Foram avaliados o desempenho produtivo e os parâmetros fisiológicos de tilápias-do-nilo alimentadas, durante 112 dias, com dietas preparatórias para o inverno, formuladas com diferentes níveis de lipídeo e vitamina C. Utilizaram-se 192 alevinos (5,57 ± 0,50 g) distribuídos em 32 tanques-rede. O experimento foi conduzido em esquema fatorial 2 ´ 3, composto de dois níveis (8,0 e 12,0%) de lipídeo e três níveis (300,0; 600,0 e 1.200,0 mg/kg de ração) de vitamina C, em comparação a duas dietas (uma isenta dos nutrientes-teste e outra com 6,0% de lipídeo e 125,0 mg de vitamina C), totalizando oito dietas isoprotéicas (32,0% de proteína digestível). A energia adicional proveniente do lipídeo é depositada na cavidade abdominal em forma de gordura visceral; a deposição desta gordura como reserva de energia foi obtida nos níveis de suplementação de lipídeo testados; a concentração hepática de vitamina C é proporcional à sua concentração na dieta; a ausência desta vitamina na dieta prejudica a eritropoiese e a síntese de colágeno.

Objectives: To study the nanoleakage pattern in the dentin hybrid layer by using different dentin adhesives. The null hypotheses tested in this study were: 1) dentin conditioning time does not affect nanoleakage within the hybrid layer; 2) the type of dentin adhesive used does not affect nanoleakage. Methods: Standardized Class V cavities were prepared in 30 intact human molars on the buccal and lingual surfaces. The specimens were randomly assigned to 2 total-etch dentin adhesives (OptiBond SOLO Plus [OPS, Kerr] and One-Step [ONS, BISCO Inc]) and 2 self-etch dentin adhesives (Clearfil SE Bond [CSE, Kuraray] and Adper Prompt L-Pop [APL, 3M ESPE]). The specimens were etched or conditioned for 15 seconds, 30 seconds or 60 seconds. Upon restoration of the Class V cavities with the proprietary resin composite, the specimens were isolated with nail polish except for a 2.0-mm rim around the restoration, and they were immersed in 50 wt% ammoniacal silver nitrate solution (pH=9.5) for 24 hours followed by 8 hours of immersion in photo-developing solution to reduce the silver ions to metallic silver. The specimens were fixed, dehydrated and processed for FESEM and TEM. Silver penetration was measured along the cervical wall, and data were analyzed with Kruskal-Wallis non-parametric tests at a significance level of 95%. Results: There were no statistically significant differences among the experimental groups for the factor conditioning time (p>0.926). There were significant differences for the variable dentin adhesive (p<0.0001). The least amount of nanoleakage within the hybrid layer occurred with CSE, while ONS resulted in the greatest penetration of silver ions. The adhesives OPS and APL ranked in the intermediary subset. Under TEM, all adhesives resulted in some degree of nanoleakage within the hybrid layer. Both spotted/reticular and water-tree nanoleakage patterns were observed. Significance: Longer conditioning times did not increase nanoleakage within the hybrid layer. Nanoleakage varied with the type of adhesive used.

The effect of lysine amino acid supplementation on the growth characteristics and morphological pattern of skeletal muscle tissue in Nile tilapia Oreochromis niloticus larvae was evaluated. There were four treatments (T) with increasing levels of lysine supplement (T1 = 0.0%: T2 =1.1%; T3 = 1.7%; T4 = 4.0%) and one treatment with a commercial diet (T5). In all treatments, morphological and histochemical muscle tissue analyses were similar. Two distinct layers were identified: a superficial red layer, more developed in the lateral line region, formed by fibres with intense to moderate NADH-TR reaction and strong acid-stable mATPase activity, and a deep white one, most of the Muscle mass, formed by fibres with weak NADH-TR reaction and strong alkali-stable mATPase activity. There was an intermediate layer between these two layers with fibres exhibiting either weak acid-stable or acid-labile mATPase activity. Body mass increase was significantly higher in T5 than in the lysine treatments (T1-T4). There was no difference in number and diameters of muscle fibres between lysine treatments. In T5, muscle fibre diameter and number were higher. The frequency of red fibres with diameters <= 8 mu m was higher in the lysine treatments, and with diameters between 16 and 24 mu m, was higher in T5. Most white fibre diameters in T5 were significantly larger than 24 mu m and in T1-T4 were between 8 and 16 mu m. This study tested the use of ventilatory frequency (VF) as an indicator of stress in the Nile tilapia, Oreochromis niloticus (L.). Firstly, we tested the relationship between VF and plasma cortisol after confinement. Confined fish showed higher VF and plasma cortisol levels, but the latter continued to increase significantly for longer time than VF. Secondly, we conducted another experiment to test the use of VF as indicator of fish stress. In four out of six treatment, we confined the fish for different intervals (30 s, 5, 15 or 30 min). The others were used as control. In one, no handling was imposed. The other control consisted of introducing the partition (the same used to perform the confinement) into the aquarium for less than 4 s, without confinement and immediately removing the partition (partition control). Ventilatory frequency was increased for the partition control as much as for the longer duration of confinement. This clearly indicates that VF is a very sensitivity response to disturbance, but of limited use because this parameter does not reflect the severity of the stimulus. Thus, although VF is a non-invasive technique that does not require sophisticated recording equipment, its usefulness is limited. (C) 2004 Elsevier B.V. All rights reserved.

Selection favors females that attend to reliable information about male genetic quality and fitness. Male nutritional condition can be a significant sign of mate quality since poor nutrition can be related to reduced sperm quality, low sperm quantity, sexually transmitted diseases, and parasites. We tested whether female Nile tilapia, Oreochromis niloticus, preferred the chemical cues of males that were well fed with high-protein diet over those fed with low-protein diet. Females do not only discriminate between males but also show a preference for well-fed males, discriminating between the odors with respect to nutritional state, suggesting that they were responding to a food-specific chemical cue. It is therefore likely that nutritional condition is related to the production of pheromones in males. Our results suggest that information about male nutritional state can be conveyed in chemical cues and that females attend to these cues during mate choice.

We investigated whether juveniles of the nocturnal fish jundia (Rhamdia quelen) and the diurnal fish Nile tilapia (Oreochromis niloticus) are able to chemically communicate stress to conspecifics. Groups of 8 fish were reared in tanks under recirculated water (water exchanged among all the tanks) for each species. Fish were handled in half of the tanks (stressor fish) and whole-body cortisol concentrations were compared among handled fish, non-handled fish exposed to water from the handled fish, and non-handled control fish held with no water communication. For each treatment cortisol concentrations were determined before exposure to the stressor (basal levels) and after 1, 2, 4, 8, and 24 h. Basal levels of cortisol confirmed fish were unstressed in the beginning of the experiment. Cortisol was increased in the stressor fish 1 h after handling. Fish receiving water from the stressor fish increased cortisol levels later (2 h after the stressor fish were handled). As the isolated control group maintained cortisol levels unchanged throughout the experiment, we concluded that some chemical factor was released by the stressed fish in the water and thus stressed the conspecifics. This pattern was similar for both unrelated species, thus suggesting that this communication might have evolved earlier in fish and reinforcing the biological value of this kind of information. 5S rDNA sequences present an intense dynamism and have proved to be valuable as genetic markers to distinguish closed related species and also in the understanding of the evolutionary dynamic of repetitive sequences in the genomes. In order to identify patterns of 5S rDNA organization and their evolution in the genome of fish species, such genomic segment was investigated in the tilapias Oreochromis niloticus and Tilapia rendalli, and in the hybrid O. urolepis hornorum x O. mossambicus. A dual 5S rDNA system was identified in the three analyzed tilapia samples. Although each 5S rDNA class was conserved among the three samples, a distinct 5S rDNA genome organization pattern could be evidenced for each sample. The presence of a dual 5S rDNA system seems to be a general trait among non-related teleost fish orders, suggesting that evolutionary events of duplication have occurred before the divergence of the main groups of teleost fishes.
