Malgré le progrès technologique et nos connaissances pharmaceutiques et médicales croissantes, le développement du médicament demeure un processus difficile, dispendieux, long et très risqué. Ce processus mérite d'être amélioré pour faciliter le développement de nouveaux traitements. À cette fin, cette thèse vise à démontrer l’utilité de principes avancés et d’outils élaborés en pharmacocinétique (PK), actuels et nouveaux. Ces outils serviront à répondre efficacement à des questions importantes lors du développement d’un médicament, sauvant ainsi du temps et des coûts. Le premier volet de la thèse porte sur l’utilisation de la modélisation et des simulations et la création d’un nouveau modèle afin d’établir la bioéquivalence entre deux formulations de complexe de gluconate ferrique de sodium en solution de sucrose pour injection. Comparé aux méthodes courantes, cette nouvelle approche proposée se libère de plusieurs présuppositions, et requiert moins de données. Cette technique bénéficie d’une robustesse scientifique tout en étant associée à des économies de temps et de coûts. Donc, même si développé pour produits génériques, elle pourra également s’avérer utile dans le développement de molécules innovatrices et « biosimilaires ». Le deuxième volet décrit l’emploi de la modélisation pour mieux comprendre et quantifier les facteurs influençant la PK et la pharmacodynamie (PD) d’une nouvelle protéine thérapeutique, la pegloticase. L’analyse a démontré qu’aucun ajustement posologique n’était nécessaire et ces résultats sont inclus dans la monographie officielle du produit. Grâce à la modélisation, on pouvait répondre à des questions importantes concernant le dosage d’un médicament sans passer par des nouvelles études ni d'évaluations supplémentaires sur les patients. Donc, l’utilisation de cet outil a permis de réduire les dépenses sans prolonger le processus de développement. Le modèle développé dans le cadre de cette analyse pourrait servir à mieux comprendre d’autres protéines thérapeutiques, incluant leurs propriétés immunogènes. Le dernier volet démontre l’utilité de la modélisation et des simulations dans le choix des régimes posologiques d’un antibiotique (TP-434) pour une étude de Phase 2. Des données provenant d’études de Phase 1 ont été modélisées au fur et à mesure qu’elles devenaient disponibles, afin de construire un modèle décrivant le profil pharmacocinétique du TP-434. Ce processus de modélisation exemplifiait les cycles exploratoires et confirmatoires décrits par Sheiner. Ainsi, en se basant sur des relations PK/PD d’un antibiotique de classe identique, des simulations ont été effectuées avec le modèle PK final, afin de proposer de nouveaux régimes posologiques susceptibles d’être efficace chez les patients avant même d'effectuer des études. Cette démarche rationnelle a mené à l’utilisation de régimes posologiques avec une possibilité accrue d’efficacité, sans le dosage inutile des patients. Ainsi, on s’est dispensé d’études ou de cohortes supplémentaires coûteuses qui auraient prolongé le processus de développement. Enfin, cette analyse est la première à démontrer l’application de ces techniques dans le choix des doses d’antibiotique pour une étude de Phase 2. En conclusion, cette recherche démontre que des outils de PK avancés comme la modélisation et les simulations ainsi que le développement de nouveaux modèles peuvent répondre efficacement et souvent de manière plus robuste à des questions essentielles lors du processus de développement du médicament, tout en réduisant les coûts et en épargnant du temps.

La démarche scientifique (ou expérimentale) en milieu scolaire nécessite des savoir-faire expérimentaux qui ne s’acquièrent habituellement qu’en présentiel, c’est-à-dire en laboratoire institutionnel où l’enseignant ou le technicien sont présents et peuvent, à tout moment, assister pleinement l’apprenant dans sa démarche d’investigation scientifique et technologique. Ils peuvent l’orienter, le diriger, susciter sa réflexion, faire des démonstrations réelles ou contrôler son expérimentation en lui montrant comment paramétrer les outils d’expérimentation assistée par ordinateur (ExAO). Pour répondre aux besoins de la formation à distance, cette recherche de développement en didactique des sciences et de la technologie propose de mettre à la disposition des apprenants et des enseignants un environnement de laboratoire informatisé, contrôlé et assisté à distance. Cet environnement, axé sur un microlaboratoire d’ExAO (MicrolabExAO), que nous avons nommé Ex@O pour le distinguer, a été testé de manière fonctionnelle, puis évalué en situation réelle par des étudiants-maîtres et des élèves de l’éducation des adultes qui ont pratiqué et expérimenté la démarche scientifique, en situation de laboratoire réel, mais à distance. Pour ce faire, nous avons couplé le logiciel MicrolabExAO à un logiciel de prise en main à distance avec outils audio et vidéo (Teamviewer). De plus, nous avons créé et inséré, dans le logiciel MicrolabExAO, une aide en ligne pour télécharger et faciliter la prise en main à distance. Puisque cet environnement Ex@O permet de multiplier les contacts des apprenants avec une expérimentation concrète, ce prototype répond bien à l’un des objectifs du Programme de formation de l’école québécoise (PFEQ) qui est de rendre l’apprenant plus actif dans ses apprentissages. Et parce que ce premier prototype d’environnement Ex@O permet d’effectuer des activités en laboratoire à distance, nous avons pu vérifier qu’il met aussi l’accent, non seulement sur les savoirs, mais également sur les savoir-faire expérimentaux en sciences et technologie, traditionnellement développés dans les locaux des laboratoires institutionnels. Notons ici que la démarche expérimentale s’acquiert très majoritairement en laboratoire en pratiquant, souvent et régulièrement, le processus inductif et déductif propre à cette démarche. Cette pratique de la démarche expérimentale, à distance, avec la technologie Ex@O qui l’accompagne, nous a permis de vérifier que celle-ci était possible, voire comparable à la réalisation, pas-à-pas, d’un protocole expérimental effectué dans un laboratoire institutionnel.

Es preciso que Colombia se muestre ante la U.E como un socio estratégico para el desarrollo de sus intereses científicos y tecnológicos, lo cual es posible exhibiendo importantes resultados en el Séptimo Programa Marco (7PM), tanto en el número de proyectos avalados como en la excelencia contenida en los mismos. Esto conducirá a fortalecer la dinámica de colaboración internacional en CyT entre estos dos actores.

Después que muchos Estados se enriquecieran tanto intelectual como económicamente de la explotación o apropiación ilegal de la biodiversidad andina de la cual los miembros de la CAN eran país de origen, surge la necesidad de proteger la riqueza de la región, razón por la cual nace la Decisión 391 en 1996 sobre un Régimen Común de Acceso a Recursos genéticos. De esta manera los países miembros de la CAN se convirtieron en los primeros con un mecanismo de control para el acceso a los recursos genéticos y partición equitativa de los beneficios; buscando con ello sentar bases para la creación de conocimientos sobre el tema, el desarrollo de capacidades científicas y tecnológicas, y la promoción del uso sostenible de los recursos. Sin embargo por razones de carácter nacional la implementación del mismo no ha progresado, generando que los casos de biopiratería aumenten e incluso sean difíciles de prevenir. Siendo este un tema de gran relevancia para Bolivia, Colombia, Ecuador, Perú y Venezuela, esta monografía examinará en las siguientes páginas la reglamentación nacional para comprender a mayor profundidad los defectos y/o cualidades de las decisiones tomadas por los Estados para la aplicación de la Decisión 391, y así mismo evidenciará las razones por las cuales la implementación de la Decisión no ha tenido éxito total sobre el control de la biopiratería, entre ellas la falta de reglamentación, la especificación de roles, la concientización ciudadana e incluso información al público en general.

Un gran número de empresas están inmersas actualmente en espacios de mercado conocidos y saturados de competidores. La innovación constituye una de las principales alternativas de las empresas para encontrar su posicionamiento estratégico y adaptarse a los cambios del entorno (Kim & Mauborgne, 2005). Igualmente, Demirci (2013) asegura que la cultura es un factor clave en la innovación, dado que está fuertemente asociada con los valores, actitudes, comportamientos y prácticas organizacionales. Esta investigación abarca el estudio de la cultura organizacional y la innovación en el marco de estrategias de cooperación inter-organizacional donde se plantea que el grado de cooperación que existe entre las empresas tiene un efecto sobre los valores culturales y la incorporación de innovaciones en cada organización. Para esto se llevó a cabo una investigación cuantitativa con un alcance de tipo descriptivo y de carácter no experimental y trans seccional, cuya unidad de análisis fueron 20 empresas de la red ParqueSoft Manizales. Para la medición de las variables de innovación se aplicó un instrumento basado en el Manual de Oslo de la OECD y Eurostat (2005) el cual contempla la innovación de producto, proceso, mercadotecnia y organización. A nivel de los valores culturales, la medición se realizó a través de un cuestionario inspirado en el modelo de Hofstede (1980). Los resultados obtenidos permiten demostrar que existe un grado de relación entre la cooperación y los valores culturales ‘distancia al poder’ y ‘tolerancia a la incertidumbre´, sin poder determinar la relación con la generación de innovación de producto, proceso, mercadotecnia y organización, así como con las otras dimensiones del modelo de valores de Hofstede.

Este documento presenta una revisión de las principales aproximaciones teóricas sobre recursos humanos en ciencia y tecnología y la modelación empírica de las carreras académicas y científicas utilizando los CVs como fuente de información principal. Adicionalmente, muestra los resultados de varios estudios realizados en Colombia basados en la teoría del capital conocimiento. Estos estudios han permitido establecer una línea de investigación sobre la evaluación del comportamiento de los recursos humanos, el tránsito hacia comunidades científicas y el estudio de las carreras académicas de los investigadores. Adicionalmente, muestran que la información contenida en la Plataforma ScienTI (Grup-Lac y Cv-Lac) permite establecer de manera concreta las capacidades científicas y tecnológicas del país.

Internal tapered connections were developed to improve biomechanical properties and to reduce mechanical problems found in other implant connection systems. The purpose of this study was to evaluate the effects of mechanical loading and repeated insertion/removal cycles on the torque loss of abutments with internal tapered connections. Sixty-eight conical implants and 68 abutments of two types were used. They were divided into four groups: groups 1 and 3 received solid abutments, and groups 2 and 4 received two-piece abutments. In groups 1 and 2, abutments were simply installed and uninstalled; torque-in and torque-out values were measured. In groups 3 and 4, abutments were installed, mechanically loaded and uninstalled; torque-in and torque-out values were measured. Under mechanical loading, two-piece abutments were frictionally locked into the implant; thus, data of group 4 were catalogued under two subgroups (4a: torque-out value necessary to loosen the fixation screw; 4b: torque-out value necessary to remove the abutment from the implant). Ten insertion/removal cycles were performed for every implant/abutment assembly. Data were analyzed with a mixed linear model (P <= 0.05). Torque loss was higher in groups 4a and 2 (over 30% loss), followed by group 1 (10.5% loss), group 3 (5.4% loss) and group 4b (39% torque gain). All the results were significantly different. As the number of insertion/removal cycles increased, removal torques tended to be lower. It was concluded that mechanical loading increased removal torque of loaded abutments in comparison with unloaded abutments, and removal torque values tended to decrease as the number of insertion/removal cycles increased. Introduction: The objective of this study was to investigate the expression of matrix metalloproteinases (MM Ps) in apical periodontitis and during the periapical healing phase after root canal treatment. Methods: Apical periodontitis was induced in dog teeth, and root canal treatment was performed in a single visit or by using an additional calcium hydroxide root canal dressing. One hundred eighty days after treatment the presence of inflammation was examined, and tissues were stained to detect bacteria. Bacterial status was correlated to the degree of tissue organization, and to further investigate molecules involved in this process, tissues were stained for MMP-1, MMP-2, MMP-8, and MMP-9. Data were analyzed by using one-way analysis of variance followed by Tukey test or Kruskal-Wallis followed by Dunn test. Results: Teeth with apical periodontitis that had root canal therapy performed in a single visit presented an intense inflammatory cell infiltrate. Periapical tissue was extremely disorganized, and this was correlated with the presence of bacteria. Higher MMP expression was evident, similar to teeth with untreated apical periodontitis. In contrast, teeth with apical periodontitis submitted to root canal treatment with calcium hydroxide presented a lower inflammatory cell infiltrate. This group had moderately organized connective tissue, lower prevalence of bacteria, and lower number of MMP-positive cells, similar to healthy teeth submitted to treatment. Conclusions: Teeth treated with calcium hydroxide root canal dressing exhibited a lower percentage of bacterial contamination, a lower MMP expression, and a more organized extracellular matrix, unlike those treated in a single visit. This suggests that calcium hydroxide might be beneficial in tissue repair processes. (J Endod 2010;36:231-237)

Objective: Using checkerboard DNA-DNA hybridisation (CDDH) assay, this randomised clinical study evaluated the contamination of metallic brackets by four cariogenic bacterial strains (Streptococcus mutans, Streptococcus sobrinus, Lactobacillus casei and Lactobacillus acidophilus) and the efficacy of 0.12% chlorhexidine gluconate (CHX) mouthwashes in reducing bacterial contamination. Methods: Thirty-nine 11-33-year-old patients under treatment with fixed orthodontic appliances were enrolled in the study and had 2 new metallic brackets bonded to premolars. Nineteen patients used a 0.12% CHX mouthwash (Periogard (R)) and 20 patients used a placebo mouthwash (control) twice a week. After 30 days, the brackets were removed and samples were obtained for analysis by CDDH. Data were analysed statistically by the Kruskal-Wallis test (alpha = 0.05) using the SAS software. Results: S. mutans, S. sobrinus, L. casei and L. acidophilus were detected in 100% of the samples from both groups. However, brackets of the control group were more heavily contaminated by S. mutans and S. sobrinus (P < 0.01). In the experimental group, although all counts decreased after rinsing with the chlorhexidine solution, there was significant difference only for S. mutans (P = 0.03). Conclusions: The use of 0.12% chlorhexidine gluconate mouthwashes can be useful in clinical practice to reduce the levels of cariogenic microorganisms in patients under treatment with fixed orthodontic appliances. Aims: To assess the prevalence of primary headaches (HA) in adults with temporomandibular disorders (TMD) who were assessed in a specialty orofacial pain clinic, as well as in controls without TMD. Methods: The sample consisted of 158 individuals with TMD seen at a university-based specialty clinic, as well as 68 controls. The Research Diagnostic Criteria for TMD were used to diagnose the TMD patients. HAs were assessed using a structured interview and classified according to the Second Edition of the International Classification for Headache Disorders. Data were analyzed by chi-square tests with a significance level of 5% and odds ratio (OR) tests with a 95% confidence interval (CI). Results: HAs occurred in 45.6% of the control group (30.9% had migraine and 14.7% had tension-type headache [TTH]) and in 85.5% of individuals with TMD. Among individuals with TMD, migraine was the most prevalent primary HA (55.3%), followed by TTH (30.2%); 14.5% had no HA. In contrast to controls, the odds ratio (OR) for HA in those with TMD was 7.05 (95% confidence interval [CI] = 3.65-13.61; P = .000), for migraine, the OR was 2.76 (95% CI = 1.50-5.06; P = .001), and for TTH, the OR was 2.51 (95% CI = 1.18-5.35; P = .014). Myofascial pain/arthralgia was the most common TMD diagnosis (53.2%). The presence of HA or specific HAs was not associated with the time since the onset of TMD (P = .714). However, migraine frequency was positively associated with TMD pain severity (P = .000). Conclusion: TMD was associated with increased primary HA prevalence rates. Purpose. To describe the occurrence of self-reported problems of accessibility to health services used by persons with disabilities in terms of social and health services variables. Methods. We performed a cross-sectional household survey designed to assess problems with accessibility to health services faced by persons with disabilities. We interviewed 333 persons in Sao Paulo city, in 2007. Variables related to the presence of accessibility problems, disabilities, gender, age, family head income, ethnicity, use of health services and others were analysed using frequencies, percentages, chi(2)-test, ANOVA and Poisson regression models. Results. 15.92% of the interviewed persons reported problems with accessibility to health services. Persons having multiple (prevalence ratios; PR = 2.91) or mobility disability (PR = 6.46) had more problems with accessibility than persons with hearing disability. Persons younger than 78 years old had more problems with accessibility; those who needed help to go to the health service (PR = 3.01) also. Conclusions. Persons with multiple or mobility disability, younger than 78 years, and those who needed help of others to go to the health service were more likely to have problems with accessibility to health services. This information could be one of the first steps to the management and/or planning of appropriate health services for persons with disabilities.

In the first part some information and characterisation about an AC distribution network that feeds traction substations and their possible influences on the DC traction load flow are presented. Those influences are investigated and mathematically modelled. To corroborate the mathematical model, an example is presented and their results are confronted with real measurements.

The objective of this study was to select the optimal operational conditions for the production of instant soy protein isolate (SPI) by pulsed fluid bed agglomeration. The spray-dried SPI was characterized as being a cohesive powder, presenting cracks and channeling formation during its fluidization (Geldart type A). The process was carried out in a pulsed fluid bed, and aqueous maltodextrin solution was used as liquid binder. Air pulsation, at a frequency of 600 rpm, was used to fluidize the cohesive SPI particles and to allow agglomeration to occur. Seventeen tests were performed according to a central composite design. Independent variables were (i) feed flow rate (0.5-3.5 g/min), (ii) atomizing air pressure (0.5-1.5 bar) and (iii) binder concentration (10-50%). Mean particle diameter, process yield and product moisture were analyzed as responses. Surface response analysis led to the selection of optimal operational parameters, following which larger granules with low moisture content and high process yield were produced. Product transformations were also evaluated by the analysis of size distribution, flowability, cohesiveness and wettability. When compared to raw material, agglomerated particles were more porous and had a more irregular shape, presenting a wetting time decrease, free-flow improvement and cohesiveness reduction. Tenofovir disoproxil fumarate (TDF) is a first-line drug used in patients with highly active retroviral disease; however, it can cause renal failure associated with many tubular anomalies that may be due to down regulation of a variety of ion transporters. Because rosiglitazone, a peroxisome proliferator-activated receptor-gamma agonist induces the expression of many of these same transporters, we tested if the nephrotoxicity can be ameliorated by its use. High doses of TDF caused severe renal failure in rats accompanied by a reduction in endothelial nitric-oxide synthase and intense renal vasoconstriction; all of which were significantly improved by rosiglitazone treatment. Low-dose TDF did not alter glomerular filtration rate but produced significant phosphaturia, proximal tubular acidosis, polyuria and a reduced urinary concentrating ability. These alterations were caused by specific downregulation of the sodium-phosphorus cotransporter, sodium/hydrogen exchanger 3 and aquaporin 2. A Fanconi`s-like syndrome was ruled out as there was no proteinuria or glycosuria. Rosiglitazone reversed TDF-induced tubular nephrotoxicity, normalized urinary biochemical parameters and membrane transporter protein expression. These studies suggest that rosiglitazone treatment might be useful in patients presenting with TFV-induced nephrotoxicity especially in those with hypophosphatemia or reduced glomerular filtration rate.

Recent literature has highlighted that the flexibility of walking barefoot reduces overload in individuals with knee osteoarthritis (OA). As such, the aim of this study was to evaluate the effects of inexpensive, flexible, non-heeled footwear (Moleca (R)) as compared with a modern heeled shoes and walking barefoot on the knee adduction moment (KAM) during gait in elderly women with and without knee OA. The gait of 45 elderly women between 60 and 70 years of age was evaluated. Twenty-one had knee OR graded 2 or 3 according to Kellgren and Lawrence`s criteria, and 24 who had no OA comprised the control group (CG). The gait conditions were: barefoot, Moleca (R), and modern heeled shoes. Three-dimensional kinematics and ground reaction forces were measured to calculate KAM by inverse dynamics. For both groups, the Moleca (R) provided peak KAM and KAM impulse similar to barefoot walking. For the OA group, the Moleca (R) reduced KAM even more as compared to the barefoot condition during midstance. On the other hand, the modern heeled shoes increased this variable in both groups. Inexpensive, flexible, and non-heeled footwear provided loading on the knee joint similar to a barefoot gait and significant overload decreases in elderly women with and without knee OA, compared to modern heeled shoes. During midstance, the Moleca (R) also allowed greater reduction in the knee joint loads as compared to barefoot gait in elderly women with knee OA, with the further advantage of providing external foot protection during gait.
