Neurofibromatosis 2 (NF2) is a dominantly inherited disorder, which predisposes to multiple tumours of the nervous system, typically schwannomas and meningiomas. Biallelic inactivation of the NF2 gene occurs both in sporadic and NF2-related schwannomas and in most meningiomas. The NF2 gene product merlin (or schwannomin) is structurally related to the ERM proteins, ezrin, radixin and moesin, which act as molecular linkers between the actin cytoskeleton and the plasma membrane. Merlin is a tumor suppressor that participates in cell cycle regulation. Merlin s phosphorylation status appears to be associated with its tumour suppressor activity, i.e. non-phosphorylated merlin functions as a tumour suppressor, whereas protein phosphorylation results in loss of functional activity. This thesis study was initiated to investigate merlin s role as a tumor suppressor and growth inhibitor. These studies show, that like many other tumor suppressors, also merlin is targeted to the nucleus at some stages of the cell cycle. Merlin s nuclear localization is regulated by cell cycle phase, contact inhibition and adhesion. In addition, a potential nuclear binding partner for merlin was identified, Human Enhancer of Invasion 10 (HEI10), a cyclin B interacting protein. Many tumor suppressors interact with microtubules and this thesis work shows that also merlin colocalizes with microtubules in mitotic structures. Merlin binds microtubules directly, and increases their polymerization in vitro and in vivo. In addition, primary mouse Schwann cells lacking merlin displays disturbed microtubule cytoskeleton. Fourth part of this thesis work began from the notion that PKA phosphorylates an unidentified site from the merlin N-terminus. Our studies show that serine 10 is a target for PKA and modulation of this residue regulates cytoskeletal organization, lamellipodia formation and cell migration. In summary, this thesis work shows that merlin s role is much more versatile than previously thought. It has a yet unidentified role in the nucleus and it participates in the regulation of both microtubules and the actin cytoskeleton. These studies have led to a better understanding of this enigmatic tumor suppressor, which eventually will aid in the design of specific drugs for the NF2 disease.

The potential for fertiliser use in Lockyer Valleys intensive vegetable production to impact on the Moreton Bay Waterways (MBW) is not well defined. Notwithstanding nutrient runoff through soil erosion of agricultural lands has been identified as a process that significantly contributes artificial fertiliser to the MBW (SEQ Healthy Waterways Draft Strategy 2006). In order to better understand this issue the present study undertakes a nutrient mass balance to evaluate nitrogen use efficiency in the intensive horticultural industry of the Lockyer Valley.

This project investigates the impact of vegetable production systems on sensitive waterways focusing on the risk of off-site nutrient movement at farm block scale under current management practices. The project establishes a series of case studies in two environmentally important Queensland catchments and conducts a broader survey of partial nutrient budgets across tropical vegetable production. It will deliver tools to growers that can improve fertiliser use efficiency delivering profitability and environmental improvements.

Hydroponic production systems offer optimal conditions for rapid growth, protection from adverse weather and greater water use efficiency. The most important limitation for hydroponic production production is water borne disease. Water borne disease can rapidly spread causing up to 100% crop failure.

A theoretical analysis of the Gifford-McMahon cycle is presented. Expressions for the ideal refrigeration produced and the figure of merit are developed. Various losses occurring in a real machine are considered and equations to account for these losses are derived. Results are presented in graphical form.

A recent work obtained closed-form solutions to the.problem of optimally grouping a multi-item inventory into subgroups with a common order cycle per group, when the distribution by value of the inventory could be described by a Pareto function. This paper studies the sensitivity of the optimal subgroup boundaries so obtained. Closed-form expressions have been developed to find intervals for the subgroup boundaries for any given level of suboptimality. Graphs have been provided to aid the user in selecting a cost-effective level of aggregation and choosing appropriate subgroup boundaries for a whole range of inventory distributions. The results of sensitivity analyses demonstrate the availability of flexibility in the partition boundaries and the cost-effectiveness of any stock control system through three groups, and thus also provide a theoretical support to the intuitive ABC system of classifying the items.

This project has shown the potential for cotton production in the region developed a range of tactics that can be deployed to minimise the impact of cloudy wet weather. These agronomic tactics have been published in a new book - NORpak - Cotton production and management guidelines for the Burdekin and NQ coastal dry tropics. This publication has been specifically targeted for local sugarcane producers who may stand to benefit by including cotton rotation crops into their current largely mono-culture production systems. This publication is available at http://www.cottoncrc.org.au/industry/Publications/Northern_Production.
