El entorno alpino de los Grisones ha sido un laboratorio donde experimentar un método arquitectónico procedente de diversas fuentes durante los años 70 en la ETHZ. Este método ha producido durante las últimas tres décadas un discurso teórico propiamente suizo alemán. La tradición continúa siendo un valor cultural para la comunidad alpina de los Grisones, que apuesta por seguir relacionando la arquitectura con su historia y su paisaje. La especificidad de cada entorno suizo alemán es un hecho obvio pero, por encima de sus rasgos característicos, hay un denominador común que les da unidad en silencio: se trata de una idea de la construcción. El fenómeno de los Grisones es territorial en su localización concreta, pero comparte el trasfondo teórico del resto de la arquitectura suizo alemana reciente. La tesis recorre transversalmente las condiciones que han dado lugar a esa trayectoria común hasta la actualidad, en que la arquitectura suizo alemana se halla precisamente en un momento de cambio generacional. La obra de Peter Märkli constituye en esta tesis un paradigma de la voluntad suizo alemana por formar parte de la continuidad cultural de la arquitectura, como una actitud de resistencia compartida frente al mundo globalizado. La tesis se articula a partir de ocho proyectos recientes, realizados por arquitectos suizo alemanes en el entorno del territorio alpino de los Grisones. Estos proyectos condensan la trayectoria arquitectónica suizo alemana durante el siglo XX, así como el fenómeno arquitectónico que ha tenido lugar en el entorno de los Grisones durante las últimas tres décadas. La tesis analiza a través de cada pareja de proyectos los episodios teóricos, artísticos y filosóficos más relevantes que se sintetizan en estas obras, así como sus referentes arquitectónicos y la dimensión trascendente de los materiales en los que la cultura suiza hunde sus raíces: la madera, la piedra y el hormigón. Las vanguardias pictóricas, el Movimiento Moderno y las corrientes humanistas y regionalistas que se fueron sucediendo durante el pasado siglo, configuran el escenario en el que tomó cuerpo la sensibilidad arquitectónica suizo alemana reciente, que emerge a través de la compleja red de relaciones establecidas entre el panorama suizo y el internacional. El mismo bagaje teórico subyace al territorio suizo alemán a nivel general, por lo que asociarlo a través de esta tesis a proyectos realizados en un entorno característico, permite acceder a una realización concreta de ese debate en un entorno tan particular como son los Grisones. La aproximación a los proyectos es de carácter fenomenológico: la experiencia directa de los mismos ha sido el sustrato fundamental del enfoque de la tesis. El trabajo de campo no se limitó a visitar los proyectos que vertebran la tesis, sino que trató de abarcar las obras más relevantes de la producción suizo alemana reciente en los Grisones, con el fin de adquirir cierto grado de sensibilidad perceptiva hacia la singularidad de su identidad territorial. La tesis hace especial hincapié en las obras de Peter Märkli y Peter Zumthor, representantes de los dos extremos del discurso suizo alemán reciente, respecto a la materialidad y al carácter de la secuencia espacial, sistemático o fluido respectivamente, pero que asimismo comparten el clasicismo formalista que subyace al fenómeno suizo alemán. Los dos proyectos que establecen el periodo 1992-2004 explorado por esta investigación constituyen dos hitos fundamentales. El Museo de La Congiunta en Giornico de Peter Märkli se construyó en 1992 y se erige a partir de entonces como un icono arquitectónico de la condensación típicamente suizo alemana de base reductiva. Este paradigma de síntesis conceptual y constructiva comenzó a forjarse en la década de 1930 de la mano del Konkrete Kunst formulado por Max Bill, que es la única manifestación artística genuinamente suiza del siglo XX y que desarrolló pictóricamente los valores de precisión, rigor, racionalismo y abstracción afines a la sensibilidad suizo alemana. En segundo lugar, el proyecto para la rehabilitación de la Villa Garbald y la construcción de la Torre Roccolo en Castasegna llevado a cabo por Miller & Maranta en 2004, supuso la consumación de la Analoge Architektur, que fue la primera corriente arquitectónica propiamente suiza, desarrollada durante la década de 1980 por Miroslav Sik. La Deutscher Tendenza es un periodo de la trayectoria suizo alemana reciente poco conocido, representado por una figura tan relevante como es Peter Zumthor. Las obras que proyectó Zumthor entre 1979 y 1985 forman parte de un periodo en que el arquitecto se encontraba profundamente influido por el neorracionalismo italiano, como ilustra la escuela de Churwalden de 1983. La arquitectura suizo alemana reciente exploró sistemáticamente los sistemas compositivos de las vanguardias pictóricas y del Movimiento Moderno, por encima de su formalismo concreto. La segunda fase de la obra de Zumthor entre 1985 y 1996 se caracteriza por la transfiguración del plano libre del sistema compositivo neoplasticista desarrollado por De Stijl, plano que adquirió grosor hasta convertirse en la caja articuladora de la fluidez de la secuencia espacial. El formalismo de Zumthor en esta fase implica la prioridad del esquema compositivo, compuesto por un perímetro regular en el que se reúnen las cajas que, como las superficies de color de las composiciones de Mondrian, articulan el espacio a su alrededor. La Residencia de Masans de Zumthor de 1993 es un proyecto canónico de este sistema compositivo, que culmina en el esquema radial de las Termas de Vals de 1996. La imagen poética se convirtió en el lenguaje propio del pensamiento de la arquitectura suizo alemana, que permitía una reflexión no verbal, a partir de unos pocos arquetipos fundamentales. Los arquitectos del Ticino buscaron en la Tendenza italiana durante la década de 1970 una alternativa al insostenible modelo territorial que estaba destruyendo su paisaje cultural. Algunos de estos arquitectos comenzaron a dar clase durante esa década en la ETHZ, por lo que transmitieron sus inquietudes al alumnado e introdujeron a Aldo Rossi en la facultad, que era uno de los máximos representantes de la Tendenza. El método analógico de Rossi, basado en la psicología analítica de Carl Jung, fue una influencia fundamental en la ETHZ durante los años 70. Rossi partía de una teoría afín al colectivo intelectual suizo que los arquitectos de la ETHZ pronto comprendieron que debían independizar del contexto italiano, desarrollando su propio método a partir de 1985 con la Analoge Architektur. El valor de la preexistencia de acuerdo a este método incipiente se hallaba en su papel articulador de la continuidad cultural de una comunidad, que Miroslav Sik definió a partir de su concepto de Altneue (literalmente, lo Nuevo-Viejo). La fusión entre lo nuevo y lo viejo era la base de la espontaneidad con la que habían crecido los asentamientos históricamente. Sik consideraba que la continuidad dependía de la comprensión de los patrones preexistentes, con el objeto de mantener su vigencia mediante la incorporación de estrategias contemporáneas, que renovarían su significado sin perder su esencia. La Casa Gugalun en Versam de Zumthor de 1994 supone una aproximación a escala doméstica a este planteamiento. La arquitectura suizo alemana reciente a menudo busca en sus referentes plásticos una manera de explorar ámbitos poco accesibles para la disciplina arquitectónica. El pensamiento arquitectónico basado en la imagen poética se implementa a partir de la tectónica de la construcción, que es el medio para comunicar una idea vertebradora del proyecto de principio a fin. La construcción como medio introduce el concepto clave de la apariencia de la arquitectura, estrechamente relacionado con la filosofía idealista de Friedrich Schiller. La apariencia debe expresar el sentido conceptual de la obra, que los arquitectos suizo alemanes denominan idea, por lo que la construcción no tiene un valor por sí misma, sino en cuanto a la apariencia. La Capilla de Oberrealta de Christian Kerez de 1993 puede ser considerada una obra escultórica en el paisaje, más próxima a las artes plásticas que a la arquitectura. La tensión que surge entre la figura ideal y la pieza sometida a una sutil deformación confiere un carácter dinámico a la composición, que intensifica el efecto perceptivo, de acuerdo a la teoría del pensamiento visual de Rudolph Arnheim. La deformación al servicio de la psicología de la percepción es un fenómeno que caracteriza la arquitectura suizo alemana reciente. El concepto de Forme Forte (forma fuerte) fue introducido por Martin Steinmann para caracterizar el objeto arquitectónico denominado internacionalmente Swiss Box. Este concepto se caracteriza por una predilección por lo monolítico, lo unitario y lo arquetípico. La condición más sustancial de una Forme Forte es, sin embargo, la de configurar un elemento estructurador del entorno. Tanto la base morfológica de una Forme Forte, como su capacidad para estructurar el lugar, se corresponden con los conceptos de arquetipo y monumento de la teoría de Rossi. La tesis explora el sentido de la deformación en la producción suizo alemana reciente, a partir de la Escuela de Paspels de Valerio Olgiati de 1998 y la Torre Roccolo de Villa Garbald de Miller&Maranta de 1998. La teoría tectónica de Gottfried Semper y la razón de la forma de Adolf Loos constituyen el sustrato teórico, que relaciona la actividad existencial de habitar con la materialidad de la arquitectura suizo alemana reciente. La teoría tectónica de Semper fundamenta las revisiones posteriores de los sistemas constructivos tradicionales suizo alemanes. Esta influencia sirvió como base a la vanguardia suiza de la Neues Bauen, que desarrolló un racionalismo funcionalista durante las décadas de 1920 y 1930, que constituye una gramática de base no simbólica. Esta gramática constituye el fundamento de la transición del orden clásico a la materialidad como instrumento compositivo durante la segunda mitad del siglo XX, consolidando el concepto de la base material de la forma. La revisión de la tradición regionalista y racionalista llevada a cabo por la generación de arquitectos suizo alemanes de la década de 1950 y 1960, constituyó una investigación lingüística sustancial. Rudolph Olgiati es la figura determinante de este periodo. La revisión de esta investigación se codificó en clave estructuralista durante los años 70 en la ETHZ, influenciada por la presencia de Roland Barthes como profesor invitado. Esta revisión constituyó la base de la articulación tectónica propiamente suizo alemana todavía vigente, como ilustra expresamente la obra de Burkhalter&Sumi en esta tesis. La obra de Gion Caminada en Vrin entre 1995 y 2002 aporta una intensa investigación sobre el Strickbau, que implementa una y otra vez variaciones sobre un sistema tradicional, ilustrando las infinitas posibilidades que ofrece un lenguaje, paradójicamente limitado por los medios disponibles en un entorno alpino aislado. Para terminar Jürg Conzett es el ingeniero con el que han desarrollado sus proyectos la mayoría de los arquitectos en el entorno de los Grisones, formado durante 8 años con Zumthor y colaborador habitual de Caminada entre otros. Conzett es el referente que relaciona la ingeniería con la arquitectura en el ámbito de esta tesis, imprescindible en el contexto de esta investigación. ABSTRACT The Alpine setting of Grisons has been a laboratory for architectural experimentation with an approach forged in the 1970s from various sources in the Swiss Federal Institute of Technology in Zurich (ETHZ). This method has produced a uniquely German-Swiss theoretical discourse over the past three decades. Tradition continues to be a cultural value for Grisons, a region committed to continue linking its architecture with the region’s history and landscape. The specificity of the German-Swiss environment is obvious, but beyond its characteristic features, there is a common denominator that gives these settings a silent unity: it is an idea of building. The phenomenon of Grisons is geographic in its specific location, but it shares the theoretical basis of the rest of the new German-Swiss architecture. This thesis transversely delves into the conditions that have shaped this common path up to the present, just as German-Swiss architecture is going through a time of generational change. In this thesis, Märkli Peter's work constitutes a paradigm of the German-Swiss will to uphold the cultural and architectural continuity of the region, exemplifying an attitude of shared resistance to the globalized world. The thesis is structured around eight recent projects carried out by German-Swiss architects in the Alpine region of Grisons. These projects represent the trajectory of twentieth century German-Swiss architectural history, as well as the architectural phenomenon that has taken place in the environs of Grisons over the past three decades. Through these projects the thesis analyzes the most relevant theoretical, artistic and philosophical references that are synthesized in the works, as well as their architectural references and the transcendental dimension of the materials in which Swiss culture is rooted: wood, stone and concrete. The artistic avant-garde, together with Modernism and the humanistic and regional movements that occurred during the last century, set the stage for present-day German-Swiss architectural sensitivity, which emerges from a complex web of relationships established between the Swiss and international panorama. This same theoretical background and experience underlies all of the German-Swiss territory in general, so associating it through this thesis to a particular context allows the description of a specific embodiment of this debate, within the unique environment of Grisons. The methodological approach to analyzing the projects was phenomenological: direct experience is the main substrate underpinning the focus of the thesis. Field work was not limited to visiting the projects featured in the thesis, but rather encompassed the most important works of recent German-Swiss construction in the Grisons in order to gain some degree of perceptual sensitivity to the uniqueness of its territorial identity. The present paper puts special emphasis on the works of Peter Märkli and Peter Zumthor, who share the formal classicist perspective of the German-Swiss context but stand on opposite sides of the philosophical spectrum with regard to the notion of materiality and the nature of space, which they conceive as systematic or fluid, respectively. The two projects that establish the boundaries of the time period 1992–2004 explored by this research represent two key milestones. The Museum La Congiunta in Giornico, by Peter Märkli, was built in 1992 and quickly established itself as an architectural icon of German-Swiss reductionism. This paradigm of conceptual and constructive synthesis began to take shape in the 1930s under the banner of Konkrete Kunst (Concrete Art), led by Max Bill. The only genuinely Swiss artistic movement of the twentieth century, Konkrete Kunst was characterized by the artistic values of precision, rigor, rationalism and abstraction, sentiments that were very close to the German-Swiss tradition. Secondly, the project for the rehabilitation of Villa Garbald and the construction of the Roccolo Tower in Castasegna, conducted by Miller&Maranta in 2004, represented the consummation of the Analoge Architektur, which was the first truly Swiss architectural movement, spearheaded in the 1980s by Miroslav Sik. The Deutscher Tendenza is a little-known period of recent German-Swiss history, represented by the important figure of Peter Zumthor. The projects that Zumthor led between 1979 and 1985 are part of a period when Italian Neo-Rationalism exercised a profound influence on the architect, as illustrated by the Churwalden School, circa 1983. Recent German-Swiss architecture systematically explored the compositional systems of the artistic avant-garde and Modernism, beyond its specific formal aspects. The second phase of Zumthor's work, between 1985 and 1996, is characterized by the transfiguration of the free plane, neoplastic compositional system developed by De Stijl, a plane that thickened until it became the box articulating fluidity from the spatial sequence. Zumthor's formalism in this phase prioritizes the compositional scheme, consisting of a regular perimeter in which the boxes that -like Mondrian’s colored surfaces- arrange the space around it. Zumthor’s Masans Residence, circa 1993, is a canonical project of this compositional system, which culminates in the radial pattern of the Therme Vals, circa 1996. The poetic image became the appropriate language of thought for German-Swiss architecture, which invited a nonverbal reflection inspired by a few fundamental archetypes. The architects of Ticino sought, through the Italian Tendenza of the 1970s, an alternative to the unsustainable territorial model that was destroying their cultural landscape. Some of these architects began to teach during that decade at ETHZ, and naturally they transmitted their unease to their students. These architects also introduced Aldo Rossi, one of the leading representatives of the Tendenza, to the school. Rossi’s analogue method, based on the analytical psychology of Carl Jung, was a major influence on the ETHZ during the 1970s. Rossi’s theoretical grounding was very much in tune with Swiss intellectualism, and it did not take long for architects from ETHZ to realize that they should break away from the Italian context, developing their own method from 1985 on with the Analoge Architektur. The value of the pre-existing conformity of this emerging method, stemmed from its role in facilitating the cultural continuity of a community, which Miroslav Sik defined in his concept of Altneue (literally, Old-new). This fusion of old and new was the basis for the spontaneity with which settlements had historically grown. Sik considered that continuity depended on understanding the existing patterns and sustaining their relevance through the incorporation of contemporary strategies, which would renew their meaning without losing their essence. Zumthor’s Gugalun House in Versam, circa 1994, is a domestic-scale approach to this philosophy. Modern German-Swiss architecture often looks to its references in visual art for a way to explore areas that are normally inaccessible to the architectural discipline. Architectural thinking based on the poetic image is achieved through a building’s tectonics, which communicate the core idea of a project from start to finish. The understanding of construction as a medium introduces the key concept of the appearance in architecture, closely related to the idealistic philosophy of Friedrich Schiller. The appearance should express the conceptual meaning of the work, which German-Swiss architects call the idea, so that the building does not have value in and of itself, but only in terms of its appearance. The Oberrealta Chapel by Christian Kerez, circa 1993, can be considered a sculpture in the landscape, closer to the visual arts than to architecture. The tension that arises between the ideal figure and the piece subjected to a subtle deformation confers a dynamic character onto the composition, intensifying the perceptual effect, according to Rudolf Arnheim’s theory of visual thought. The deformation in the service of the psychology of perception is a phenomenon that characterizes recent German-Swiss architecture. Martin Steinmann introduced the Forme Forte (Strong Form) concept to describe the architectural object known internationally as the Swiss Box. This concept is characterized by a predilection for all things monolithic, unitary and archetypal. The most substantial condition of a Forme Forte, however, is the configuration of a structuring element in the environment. Both the morphological basis of a Forme Forte and its ability to frame the place, correspond to the concepts of archetype and monument put forward by Rossi’s theory. The present thesis explores the sense of deformation in recent German-Swiss production, based on the School of Paspels by Valerio Olgiati, circa 1998 and the Roccolo Tower in Villa Garbald, by Miller&Maranta, circa 1998. Gottfried Semper’s tectonic theory and Adolf Loos’s reason for form constitute the theoretical foundation that links the existential activity of dwelling to the materiality of recent German-Swiss architecture. Semper’s tectonic theory laid the foundation for subsequent revisions of the German-Swiss traditional building systems. This influence was the basis for the Swiss avant-garde of Neues Bauen, which developed a functional rationalism during the 1920s and 30s that served as a non-symbolic grammatical foundation for the transition from classical order to materiality as a compositional tool. During the second half of the twentieth century, this architectural grammar helped consolidate the concept of the material base of the form. The revision of the regionalist and rationalist tradition, carried out by the generation of German-Swiss architects of the 1950s and 60s, constituted a substantial linguistic investigation. Rudolph Olgiati is the key figure of this period. The research was codified in terms of structuralism in the 1970s in ETHZ and influenced by the presence of Roland Barthes as a visiting professor. This revision was the basis of the uniquely German-Swiss tectonic design still in use today, as specifically illustrated by the work of Burkhalter & Sumi in this thesis. Gion Caminada's work in Vrin between 1995 and 2002 offers an extensive study on the Strickbau, which implements variations on a traditional system again and again, illustrating the endless possibilities of a language that paradoxically seems limited by the available resources in a remote Alpine setting. Finally, Jürg Conzett is the engineer with whom most architects in the Grisons region have developed their projects. Trained under Zumthor for 8 years, and a regular collaborator of Caminada, among others, Conzett is the reference point linking engineering with architecture in this thesis, essential in the context of this research.

La Mezquita-Catedral de Córdoba es un edificio vivo. Un edificio que ha sido transformado sucesivamente por hombres de razas, culturas y religiones distintas durante sus más de 1.200 años de vida y que, a pesar de ello, no ha dejado de estar en uso ni uno solo de esos días de esa larga vida. De esta forma, el edificio se muestra ante el visitante como un complejo objeto arquitectónico, resultado de una continua transformación. La capacidad de la transformación de los edificios es algo inherente a su propia condición arquitectónica, no es un hecho exclusivo de la Mezquita-Catedral. Sin embargo, en este edificio esa transformación se produce con una gran intensidad y sin pérdida de su autenticidad. Tradicionalmente, los edificios se han adaptado a los nuevos requerimientos de cada época en un proceso que ha buscado en el propio edificio las leyes o principios que habían de regir la intervención. De esta forma, tanto las sucesivas ampliaciones de la Mezquita de Abd al-Rahman I como las siguientes intervenciones cristianas debieron asumir lo preexistente como material de trabajo. Así, los arquitectos del califa al-Hakam II dialogaron con sus antecesores complejizando el espacio que recibieron, así como los Hernán Ruiz consiguieron un nuevo organismo resultante de la introducción de su arquitectura luminosa en la trama hispanomusulmana. El siglo XIX confirmó el deseo por descubrir las huellas de un pasado esplendoroso que la intervención barroca había silenciado bajo un tratamiento homogéneo del espacio. La recuperación de esas huellas supuso, hace exactamente dos siglos, el inicio de la última gran etapa en la transformación del edificio, la de la restauración. La fábrica es considerada como objeto a conservar y los esfuerzos desde ese momento se centraron en la recuperación de la arquitectura omeya latente. De este modo, la práctica de la restauración como disciplina se encontró absolutamente influenciada por la Arqueología como única fuente de conocimiento. Las intervenciones buscaban lo original como modo de recuperar espacial y formalmente aquel pasado, concentrándose en los lugares del edificio considerados como esenciales. La declaración del edificio como monumento nacional en 1882 propició que el Estado se hiciera cargo de su mantenimiento y conservación, sustituyendo en esa tarea a los Obispos y Cabildos del siglo XIX, que tuvieron un entendimiento muy avanzado para su época. La llegada del arquitecto Velázquez Bosco en las últimas décadas del siglo XIX supuso un cambio trascendental en la historia del edificio, puesto que recibió un edificio con importantes deterioros y consiguió poner las bases del edificio que hoy contemplamos. El empeño por la recuperación material y espacial devolvió a la Mezquita-Catedral buena parte de su imagen original, reproduciendo con exactitud los modelos hallados en las exploraciones arqueológicas. La llegada de Antonio Flórez tras la muerte de Velázquez Bosco supuso la traslación al edificio del debate disciplinar que se desarrolló en las dos primeras décadas del siglo XX. Flórez procuró un nuevo entendimiento de la intervención, considerando la conservación como actuación prioritaria. En 1926 el Estado reformó la manera en que se atendía al patrimonio con la creación de un sistema de zonas y unos arquitectos a cargo de ellas. La existencia de un nuevo marco legislativo apuntaló esa nueva visión conservativa, avalada por la Carta de Atenas de 1931. Este modelo restauración científica huía de la intervención en estilo y valoraba la necesidad de intervenir de la manera más escueta posible y con un lenguaje diferenciado, basándose en los datos que ofrecía la Arqueología. Por tanto, se continuaba con la valoración del edificio como documento histórico, buscando en este caso una imagen diferenciada de la intervención frente a la actitud mimética de Velázquez. Resulta destacable la manera en la que el historiador Manuel Gómez-Moreno influyó en varias generaciones de arquitectos, arqueólogos e historiadores, tanto en el entendimiento científico de la restauración como en la propia estructura administrativa. La labor desarrollada en el edificio por José Mª Rodríguez Cano primero y Félix Hernández a continuación estuvo influida de manera teórica por el método de Gómez-Moreno, aunque en muchos aspectos su labor no representó una gran diferencia con lo hecho por Velázquez Bosco. La búsqueda de lo original volvió a ser recurrente, pero la carga económica del mantenimiento de un edificio tan extenso conllevó la no realización de muchos de los proyectos más ambiciosos. Esta obsesiva búsqueda de la imagen original del edificio tuvo su última y anacrónica etapa con la intervención de la Dirección General de Arquitectura en los 70. Sin embargo, el agotamiento del modelo científico ya había propiciado un nuevo escenario a nivel europeo, que cristalizó en la Carta de Venecia de 1964 y en una nueva definición del objeto a preservar, más allá del valor como documento histórico. Esta nueva posición teórica tuvo su traslación al modelo restaurador español en el último cuarto de siglo XX, coincidiendo con la Transición. El arquitecto Dionisio Hernández Gil defendió una interpretación distinta a la de los arqueólogos y de los historiadores, que había prevalecido durante todo el siglo. En opinión de Hernández Gil, los problemas de intervención debían enfocarse fundamentalmente como problemas de Arquitectura, abandonando la idea de que solamente podían ser resueltos por especialistas. Esta convicción teórica fue defendida desde la nueva Administración y deparó la utilización de unos criterios de intervención particularizados, provenientes del análisis multifocal de cada situación y no sólo desde el valor de los edificios como documentos históricos. Y este cambio tuvo su traslación a la Mezquita-Catedral con la práctica de Gabriel Ruiz Cabrero y Gabriel Rebollo. En consecuencia con esa nueva perspectiva, aceptaron el edificio que recibieron, sustituyendo la búsqueda de aquella página original por la aceptación de cada una de las páginas de su historia y el respeto a las técnicas constructivas del pasado. La búsqueda de soluciones específicas desde el propio objeto arquitectónico significó la renovada atención a la potente estructura formal-constructiva como origen de toda reflexión. Considerar la Mezquita-Catedral en primer lugar como Arquitectura implicaba la atención a todo tipo de factores además de los históricos, como medio para preservar su autenticidad. Esta tesis pretende demostrar que la práctica de la restauración realizada en la Mezquita-Catedral a lo largo del siglo XX ha evolucionado desde la búsqueda de lo original hasta la búsqueda de lo auténtico, como reflejo de una visión basada en lo arqueológico frente a una renovada visión arquitectónica más completa, que incluye a la anterior. La consideración de la intervención en este edificio como otra página más de su historia y no como la última, significa la reedición de un mecanismo recurrente en la vida del edificio y un nuevo impulso en ese proceso de continua transformación. ABSTRACT The Mosque-Cathedral of Cordoba is a living building. A building transformed by men of different races, cultures and religions during more than 1.200 years old and that, nevertheless, it has continued to be in use all days in that long life. Thus, the building shows to the visitor as a complex architectural object, the result of continuous transformation. This transformation capacity of the buildings is inherent in their own architectural condition, it’s not an exclusive fact of the Mosque-Cathedral. However, in this building that transformation happens with a great intensity, without losing their authenticity. Traditionally, buildings have been adapted to the new requirements of times in a process that looked for laws or principles in order to guide the intervention. Thus, both the successive enlargements of the Mosque of Abd al-Rahman and Christian interventions must assume the preexistence as a working material. So, the architects of the caliph al-Hakam II spoke to their predecessors, complexing the receiving space, as well as Hernan Ruiz got a new organism as result the introduction of his luminous architecture into hispanic-muslim weft. The nineteenth century confirmed the desire to discover the traces of a glorious past that Baroque intervention had silenced, under a uniform space treatment. Exactly two centuries ago, the recovery of these traces meant the start of the last major phase in the transformation of the building: the restoration. The building was considered subject to conserve and since then, efforts focused on the recovery of latent Umayyad architecture. Thus, the practice of restoration as a discipline was absolutely influenced by Archaeology as the only source of knowledge. Interventions were seeking the original as the way to recover that past in a space and formal way, concentrating on essential sites of the building. The statement as a national monument in 1882 prompted the State take charge of its maintenance and preservation, replacing to the nineteenth century Bishops and Cabildos, which had a very advanced understanding for that time. The arrival of the architect Velazquez Bosco in the last decades of the nineteenth century involved a momentous change in the history of the building, since he received a building with significant damage and he achieved the foundations of the building that we can see today. Efforts to a material and space recover returned the Mosque-Cathedral to its original image, accurately reproducing the models found in archaeological explorations. The arrival of Antonio Florez after Velazquez’s death involved the translation of discipline debate, which was developed in the first two decades of the twentieth century. Florez tried a new understanding of the intervention, considering conservation as a priority action. In 1926, the State reformed the way in which heritage was attended, creating a zones system with a few architects in charge of them. The existence of a new legislative framework, underpinned this new conservative vision, supported by the Athens Charter of 1931. This scientific restoration model fleeing from intervention in style and it appreciated the need to intervene in the most concise way, with a distinct language based on the data offered by Archaeology. Therefore, it continued with the appraisement of the building as a historical document, seeking in this case a differentiated image of intervention, against Velazquez mimetic attitude. It is remarkable the way in which the historian Manuel Gomez-Moreno influenced several generations of architects, archaeologists and historians, both in the scientific understanding of the restoration and the administrative structure. The work of Jose Maria Rodriguez Cano first and then Felix Hernandez was theoretically influenced by the Gomez-Moreno’s method, although in many respects their work did not represent a great difference to Velazquez Bosco. The search of the original returned to recur, but the economic charge of maintaining such a large building led to the non-realization of many of the most ambitious projects. This obsessive search for the original image of the building had its last and anachronistic stage with the intervention of the Department of Architecture at 70’s. However, the exhaustion of the scientific model had already led to a new scenario at European level, which crystallized in the Venice Charter of 1964 and a new definition of the object to be preserved beyond the value as a historical document. This new theoretical position had its translation to Spanish restaurateur model in the last quarter of the twentieth century, coinciding with the Transition. The architect Dionisio Hernandez Gil defended a different interpretation from archaeologists and historians, that had prevailed throughout the century. According to Hernandez Gil, the problems of intervention should focus primarily as architectural issues, abandoning the idea that they could only be determined by specialist. This theoretical conviction was defended from the new administration and led to the use of particularized criteria, from a multifocal analysis of each situation. And this change had its translation to the Mosque with the practice of Gabriel Ruiz Cabrero and Gabriel Rebollo. Consistent with this new perspective, they accepted the receiving building, replacing the search on original page for acceptance of all historical pages and respecting the constructive techniques of the past. The search for specific solutions from the architectural object meant the renewed attention to the powerful formal-constructive structure as the origin of all thought. Consider the Mosque-Cathedral as Architecture, involved the attention to all kinds of factors in addition to the historical, as a means to preserve its authenticity. This thesis aims to demonstrate that the practice of restoration in the Mosque-Cathedral throughout the twentieth century has evolved from the search of the original to the search for the authentic, reflecting a vision based on the archaeological against a renewed more complete architectural vision, including the above. Consideration of intervention in this building as another page in its history and not the last one, means the reissue of an own mechanism and a new impetus in that continuous transformation process.

La determinación del valor del suelo implicado en los procesos de urbanización se mueve en una compleja dialéctica en la que algunos fenómenos de naturaleza morfológica como el modelado del espacio social, o el que se deduce de las determinaciones del planeamiento (aprovechamiento especialmente) se enfrentan a instrumentos de regulación que se rigen por normas de carácter administrativo (urbanísticas y expropiatorias, fiscales, financieras-hipotecarias etc.). En ese marco en el que las discrepancias son frecuentes se pretende analizar las posibles actuaciones encaminadas a influir sobre la formación de ese valor en el sentido de limitar las expectativas de aprovechamiento tradicionalmente vinculadas al sistema urbanístico establecido, destacando al respecto la implantación de criterios de sostenibilidad y participación ciudadana, así como que la utilización de normas administrativas de valoración con la finalidad de reducir tales expectativas y tratar de influir sobre el sistema urbanístico en su conjunto, no se considera suficiente para conseguir el logro de tales objetivos. Añadiendo, respecto del conjunto de normas administrativas de determinación del valor del suelo, que si bien las reglas urbanísticas y expropiatorias establecen actualmente reglas objetivas de valoración, a través de la normativa de valoración destinada a entidades financieras, o de carácter fiscal, se introducen criterios subjetivos que tratan de aproximarse al mercado, incluso en el caso del valor catastral, estructuralmente reglado. Esta separación de criterios ha llevado a un enfrentamiento al que ha venido a dar respuesta la reciente sentencia del Tribunal Supremo de 30 de mayo de 2014, que impone los criterios urbanísticos de consideración de situaciones básicas de suelo, frente a los fiscalmente regulados en relación con la normativa catastral en cuanto a la consideración de suelo urbanizable sin desarrollar, que pasa a ser considerado como en situación rural, siendo por tanto aplicables sobre el mismo las reglas de valoración que la normativa catastral establece para esta categoría de suelo (rústico) que no considera, incluso con la posterior modificación, de junio de 2015, incorporada para adaptarse a la mencionada sentencia, expectativas de aprovechamiento. Pero como se ha señalado, la exclusiva utilización de la normativa urbanística de valoración no se considera suficiente para evitar los efectos que sobre el valor de suelo produce la asignación de tales aprovechamientos, por lo que acuerdo con la actual legislación de suelo, deben asimismo potenciarse actuaciones encaminadas a un desarrollo sostenible así como a la participación ciudadana, la transparencia y el control, con la finalidad de lograr un mayor rigor en la utilización del suelo que ha de traducirse en su valoración. ABSTRACT Determining the land value involved in the urbanisation process implies a complex dialectic in which certain morphological natural phenomena like the modelling of social space or that deduced from planning determinations (especially land use) face regulatory instruments governed by administrative norms (urban planning and expropriator, tax, financial-mortgage etc.). In this context where discrepancies are common, the purpose is to analyse the possible actions geared to influence the formation of that value in the sense of limiting land use expectations traditionally linked to the established urban planning system, highlighting the implementation of sustainability criteria and citizen participation, as well as to consider that the administrative standards of evaluation for reducing said expectations, and trying to influence the urban system as a whole, are not considered sufficient for achieving these objectives. We might add, with respect to the set of administrative norms for determining the land value, that while urban and expropriations norms currently provide objective rules of valuation, subjective criteria are introduced trying to approach the market, including the structurally regulated cadastral value through valuation rules aimed at financial or fiscal institutions. This separation of criteria has led to a confrontation to which the recent ruling of the Supreme Court of May 30, 2014 responds. The aforementioned judgement imposes the urban core criteria for basic land use situations against those fiscally regulated in relation to cadastral regulations regarding the consideration of undeveloped building land, which is considered to be in a rural situation, making it subject to the same rules that cadastral valuation norms established for this category of land use (rustic) which do not consider, even with the subsequent amendment of June 2015, introduced to fit that judgment, use expectations. But as noted, the exclusive use of valuation planning regulations is not considered sufficient to avoid the effects on the land value produced by the allocation of said uses, so that according to the current land use legislation, actions aimed at sustainable development as well as citizen participation, transparency and control, shall be enhanced in order to achieve greater rigor in the use of land which should be translated in their valuation.

Although protein degradation is enhanced in muscle-wasting conditions and limits the rate of muscle growth in domestic animals, the proteolytic system responsible for degrading myofibrillar proteins in skeletal muscle is not well defined. The goals of this study were to evaluate the roles of the calpains (calcium-activated cysteine proteases) in mediating muscle protein degradation and the extent to which these proteases participate in protein turnover in muscle. Two strategies to regulate intracellular calpain activities were developed: overexpression of dominant-negative m-calpain and overexpression of calpastatin inhibitory domain. To express these constructs, L8 myoblast cell lines were transfected with LacSwitch plasmids, which allowed for isopropyl β-d-thiogalactoside-dependent expression of the gene of interest. Inhibition of calpain stabilized fodrin, a well characterized calpain substrate. Under conditions of accelerated degradation (serum withdrawal), inhibition of m-calpain reduced protein degradation by 30%, whereas calpastatin inhibitory domain expression reduced degradation by 63%. Inhibition of calpain also stabilized nebulin. These observations indicate that calpains play key roles in the disassembly of sarcomeric proteins. Inhibition of calpain activity may have therapeutic value in treatment of muscle-wasting conditions and may enhance muscle growth in domestic animals.

In human beings of both sexes, dehydroepiandrosterone sulfate (DHEAS) circulating in blood is mostly an adrenally secreted steroid whose serum concentration (in the micromolar range and 30–50% higher in men than in women) decreases with age, toward ≈20–10% of its value in young adults during the 8th and 9th decades. The mechanism of action of DHEA and DHEAS is poorly known and may include partial transformation into sex steroids, increase of bioavailable insulin-like growth factor I, and effects on neurotransmitter receptors. Whether there is a cause-to-effect relationship between the decreasing levels of DHEAS with age and physiological and pathological manifestations of aging is still undecided, but this is of obvious theoretical and practical interest in view of the easy restoration by DHEA administration. Here we report on 622 subjects over 65 years of age, studied for the 4 years since DHEAS baseline values had been obtained, in the frame of the PAQUID program, analyzing the functional, psychological, and mental status of a community-based population in the south-west of France. We confirm the continuing decrease of DHEAS serum concentration with age, more in men than in women, even if men retain higher levels. Significantly lower values of baseline DHEAS were recorded in women in cases of functional limitation (Instrumental Activities of Daily Living), confinement, dyspnea, depressive symptomatology, poor subjective perception of health and life satisfaction, and usage of various medications. In men, there was a trend for the same correlations, even though not statistically significant in most categories. No differences in DHEAS levels were found in cases of incident dementia in the following 4 years. In men (but not in women), lower DHEAS was significantly associated with increased short-term mortality at 2 and 4 years after baseline measurement. These results, statistically established by taking into account corrections for age, sex, and health indicators, suggest the need for further careful trials of the administration of replacement doses of DHEA in aging humans. Indeed, the first noted results of such “treatment” are consistent with correlations observed here between functional and psychological status and endogenous steroid serum concentrations.

The increasing resistance of the malaria parasite Plasmodium falciparum to currently available drugs demands a continuous effort to develop new antimalarial agents. In this quest, the identification of antimalarial effects of drugs already in use for other therapies represents an attractive approach with potentially rapid clinical application. We have found that the extensively used antimycotic drug clotrimazole (CLT) effectively and rapidly inhibited parasite growth in five different strains of P. falciparum, in vitro, irrespective of their chloroquine sensitivity. The concentrations for 50% inhibition (IC50), assessed by parasite incorporation of [3H]hypoxanthine, were between 0.2 and 1.1 μM. CLT concentrations of 2 μM and above caused a sharp decline in parasitemia, complete inhibition of parasite replication, and destruction of parasites and host cells within a single intraerythrocytic asexual cycle (≈48 hr). These concentrations are within the plasma levels known to be attained in humans after oral administration of the drug. The effects were associated with distinct morphological changes. Transient exposure of ring-stage parasites to 2.5 μM CLT for a period of 12 hr caused a delay in development in a fraction of parasites that reverted to normal after drug removal; 24-hr exposure to the same concentration caused total destruction of parasites and parasitized cells. Chloroquine antagonized the effects of CLT whereas mefloquine was synergistic. The present study suggests that CLT holds much promise as an antimalarial agent and that it is suitable for a clinical study in P. falciparum malaria.

Epidemiological evidence has suggested that some pediatric leukemias may be initiated in utero and, for some pairs of identical twins with concordant leukemia, this possibility has been strongly endorsed by molecular studies of clonality. Direct evidence for a prenatal origin can only be derived by prospective or retrospective detection of leukemia-specific molecular abnormalities in fetal or newborn samples. We report a PCR-based method that has been developed to scrutinize neonatal blood spots (Guthrie cards) for the presence of numerically infrequent leukemic cells at birth in individuals who subsequently developed leukemia. We demonstrate that unique or clonotypic MLL-AF4 genomic fusion sequences are present and detectable in neonatal blood spots from individuals who were diagnosed with acute lymphoblastic leukemia at ages 5 months to 2 years and, therefore, have arisen during fetal hematopoiesis in utero. This result provides unequivocal evidence for a prenatal initiation of acute leukemia in young patients. The method should be applicable to other fusion genes in children with common subtypes of leukemia and will be of value in attempts to unravel the natural history and etiology of this major subtype of pediatric cancer.

The yeast Candida albicans has a distinguishing feature, dimorphism, which is the ability to switch between two morphological forms: a budding yeast form and a multicellular invasive filamentous form. This ability has been postulated to contribute to the virulence of this organism. Studies on the morphological transition from a filamentous to a budding yeast form in C. albicans have shown that this organism excretes an autoregulatory substance into the culture medium. This substance was extracted and purified by normal-phase and reversed-phase HPLC. The autoregulatory substance was structurally identified as 3,7,11-trimethyl-2,6,10-dodecatrienoate (farnesoic acid) by NMR and mass spectrometry. Growth experiments suggest that this substance does not inhibit yeast cell growth but inhibits filamentous growth. These findings have implications for developmental signaling by the fungus and might have medicinal value in the development of antifungal therapies.

We have used a fluorescence recovery after photobleaching (FRAP) technique to measure radial diffusion of myoglobin and other proteins in single skeletal and cardiac muscle cells. We compare the radial diffusivities, Dr (i.e., diffusion perpendicular to the long fiber axis), with longitudinal ones, Dl (i.e., parallel to the long fiber axis), both measured by the same technique, for myoglobin (17 kDa), lactalbumin (14 kDa), and ovalbumin (45 kDa). At 22°C, Dl for myoglobin is 1.2 × 10−7 cm2/s in soleus fibers and 1.1 × 10−7 cm2/s in cardiomyocytes. Dl for lactalbumin is similar in both cell types. Dr for myoglobin is 1.2 × 10−7 cm2/s in soleus fibers and 1.1 × 10−7 cm2/s in cardiomyocytes and, again, similar for lactalbumin. Dl and Dr for ovalbumin are 0.5 × 10−7 cm2/s. In the case of myoglobin, both Dl and Dr at 37°C are about 80% higher than at 22°C. We conclude that intracellular diffusivity of myoglobin and other proteins (i) is very low in striated muscle cells, ≈1/10 of the value in dilute protein solution, (ii) is not markedly different in longitudinal and radial direction, and (iii) is identical in heart and skeletal muscle. A Krogh cylinder model calculation holding for steady-state tissue oxygenation predicts that, based on these myoglobin diffusivities, myoglobin-facilitated oxygen diffusion contributes 4% to the overall intracellular oxygen transport of maximally exercising skeletal muscle and less than 2% to that of heart under conditions of high work load.

Human monoclonal antibodies have considerable potential in the prophylaxis and treatment of viral disease. However, only a few such antibodies suitable for clinical use have been produced to date. We have previously shown that large panels of human recombinant monoclonal antibodies against a plethora of infectious agents, including herpes simplex virus types 1 and 2, can be established from phage display libraries. Here we demonstrate that facile cloning of recombinant Fab fragments against specific viral proteins in their native conformation can be accomplished by panning phage display libraries against viral glycoproteins "captured" from infected cell extracts by specific monoclonal antibodies immobilized on ELISA plates. We have tested this strategy by isolating six neutralizing recombinant antibodies specific for herpes simplex glycoprotein gD or gB, some of which are against conformationally sensitive epitopes. By using defined monoclonal antibodies for the antigen-capture step, this method can be used for the isolation of antibodies to specific regions and epitopes within the target viral protein. For instance, monoclonal antibodies to a nonneutralizing epitope can be used in the capture step to clone antibodies to neutralizing epitopes, or antibodies to a neutralizing epitope can be used to clone antibodies to a different neutralizing epitope. Furthermore, by using capturing antibodies to more immunodominant epitopes, one can direct the cloning to less immunogenic ones. This method should be of value in generating antibodies to be used both in the prophylaxis and treatment of viral infections and in the characterization of the mechanisms of antibody protective actions at the molecular level.

Técnicas analíticas empregadas para a quantificação do teor de lignina em plantas forrageiras, atualmente em uso, são questionáveis quanto às suas acurácias. O método lignina detergente ácido (LDA), que é um dos métodos mais utilizado em Ciência Animal e Agronomia, apresenta algumas falhas, particularmente devido à parcial solubilização da lignina durante a preparação da fibra em detergente ácido (FDA). A lignina Klason (LK), outro método muito usado, apresenta o inconveniente de mensurar a proteína da parede celular como sendo lignina. Em ambos os procedimentos recomenda-se também mensurar cinzas nos resíduos de lignina. A quantificação da concentração de lignina pelo método espectrofotométrico lignina brometo de acetila (LBA) vem ganhando interesse de pesquisadores no Brasil e no exterior. Nesta metodologia, a lignina da planta contida na preparação parede celular (PC) é solubilizada numa solução a 25% de brometo de acetila em ácido acético e a absorbância mensurada é com luz UV a 280 nm. O valor da absorbância é inserido numa equação de regressão e a concentração de lignina é obtida. Para que esta técnica analítica seja mais aceita pelos pesquisadores, ela deve ser, obviamente, convincente e atrativa. O presente trabalho analisou alguns parâmetros relacionados à LBA em 7 gramíneas e 6 leguminosas, em dois estádios de maturidade. Dentre as diferentes temperaturas de pré-secagem, os resultados indicaram que os procedimentos de 55°C com ventilação e liofilização podem ser utilizados com a mesma eficácia. As temperaturas de 55°C sem ventilação e 80°C sem ventilação não são recomendadas, pois aumentaram os valores de FDA e LDA, possivelmente devido ao surgimento de artefatos de técnica como os compostos de Maillard. No método LBA os valores menores das amostras de leguminosas chamaram a atenção e colocaram em questão se a lignina destas plantas seria menos solúvel no reagente brometo de acetila. Dentre algumas alterações na metodologia da técnica LBA, a utilização do moinho de bolas (para diminuir o tamanho particular) nas amostras de PC não mostrou efeito; a hipótese era melhorar a solubilização da lignina usando partículas menores. O uso de um ultrasonicador, que aumenta a vibração das moléculas e assim, facilitaria a solubilização da lignina no reagente brometo de acetila, melhorou a solubilização da lignina em cerca de 10%, tanto nas gramíneas como nas leguminosas. Foi acoplado um ensaio biológico como referência, a degradabilidade in vitro da matéria seca (DIVMS); e como a lignina está intimamente associada à estrutura fibrosa da parede celular, também foi feito um ensaio de degradabilidade in vitro da fibra em detergente neutro (DIVFDN). Os resultados confirmaram o efeito da maturidade, reduzindo a degradabilidade nas plantas mais maduras, e que o teor de lignina de leguminosas é realmente inferior ao de gramíneas. Os resultados de degradabilidade apresentaram coeficientes de correlação mais elevados com o método LBA, quando foi empregada a técnica do ultrasom; o método LK mostrou os menores coeficientes. Também testou-se, com sucesso, a utilização da FDN, como preparação fibrosa, ao invés de PC. A razão é simples: enquanto que a FDN é amplamente conhecida, a preparação PC não o é. Inquestionável que esta manobra facilitará substancialmente a divulgação desse método, tornando-a mais aceitável pela comunidade científica
