Our objective was to evaluate the concentrations of serum 25-hydroxyvitamin D [25(OH)D], serum calcium, serum phosphorus, alkaline phosphatase, and parathormone (PTH) in patients with polyarticular juvenile idiopathic arthritis (JIA) and to associate them with disease duration and activity, bone mineral density and use of medications. In a cross-sectional and controlled study, 30 patients with polyarticular JIA were evaluated and compared to 30 healthy individuals matched for age and gender. Clinical status, anthropometry, laboratory markers in both patients and controls, and bone mineral density, only in the patients, were measured. Of the 30 patients included in the study, 23 (76.7%) were female and 16 (53.3%) non-Caucasian; mean age was 14 years (range = 4 to 20 years). Mean disease duration was 5 years (range = 1 to 12 years). The mean concentrations of serum albumin-corrected calcium (9.04 ± 0.41 mg/dL) and alkaline phosphatase (153.3 ± 100.1 IU) were significantly lower in patients with JIA than in controls (P < 0.0001 and P = 0.001, respectively). No differences in 25(OH)D, PTH or serum phosphorus were observed between JIA and control subjects. Regarding 25(OH)D concentration, 8 patients (26.7%) and 5 controls (16.7%) had 25(OH)D concentrations compatible with deficiency (lower than 20 ng/mL) and 14 patients (46.7%) and 18 controls (60%) had concentrations compatible with insufficiency (20-32 ng/mL). These values were not associated with disease activity, use of medications or bone mineral density. We observed a high frequency of 25(OH)D insufficiency and deficiency in the study sample. The compromised bone metabolism emphasizes the importance of follow-up of JIA patients.

The purpose of this study was to investigate the effect of supplementary vitamin D therapy in addition to amitriptyline on the frequency of migraine attacks in pediatric migraine patients. Fifty-three children 8-16 years of age and diagnosed with migraine following the International Headache Society 2005 definition, which includes childhood criteria, were enrolled. Patients were classified into four groups on the basis of their 25-hydroxyvitamin D [25(OH)D] levels. Group 1 had normal 25(OH)D levels and received amitriptyline therapy alone; group 2 had normal 25(OH)D levels and received vitamin D supplementation (400 IU/day) plus amitriptyline; group 3 had mildly deficient 25(OH)D levels and received amitriptyline plus vitamin D (800 IU/day); and group 4 had severely deficient 25(OH)D levels and was given amitriptyline plus vitamin D (5000 IU/day). All groups were monitored for 6 months, and the number of migraine attacks before and during treatment was determined. Calcium, phosphorus alkaline phosphatase, parathormone, and 25(OH)D levels were also determined before and during treatment. Results were compared between the groups. Data obtained from the groups were analyzed using one-way analysis of variance. The number of pretreatment attacks in groups 1 to 4 was 7±0.12, 6.8±0.2, 7.3±0.4, and 7.2±0.3 for 6 months, respectively (all P>0.05). The number of attacks during treatment was 3±0.25, 1.76±0.37 (P<0.05), 2.14±0.29 (P<0.05), and 1.15±0.15 (P<0.05), respectively. No statistically significant differences in calcium, phosphorus, alkaline phosphatase, or parathormone levels were observed (P>0.05). Vitamin D given in addition to anti-migraine treatment reduced the number of migraine attacks.

The objective of this study was to evaluate the effect of tamoxifen on the plasma concentration of NT-pro-B-type natriuretic peptide (NT-proBNP) in women undergoing chemotherapy for breast cancer and to correlate changes in NT-proBNP with the left ventricular ejection fraction (LVEF). Over a period of 12 months, we followed 60 women with a diagnosis of breast cancer. The patients were separated into a group that received only chemotherapy (n=23), a group that received chemotherapy + tamoxifen (n=21), and a group that received only tamoxifen (n=16). Plasma levels of NT-proBNP were assessed at 0 (T0), 6 (T6), and 12 (T12) months of treatment, and echocardiography data were assessed at T0 and T12. Plasma NT-proBNP levels were increased in the chemotherapy-only group at T6 and T12, whereas elevated NT-proBNP levels were only found at T6 in the chemotherapy + tamoxifen group. At T12, the chemotherapy + tamoxifen group exhibited a significant reduction in the peptide to levels similar to the group that received tamoxifen alone. The chemotherapy-only group exhibited a significant decrease in LVEF at T12, whereas the chemotherapy + tamoxifen and tamoxifen-only groups maintained levels similar to those at the beginning of treatment. Treatment with tamoxifen for 6 months after chemotherapy significantly reduced the plasma levels of NT-proBNP and did not change LVEF in women with breast cancer.

In DNA vaccines, the gene of interest is cloned into a bacterial plasmid that is engineered to induce protein production for long periods in eukaryotic cells. Previous research has shown that the intramuscular immunization of BALB/c mice with a naked plasmid DNA fragment encoding the Mycobacterium leprae 65-kDa heat-shock protein (pcDNA3-Hsp65) induces protection against M. tuberculosis challenge. A key stage in the protective immune response after immunization is the generation of memory T cells. Previously, we have shown that B cells capture plasmid DNA-Hsp65 and thereby modulate the formation of CD8+ memory T cells after M. tuberculosis challenge in mice. Therefore, clarifying how B cells act as part of the protective immune response after DNA immunization is important for the development of more-effective vaccines. The aim of this study was to investigate the mechanisms by which B cells modulate memory T cells after DNA-Hsp65 immunization. C57BL/6 and BKO mice were injected three times, at 15-day intervals, with 100 µg naked pcDNA-Hsp65 per mouse. Thirty days after immunization, the percentages of effector memory T (TEM) cells (CD4+ and CD8+/CD44high/CD62Llow) and memory CD8+ T cells (CD8+/CD44high/CD62Llow/CD127+) were measured with flow cytometry. Interferon γ, interleukin 12 (IL-12), and IL-10 mRNAs were also quantified in whole spleen cells and purified B cells (CD43−) with real-time qPCR. Our data suggest that a B-cell subpopulation expressing IL-10 downregulated proinflammatory cytokine expression in the spleen, increasing the survival of CD4+ TEM cells and CD8+ TEM/CD127+ cells.

O coração da palmeira Bactris gasipaes H.B.K. (pupunha) foi utilizado, sob forma desidratada, na formulação de uma sopa-creme para merenda escolar. Análises químicas do palmito e do coração da palmeira (açúcares redutores, lipídeos, proteínas totais, aminograma, fibras, calorias e minerais) foram feitas tanto na forma in natura como na forma processada, bem como na de sopa-creme. Não foram encontradas diferenças relevantes em relação a essas determinações, podendo-se ressaltar a boa qualidade aminoacídica do coração da palmeira.

Neste trabalho, foram determinados os lipídios totais e a composição de ácidos graxos da cabeça in natura de três espécies de peixes do gênero Brycon, matrinxã (B. cephalus), piraputanga (B. microlepis) e piracanjuba (B. orbignyanus), criados em cativeiros (açudes e gaiolas) e, nativos coletados nos rios Cuiabá-Manso (piraputanga) e no rio Paraná (piracanjuba). Os teores de lipídios totais nas cabeças variaram de 14,26 a 22,00%. Com relação aos ácidos graxos, todas as espécies apresentaram como predominantes os ácidos oléico-C18:1ômega9 (40,21 a 44,41%), seguido dos ácidos palmítico-C16:0 (22,04 a 27,04%), esteárico-C18:0 (7,78 a 12,11%) e linoléico-18:2ômega6 (5,27 a 14,68%). O percentual dos ácidos alfa-linolênico (18:3ômega3), araquidônico (20:4ômega6), eicosapentaenóico (20:5ômega3) e docosahexaenóico (22:6ômega3) foram inferiores a 2,52%. A somatória dos ácidos graxos polinsaturados (AGPI) variou de 10,30 a 19,32%, e dos ácidos graxos saturados (AGS) de 32,42 a 39,41%. A espécie piraputanga nativa apresentou maior proporção de ácidos ômega3 (3,67%) e menor de ácidos ômega6 (6,02%).

The aim of this study was to introduce the tangential microfiltration (TMF) technique on the production of orange juice (TMFJ), and compare it with pasteurised juice (control) as regards chemical composition and sensorial characteristics. We used a TMF pilot equipped with four monotubular ceramic membranes (0.1, 0.2, 0.8 and 1.4mm) arranged in series with a filtering area of 0.005 m² each. Commercial flash-pasteurised orange juice was used as the initial product. Experiments were divided into three parts: a) the characterisation of the TMF pilot; b) optimisation of operational conditions; c) production of the TMFJ. In the second part, membrane with 0.8-mm pores presented best flux followed by those with 1.4-, 0.1-, and 0.2-mm pores. However, to guarantee permeate sterility, we chose the membrane with 0.1-mm pores for TMFJ production. Initially, the orange juice was sieved in order to separate part of the pulp, being subsequently submitted to TMF. A mixture of retentate and pulp was made, and was subsequently pasteurised. We obtained the TMFJ by adding the permeate to the mixture. TMFJ presented soluble solids content (°Brix), pulp, pH, and titrable acidity similar to the initial pasteurised juice (control). Nevertheless, 28% of vitamin C was lost during the TMFJ production. According to the juice taster panel, the control juice presented best sensorial characteristics (greater aroma intensity and fruity flavour) when compared with the TMJF.

Amostras de hortaliças dos tipos A e B foram analisadas quanto ao teor total de cobre através de espectroscopia de absorção atômica em chama, e o método de extração seqüencial foi aplicado com os seguintes extratores: solução de CaCl(2)1,0M; solução de ácido acético 0,1M; solução de ácido acético 0,5M / acetato de amônio 5% [pH=5,0]; solução de NaOH 0,1M e solução de HCl 0,5M. Na análise da rejeição de resultados foi aplicado o teste de Grubbs e na comparação dos dados o teste t de Student foi utilizado. Em média 25,4% do teor total de cobre foram extraídos com solução de CaCl2 1,0M, sendo o máximo de 50,5% e o mínimo de 8,3%. Em média 14,8% do teor total de cobre, sendo que 32,6% e 6,4%, máximo e mínimo respectivamente, foram extraídos com solução de ácido acético 0,1M. Com relação ao uso de solução de ácido acético 0,5M / acetato de amônio 5% [pH=5,0], o mínimo extraído obteve valor de 6,2% e o máximo 27,6%, com média de 13,7%. O extrator de NaOH 0,1M foi o que teve menor extração, cerca de 10,5% e com extrator de HCl 0,5M a extração foi em média de 12,6%. A fração extraída de cobre nas amostras foi de no mínimo 34,5% e no máximo 100%. Observa-se que para a maioria das amostras, o cobre se encontra sob a forma de, no mínimo, 6 espécies químicas distintas.

Objetivou-se avaliar as alterações de b-caroteno, ácido ascórbico e antocianinas totais na polpa de frutos de clones de aceroleira conservada por congelamento. Os frutos dos clones BRS 152 (Sertaneja) BRS 235 (Apodi), BRS 236 (Cereja), BRS 237 (Roxinha), BRS 238 (Frutacor) e II 47/1 foram colhidos no estádio de maturação comercial (vermelho maduro) em Limoeiro do Norte, Ceará, Brasil, transportados para a Planta Piloto de Processamento de Frutos da Embrapa Agroindústria Tropical, despolpados, acondicionada a polpa em sacos de polietileno (100 g), congelada, mantida em freezer a -20 °C, e avaliada a cada 30 dias durante 12 meses. O experimento foi realizado em delineamento experimental inteiramente casualizado em esquema fatorial 6 x 13 (clones x tempo), com 3 repetições. A concentração de beta-caroteno foi estável no clone Cereja, enquanto, nos demais, houve decréscimo durante todo o período do experimento. Houve pequeno decréscimo no teor de ácido ascórbico em todos os clones estudados durante o armazenamento, provavelmente devido à alta acidez da polpa, que auxilia na manutenção deste nutriente. O teor de antocianinas totais foi estável nos clones Frutacor e Sertaneja, enquanto nos demais houve diminuição. O clone II 47/1 foi dentre os estudados o que apresentou maiores teores de ácido ascórbico e antocianinas totais, mantendo estas características durante todo o armazenamento. De um modo geral, os clones em que se determinou menor teor de beta-caroteno foram observadas as mais elevadas concentrações de antocianinas totais.

O camu-camu é um fruto com notável potencial nutricional, pelo seu alto conteúdo em ácido ascórbico. Sendo assim, objetivou-se avaliar a estabilidade do ácido ascórbico e pigmentos presentes no néctar de camu-camu, armazenados sob diferentes condições de luminosidade e temperatura. Os frutos foram despolpados e avaliados quanto às características físico-químicas. O néctar, obtido a partir da polpa, foi acondicionado em garrafas PET e armazenado em temperatura ambiente e sob refrigeração na presença e ausência de luz, sendo avaliado por 120 dias quanto à estabilidade de ácido ascórbico e antocianinas. O teor de ácido ascórbico nos néctares armazenados sob luz não diferiu estatisticamente dos armazenados protegidos da luz (343,25 e 340,48 mg.100 g -1), respectivamente, nos armazenados sob refrigeração, e (330,48 e 333,56 mg.100 g -1) nos armazenados em temperatura ambiente. Constatou-se que esta vitamina em néctares armazenados por 120 dias em temperatura de refrigeração apresentou boa estabilidade, com perda de apenas 12 a 14%. Quanto às antocianinas, a temperatura ambiente contribuiu negativamente, ocasionando uma degradação mais acelerada, no entanto, a exposição à luz não teve efeito. Nestas condições experimentais, conclui-se que o fator luminosidade tem pouca influência sobre o ácido ascórbico e antocianinas no néctar de camu-camu, e que a temperatura ambiente de armazenamento é fator negativo na estabilidade destes pigmentos.

This study is integrated within a research line dedicated to evaluate different options to introduce mandarin in Spanish agro-food industries and in both national and international markets. The effects of different preservation liquids (sucrose solution, sucrose and ascorbic acid solution, grape, and grapefruit juices), soluble solids (10, 12, 14 ºBrix), and thermal processing (30 seconds at 98 ºC) on vitamin C, color, carotenoids and sensory quality of canned Satsuma mandarin segments were studied. The liquid preservation that presented the best results was grapefruit juice, which provided the highest vitamin C (323 mg.L-1) and total carotenoids (16.4 mg.L-1) contents (the main carotenoid found was β-cryptoxanthin), intense orange color, high thermal stability, and very good sensory quality (e.g. intense floral and fresh-mandarin aromas and low levels of off-flavors).

The objective of this work was to verify the availability of iron in the presence of vitamin A as components of foods and in combinations with medicines. The iron available was measured in the presence of vitamin A in foods - common bean (B), beef liver (Li) and carrot (C) - and medicines - Fer-In-Sol® (Fer) (Mead Johnson), Arovit® (A) (Roche) and Neutrofer® (N) (Sigma Pharma) - as well as in combinations of both. β-carotene, vitamin A, total iron, heme and non heme iron, percentage of dialyzable iron and amount of dialyzable iron was determined. Vitamin A and β-carotene had a positive effect on the percentage of iron dialysis. Carrot and liver had a better percentage of dialyzable iron than their respective medicine at similar concentrations. Therefore, we can conclude that there has been an influence of vitamin A over the dialysis of iron, being the mixtures containing liver the ones which achieved the highest concentrations of dialyzable iron, and also that, according to the amounts needed to obtain the daily recommended intake of iron, they are good for consumption.

The physiological state of a fruit is closely related to ripening and climatic conditions during the growing period when the fruit undergo changes in color, texture, and flavor. The ripening of the fruit can involve a complex series of biochemical reactions with alteration in enzymes activities, phenols, tannins, and ascorbic acid. The activity of enzymes (carboximethylcellulase, polygalacturonase, and pectinlyase), the total concentration of phenolic compounds, condensed tannins, and vitamin C in five stages of maturation were studied. Significant changes were observed between the maturity stages. The phenolic compounds were higher at green stage (705.01 ± 7.41); tannins were higher at green/purple stage (699.45 ± 0.22). The results showed that the ascorbic acid levels of the pulp varied significantly from 50.81 ± 1.43 to 6.61 ± 1.04 mg.100 g-1 during maturation. The specific activity of pectin lyase was higher at green stage (1531.90 ± 5.83). The specific activity of polygalacturonase was higher at mature stage (1.83 ± 0.0018). The specific activity of carboximetilcelulose was higher at ripe mature stage (4.61 ± 0.0024). The low ascorbic acid content found in jambolan fruit indicates that this fruit is not a rich source of this nutrient; however, other characteristics can make jambolan products fit for human consumption.

This paper aims to evaluate the physicochemical characteristics, sensory acceptance, and purchase intent of acerola nectar sweetened with sucrose and other sweeteners (neotame, sucralose and stevia extracts with 40%, 60%, 80%, and 95% rebaudioside A). The analyses were carried out for pH, soluble solids, total titratable acidity, ascorbic acid, and colorimetry (L*a*b). The acceptance test was performed by 120 consumers who evaluated the appearance, aroma, flavor, texture, and overall impression of the samples using a 9-cm unstructured hedonic scale. Furthermore, the consumers were asked to rate overall purchase intent along the scale anchored with (1) "would definitely not purchase" to (5) "would definitely purchase." The results were evaluated using analysis of variance/Tukey test and the internal preference mapping technique. The acerola nectar samples did not differ significantly (p>0.05) between themselves in terms of vitamin C content and total titratable acidity. As for appearance and aroma, there was no significant difference (p>0.05) between the samples, and as for flavor and overall impression, the most accepted samples were those with sucrose and sucralose. The internal preference mapping indicated that the most accepted samples were those with sucrose, sucralose, and neotame were. The highest frequency of positive purchase intent scores was observed for sucrose and sucralose.
