[目的 ]探讨蓝氏贾第鞭毛虫种内系统发育及遗传多样性。 [方法 ]对不同来源虫株的磷酸丙糖异构酶(tim)基因进行 PCR扩增、序列测定后 ,用简约法和 NJ法构建分子系统树进行系统学分析。 [结果 ]在所测序列中共有 12 4个位点存在变异 (2 3% ) ,且大多数为发生在第三密码子的同义突变 ,两种构树方法所得两树的分枝结构相似 ,均将受试的 16株蓝氏贾第鞭毛虫分为明显的两组。 [结论 ]tim基因可作为研究蓝氏贾第鞭毛虫群体遗传结构一个有效的遗传标记

综述了timeless基因的发现、多态性和重要功能。timeless是最先被发现的两个生物钟基因之一。生物钟的昼夜节律由PER、TIM、CLOCK和CYCLE 4个生物钟齿轮组成的正负反馈回路进行调节。其中TIM可以受光因子调控,它还可以与PER形成异二聚体,通过正负调控方式调节果蝇的昼夜节律行为。

1. The stripe-backed weasel Mustela strigidorsa is one of the rarest and least-known mustelids in the world. Its phylogenetic relationships with other Mustela species remain controversial, though several unique morphological features distinguish it from congeners. 2. It probably lives mainly in evergreen forests in hills and mountains, but has also been recorded from plains forest, dense scrub, secondary forest, grassland and farmland. Known sites range in altitude from 90 m to 2500 m. Data are insufficient to distinguish between habitat and altitudes which support populations, and those where only dispersing animals may occur. 3. It has been confirmed from many localities in north-east India, north and central Myanmar, south China, north Thailand, north and central Laos, and north and central Vietnam. Given the limited survey effort, the number of recent records shows that the species is not as rare as hitherto believed. Neither specific nor urgent conservation needs are apparent.

Objective To confirm the genetic relation between Giardia lamblia (G. lamblia) isolates from different geographic regions of China and other countries. Methods Genomic DNA were extracted from the trophozoites or cysts of Giardia lamblia. The triose phosphate isomerase (tim) gene was amplified using polymerase chain reaction (PCR) technique. PCR products were digested with endonuclease and sequenced. The data of sequencing were analyzed with the DNAstar software and compared with that of the isolates acquired from GenBank. Results Of nine isolates of Giardia lamblia from China (C1, C2, CH2 and CH3), Cambodia (CAM), Australia (A1 and A2) and America (BP and CDC), respectively, 3 (A1, A2 and CAM) fit into Group 1 (WB), 2 (CH2 and CH3) into Group 2, and 4 (C1, C2, BP and CDC) into Group 3 (GS). The results confirmed the genetic relatedness of G. lamblia isolates from all over the world. Conclusion Genotyping isolates of G. Lamblia provides important information for establishing the phylogenetic relationship or for the epidemiological evaluation of the spreading of this organism.

Experiments are conducted to examine the mechanisms behind the coupling between corner separation and separation away from the corner when holding a high-Machnumber M∞ = 1.5 normal shock in a rectangular channel. The ensuing shock wave interaction with the boundary layer on the wind tunnel floor and in the corners was studied using laser Doppler anemometry, Pitot probe traverses, pressure sensitive paint and flow visualization. The primary mechanism explaining the link between the corner separation size and the other areas of separation appears to be the generation of compression waves at the corner, which act to smear the adverse pressure gradient imposed upon other parts of the flow. Experimental results indicate that the alteration of the -region, which occurs in the supersonic portion of the shock wave/boundary layer interaction (SBLI), is more important than the generation of any blockage in the subsonic region downstream of the shock wave. © Copyright 2012 Cambridge University Press.

Experiments have been conducted to examine the mechanisms behind the coupling between corner separation and centreline separation when holding a normal shock in a rectangular channel. The study has focused on a M ∞ = 1.5 normal shock held in a wind tunnel with a parallel rectangular cross-section. The primary mechanism explaining the link between the corner separation size and the centreline separation appears to be the generation of compression waves which act to smear the adverse pressure gradient imposed upon other parts of the flow. In addition, the origin of the λ-foot leading leg appears to be depended upon the size of the corner separations. Experimental results indicate that the alteration of the λ-region, which occurs in the supersonic portion of the SBLI, is more important than the generation of any blockage in the subsonic region downstream of the shock wave. 糖酵解作为细胞的基本能量代谢途径广泛存在于各类生物中。真核生物的糖 酵解一般都在细胞质中进行。然而,有报道发现少数物种如原生生物的动基体类、 少数藻类(如硅藻)中糖酵解途径却并非发生在胞质中,而是分别发生在糖酵解 体和线粒体中。在处于关键进化地位的单细胞绿藻—衣藻中,其糖酵解途径的亚 细胞定位一直存在争议。本文针对衣藻糖酵解途径的亚细胞定位问题开展了如下 两方面的研究工作,获得了一些重要结果: 1 衣藻糖酵解途径亚细胞定位的实验研究 本实验室曾利用生物信息学方法 对衣藻糖酵解途径的酶进行了鉴定和定位预测的理论分析,结果发现该途径的前 7 步主要发生在叶绿体,后3 步则主要发生在胞质。据此,本文选取了糖酵解途 径中具有代表性的几个酶(PFKa、PFKb、NAD+-GAPDH 和TIM)进行了GFP 定 位实验,以期通过实验验证该结论。结果表明,参与糖酵解途径第3、5、6 步反 应的酶(分别为PFK、TIM 和NAD+-GAPDH)确实定位到叶绿体。本文的研究 结果在一定程度上证实了以往的生物信息学理论分析结果--衣藻糖酵解途径的 前七步发生在叶绿体中。 2 衣藻叶绿体内糖酵解与卡尔文循环二者之间协调机制的研究 既然衣藻糖 酵解途径发生在叶绿体中,这势必与同样发生在叶绿体中的卡尔文循环途径发生 冲突。因为这两个途径有五步反应是逆向重叠的,它们之间是如何协调的呢?为 了探讨这一问题,本文开展了如下研究:首先,我们利用生物信息学方法对衣藻 全基因组中参与上述五步逆向重叠反应的酶进行了鉴定,并对它们的定位进行了 预测。结果发现,第一步有两个定位到叶绿体的PFK 催化糖酵解途径的反应, 其逆向反应则是由FBP 来催化;第四步在叶绿体中有两个利用不同辅酶(NAD+ 和NADP+)的GAPDH;第二步有两个拷贝的FBA(FBAa 和FBAb)定位到叶 绿体;而参与第三步与第五步的TIM 和PGK 仅有一个拷贝且是叶绿体定位。其 次,我们对这些参与五步逆向重叠反应且定位到叶绿体的酶进行了real time RT-PCR 实验,得到了它们的转录表达谱。PFK、FBAb 和NAD+-GAPDH 在整个 光照和黑暗中均表达恒定波动不大,但相对于PFK 和NAD+-GAPDH,FBAb 的 表达量却极低;而FBP、FBAa 和NADP+-GAPDH 在黑暗条件下表达量低且恒定, 而进入光照后表达量急剧增长,1 小时之后即能达到最高,表明这些酶的表达是 受光调节的;PGK 的转录表达情况则与FBP、FBAa 和NADP+-GAPDH 类似,也 是光照条件下表达量剧增,说明它也是受光调节的;TIM 在光照条件下也是有上 调趋势的,只是幅度较小,推测可能与其作为异构酶催化效率高有关。因此我们 认为PFK、FBAb 和NAD+-GAPDH 是专职参与糖酵解途径的,而FBP、FBAa 和 NADP+-GAPDH 是专职催化卡尔文循环反应的,通过光对它们表达的调节而 来协调它们各自参与的反应;而TIM 和PGK 则是两个代谢途径所共有的,它们 是通过光照、底物浓度等综合因素来调节它们所参与的反应方向进而达到两个途 径之间的协调。 该研究工作不仅对衣藻糖酵解途径的亚细胞定位进行了实验验证,还首次揭 示了衣藻同处叶绿体中的糖酵解与卡尔文循环两个途径之间的协调机制。

We present results on the system size dependence of high transverse momentum di-hadron correlations at root s(NN) = 200 GeV as measured by STAR at RHIC. Measurements in d + Au, Cu + Cu and Au + Au collisions reveal similar jet-like near-side correlation yields (correlations at small angular separation Delta phi similar to 0, Delta eta similar to 0) for all systems and centralities. Previous measurements have shown Chat the away-side (Delta phi similar to pi) yield is suppressed in heavy-ion collisions. We present measurements of the away-side Suppression as a function of transverse momentum and centrality in Cu + Cu and Au + Au collisions. The suppression is found to be similar in Cu + Cu and An + An collisions at a similar number of participants. The results are compared to theoretical calculations based on the patron quenching model and the modified fragmentation model. The observed differences between data and theory indicate that the correlated yields presented here will further constrain dynamic energy loss models and provide information about the dynamic density profile in heavy-ion collisions. The contribution of B meson decays to nonphotonic electrons, which are mainly produced by the semileptonic decays of heavy-flavor mesons, in p + p collisions at root s = 200 GeV has been measured using azimuthal correlations between nonphotonic electrons and hadrons. The extracted B decay contribution is approximately 50% at a transverse momentum of p(T) >= 5 GeV/c. These measurements constrain the nuclear modification factor for electrons from B and D meson decays. The result indicates that B meson production in heavy ion collisions is also suppressed at high p(T).

200 GeV corresponding to baryon chemical potentials (mu(B)) between 200 and 20 MeV. Our measurements of the products kappa sigma(2) and S sigma, which can be related to theoretical calculations sensitive to baryon number susceptibilities and long-range correlations, are constant as functions of collision centrality. We compare these products with results from lattice QCD and various models without a critical point and study the root s(NN) dependence of kappa sigma(2). From the measurements at the three beam energies, we find no evidence for a critical point in the QCD phase diagram for mu(B) below 200 MeV.
