O tempo de vida pós-colheita de frutas e hortaliças está diretamente relacionado à temperatura de armazenamento do produto. Em condições controladas de temperatura e umidade relativa do ar, as reações metabólicas podem ser retardadas, proporcionando melhor conservação do produto. Foram realizadas avaliações das câmaras frias destinadas à estocagem de frutas e hortaliças na Companhia de Entrepostos e Armazéns Gerais de São Paulo, CEAGESP - São Paulo - SP, com o objetivo de identificar a situação das câmaras frias utilizadas nesse entreposto. As condições de estocagem dos produtos foram avaliadas por meio dos parâmetros: temperatura do ar, umidade relativa, isolamento, equipamento frigorífico (condensador, compressor, evaporador), piso, dimensões da câmara e da porta. Avaliou-se a eficiência de uso por meio do cálculo para a determinação da carga térmica. Observou-se que maçã e pêra possuem os maiores volumes comercializados, utilizando-se de estocagem pelo frio (63,25%), seguido pela banana (24,10%). Baseando-se no volume médio de comercialização dos permissionários, constatou-se que 73,91% possuem motores superdimensionados a sua capacidade calorífica de uso.

The study aimed to evaluate chemical, microbiological and hydro-physical changes of a Dystrophic Yellow Latosol, receiver of different levels of manipueira (cassava wastewater) application, in the cultivation of 'Terra Maranhão' banana. The experimental design was a randomized block with three replications in a factorial scheme 3 x 4, in which it was considered three soil depths and four levels of manipueira. It was evaluated the weighted mean diameter of the aggregate, the percentage of aggregation at different periods, soil density, particle density, porosity and soil saturated hydraulic conductivity, in addition to pH of P (mg dm -3), K (mg dm-3), Ca (cmolc dm-3), Mg (cmolc dm-3), Ca+Mg (cmolc dm-3), Al (cmolc dm-3), Na (cmolc dm -3), H+Al (cmolc dm-3), CEC (cmolc dm-3), V%, OM (g kg-1), soil microbial biomass (Ug Cg-1 dry soil), acid phosphatase (Ug PNP g-1 h-1). The use of manipueira influenced some physical characteristic of the soil, but it was not possible to specify the effect of increasing application dosage. Therefore, the application did not affect the biological indicators assessed in the soil or its pH. The use of manipueira as a fertilizer in the doses used in this study showed low increase of K, P, H+Al and Al in the soil and a good increase of Mg, Ca and Ca+Mg, Na, CEC and V%.

Both healthy eyes of 10 six-year-old male and female mongrel dogs were studied. With a contact specular microscope the corneal endothelium was examined. Endothelial cells were analyzed in the central and peripheral cornea. Morphological analysis with regard to polymegathism and pleomorphism was performed. Three images of each region with at least 100 cells were obtained. The analysis showed that polygonal cells formed a mosaic-like pattern uniform in size and shape. The predominant number of cells was hexagonal. The polymegathism index was 0.22. The study demonstrates that the morphology of the normal corneal endothelial cells of dogs is similar to that found in the human cornea.

O objetivo deste experimento, realizado na cultura da banana (Musa spp.) no Vale do Ribeira, no município de Registro-SP, foi fazer um cadastramento fitossociológico das espécies de plantas daninhas sob duas formas de manejo do solo. A amostragem das espécies daninhas foi feita em duas áreas distintas de manejo, sendo uma com a cultura implantada em área de várzea drenada a 7 m de altitude e a outra com a bananicultura em área de sequeiro a 16 m de altitude. Na amostragem de um hectare, utilizou-se o método do quadrado inventário para cálculo de frequência, frequência relativa, densidade, densidade relativa, abundância, abundância relativa, índice de valor de importância e índice de importância relativa. Em ambas as áreas foram identificadas 10 famílias, distribuídas em 18 gêneros e 21 espécies. Na área de várzea drenada, 38% das famílias identificadas são monocotiledôneas e 62% dicotiledôneas, num total de 15 espécies, distribuídas em nove famílias. Na área de cultivo em sistema de sequeiro, foram identificados 50% de famílias monocotiledôneas e 50% de dicotiledôneas, num total de 11 espécies, distribuídas em seis famílias. As famílias com maior representatividade foram Poaceae, com sete espécies, seguida de Asteraceae, com três. Com o estudo realizado, verificou-se ainda grande diversidade de espécies nas áreas selecionadas.

Begomoviruses are whitefly-transmitted, single-stranded DNA viruses that are often associated with weed plants. The aim of this study was to further characterize the diversity of begomoviruses infecting weeds (mostly Sida spp.) in Brazil. Total DNA was extracted from weed samples collected in Viçosa (Minas Gerais state) and in some municipalities of Alagoas state in 2009 and 2010. Viral genomes were amplified by RCA, cloned and sequenced. A total of 26 DNA-A clones were obtained. Sequence analysis indicated the presence of 10 begomoviruses. All viral isolates from Blainvillea rhomboidea belonged to the same species, Blainvillea yellow spot virus (BlYSV ), thereby suggesting that BlYSV may be the only begomovirus present in this weed species. Four isolates represent new species, for which the following names are proposed: Sida yellow blotch virus (SiYBV), Sida yellow net virus (SiYNV), Sida mottle Alagoas virus (SiMoAV) and Sida yellow mosaic Alagoas virus (SiYMAV). Recombination events were detected among the SiYBV isolates and in the SiYNV isolate. These results constitute further evidence of the high species diversity of begomoviruses in Sida spp. However, the role of this weed species as a source of begomoviruses infecting crop plants remains to be determined.

Este trabalho teve como objetivo caracterizar a florística, estrutura e fisionomia de três florestas em distintas situações fisiográficas da planície litorânea. Foram amostradas uma floresta sobre turfeira pouco profunda (floresta turfosa rasa), uma floresta sobre turfeira profunda (floresta turfosa profunda) e uma floresta em solo firme e seco (floresta sobre morrote), todas situadas na área de floresta contínua do Parque Estadual da Campina do Encantado, Pariquera-Açu SP. Foi realizado o levantamento fitossociológico através de blocos de parcelas contíguas de 10 × 10 m -- totalizando 1,04 ha -- e amostradas todas as árvores com DAP > ou = 4,8 cm. Foram encontradas ao todo 144 espécies, sendo 112 na floresta sobre morrote, 46 na floresta turfosa rasa e cinco na floresta turfosa profunda. A similaridade entre as três florestas foi baixa, com poucas espécies em comum, principalmente em relação às florestas turfosas e a floresta sobre morrote. A floresta sobre morrote apresentou alta diversidade, típica de ambientes florestais tropicais (H' = 4,06 nat.ind.-1). A floresta turfosa rasa apresentou o valor de diversidade dentro do esperado para formações alagáveis das planícies litorâneas (H' = 2,98 nat.ind.-1). A diversidade na floresta turfosa profunda foi muito baixa (H' = 0,82 nat.ind.-1), sendo a menor já registrada para florestas sem perturbação antrópica das regiões sul e sudeste brasileira. A heterogeneidade vegetacional em áreas de floresta contínua, condicionada aos fatores fisiográficos das planícies litorâneas, indica a necessidade de se proteger o maior número de situações ambientais possíveis, objetivando a conservação da biodiversidade in situ.

Espinhaço and Mantiqueira are two mountain ranges of great importance in Brazil. In the uppermost parts of these areas, unique ecosystems occur, generally associated with rock outcrops, they are collectively called High Altitude Rocky Complexes. These environments show distinct soil and biota characteristics in relation to the surrounding biome. The soils are generally shallow, coarse textured, with high Al3+ and varying amounts of organic matter. Entisols, Inceptsols and Histosols are dominant, directly associated with the rock outcrops, and forming a complex mosaic of soils. Some of these soils are endemic, based on peculiar conditions of parent materials, topography and vegetation, and this pedodiversity is important for detecting unique and endangered soils. In these soils, organic matter is highly humified, with a great amount of soluble forms and conspicuous presence of charcoal. Spodic horizons and dark water rivers are typically associated with quartzite and quartzite outcrops, formed by illuviation of organic compounds, being less common in granitic rocks. The very low nutrient content of these soils and other environmental limitations required the development of specific physiological and morphological plant adaptations. Most high altitude environments are unstable under current climatic conditions, and anthropic interventions may be accelerating this process. Detailed soil surveys are necessary for a better understanding of the role of these soils in ecological processes and for the development of adequate conservation policies.

The Guayana Shield, located in north-eastern South America, consists of a highly complex and composite mosaic of landscape elements. Amongst these, inselbergs are very conspicuous, because of their peculiar shape and their unusual associated vegetation. Geologically, these rock outcrops are part of the underlying ancient igneous-metamorphic basement and occur mainly in the lowlands of the periphery of the shield. As azonal habitats, inselbergs harbour a highly specialized flora. The characteristic vegetation is composed of lithophytic and savanna-like plant communities, as well as low dry forests. As a whole, the vegetation of an inselberg may be interpreted as a mosaic of marginal habitats. Therefore a large number of taxa find suitable niches in a quite condensed space. Gradients of soil depth and water availability are the main factors determining the floristic composition. A preliminary floristic inventory of the Venezuelan inselberg flora comprises 614 vascular plant species. 24% of them are endemic to the Guayana region, 15% are endemic to inselbergs of the Guayana region. The distribution patterns of these latter, eco-endemic species allows to distinguish a northern and a southern inselberg district. The two districts overlap in the Átures area, in the surroundings of Puerto Ayacucho, where a true centre of endemism is located. The distinction into two districts is emphasised by different phytogeographic relations. The southern inselberg district shows connections to the "tepui" flora, whereas the northern district reveals phytogeographic relations to the Caribbean region as well as to the Brazilian Shield. Possible explanations for the floristic interchange across the equator are discussed.

Granitic rock outcrops of the Brazilian southeast are either coastal or inland. The latter can often have high altitudes, such as in the summits of "Serra do Mar" and "Serra da Mantiqueira", where they are known as "Campos de Altitude". The landscape on these high altitude plateaux is often a mosaic of shrubs and treelets within a bunchgrass matrix, with sparse pteridophytes and other herbs, interspersed with variable extensions of rock outcrops. Despite the pervasiveness of rock outcrops in the Brazilian landscape, studies on the structural analysis of the vegetation on such formations are scarce. This study aimed to analyze the structure of the vegetation on a highland granitic rock outcrop in "Parque Estadual da Serra do Brigadeiro", state of Minas Gerais, Southeast Brazil (42°20' - 42°40' S and 20°20' - 21°00' W, 1,722 m of elevation ). Quantitative parameters of absolute and relative frequency and dominance (cover) were estimated. The group analysis used the Jaccard similarity index. Trilepis lhotzkiana, Panicum sp. 1, and Vellozia variegata presented the highest relative frequencies, relative dominances and importance values. These three species, along with Dyckia bracteata, Rhynchospora emaciate, and Tibouchina cf. manicata, represented 98.3% of the relative dominance. The remaining 1.7% referred to 22 remaining species. The distinction among quadrats within formed groups by cluster analysis was due to the great number of low frequency species.

The paper proposed Cyanoaggregatum brasiliense, a new genus and species from the plankton of a subtropical brackish coastal lagoon from Rio Grande do Sul State, South Brazil. It differs from all other members of Chroococcales by its characteristic arrangement of cells in irregular groups distributed in a single, flat or slightly curved layer, on irregular rows, slightly distant from each other, forming a mosaic-like pattern slightly below the surface of the mucilaginous colonial envelope. The cell division in one plane perpendicular to the long axis and the lack of pseudo-filaments indicate its classification in the family Synechococcaceae, sub-family Aphanothecoideae. The general characteristics, the diagnostic criteria and the taxonomic position are discussed, and a comparison between Cyanoaggregatum and its most closely related genera is presented. Physical and chemical data on the studied lagoon and geographical distribution are presented.

In coastal waters, physico-chemical and biological properties and constituents vary at different time scales. In the study area of this thesis, within the Archipelago Sea in the northern Baltic Sea, seasonal cycles of light and temperature set preconditions for intra-annual variations, but developments at other temporal scales occur as well. Weather-induced runoffs and currents may alter water properties over the short term, and the consequences over time of eutrophication and global changes are to a degree unpredictable. The dynamic characteristics of northern Baltic Sea waters are further diversified at the archipelago coasts. Water properties may differ in adjacent basins, which are separated by island and underwater thresholds limiting water exchange, making the area not only a mosaic of islands but also one of water masses. Long-term monitoring and in situ observations provide an essential data reserve for coastal management and research. Since the seasonal amplitudes of water properties are so high, inter-annual comparisons of water-quality variables have to be based on observations sampled at the same time each year. In this thesis I compare areas by their temporal characteristics, using both inter-annual and seasonal data. After comparing spatial differences in seasonal cycles, I conclude that spatial comparisons and temporal generalizations have to be made with caution. In classifying areas by the state of their waters, the results may be biased even if the sampling is annually simultaneous, since the dynamics of water properties may vary according to the area. The most comprehensive view of the spatiotemporal dynamics of water properties would be achieved by means of comparisons with data consisting of multiple annual samples. For practical reasons, this cannot be achieved with conventional in situ sampling. A holistic understanding of the spatiotemporal features of the water properties of the Archipelago Sea will have to be based on the application of multiple methods, complementing each other’s spatial and temporal coverage. The integration of multi-source observational data and time-series analysis may be methodologically challenging, but it will yield new information as to the spatiotemporal regime of the Archipelago Sea.

We review studies from our laboratories using different molecular tools to characterize the ancestry of Brazilians in reference to their Amerindian, European and African roots. Initially we used uniparental DNA markers to investigate the contribution of distinct Y chromosome and mitochondrial DNA lineages to present-day populations. High levels of genetic admixture and strong directional mating between European males and Amerindian and African females were unraveled. We next analyzed different types of biparental autosomal polymorphisms. Especially useful was a set of 40 insertion-deletion polymorphisms (indels) that when studied worldwide proved exquisitely sensitive in discriminating between Amerindians, Europeans and Sub-Saharan Africans. When applied to the study of Brazilians these markers confirmed extensive genomic admixture, but also demonstrated a strong imprint of the massive European immigration wave in the 19th and 20th centuries. The high individual ancestral variability observed suggests that each Brazilian has a singular proportion of Amerindian, European and African ancestries in his mosaic genome. In Brazil, one cannot predict the color of persons from their genomic ancestry nor the opposite. Brazilians should be assessed on a personal basis, as 190 million human beings, and not as members of color groups.

Mutations in Bruton's tyrosine kinase (BTK) gene are responsible for X-linked agammaglobulinemia (XLA), which is characterized by recurrent bacterial infections, profound hypogammaglobulinemia, and decreased numbers of mature B cells in peripheral blood. We evaluated 5 male Brazilian patients, ranging from 3 to 10 years of age, from unrelated families, whose diagnosis was based on recurrent infections, markedly reduced levels of IgM, IgG and IgA, and circulating B cell numbers <2%. BTK gene analysis was carried out using PCR-SSCP followed by sequencing. We detected three novel (Ala347fsX55, I355T, and Thr324fsX24) and two previously reported mutations (Q196X and E441X). Flow cytometry revealed a reduced expression of BTK protein in patients and a mosaic pattern of BTK expression was obtained from mothers, indicating that they were XLA carriers.

A hidrólise do amido e a síntese de açúcares durante o amadurecimento da banana são transformações bioquímicas importantes, havendo evidências de que ocorrem de forma homogênea no fruto. Para confirmar este fato, amostras de banana nanicão (Musa spp.) colhidas aos 110 dias pós-antese, foram coletadas no decorrer do amadurecimento e foram determinados os teores de amido, hexoses e sacarose e a atividade das enzimas sacarose-fosfato sintase (SPS) e sacarose sintase (SS) em diferentes partes do fruto. Observou-se que na banana verde, existe mais amido na porção periférica (18%) do que na central (13%). Porém, a sua velocidade de degradação durante o amadurecimento é a mesma, o que resulta em teores diferenciados de amido residual na banana madura. Também o aparecimento e acúmulo de sacarose foi simultâneo nas duas regiões e coincidente com os valores máximos de atividade da SPS. Utilizando-se de técnica de identificação por anticorpos específicos para SS e SPS em tecidos verde e maduro, observou-se uma distribuição homogênea das enzimas e aparente correlação entre a cor desenvolvida e a variação de atividade.
