) in order to determine some classes of secondary metabolites. Capillary electrophoresis protocols were highly specific, which makes it possible to identify 5 classes of compounds using the same crude extract samples and analyze them fartly. Some of the compounds identified show activity in the inhibition of seeds germination.

Det fÃrefaller självklart att begrepp som lojalitet, respekt, rättigheter eller altruism beskriver nÃ¥got entydigt gott. Men tänk om det i själva verket är sÃ¥ att de aktualiseras bara där vi avvisat godheten? Tänk om vÃ¥r moral och hela vÃ¥rt liv genomsyras av en djup fÃrvirring, av en ovilja att se vad som egentligen pÃ¥gÃ¥r mellan oss? I min avhandling ställer jag de här obekväma frÃ¥gorna. UtgÃ¥ngspunkten är en diskussion om vänskap, som jag beskriver som en fÃrbehÃ¥llslÃs, helhjärtad Ãppenhet mellan människor. Denna Ãppenhet â som lika väl kan kallas godhet eller kärlek â är det svÃ¥raste som finns. Samtidigt finns Ãppenheten alltid där mellan oss, om sÃ¥ bara som en anad mÃjlighet som skrämmer oss, som vi sluter oss fÃr och inte vill veta av. Vi lever i en ständig spänning mellan Ãppenhet och avvisande, och denna spänning yttrar sig i allt vi gÃr, känner och tänker. Det är vad jag vill visa. Bland de filosofer jag diskuterar kan nämnas Aristoteles, Kant, Kierkegaard, Nietzsche och Emmanuel Levinas.

Neurofibromatosis type 1 (NF1) is an autosomal dominant cancer predisposition syndrome that affects about 1 in 3500 individuals worldwide. NF1 is caused by mutations in the NF1 gene that encodes the tumor suppressor protein neurofibromin, an inactivator of the Ras oncogene. The hallmarks of NF1 include pigmentary lesions of the skin, Lisch nodules of the iris and cutaneous neurofibromas. Cutaneous neurofibromas are benign tumors composed of all the cell types of normal peripheral nerve. The traditional view of neurofibroma development has been that cutaneous neurofibromas arise from the disruption of the small nerve tributaries of the skin and subsequent proliferation of the resident cells. The second hit mutation in the NF1 gene has been considered as a prerequisite for neurofibroma development. The second hit is detectable in a subpopulation of primary Schwann cells cultured from neurofibromas. This thesis challenges the traditional concept of neurofibroma development. The results show that cutaneous neurofibromas are intimately associated with hair follicular structures and contain multipotent precursor cells (NFPs), suggesting that neurofibromas may arise from the multipotent cells which reside in hair follicles. Furthermore, this study presents that neurofibroma-derived Schwann cells that harbor bi-allelic inactivation in the NF1 gene express HLA class II genes and may act as nonprofessional antigen presenting cells. The CD4- and FoxP3-positive cells detected in cutaneous neurofibromas suggest that these cells may represent regulatory T cells (Tregs) which interact with HLA II âpositive cells and aid the tumor cells in hiding from the immune system and are thus mediators of immune tolerance. This thesis also investigated neurofibroma development in the oral cavity and the use of different biomarkers to characterize cellular differentiation in neurofibromas. The results revealed that oral neurofibromas are not rare, but they usually appear as solitary lesions contrary to multiple cutaneous neurofibromas and present high heterogeneity within and between tumors. The use of class III beta-tubulin as a marker for neuronal differentiation led to an unexpected finding showing that multiple cell types express class III beta-tubulin during mitosis. The increased understanding of the multipotency of tumor cells, cellular differentiation and ability to hide from immune system will aid in the development of future treatments. Specifically, targeting Tregs in NF1 patients could provide a novel therapeutic approach to interfere with the development of neurofibromas.

Den evolutionära fÃrklaringen till den allmänt utbredda incestaversionen, dvs. motviljan fÃr sex med nära släktingar, fÃrsÃker besvara sÃ¥väl frÃ¥gan om varfÃr incestaversionen gynnats i det naturliga urvalet som frÃ¥gan om hur denna aversion regleras pÃ¥ individuell nivÃ¥. Eftersom inavlade barn har en fÃrsämrad biologisk duglighet än andra barn, medfÃr kostnaderna av denna icke-optimala reproduktion ett selektionstryck mot inavel. I en serie studier visade vi att eftersom kvinnor i allmänhet satsar mer biologiska resurser pÃ¥ sina barn än vad män gÃr, känner kvinnor starkare incestaversion än män och eftersom det endast är fertila kvinnor som riskerar satsa resurser i en inavlad avkomma har även fertila kvinnor hÃgre incestaversion än icke-fertila kvinnor. Vi visade ocksÃ¥ att de biologiska kostnaderna av inavel inte begränsas till enbart de individer som har incest. Eftersom alla vÃ¥ra biologiska släktingar sannolikt delar vÃ¥ra alleler speglar incestaversionen även de biologiska kostnader som incest mellan vÃ¥ra släktingar medfÃr Ã¥t oss. Den psykologiska mekanism med vilken incestsituationer bedÃms har hittills varit okänd. I vÃ¥ra studier testades teorin om självreflekterande empati. Enligt den teorin bedÃms sÃ¥dana situationer emotionellt genom att man själv fÃreställer sig ha sex med motsvarande släkting och känslan som väcks i denna process ger därefter emotionell information till bedÃmningen av andras incest. I tre delstudier fann vi att självreflektion var positivt associerat med styrkan av aversion gentemot andras incest, vilket stÃder teorin om självreflekterande empati.

Morphological and anatomical features of roots, stems, leaves, and scapes were studied in Heliconia angusta and H. velloziana from the Atlantic forest in the southeastern of Brazil. Morphologically H. angusta and H. velloziana show differences in their sizes, blade shapes, number and shape of inflorescence bracts. On the other hand, they have common anatomical characteristics such as: roots with air-canals in the cortex; rhizomes with isolated fiber bundles, collateral vascular bundles, and uniseriate endodermis and pericycle; leaves presenting air-canals and collateral vascular bundles forming arcs, and thin-walled epidermal cells; scapes with collateral vascular and fiber bundles in the cortex. The distribution of the fiber bundles in the leaves and in the scapes was different for each species, having a taxonomical value, H. velloziana presenting continuous fiber bundles. Air-canals in roots and leaves with narrow mesophyll might be related to the moist understorey of the Atlantic forest habitats.
