Fluorescent in situ hybridization (FISH) with commercial probes covering the elastin gene (ELN) was used to determine the frequency of the 7q11.23 deletion in 18 children clinically diagnosed with Williams-Beuren syndrome (WBS). A de novo deletion was detected in 15 of the children (83%). Diagnostic investigation for WBS started late in childhood (median = 5.8 years). All the children showed facial features typical of the syndrome, mental retardation and developmental delay. Over-friendliness was observed in the majority of cases. Clinodactyly of the 5th finger (n = 13), cardiovascular disease (n = 9), loquacity (n = 9), low birthweight (n = 8), and failure to thrive (n = 9) were observed only in those children with the deletion. Respiratory problems (n = 9), though not previously reported in the literature, was a common finding in the group studied. Our results confirmed that FISH is useful in identifying 7q11.23 deletions in cases of WBS. Clinical manifestations were more evident in the deletion-positive children.

RESUMO - O objetivo deste trabalho foi comparar o sistema de monitoramento computadorizado da produção de gás in vitro com os métodos in vivo e in situ. Nas comparações, foram utilizadas silagens de milho com alto/baixo teores de matéria seca, com/sem inoculante. A digestibilidade das silagens com alto teor de matéria seca (MS), com/sem inoculação, não apresentou diferenças entre os métodos analisados. Quando avaliadas separadamente do efeito do inóculo, essas silagens diferiram nos métodos in vitro/gás e in situ, mas não no in vivo. Entretanto, quando analisadas somente sob efeito do inoculante, apenas no método in situ houve diferença. Não foram encontradas diferenças na digestibilidade da fibra em detergente neutro (FDN) das silagens com alta ou baixa MS, inoculadas ou não, bem como em relação ao pH ao final da digestão. em conclusão, o desaparecimento da MS e FDN quantificada pelo resíduo no sistema in vitro/gás foi semelhante aos demais métodos avaliados.

Objetivou-se no presente trabalho verificar a degradação ruminal e a digestibilidade intestinal e total da matéria seca (MS) e da proteína bruta (PB) do farelo de soja, do grão de milho, do melaço em pó, da farinha de peixe, da farinha de penas e do feno de alfafa, por intermédio da técnica de degradabilidade ruminal in situ associada à técnica do saco de náilon móvel. As amostras dos alimentos foram moídas e colocadas em duplicata em sacos de náilon de 10x5 cm (48 micras) nas quantidades de 15 e 5 mg de MS/cm² para os alimentos concentrados e feno de alfafa, respectivamente. Os sacos de náilon permaneceram incubados no rúmen de bois holandeses por 0; 2; 6; 8; 24 e 48 h; e 0; 8; 12; 24; 48; 72 e 96 horas, respectivamente, sendo depois retirados e sua duplicata inserida no duodeno através de uma cânula. Posteriormente, os sacos foram coletados junto com as fezes. Os valores de degradabilidade efetiva da PB para uma velocidade de passagem de 5%/hora, para o melaço em pó, grão de milho, farelo de soja, farinha de peixe, farinha de penas e feno de alfafa, foram de 100,00; 62,50; 57,90; 39,30; 34,20 e 60,90%, respectivamente; a digestibilidade intestinal de 100,00; 96,05; 99,79; 98,19; 96,07 e 94,64%, respectivamente; e a digestibilidade total de 100,00; 97,86; 99,87; 98,88; 97,35 e 98,09%, respectivamente. Verificou-se que as proteínas do melaço foram totalmente solúveis no rúmen, sendo as do milho, feno e farelo de soja bastante degradadas, além de possuírem um aproveitamento quase total no intestino. As proteínas das farinhas de peixe e de penas apresentaram baixa solubilidade ruminal e alta digestibilidade intestinal, sendo a farinha de peixe levemente mais digerida no intestino do que a farinha de penas.

To validate the use of the Cornell Net Carbohydrate and Protein System (CNCPS) under Brazilian conditions, the chemical composition, the potential and the effective degradabilities of the carbohydrate and protein subfractions, and the solid phase passage rate were determined for corn silage in diets with forage:concentrate ratios of 80:20 (DI) and 60:40 (D2) using Nellore cattle. The chromium mordant technique was used to determine the particulate passage rate of solids. For diet 2, there was a reduction in the potentially degradable dry matter (DM), and however not significant, a reduction in the degradation rates of neutral detergent Fiber (NDF, 49%) and neutral detergent insoluble nitrogen (NDIN, 32%), and an increase in the degradation rare of starch (ST, 25%). The use of lag time increased the effective DM degradation (EDDM) of corn silage in both diets (23% and 53% for D1 and D2, respectively). The concentrate ratios did not influence the particulate passage rate of the diets. The higher values related ro the availability of the protein subfractions may indicate a underestimation by CNCPS, Rnd(or) could be attributed to the Zebu animals used in this study.

Background. Ductal carcinoma in situ (DCIS) of the breast has been diagnosed increasingly since the advent of mammography. However, the natural history of these lesions remains uncertain. Ductal carcinoma in situ of the breast does not represent a single entity but a heterogeneous group with histologic and clinical differences. The histologic subtype of DCIS seems to have an influence on its biologic behavior, but there are few studies correlating subtype with biologic markers.Methods. The authors studied a consecutive series of 40 cases of DCIS and after its histologic categorization verified its relationship with ploidy using image analysis and analyzing estrogen receptor (ER), progesterone receptor (PR), p53 and c-erbB-2 expression using immunohistochemistry.Results. The three groups proposed according to the grade of malignancy were correlated significantly with some of the additional parameters studied, including aneuploidy and c-erB-2 expression. Aneuploidy was detected in 77.5% of cases of DCIS mainly in high and intermediate grade subtypes (100% and 80% vs. 35.7% in low grade) whereas immunoreactivity for c-erbB-2 was detected in 45% of cases of DCIS mainly in the high grade group. Expression of ER and PR were observed frequently in this study (63.9% and 65.7% respectively), but without correlation with the histologic subtype of DCIS, although we found a somewhat significant association between high grade DCIS and lack of ER. p53 protein expression was detected in 36.8% of these cases, but no relationship between this expression and histologic subtype or grading of DCIS was found.Conclusions. These results provide further evidence for the morphologic and biologic heterogeneity of DCIS. Besides histologic classification and nuclear grading, some biologic markers such as aneuploidy and c-erbB-2 expression constitute additional criteria of high grade of malignancy.

Polyaniline (PAni) thin films were deposited onto BK7 glass substrates using the in situ deposition technique. The control of the time and the aniline concentration in the PAni polymerization reaction on the film deposition allowed us to prepare films with different thickness, down to similar to 25 nm. The film growth process was monitored by measuring the UV-vis spectra and the AFM height profiles of the film surface. The curves of adsorption kinetics were analyzed with the Avrami's model, yielding an exponent it n = 3, thus indicating nucleation of spheroids at the initial stages of polymerization that grow through a diffusion process. AFM images of the surface height profiles corroborate this hypothesis, with spheroids growing with no preferred orientation during the in situ deposition. (C) 2007 Elsevier B.V. All rights reserved.

Objective: This in situ blind crossover study investigated the effect of calcium (Ca) rinse prior to the use fluoride (F) dentifrice on remineralisation of artificially demineralised enamel and on the composition of biofilm. Design: During four phases of 14 days, 10 volunteers wore appliances containing two artificially demineralised bovine enamel blocks. Three times a day, they rinsed with 10 mL, of Ca (150 mM) or placebo rinse (1 min). A slurry (1:3, w/v) of F (1030 ppm) or placebo dentifrice was dripped onto the blocks. During I min, the volunteers brushed their teeth with the respective dentifrice. The appliance was replaced into the mouth and the volunteers rinsed with water. The biofilm formed on the blocks was analysed for F and Ca. Enamel alterations were evaluated by the percentage of surface microhardness change (%SMHC), cross-sectional microhardness (% mineral volume) and alkali-soluble F analysis. Data were analysed by ANOVA (p < 0.05). Results: the use of the Ca pre-rinse before the F dentifrice produced a six- and four-fold increase in biofilm F and Ca concentrations, respectively. For enamel, the remineralisation was significantly improved by the Ca pre-rinse when compared to the other treatments. There was a significantly higher concentration of alkali-soluble F in enamel when the F dentifrice was used, but the Ca pre-rinse did not have any significant additive effect. Conclusions: According to our protocol, the Ca pre-rinse significantly increased biofilm F concentration and, regardless the use of F dentifrice, significantly enhanced the remineralisation of artificially demineralised enamel. The two-dimensional hybrid organic-inorganic materials Zn-2-Cr and Zn-2-Al-LDHs (Layered Double Hydroxides) containing 4-(1H-pyrrol-1yl)benzoate anions as the interlayer anions were synthesized by the co-precipitation method at constant pH followed by subsequent hydrothermal treatment for 72 h. The materials were characterized by PXRD, C-13 CP-MAS NMR, ESR, TGA, and TEM. The basal spacing found by the X-ray diffraction technique is coincident with the formation of bilayers of the intercalated anions. Solid-state C-13 NMR and ESR data strongly suggest the partial in situ polymerization of the 4-(1H-pyrrol-1yl)benzoate anions during coprecipitation. (c) 2006 Elsevier Ltd. All rights reserved.

We have pointed Out that. zinc-based particles obtained from zinc acetate sol-gel route is a mixture of quantum-sized ZnO nanoparticles, zinc acetate, and zinc hydroxide double salt (Zn-HDS). Aiming the knowledge of the mechanisms involved in the formation of ZnO and Zn-HDS phases, the thermohydrolysis of ethanolic zinc acetate solutions induced by lithium hydroxide ([LiOH]/[Zn2+] = 0.1) or water ([H2O]/[Zn2+] = 0.05) addition was investigated at different isothermal temperatures (40, 50, 60 and 70 degrees C) by in situ measurements of turbidity, UV-vis absorption spectra and extended X-ray absorption fine structures (EXAFS). Only the growth of ZnO nanoparticles was observed in sol prepared with LiOH, while a two-step process was observed in that prepared with water addition, leading the fast growth of Zn-HDS and the formation of ZnO nanoparticles at advanced stage. A mechanism of dissolution/reprecipitation governed by the water/ethanol proportion is proposed to account for relative amount of ZnO. (c) 2007 Elsevier Ltd. All rights reserved.

In order to compare the in situ degradability of tanzania grass samples obtained as by extrusa or hand plucked, three ruminal fistulated cows were used in a completely randomized block design with split-plot scheme. Five grams of extrusa or hand harvested grasses were placed in nylon bags rumen incubated during 3, 6, 12, 24, 48, 96 and 120 hours. The degradability of DM, CP, NDF and ADF were, in this sequence, 62.59, 80.88, 50.73 and 46.65%, for hand-harvested grass; and 79.53, 90.97, 71.21 and 65.68%, for extrusa. In situ degradability data of hand harvested samples were not reliable.

This double-blind crossover in situ study evaluated the effect of rinsing with water immediately after the application of acidulated phosphate fluoride (APF) on enamel demineralization. APF application was followed by: no rinsing or consuming of liquids or solids for the next 30 min; immediately washing with water jet or drinking of a glass of water. All treatments showed a statistically significant decrease in enamel mineral loss compared to the control (p < 0.05), but the differences among treatments were not significant (p > 0.05). The data suggest that the recommendation of asking patients to refrain from drinking water following professional APF application does not seem to have an influence on the anticaries effect of fluoride. The chemical composition of corn and cottonseed meal was determined in accordance with the Cornell Net Carbohydrate and Protein System (CNCPS) in diets with forage:concentrate ratios of 80:20 (D1) and 60:40(D2) using Nellore cattle. The potential and effective degradabilities of the carbohydrate and protein subfractions were also determined. For corn. diet 2 resulted in an increase for potentially degradable dry matter (DM). An increase was observed in the effective degradation of DM (EDDM) when lag time was not considered, however, the use of Eng time increased the EDDM for both diets, 23 and 18% for DI and D2. respectively. For cottonseed meal, diet 2 determined a reduction in the potentially degradable DM: therefore, the use of lag time increased the EDDM for bath diets, 14% for D1 and 15% for D2. The availability of corn starch (fraction B1) was lower than that determined by CNCPS. The higher values for the degradability of corn and cottonseed meal protein subfractions may indicate an underestimation by the model, and, or, could be attributed to the Zebu animals used in this study.
