Small heat shock proteins are ubiquitous molecular chaperones that form the first line of defence against the detrimental effects of cellular stress. Under conditions of stress they undergo drastic conformational rearrangements in order to bind to misfolded substrate proteins and prevent cellular protein aggregation. Owing to the dynamic nature of small heat shock protein oligomers, elucidating the structural basis of chaperone action and oligomerization still remains a challenge. In order to understand the organization of sHSP oligomers, we have determined crystal structures of a small heat shock protein from Salmonella typhimurium in a dimeric form and two higher oligomeric forms: an 18-mer and a 24-mer. Though the core dimer structure is conserved in all the forms, structural heterogeneity arises due to variation in the terminal regions.

For brittle solids containing numerous small cracks, a micromechanical damage theory is presented which accounts for the interactions between different small cracks and the effect of the boundary of a finite solid, and includes growth of the pre-existing small cracks. The analysis is based on a superposition scheme and series expansions of the complex potentials. The small crack evolution process is simulated through the use of fracture mechanics incorporating appropriate failure criteria. The stress-strain relations are obtained from the micromechanics analysis. Typical examples are given to illustrate the potential capability of the proposed theory. These results show that the present method provides a direct and efficient approach to deal with brittle finite solids containing multiple small cracks. The stress-strain relation curves are evaluated for a rectangular plate containing small cracks.

A perturbation method is used to examine the linear instability of thermocapillary convection in a liquid bridge of floating half-zone filled with a small Prandtl number fluid. The influence of liquid bridge volume on critical Marangoni number and flow features is analyzed. The neutral modes show that the instability is mainly caused by the bulk flow that is driven by the nonuniform thermocapillary forces acting on the free surface. The hydrodynamic instability is dominant in the case of small Prandtl number fluid and the first instability mode is a stationary bifurcation. The azimuthal wave number for the most dangerous mode depends on the liquid bridge volume, and is not always two as in the case of a cylindrical liquid bridge with aspect ratio near 0.6. Its value may be equal to unity when the liquid bridge is relatively slender.

用去离子水及有机液体在内径约为25μm的石英圆管内进行了流量特性实验.液体分子量范围为18~160,动力黏性系数的范围为0.5~1 mPa.s.实验雷诺数范围为Re<8.所用有机液体为:四氯化碳、乙基苯及环己烷都是非极性液体,其分子结构尺度小于1 nm.实验结果表明,在定常层流条件下,圆管内的液体流量与两端压力差成正比,其压力-流量关系仍符合经典的Hagen-Poiseuille流动.这说明非极性小分子有机液体在本实验所用微米尺度管道中其流动规律仍符合连续介质假设.鉴于微尺度流动实验的特殊性,文中还介绍了微流动实验装置,分析了微尺度流动测量误差来源及提高测量精度的措施.

Microstructures and mechanical properties have been studied in aluminium containing a fine dispersion of alumina particles, deformed by cold-rolling to strains between 1.4 and 3.5. The microstructure was characterised by TEM. The deformation structures evolved very rapidly, forming a nanostructured material, with fine subgrains about 0.2 μm in diameter and a fraction of high-angle boundaries which was already high at a strain of 1.4, but continued to increase with rolling strain. The yield stress and ductility of the rolled materials were measured in tension, and properties were similar for all materials. Yield stress measurements were correlated with estimates made using microstructural models. The role of small particles in forming and stabilising the deformation structure is discussed. This nanostructured cold-deformed alloy has mechanical properties which are usefully enhanced at comparatively low cost. This gives it, and similar particle-strengthened alloys, good potential for commercial exploitation. © 2002 Acta Materialia Inc. Published by Elsevier Science Ltd. All rights reserved.

Squeeze-film effects of perforated plates for small amplitude vibration are analyzed through modified Reynolds equation (MRE). The analytical analysis reckons in most important influential factors: compressibility of the air, border effects, and the resistance caused by vertical air flow passing through perforated holes. It is found that consideration of air compressibility is necessary for high operating frequency and small ratio of the plate width to the attenuation length. The analytical results presented in this paper agree with ANSYS simulation results better than that under the air incompressibility assumption. The analytical analysis can be used to estimate the squeeze-film effects causing damping and stiffness added to the system. Since the value of Reynolds number involved in this paper is low (< 1), inertial effects are neglected.

Resumen: El Modelo de Red Causal propone que la estructura causal de una historia y su representación en la memoria episódica se asemejan a una red, en la que los acontecimientos resultan de una combinación de antecedentes causales, que a su vez tienen múltiples consecuencias. El estudio de la comprensión de textos según este modelo ha tendido a llevarse a cabo utilizando textos experimentales en inglés. En razón de ello, el objetivo de este trabajo consistió en presentar la aplicación del Modelo de Red Causal a un texto narrativo natural en español, a fin de abogar por su utilidad para examinar los procesos cognitivos involucrados en la comprensión textual.

El estudio surge como respuesta a la escasa información sobre abastecimientos de agua cuando se utilizan pozos y equipos de bombeo, además tiene la finalidad de generar los elementos necesarios al parque de ciencias Estelimar e interesa dos en este tipo de investigación para que pueda aprovecharse sosteniblemente el recurso agua, usar eficientemente el equipo de bombeo al mismo tiempo reducir los costos de energía que se generan con el abastecimiento de agua en estas condiciones. Los objetivos de la investigación fueron: caracterizar el acuífero existente, describir las red hidráulica, conocer el punto de operación de la bomba así mismo de terminar los costos de energía generados en el abastecimiento de los deposito demandantes. Para caracterizar el acuífero existente se realizó una prueba de bombeo a caudal constante en el pozo Estelimar que comprendió 7 horas de extracción y 8 de recuperación también se analizó el perfil estratigráfico de un pozo situado a 100m del sitio en estudio. La red hidráulica se describió con el apoyo de un mapa elaborado por técnicos de Estelimar acompañados de observaciones directas en las instalaciones de la red. En el caso del punto de operación de la bomba fue necesario aforar todas las salidas hacia los de pósitos demandantes, mediciones de voltaje e intensidad de corriente y un levantamiento topográfico por taquimetría. Los costo de energía fueron determinados utilizando además de las descarga de agua obtenidas del aforo las capacidades de cada depósito. Finalmente se concluye que el acuífero afectado por el pozo estelimar presenta características de semiconfinado estimándose su transmisividad en 423.36 m2/día, el nivel estático registrado fue de 26.60m produciéndose un rebajamiento de 27.30m, se identificaron tres codos de 90º, seis tee, tres válvulas de compuertas y un codo de 45º, la tubería de succión es de hierro y el de descarga PVC, ambos de 2 pulg de diámetro, el punto en que opera la bomba con un 63% de eficiencia es de 1.03l/s venciendo una carga de trabajo de 45.50m, los costos de energía registrados para los meses de abril,mayo, junio y julio del 2008 están alrededor de C$8233.54 (Còrdobas).

The rationale behind this work is to design an implant device, based on a ferromagnetic material, with the potential to deform in vivo promoting osseointegration through the growth of a healthy periprosthetic bone structure. One of the primary requirements for such a device is that the material should be non-inflammatory and non-cytotoxic. In the study described here, we assessed the short-term cellular response to 444 ferritic stainless steel; a steel, with a very low interstitial content and a small amount of strong carbide-forming elements to enhance intergranular corrosion resistance. Two different human cell types were used: (i) foetal osteoblasts and (ii) monocytes. Austenitic stainless steel 316L, currently utilised in many commercially available implant designs, and tissue culture plastic were used as the control surfaces. Cell viability, proliferation and alkaline phosphatase activity were measured. In addition, cells were stained with alizarin red and fluorescently-labelled phalloidin and examined using light, fluorescence and scanning electron microscopy. Results showed that the osteoblast cells exhibited a very similar degree of attachment, growth and osteogenic differentiation on all surfaces. Measurement of lactate dehydrogenase activity and tumour necrosis factor alpha protein released from human monocytes indicated that 444 stainless steel did not cause cytotoxic effects or any significant inflammatory response. Collectively, the results suggest that 444 ferritic stainless steel has the potential to be used in advanced bone implant designs. © 2011 Elsevier Ltd.

Cell adhesion is crucial to many biological processes, such as inflammatory responses, tumor metastasis and thrombosis formation. Recently a commercial surface plasmon resonance (SPR)-based BIAcore biosensor has been extended to determine cell binding mediated by surface-bound biomolecular interactions. How such cell binding is quantitatively governed by kinetic rates and regulating factors, however, has been poorly understood. Here we developed a novel assay to determine the binding kinetics of surface-bound biomolecular interactions using a commercial BIAcore 3000 biosensor. Human red blood cells (RBCs) presenting blood group B antigen and CM5 chip bearing immobilized anti-B monoclonal antibody (mAb) were used to obtain the time courses of response unit, or sensorgrams, when flowing RBCs over the chip surface. A cellular kinetic model was proposed to correlate the sensorgrams with kinetic rates. Impacts of regulating factors, such as cell concentration, flow duration and rate, antibody-presenting level, as well as pH value and osmotic pressure of suspending medium were tested systematically, which imparted the confidence that the approach can be applied to kinetic measurements of cell adhesion mediated by surface-bound biomolecular interactions. These results provided a new insight into quantifying cell binding using a commercial SPR-based BIAcore biosensor.

Numerical simulations were conducted to study thermocapillary flows in short half-zone liquid bridges of molten tin with Prandtl number Pr = 0.009, under ramped temperature difference. The spatio-temporal structures in the thermocapillary flows in short half-zone liquid bridges with aspect ratios As = 0.6, 0.8, and 1.0 were investigated. The first critical Marangoni numbers were compared with those predicted by linear stability analyses (LSA). The second critical Marangoni numbers for As = 0.6 and 0.8 were found to be larger than that for As = 1.0. The time evolutions of the thermocapillary flows exhibited unusual features such as a change in the azimuthal wave number during the three-dimensional stationary (non-oscillating) flow regime, a change in the oscillation mode during the three-dimensional oscillatory flow regime, and the decreasing and then increasing of amplitudes in a single oscillation mode. The effects of the ramping rate of the temperature difference on the flow modes and critical conditions were studied as well. Resumen: El objetivo de este trabajo es analizar el comportamiento del protocolo OLSR sobre una red mallada configurada con firmware OpenWrt utilizando distintos equipos de hardware. Se presentan los resultados empíricos de varias pruebas utilizando el mismo escenario. El escenario que se presenta es una red de mundo real (no de laboratorio) con pruebas reales y no simulaciones. OpenWrt es un software perfectamente válido que puede ser utilizado en una gran variedad de dispositivos y su configuración para utilizarlo con protocolo OLSR es sencilla de realizar y no presenta problemas de funcionamiento con dicho protocolo.
