This paper examines theoretical and methodological implications of Clifford Geertz's approach to religion as he formulated it in 'Religion as a cultural system' (Geertz 1966), where religion and culture seem to be defined as functional equivalents. The paper considers religious symbols in the public space, using two examples from contemporary reality - one being a certain expression spoken by the copilot of Egypt Air Flight 990, the other being the headscarf controversy in France - in order to explore how the anthropologist relates the microsituations he observes to an all-embracing context

RESUME L'architecture nucléaire ainsi que l'ultrastructure des microtubules ont été abondamment étudiées par des méthodes cytochimiques utilisant des échantillons fixés chimiquement, enrobés dans des résines ou fixés à basse température. Les échantillons fixés à basse température pouvant aussi avoir été substitués, déshydratés et enrobés dans des résines pour la plupart hydrophiles. Ici, nous avons étendu ces études en utilisant la microscopie électronique effectuée sur des sections hydratées (CEMOVIS) permettant d'observer les échantillons dans un état le plus proche de leur état natif. De plus, nous avons effectué de la tomographie électronique sur des sections hydratées (TOVIS) afin d'obtenir une vision tridimensionnelle de : 1) la périphérie du noyau et de la région périchromatinienne et 2) de la lumière des microtubules. Concernant l'architecture nucléaire Nos observations montrent que le nucléole et la chromatine condensée sont facilement visualisés grâce à la texture spécifique qu'ils arborent. Au contraire, la visualisation de domaines nucléaires importants et spécialement ceux qui contiennent des ribonucléoprotéines, est rendue difficile, à cause du faible contraste qui caractérise l'espace interchromatinien. Ceci est essentiellement dû à la quantité d'information présente dans le volume de la section qui semble être superposée, lorsque observée sur des micrographies en deux dimensions. La tomographie nous a permis de mieux visualiser les différentes régions du noyau. Les mottes de chromatine condensée sont décorées à leur périphérie (région périchromatinienne), par nombre de fibrilles et granules. Des tunnels d'espace interchromatinien sont occasionnellement observés en train de traverser des régions de chromatine condensée favorisant l'accès aux pores nucléaires. Enfin, nous avons pu, au niveau d'un pore unique, observer la plupart des structures caractéristiques du complexe de pore nucléaire. Concernant l'ultrastructure des microtubules: Nous avons démontré que la polarité d'un microtubule observé in situ en section transversale, par CEMOVIS, est directement déduite de l'observation de la chiralité de ses protofilaments. Cette chiralité, a été établie précédemment comme étant liée à la morphologie des sous unités de tubuline. La tomographie électronique effectuée sur des sections hydratées, nous a permis d'observer les microtubules dans leur contexte cellulaire avec une résolution suffisante pour visualiser des détails moléculaires, comme les monomères de tubuline. Ainsi, des molécules n'ayant pas encore été caractérisées, ont été observées dans la lumière des microtubules. Ces observations ont été effectuées autant sur des cellules observées en coupe par CEMOVIS que sur des cellules congelées dans leur totalité par immersion dans un bain d'éthane liquide. Enfin, nous avons montré que les microtubules étaient aussi de formidables objets, permettant une meilleure compréhension des artéfacts de coupe occasionnés lors de la préparation des échantillons par CEMOVIS. Les buts des études qui seront menées â la suite de ce travail seront de 1) essayer de localiser des domaines nucléaires spécifiques par des approches cytochimiques avant la congélation des cellules. 2) Appliquer des méthodes de moyennage afin d'obtenir un modèle tridimensionnel de la structure du complexe de pore nucléaire dans son contexte cellulaire. 3) Utiliser des approches biochimiques afin de déterminer la nature exacte des particules qui se trouvent dans la lumière des microtubules. ABSTRACT Nuclear architecture as well as microtubule ultrastructure have been extensively investigated by means of different methods of ultrastructural cytochemistry using chemically fixed and resin embedded samples or following cryofixation, cryosubstitution and embedding into various, especially partially hydrophilic resins. Here, we extend these studies using cryoelectron microscopy of vitreous sections (CEMOVIS) which allows one to observe the specimen as close as possible to its native state. Furthermore, we applied cryoelectron tomography of vitreous sections (TOVIS) in order to obtain athree-dimensional view of: 1) the nuclear periphery, and of the perichromatin region, and 2) the microtubule lumen. Concerning the nuclear architecture: Our observations show that nucleoli and condensed chromatin are well recognisable due to their specific texture. Conversely, the visualisation of other important nuclear domains, especially those containing ribonucleoproteins, is seriously hampered by a generally low contrast of the interchromatin region. This is mainly due to the plethora of information superposed in the volume of the section observed on two-dimensional micrographs. Cryoelectron tomography allowed us to better visualise nuclear regions. Condensed chromatin clumps are decorated on their periphery, the perichromatin region, by numerous fibrils and granules. Tunnels of interchromatin space can occasionally be found as crossing condensed chromatin regions, thus, allowing the access to nuclear pores. Finally, we were able to use TOVIS to directly distinguish most of the nuclear pore complex structures, at the level of a single pore. Concerning the microtubule ultrastructure: We have demonstrated that the polarity of across-sectioned microtubule observed in situ by CEMOVIS wás directly deducible from the visualisation of the tubulin protofiíaments' chirality. This chirality has been established before as related to the shape. of the tubulin subunits. Cryoelectron tomography allowed us to observe microtubules in their cellular context at a resolution sufficient to resolve molecular details such as their tubulin monomers. In this way, uncharacterized molecules were visualised in the microtubule lumen. These observations were made either on samples prepared by CEMOVIS or plunge freezing of whole cells. Finally, we have shown that microtubules are also relevant objects for the understanding of cutting artefacts, when performing CEMOVIS. The goals of our further studies will be to: 1) try to speciifically target different nuclear domains by cytochemical approaches in situ, prior to cryofixation. 2) Apply averaging methods in order to obtain a three-dimensional model of the nuclear pore complex at work, in its cellular context. 3) Use biochemical analysis combined in a second time to immunocytochemical approaches, to determine the exact nature of the microtubule's luminal particles.

We identify a number of meanings of "Open", as part of the motivating rationale for a social media space tuned for learning, called SocialLearn. We discuss why online social learning seems to be emerging so strongly at this point, explore features of social learning, and identify some of the dimensions that we believe characterize the social learning design space, before describing the emerging design concept and implementation.

The geometry and connectivity of fractures exert a strong influence on the flow and transport properties of fracture networks. We present a novel approach to stochastically generate three-dimensional discrete networks of connected fractures that are conditioned to hydrological and geophysical data. A hierarchical rejection sampling algorithm is used to draw realizations from the posterior probability density function at different conditioning levels. The method is applied to a well-studied granitic formation using data acquired within two boreholes located 6 m apart. The prior models include 27 fractures with their geometry (position and orientation) bounded by information derived from single-hole ground-penetrating radar (GPR) data acquired during saline tracer tests and optical televiewer logs. Eleven cross-hole hydraulic connections between fractures in neighboring boreholes and the order in which the tracer arrives at different fractures are used for conditioning. Furthermore, the networks are conditioned to the observed relative hydraulic importance of the different hydraulic connections by numerically simulating the flow response. Among the conditioning data considered, constraints on the relative flow contributions were the most effective in determining the variability among the network realizations. Nevertheless, we find that the posterior model space is strongly determined by the imposed prior bounds. Strong prior bounds were derived from GPR measurements and helped to make the approach computationally feasible. We analyze a set of 230 posterior realizations that reproduce all data given their uncertainties assuming the same uniform transmissivity in all fractures. The posterior models provide valuable statistics on length scales and density of connected fractures, as well as their connectivity. In an additional analysis, effective transmissivity estimates of the posterior realizations indicate a strong influence of the DFN structure, in that it induces large variations of equivalent transmissivities between realizations. The transmissivity estimates agree well with previous estimates at the site based on pumping, flowmeter and temperature data.

Background. We describe the diversity of two kinds of mycobacteria isolates, environmental mycobacteria and Mycobacterium bovis collected from wild boar, fallow deer, red deer and cattle in Doñana National Park (DNP, Spain), analyzing their association with temporal, spatial and environmental factors. Results. High diversity of environmental mycobacteria species and M. bovis typing patterns (TPs) were found. When assessing the factors underlying the presence of the most common types of both environmental mycobacteria and M. bovis TPs in DNP, we evidenced (i) host species differences in the occurrence, (ii) spatial structuration and (iii) differences in the degree of spatial association of specific types between host species. Co-infection of a single host by two M. bovis TPs occurred in all three wild ungulate species. In wild boar and red deer, isolation of one group of mycobacteria occurred more frequently in individuals not infected by the other group. While only three TPs were detected in wildlife between 1998 and 2003, up to 8 different ones were found during 2006-2007. The opposite was observed in cattle. Belonging to an M. bovis-infected social group was a significant risk factor for mycobacterial infection in red deer and wild boar, but not for fallow deer. M. bovis TPs were usually found closer to water marshland than MOTT. Conclusions. The diversity of mycobacteria described herein is indicative of multiple introduction events and a complex multi-host and multi-pathogen epidemiology in DNP. Significant changes in the mycobacterial isolate community may have taken place, even in a short time period (1998 to 2007). Aspects of host social organization should be taken into account in wildlife epidemiology. Wildlife in DNP is frequently exposed to different species of non-tuberculous, environmental mycobacteria, which could interact with the immune response to pathogenic mycobacteria, although the effects are unknown. This research highlights the suitability of molecular typing for surveys at small spatial and temporal scales.

We present herein a topological invariant of oriented alternating knots and links that predicts the three-dimensional (3D) writhe of the ideal geometrical configuration of the considered knot/link. The fact that we can correlate a geometrical property of a given configuration with a topological invariant supports the notion that the ideal configuration contains important information about knots and links. The importance of the concept of ideal configuration was already suggested by the good correlation between the 3D writhe of ideal knot configurations and the ensemble average of the 3D writhe of random configurations of the considered knots. The values of the new invariant are quantized: multiples of 4/7 for links with an odd number of components (including knots) and 2/7 plus multiples of 4/7 for links with an even number of components.

Compositional data analysis motivated the introduction of a complete Euclidean structure in the simplex of D parts. This was based on the early work of J. Aitchison (1986) and completed recently when Aitchinson distance in the simplex was associated with an inner product and orthonormal bases were identified (Aitchison and others, 2002; Egozcue and others, 2003). A partition of the support of a random variable generates a composition by assigning the probability of each interval to a part of the composition. One can imagine that the partition can be refined and the probability density would represent a kind of continuous composition of probabilities in a simplex of infinitely many parts. This intuitive idea would lead to a Hilbert-space of probability densitiesby generalizing the Aitchison geometry for compositions in the simplex into the set probability densities

Traffic Engineering objective is to optimize network resource utilization. Although several works have been published about minimizing network resource utilization in MPLS networks, few of them have been focused in LSR label space reduction. This letter studies Asymmetric Merged Tunneling (AMT) as a new method for reducing the label space in MPLS network. The proposed method may be regarded as a combination of label merging (proposed in the MPLS architecture) and asymmetric tunneling (proposed recently in our previous works). Finally, simulation results are performed by comparing AMT with both ancestors. They show a great improvement in the label space reduction factor

Most network operators have considered reducing LSR label spaces (number of labels used) as a way of simplifying management of underlaying virtual private networks (VPNs) and therefore reducing operational expenditure (OPEX). The IETF outlined the label merging feature in MPLS-allowing the configuration of multipoint-to-point connections (MP2P)-as a means of reducing label space in LSRs. We found two main drawbacks in this label space reduction a)it should be separately applied to a set of LSPs with the same egress LSR-which decreases the options for better reductions, and b)LSRs close to the edge of the network experience a greater label space reduction than those close to the core. The later implies that MP2P connections reduce the number of labels asymmetrically
