A long-standing controversy is whether autophagy is a bona fide cause of mammalian cell death. We used a cell-penetrating autophagy-inducing peptide, Tat-Beclin 1, derived from the autophagy protein Beclin 1, to investigate whether high levels of autophagy result in cell death by autophagy. Here we show that Tat-Beclin 1 induces dose-dependent death that is blocked by pharmacological or genetic inhibition of autophagy, but not of apoptosis or necroptosis. This death, termed "autosis," has unique morphological features, including increased autophagosomes/autolysosomes and nuclear convolution at early stages, and focal swelling of the perinuclear space at late stages. We also observed autotic death in cells during stress conditions, including in a subpopulation of nutrient-starved cells in vitro and in hippocampal neurons of neonatal rats subjected to cerebral hypoxia-ischemia in vivo. A chemical screen of ~5,000 known bioactive compounds revealed that cardiac glycosides, antagonists of Na(+),K(+)-ATPase, inhibit autotic cell death in vitro and in vivo. Furthermore, genetic knockdown of the Na(+),K(+)-ATPase α1 subunit blocks peptide and starvation-induced autosis in vitro. Thus, we have identified a unique form of autophagy-dependent cell death, a Food and Drug Administration-approved class of compounds that inhibit such death, and a crucial role for Na(+),K(+)-ATPase in its regulation. These findings have implications for understanding how cells die during certain stress conditions and how such cell death might be prevented.

The CbrA/B system in pseudomonads is involved in the utilization of carbon sources and carbon catabolite repression (CCR) through the activation of the small RNAs crcZ in Pseudomonas aeruginosa, and crcZ and crcY in Pseudomonas putida. Interestingly, previous works reported that the CbrA/B system activity in P. aeruginosa PAO1 and P. putida KT2442 responded differently to the presence of different carbon sources, thus raising the question of the exact nature of the signal(s) detected by CbrA. Here, we demonstrated that the CbrA/B/CrcZ(Y) signal transduction pathway is similarly activated in the two Pseudomonas species. We show that the CbrA sensor kinase is fully interchangeable between the two species and, moreover, responds similarly to the presence of different carbon sources. In addition, a metabolomics analysis supported the hypothesis that CCR responds to the internal energy status of the cell, as the internal carbon/nitrogen ratio seems to determine CCR and non-CCR conditions. The strong difference found in the 2-oxoglutarate/glutamine ratio between CCR and non-CCR conditions points to the close relationship between carbon and nitrogen availability, or the relationship between the CbrA/B and NtrB/C systems, suggesting that both regulatory systems sense the same sort or interrelated signal.

Members of the tumor necrosis factor (TNF) family play key roles in the regulation of inflammation, immune responses and tissue homeostasis. Here we describe the identification of the chicken homologue of mammalian B cell activating factor of the TNF family (BAFF/BLyS). By searching a chicken EST database we identified two overlapping cDNA clones that code for the entire open reading frame of chicken BAFF (chBAFF), which contains a predicted transmembrane domain and a putative furin protease cleavage site like its mammalian counterparts. The amino acid identity between soluble chicken and human BAFF is 76%, considerably higher than for most other known cytokines. The chBAFF gene is most strongly expressed in the bursa of Fabricius. Soluble recombinant chBAFF produced by human 293T cells interacted with the mammalian cell-surface receptors TACI, BCMA and BAFF-R. It bound to chicken B cells, but not to other lymphocytes, and it promoted the survival of splenic chicken B cells in culture. Furthermore, bacterially expressed chBAFF induced the selective expansion of B cells in the spleen and cecal tonsils when administered to young chicks. Our results suggest that like its mammalian counterpart, chBAFF plays an important role in survival and/or proliferation of chicken B cells.

Genes underlying mutant phenotypes can be isolated by combining marker discovery, genetic mapping and resequencing, but a more straightforward strategy for mapping mutations would be the direct comparison of mutant and wild-type genomes. Applying such an approach, however, is hampered by the need for reference sequences and by mutational loads that confound the unambiguous identification of causal mutations. Here we introduce NIKS (needle in the k-stack), a reference-free algorithm based on comparing k-mers in whole-genome sequencing data for precise discovery of homozygous mutations. We applied NIKS to eight mutants induced in nonreference rice cultivars and to two mutants of the nonmodel species Arabis alpina. In both species, comparing pooled F2 individuals selected for mutant phenotypes revealed small sets of mutations including the causal changes. Moreover, comparing M3 seedlings of two allelic mutants unambiguously identified the causal gene. Thus, for any species amenable to mutagenesis, NIKS enables forward genetics without requiring segregating populations, genetic maps and reference sequences.

Constitutive activation of the nuclear factor-κ B (NF-κB) pathway is a hallmark of the activated B-cell-like (ABC) subtype of diffuse large B-cell lymphoma (DLBCL). Recurrent mutations of NF-κB regulators that cause constitutive activity of this oncogenic pathway have been identified. However, it remains unclear how specific target genes are regulated. We identified the atypical nuclear IκB protein IκB-ζ to be upregulated in ABC compared with germinal center B-cell-like (GCB) DLBCL primary patient samples. Knockdown of IκB-ζ by RNA interference was toxic to ABC but not to GCB DLBCL cell lines. Gene expression profiling after IκB-ζ knockdown demonstrated a significant downregulation of a large number of known NF-κB target genes, indicating an essential role of IκB-ζ in regulating a specific set of NF-κB target genes. To further investigate how IκB-ζ mediates NF-κB activity, we performed immunoprecipitations and detected a physical interaction of IκB-ζ with both p50 and p52 NF-κB subunits, indicating that IκB-ζ interacts with components of both the canonical and the noncanonical NF-κB pathway in ABC DLBCL. Collectively, our data demonstrate that IκB-ζ is essential for nuclear NF-κB activity in ABC DLBCL, and thus might represent a promising molecular target for future therapies.

The cortical collecting duct (CCD) plays a key role in regulated K(+) secretion, which is mediated mainly through renal outer medullary K(+) (ROMK) channels located in the apical membrane. However, the mechanisms of the regulation of urinary K(+) excretion with regard to K(+) balance are not well known. We took advantage of a recently established mouse CCD cell line (mCCD(cl1)) to investigate the regulation of K(+) secretion by mineralocorticoid and K(+) concentration. We show that this cell line expresses ROMK mRNA and a barium-sensitive K(+) conductance in its apical membrane. As this conductance is sensitive to tertiapin-Q, with an apparent affinity of 6 nM, and to intracellular acidification, it is probably mediated by ROMK. Overnight exposure to 100 nM aldosterone did not significantly change the K(+) conductance, while it increased the amiloride-sensitive Na(+) transport. Overnight exposure to a high K(+) (7 mM) concentration produced a small but significant increase in the apical membrane barium-sensitive K(+) conductance. The mRNA levels of all ROMK isoforms measured by qRT-PCR were not changed by altering the basolateral K(+) concentration but were decreased by 15-45% upon treatment with aldosterone (0.3 or 300 nM for 1 and 3 h). The paradoxical response of ROMK expression to aldosterone could possibly work as a preventative mechanism to avoid excessive K(+) loss which would otherwise result from the increased electrogenic Na(+) transport and associated depolarization of the apical membrane in the CCD. In conclusion, mCCD(cl1) cells demonstrate a significant K(+) secretion, probably mediated by ROMK, which is not stimulated by aldosterone but increased by overnight exposure to a high K(+) concentration.

Distribuição espacial de Aphis gossypii (Glover) (Hemiptera, Aphididae) e Bemisia tabaci (Gennadius) biótipo B (Hemiptera, Aleyrodidae) em algodoeiro Bt e não-Bt. O estudo da distribuição espacial de adultos de Bemisia tabaci e de Aphis gossypii nas culturas do algodoeiro Bt e não-Bt é fundamental para a otimização de técnicas de amostragens, além de revelar diferenças de comportamento de espécies não-alvo dessa tecnologia Bt entre as duas cultivares. Nesse sentido, o experimento buscou investigar o padrão da distribuição espacial dessas espécies de insetos no algodoeiro convencional não-Bt e no cultivar Bt. As avaliações ocorreram em dois campos de 5.000 m² cada, nos quais se realizou 14 avaliações com contagem de adultos da mosca-branca e colônias de pulgões. Foram calculados os índices de agregação (razão variância/média, índice de Morisita e Expoente k da Distribuição Binomial Negativa) e realizados os testes ajustes das classes numéricas de indivíduos encontradas e esperadas às distribuições teóricas de freqüência (Poisson, Binomial Negativa e Binomial Positiva). Todas as análises mostraram que, em ambas as cultivares, a distribuição espacial de B. tabaci ajustou-se a distribuição binomial negativa durante todo o período analisado, indicando que a cultivar transgênica não influenciou o padrão de distribuição agregada desse inseto. Já com relação às análises para A. gossypii, os índices de agregação apontaram distribuição agregada nas duas cultivares, mas as distribuições de freqüência permitiram concluir a ocorrência de distribuição agregada apenas no algodoeiro convencional, pois não houve nenhum ajuste para os dados na cultivar Bt. Isso indica que o algodão Bt alterou o padrão normal de dispersão dos pulgões no cultivo.

The chicken represents the best-characterized animal model for B cell development in the so-called gut-associated lymphoid tissue (GALT) and the molecular processes leading to B cell receptor diversification in this species are well investigated. However, the mechanisms regulating B cell development and homeostasis in GALT species are largely unknown. Here we investigate the role played by the avian homologue of B cell-activating factor of the tumor necrosis factor family (BAFF). Flow cytometric analysis showed that the receptor for chicken B cell-activating factor of the tumor necrosis factor family (chBAFF) is expressed by mature and immature B cells. Unlike murine and human BAFF, chBAFF is primarily produced by B cells both in peripheral lymphoid organs and in the bursa of Fabricius, the chicken's unique primary lymphoid organ. In vitro and in vivo studies revealed that chBAFF is required for mature B cell survival. In addition, in vivo neutralization with a decoy receptor led to a reduction of the size and number of B cell follicles in the bursa, demonstrating that, in contrast to humans and mice, in chickens BAFF is also required for the development of immature B cells. Collectively, we show that chBAFF has phylogenetically conserved functions in mature B cell homeostasis but displays unique and thus far unknown properties in the regulation of B cell development in birds.

OBJECTIVE: To investigate the merits of vaccination against hepatitis B virus (HBV) in HIV-positive individuals with isolated antibodies to hepatitis B core antigen (anti-HBc). METHODS: HIV-positive patients with isolated anti-HBc and CD4 counts >200 cells/mm(3) received HBV vaccination. An antibody titre to hepatitis B surface antigen (anti-HBs titres) ≥10 IU/L one month post-vaccination was termed an anamnestic response; a titre <10 IU/L was termed a primary response. Patients with primary responses received a 3-dose vaccine course. Anti-HBs titres in all responders were measured 12 and 24 months post-vaccination. RESULTS: 37 patients were studied: 19 (51%) were co-infected with hepatitis C; median CD4 count was 443 cells/mm(3). 8/37 patients (22%) elicited an anamnestic response. 29/37 patients (78%) elicited a primary response. After a 3-dose vaccine course, 15/25 primary responders (60%) achieved anti-HBs titres ≥10 IU/L. HIV acquisition through injecting drug use was the only independent predictor of an anamnestic response (OR 22.9, CI 1.71-306.74, P=0.018). Median anti-HBs titres for anamnestic and primary responders were 51 IU/L (13-127) and 157 IU/L (25-650) respectively. Of all responders, 12/23 (52%) retained anti-HBs titres ≥10 IU/L at 24 months. Anti-HBs duration was not significantly different between anamnestic and primary responders. CONCLUSIONS: 23/37 HIV-positive patients (62%) with isolated anti-HBc achieved anti-HBs titres ≥10 IU/L after 1-3 vaccine doses. O conhecimento da erodibilidade dos solos (fator K) é de fundamental importância para a aplicação da equação universal de perdas de solo (EUPS). O objetivo deste trabalho foi verificar a adequação de diferentes métodos indiretos para estimativa da erodibilidade de solos com horizonte B textural (Bt) de várias regiões geográficas do Brasil. Coletaram-se amostras superficiais de 22 solos com Bt, cujo fator K foi determinado diretamente no campo. Realizaram-se diversas análises químicas e físicas, de modo que o fator K pudesse ser estimado por treze diferentes métodos indiretos. Correlacionaram-se os valores dos fatores K medidos no campo com os obtidos indiretamente. Verificou-se que nenhum dos métodos indiretos testados é adequado para a estimativa do fator K de todo o conjunto de solos com Bt estudados.

Os métodos existentes para estimar a erodibilidade dos solos (fator K) não se têm mostrado adequados quando aplicados a solos brasileiros. Este trabalho teve por objetivo o desenvolvimento de método indireto para estimar a erodibilidade, visando obter esse fator, a partir de propriedades do solo facilmente determináveis. Assim, os horizontes A e Bt (B textural) de 22 solos de várias regiões geográficas do Brasil foram submetidos a diversas análises químicas e físicas, e a descrição morfológica do perfil foi considerada. Tais resultados (variáveis independentes) foram submetidos à análise de regressão linear múltipla em "stepwise", e relacionados com a erodibilidade (fator K) medida diretamente no campo (variável dependente). Obtiveram-se várias equações com graus crescentes de complexidade e precisão. A partir das variáveis selecionadas, tanto do horizonte A quanto do Bt, e da distribuição do tamanho das partículas, obtida com e sem dispersante químico, foi possível estimar, satisfatoriamente, a erodibilidade desses solos. As informações contidas na descrição morfológica dos perfis de solo, especialmente as referentes à estrutura, bem como os teores de carbono orgânico, foram de grande importância para a determinação indireta da erodibilidade (fator K).

El crucero se desarrolló en dos etapas, la primera en primavera 2010, de 4° a 11°25’S y la segunda en verano 2011, de 10°10’ a 17°23’S, entre 50 a 200 mn de la costa. La captura del recurso en la primera etapa fue 35.079,6 kg (25.669,1 kg de producción) y en la segunda 165.955,7 kg (123.229,5 kg de producción). La captura por unidad de esfuerzo por día de trabajo fluctuó de 2,9 a 4.292,6 kg/hora; 0,68 a 948,4 kg/línea; 0,07 a 99,8 kg/ línea*hora y 0,0017 a 2,4957 kg/pot*hora. La longitud del manto varió de 17 a 119 cm. Se registró hembras en estadio desovante III (51,5%), madurante II (21,6%); y machos en estadio evacuación III (85,1%) y virginales I (9,7%). Los grupos tróficos más importantes fueron: cefalópodos (% IRI= 66,4), crustáceos (% IRI= 23,7), peces (% IRI= 9,9). Las hembras presentaron: L∞ =111,233 cm LM, K=0,016, t0= 235 y los machos L∞ =99,718 cm LM, K=0,167 y t0=228,4; esta especie tiene una longevidad próxima o poco mayor a un año.

T cell stimulation requires the input and integration of external signals. Signaling through the T cell receptor (TCR) is known to induce formation of the membrane-tethered CBM complex, comprising CARMA1, BCL10, and MALT1, which is required for TCR-mediated NF-κB activation. TCR signaling has been shown to activate NOTCH proteins, transmembrane receptors also implicated in NF-κB activation. However, the link between TCR-mediated NOTCH signaling and early events leading to induction of NF-κB activity remains unclear. In this report, we demonstrate a novel cytosolic function for NOTCH1 and show that it is essential to CBM complex formation. Using a model of skin allograft rejection, we show in vivo that NOTCH1 acts in the same functional pathway as PKCθ, a T cell-specific kinase important for CBM assembly and classical NF-κB activation. We further demonstrate in vitro NOTCH1 associates physically with PKCθ and CARMA1 in the cytosol. Unexpectedly, when NOTCH1 expression was abrogated using RNAi approaches, interactions between CARMA1, BCL10, and MALT1 were lost. This failure in CBM assembly reduced inhibitor of kappa B alpha phosphorylation and diminished NF-κB-DNA binding. Finally, using a luciferase gene reporter assay, we show the intracellular domain of NOTCH1 can initiate robust NF-κB activity in stimulated T cells, even when NOTCH1 is excluded from the nucleus through modifications that restrict it to the cytoplasm or hold it tethered to the membrane. Collectively, these observations provide evidence that NOTCH1 may facilitate early events during T cell activation by nucleating the CBM complex and initiating NF-κB signaling.

BAFF deficiency in mice impairs B cell development beyond the transitional stage 1 in the spleen and thus severely reduces the size of follicular and marginal zone B cell compartments. Moreover, humoral immune responses in these mice are dramatically impaired. We now addressed the question whether the decrease in mature B cell numbers and the reduced humoral immune responses in BAFF-deficient mice could be overcome by the injection of recombinant BAFF. We therefore engineered a recombinant protein containing the human IgG1 Fc moiety fused to receptor-binding domain of human BAFF (Fc-BAFF). At 1 week after the second injection of this fusion protein a complete rescue of the marginal zone B cell compartment and a 50% rescue of the follicular B cell compartment was observed. Moreover these mice mounted a T cell-dependent humoral immune response indistinguishable from wild-type mice. By day 14 upon arrest of Fc-BAFF treatment mature B cell numbers in the blood dropped by 50%, indicating that the life span of mature B cells in the absence of BAFF is 14 days or less. Collectively these findings demonstrate that injection of Fc-BAFF in BAFF-deficient mice results in a temporary rescue of a functional mature B cell compartment.

The Miocene Paine Granite in the Torres del Paine Intrusive Complex, southern Chile, is an extraordinary example of an upper crustal mafic and granitic intrusion. The granite intruded as a series of three sheets, each one underplating the previous sheet along the top of the basal Paine Mafic Complex. High-precision U/Pb geochronology on single zircons using isotope dilution-thermal ionization mass spectrometry yields distinct ages of 12.59 +/- 0.02 Ma and 12.50 +/- 0.02 Ma, respectively, for the first and last sheet of the laccolith. This age relationship is consistent with field observations. The zircon ages define a time frame of 90 +/- 40 k.y. for the emplacement of a >2000-m-thick granite laccollith. These precise U-Pb zircon ages permit identification of the pulses in a 20 k.y. range. The data obtained for the Paine Granite fill the gap between 100 k.y. and 100-1000 yr pulses described in the literature for crustal magma chambers.
