The sliding clamp of the Escherichia coli replisome is now understood to interact with many proteins involved in DNA synthesis and repair. A universal interaction motif is proposed to be one mechanism by which those proteins bind the E. coli sliding clamp, a homodimer of the beta subunit, at a single site on the dimer. The numerous beta(2)-binding proteins have various versions of the consensus interaction motif, including a related hexameric sequence. To determine if the variants of the motif could contribute to the competition of the beta-binding proteins for the beta(2) site, synthetic peptides derived from the putative beta(2)-binding motifs were assessed for their abilities to inhibit protein-beta(2) interactions, to bind directly to beta(2), and to inhibit DNA synthesis in vitro. A hierarchy emerged, which was consistent with sequence similarity to the pentameric consensus motif, QL(S/D)LF, and peptides containing proposed hexameric motifs were shown to have activities comparable to those containing the consensus sequence. The hierarchy of peptide binding may be indicative of a competitive hierarchy for the binding of proteins to beta(2) in various stages or circumstances of DNA replication and repair.

1. Protein utilisation and turnover were measured in male chickens sampled from a line selected for high breast yield and a randombred control line (lines QL and CL, experiment 1) and in male chickens sampled from lines selected for either high or low abdominal fatness (lines FL and LL, experiment 2). In each experiment, 18 birds per line were given iso-energetic (12.9 MJ ME/kg) diets containing either 120 or 220 g CP/kg from 21 to 29 d (experiment 1) and 33 to 43 d (experiment 2). 2. Measurements were made of growth rate, food intake, body composition, excreta production and N-tau-methylhistidine excretion as a measure of myofibrillar protein breakdown, and fractional rates (%/d) of protein deposition, breakdown and synthesis were calculated. 3. In experiment 1, there were no significant differences between the line means for the fractional measures of protein turnover, but there was marked differential response in the two lines in the fractional rates of protein deposition, breakdown and synthesis, to increase in protein intake. The positive slope of the regressions of fractional (%/d) protein deposition and synthesis rates on protein intake (g/d/kg BW) were approximately 1.4- and 2.0-fold higher respectively in the QL than the CL line birds, and the negative slope of the regression of fractional breakdown rate on protein intake was approximately threefold greater in the CL than the QL line birds. 4. In experiment 2, fractional deposition rate was 6.2% lower, but fractional breakdown rate 9.4% higher in the LL than the FL birds, whilst there was essentially no difference in response of the FL and LL birds in the components of protein turnover to increase in protein intake. Line differences in deposition and breakdown rates were thus a reflection of the considerably higher (20%) food and hence protein intake in the FL than the LL birds. 5. The differential line responses in protein turnover in the two experiments suggest that selection for increased breast muscle yield and for reduced body fatness manipulate different physiological pathways in relation to protein turnover, but neither selection strategy results in an improvement in net protein utilisation at typical levels of protein intake by birds on commercial broiler diets, through a reduction in protein breakdown rate.

As a result of their relative concentration towards the respective Atlantic margins, the silicic eruptives of the Parana (Brazil)-Etendeka large igneous province are disproportionately abundant in the Etendeka of Namibia. The NW Etendeka silicic units, dated at similar to132 Ma, occupy the upper stratigraphic levels of the volcanic sequences, restricted to the coastal zone, and comprise three latites and five quartz latites (QL). The large-volume Fria QL is the only low-Ti type. Its trace element and isotopic signatures indicate massive crustal input. The remaining NW Etendeka silicic units are enigmatic high-Ti types, geochemically different from low-Ti types. They exhibit chemical affinities with the temporally overlapping Khumib high-Ti basalt (see Ewart et al. Part 1) and high crystallization temperatures (greater than or equal to980 to 1120degreesC) inferred from augite and pigeonite phenocrysts, both consistent with their evolution from a mafic source. Geochemically, the high-Ti units define three groups, thought genetically related. We test whether these represent independent liquid lines of descent from a common high-Ti mafic parent. Although the recognition of latites reduces the apparent silica gap, difficulty is encountered in fractional crystallization models by the large volumes of two QL units. Numerical modelling does, however, support large-scale open-system fractional crystallization, assimilation of silicic to basaltic materials, and magma mixing, but cannot entirely exclude partial melting processes within the temporally active extensional environment. The fractional crystallization and mixing signatures add to the complexity of these enigmatic and controversial silicic magmas. The existence, however, of temporally and spatially overlapping high-Ti basalts is, in our view, not coincidental and the high-Ti character of the silicic magmas ultimately reflects a mantle signature.

A obesidade é uma doença crônica que vem acometendo, progressivamente, cada vez mais pessoas no mundo. Por ser uma patologia de difícil controle que favorece a eclosão de outros agravos à saúde, é premente a necessidade de realização de pesquisas que possam contribuir para o aperfeiçoamento dos tratamentos, bem como para a melhoria da qualidade de vida e eficácia adaptativa. Sendo assim, a presente pesquisa visou avaliar a percepção da qualidade de vida (QV), a eficácia adaptativa (EDAO) e o funcionamento global (AGF) de pessoas com obesidade, relacionando os resultados obtidos na avaliação da percepção da QV com os da eficácia adaptativa, bem como aos do funcionamento global (AGF). Para tanto, utilizou-se o questionário WHOQOL-100 versão em português para avaliação da percepção de qualidade de vida, a Entrevista Diagnóstica Preventiva Escala Diagnóstica Adaptativa Operacionalizada (EDAO) para a eficácia adaptativa e a Escala de Avaliação do Funcionamento Global (AGF) para o funcionamento global. Este estudo contou com a participação de trinta mulheres obesas (Índice de Massa Corporal IMC >=30 kg/m2), com idade média de 52,33 anos, que utilizavam os serviços de um ambulatório situado na região do Grande ABC, estado de SP. A maioria das participantes se encontrava no grau I de obesidade - 46,70%, situava-se no grau II 33,30% e 20,00% no grau III. O aumento de peso da maioria teve início nas gestações (43,30%), o segundo período onde ocorreu o início do descontrole do peso corporal foi entre os 40 aos 50 anos (20,00%). Na avaliação geral da QV, observou-se que no domínio VI Aspectos Espirituais/Religião/Crenças Pessoais foi encontrado o maior escore médio (16,17 - DP=2,95 [equivalente a 80,83% do escore máximo que poderia ser obtido]), comparando-o com os demais domínios avaliados. Em oposição, o domínio I Físico foi o que apresentou o menor escore médio (11,77 - DP=2,78 - 58,83%). Todas as participantes se encontravam em ineficácia adaptativa: Grupo 2 Ineficaz Leve (26,7%), no Grupo 3 Ineficaz Moderada (33,3%) e no Grupo 4 Ineficaz Severa (30,0%). Quanto à avaliação do funcionamento global (AGF), notou-se que 36,67% se situavam no intervalo entre 51-60 pontos. 23,33% das participantes no intervalo entre 31-50 pontos. Apenas 23,33% tiveram pontuação acima de 70 pontos. Relacionando os resultados das avaliações, foram encontradas correlações fortes, positivas e significativas entre a avaliação da percepção de qualidade de vida, da eficácia adaptativa e do funcionamento Global.

Background Food allergy has been shown to severely affect quality of life (QoL) in children and their families. The Anaphylaxis Campaign UK supports families with allergic children and as part of that support ran an activity holiday for those with food allergy. This study investigated the effectiveness of this activity holiday for reducing anxiety and improving QoL and food allergy management for these children. Methods Measures were taken at baseline, at the start of the activity holiday, at the end of the holiday, at 3 and 6 months follow-up. Children (n = 24) completed a paediatric food allergy–specific QoL questionnaire (PFA-QL), a generic QoL questionnaire (PedsQL™), the Spence Children's Anxiety Scale (SCAS) and the Children's Health Locus of Control (CHLC) scale at all stages of the study. Results There were significant improvements in social QoL, food allergy–specific QoL, total CHLC and internal locus of control scores over time (p > 0.05). There were significant decreases in powerful others locus of control, total anxiety and obsessive compulsive disorder scores (p < 0.05). Greater anxiety significantly correlated with poorer QoL at all time points; no correlations with locus of control were significant at the 3- and 6-month follow-up. Conclusions The activity holiday was of significant benefit to the children who took part, providing support for the need for activity holidays such as this for children with severe food allergy. Ways in which adaptive locus of control and improved quality of life can be facilitated need to be further explored.

OBJECTIVES: Exposure to active mobile phones (MP) has been shown to affect human neural function as shown by the electroencephalogram (EEG). Although it has not been determined whether such effects are harmful, a number of devices have been developed that attempt to minimize these MP-related effects. One such device, the Q Link Ally® (QL; Clarus Products, International, L.L.C., San Rafael, CA), is argued to affect the human organism in such a way as to attenuate the effect of MPs. The present pilot study was designed to determine whether there is any indication that QL does alter MP-related effects on the human EEG. DESIGN: Twenty-four (24) subjects participated in a single-blind, fully counterbalanced crossover design in which subjects' resting EEG and phase-locked neural responses to auditory stimuli were assessed under conditions of either active MP or active MP plus QL. RESULTS: The addition of QL to the MP condition increased resting EEG in the gamma range and did so as a function of exposure duration, and it attenuated MP-related effects in the delta and alpha range (at trend-level). The addition of the QL also affected phase-locked neural responses, with a laterality reversal in the alpha range and an alteration to changes over time in the delta range, a reduction of the MP-related beta decrease over time at fronto-posterior sites, and a global reduction in the gamma range that increased as a function of exposure duration. No unambiguous relations were found between these changes and either performance or psychologic state. CONCLUSIONS: This pilot study suggests that the addition of the QL to active MP-exposure does affect neural function in humans, altering both resting EEG patterns and the evoked neural response to auditory stimuli, and that there is a tendency for some MP-related changes to the EEG to be attenuated by the QL.

During the oil refining process a huge discard volume of water occurs, which carries the contaminants from the process. A class of contaminant compounds resulting from the petrochemical industry are the Polyaromatic Hydrocarbons (PAH's). To evaluate the biodegradation of Dibenzothiophene in refinery water a synthetic wastewater was prepared to be treated using activated sludge. For this, a 2 3 Composite Design (plus 3 central points and six axial points) was carried out. The planning had as independent variables (factors) the initial concentration of DBT, pH and time of biodegradation. Biodegradation of DBT was assayed following the parameters COD, pH, temperature, SS, VSS, FVS, SVI. Concerned to the chromatographic conditions, a methodology was validated in order to verify the presence of DBT and its metabolite, 2-HBF, in the final wastewater treated by activated sludge system using a liquid - liquid extraction coupled to HPLC / UV analysis. The parameters used for validation were DL, QL, linearity, recovery and repeatability. As for optimization, the results indicated that the studied methodology can be used in monitoring the DBT degradation and 2- HBF by activated sludge, as they showed excellent linearity values, coefficients of variation, so as satisfactory recovery percentage. COD reduction efficiency tests showed an average percentage of 64.4%. The increasing trend for the results for the TSS and VSS tests showed that the activated sludge was well tailored. The best operating conditions for the reduction of COD were observed when operated with median concentrations of DBT, a higher time to biodegradation, and pH in both the acidic range as the basic one. The biodegradability of the DBT was confirmed by determining the presence of HBF-2. The highest concentrations of HBF-2 were obtained in extreme concentrations of DBT and pH, and higher biodegradation times.

Based on the quantitative study of diatoms and radiolarians, summer sea-surface temperature (SSST) and sea ice distribution were estimated from 122 sediment core localities in the Atlantic, Indian and Pacific sectors of the Southern Ocean to reconstruct the last glacial environment at the EPILOG (19.5-16.0 ka or 23 000-19 000 cal yr. B.P.) time-slice. The statistical methods applied include the Imbrie and Kipp Method, the Modern Analog Technique and the General Additive Model. Summer SSTs reveal greater surface-water cooling than reconstructed by CLIMAP (Geol. Soc. Am. Map Chart. Ser. MC-36 (1981) 1), reaching a maximum (4-5 °C) in the present Subantarctic Zone of the Atlantic and Indian sector. The reconstruction of maximum winter sea ice (WSI) extent is in accordance with CLIMAP, showing an expansion of the WSI field by around 100% compared to the present. Although only limited information is available, the data clearly show that CLIMAP strongly overestimated the glacial summer sea ice extent. As a result of the northward expansion of Antarctic cold waters by 5-10° in latitude and a relatively small displacement of the Subtropical Front, thermal gradients were steepened during the last glacial in the northern zone of the Southern Ocean. Such reconstruction may, however, be inapposite for the Pacific sector. The few data available indicate reduced cooling in the southern Pacific and give suggestion for a non-uniform cooling of the glacial Southern Ocean.

La tesis El animal como mediador en los discursos escultóricos actuales, aúna la investigación sobre los procesos de creación en torno a la animalística durante los siglos XX y XXI (documental, gráfica, histórica y conceptual) con el desarrollo de la creación artística personal como sustrato. Se trata de una investigación marcadamente transversal, que recurre a disciplinas tales como: lenguajes artísticos (en especial la escultura), biología, antropología, filosofía, crítica artística, historia del arte, zoología y ética. Todas estas ramas resultan necesarias para abordar un tema de tanta complejidad como lo es la diversidad de discursos que el arte actual establece en relación al tema. La tesis busca como objetivo establecer un marco desde el que discutir, analizar y proponer el quehacer artístico personal, dialogado y contextualizado, con el arte animalista del momento. Tras un profundo estudio del panorama, que se encuentra muy escasamente desarrollado en España, se establecen siete territorios discursivos (incluido el propio personal) cuyo denominador común es recurrir al animal; en unos casos como instrumento para transmitir el discurso de los artistas y, en otros, como eje central de las obras. En ellos se otorga especial interés a aspectos como: la metodología chamánica, empleada por los actuales artistas con un afán sanador y una preocupación por la relación arte/vida; las consideraciones relativas al símbolo y la transcendencia; la mediación del animal para los discursos políticos sociales; el valor terapéutico, incluyendo en ellos los procesos catárticos; y la metáfora que suponen para los alegatos ecológicos. Todo esto sin olvidar la enorme sugerencia formal que los animales aportan como objetos estéticos, y como sugerencias para la creación de mundos fantásticos. Las inquietantes e interesantes propuestas que se han analizado determinan, en último extremo, el posicionamiento de la autora de la tesis dentro de un discurso a favor del animal, su dignidad, su valor sanador y su poder como aliado. Tras el estudio de 94 artistas reconocidos en este campo, se alcanza a ofrecer un horizonte clarificado de los mecanismos que operan en la animalística, dividiendo su estudio en los siguientes 7 apartados: En un primer capítulo, dedicado al símbolo de la trascendencia, se analiza la reutilización del símbolo, lo que conduce a determinar el comportamiento simbólico y el sentimiento poético; se recorren los diferentes procedimientos de anamnesis y viajes de iniciación propios de las tradiciones religiosas; y se observa el papel que juega el animal como espíritu auxiliar, recorriendo dentro de la escultura un camino hacia la abstracción y por tanto hacia la captación de la esencia. En estos desarrollos la materia es utilizada como vehículo de lo sagrado, de tal modo que la creación artística aspira a la inmortalidad y a la trascendencia del ser...
