El litoral ha constituido a lo largo de la historia una importante fuente de recursos económicos además de un punto estratégico para la defensa del territorio. El comercio, la pesca o la industria de la sal han propiciado la formación de poblaciones y ciudades al borde del mar. La costa funcionó como un ámbito defensivo, peligroso e insalubre durante siglos donde el mar modulaba el frente costero sin grandes interacciones con un entorno prácticamente deshabitado. A mediados del siglo XIX, las innovaciones técnicas y la pacificación definitiva del Mediterráneo permiten poner en valor sus características naturales y de oportunidad. Sin perder del todo su valor defensivo, el litoral resurge como recurso productivo y como lugar de ocio y disfrute de la población. El valor estratégico de la costa quedará también reflejado en el marco normativo. El derecho civil moderno recuperará el concepto de Dominio público Marítimo Terrestre a través de la Ley de Aguas de 1866 que regulará también los usos y las limitaciones en la propiedad privada litoral. Desde ese momento, las transformaciones económicas, sociales, jurídicas y ambientales van a provocar un cambio significativo en la relación entre la sociedad y la costa que dará paso a la construcción del espacio litoral que hemos heredado. Desde la triple perspectiva del litoral como sistema físico ambiental, sistema económico, productivo y cultural, y sistema administrativo y legal, el principal objetivo de la tesis será reconstruir el proceso de transformación del litoral consecuencia de la evolución en la forma de producción del espacio, de área defensiva a recurso productivo, y desde mediados del siglo XIX hasta principios del siglo XXI. Sin embargo, la construcción del litoral no ha sido homogénea ni constante a lo largo del tiempo. Ha estado sujeta a los distintos vaivenes económicos y sociales pero también a los cambios en el modelo territorial definido por el marco legal vigente, así como a los reajustes del propio sistema físico. Como instrumento sociopolítico, el marco legal regula las presiones del sistema económico sobre el medio, apostando por una visión frente a otra, y posibilitando el grado de transformación final. Así, el conocimiento sobre el territorio en el que se interviene y la definición del modelo de litoral por parte de los poderes públicos irán configurando el espacio físico, económico y social desarrollado en la costa. Para la reconstrucción del proceso de construcción del litoral, la tesis define cuatro fases diferentes y aplica las hipótesis y la metodología a la costa de Cartagena. Se presenta gráficamente la evolución en los tres sistemas en cada una de las fases, y se recompone el relato histórico a través de los hitos más relevantes para el proceso. En cada una de las fases, el nuevo modelo intentará dar respuesta a todo aquello que no funcionó o se quedó a medias en el periodo anterior. Las crisis económicas provocan la ralentización de la actividad productiva y, consecuentemente, de las transformaciones en el territorio. Servirán para establecer, en principio, un cambio de paradigma en la lectura y gestión del litoral que acabará traduciéndose en un nuevo texto legislativo en materia de costas (1969, 1988 y 2013). La reforma de la normativa responde a una nueva forma de entender, ordenar, gestionar e intervenir en el territorio, donde se modulan las pautas pero también la intensidad en la intervención. Pero nace condicionada por el litoral heredado: los derechos generados durante la vigencia del marco legal anterior; el modelo económico y sus presiones y expectativas sobre el litoral; y el medio físico en el que se acumulan los procesos de degradación no resueltos y los nuevos riesgos. Así, las conclusiones de la tesis ponen de manifiesto la necesidad de una visión compleja e integral sobre el litoral, en la que el urbanismo y la ordenación del territorio serán fundamentales para afrontar los nuevos retos en su construcción a futuro. ABSTRACT The littoral has been throughout history an important source of economic resources and a strategic point for the defense of territory. Trade, fishing or salt industry have led to the formation of towns and cities on the edge of the sea. The coast served as a defensive, dangerous and unhealthy place where the sea modulated for centuries the waterfront without major interactions with a virtually uninhabited environment. In the mid-nineteenth century, technical innovations and the final pacification of the Mediterranean allowed to value its natural features and opportunities. Without entirely losing its defensive value, the coast emerges as a productive resource and as a place of leisure and enjoyment of people. The strategic value of the coast will also be reflected in the legal framework. The concept of maritime-terrestrial public domain will be recovered by the modern civil law and the law of waters of 1866 governs the uses and limitations of private ownership of the coast. Since then, the economic, social, legal and environmental changes will to cause a significant change in the relationship between society and the coast that will give way to the construction of littoral space inherited. From the triple perspective of the littoral as a physical environmental system, an economic, productive and cultural system, and an administrative and legal system, the main objective of the thesis is to rebuild its process of transformation, as a result of the evolution in the way that space is produced, from defensive zone to productive resource, and from the middle of the nineteenth century until the beginning of the twenty-first century. However, the construction of the coast has not been uniform nor constant over time. It has been subject to different economic and social fluctuations and also to changes in the territorial model defined by the legal framework in force, as well as to readjustments of the physical system itself. As socio-political instrument, the legal framework regulates the pressure of the economic system on the environment, it bets on a vision over another and facilitates the final degree of transformation. Thus, the knowledge on the territory that is being intervened and the definition of the model of shoreline by public authorities will configure the physical, economic and social space developed on the coast. For the reconstruction of the littoral building process, the thesis defines four different phases and applies the hypothesis and methodology to the coast of Cartagena. It introduces graphically the evolution of the three systems in each of the phases and it recomposes the historical account through of the most important milestones for the process. In each phase, the new model will attempt to answer everything that did not work or was half in the previous period. Economic crises cause a slowdown in productive activity and, consequently, in the changes of territory. They serve to establish, in principle, a paradigm shift for reading and managing the littoral, eventually resulting in new legal texts on coasts matter (1969, 1988 and 2013). The reform of legislation responds to a new way for understanding, arranging, managing and intervening on the territory, where the guidelines and also the intervention intensity are modulated. But it is born conditioned by the inherited coast: the rights generated under previous legal framework; the economic model and its pressures and expectations on the littoral; and the physical environment which accumulates degradation processes unresolved and new risks. Thus, the thesis conclusions highlight the need for a complex and comprehensive view on the littoral, where urban planning and land-use planning will be key to meet the future challenges in its construction.

The spatial complexity of the distribution of organic matter, chemicals, nutrients, pollutants has been demonstrated to have multifractal nature (Kravchenco et al. [1]). This fact supports the possibility of existence of some emergent heterogeneity structure built under the evolution of the system. The aim of this note is providing a consistent explanation to the mentioned results via an extremely simple model.

As condições inadequadas vivenciadas nas organizações afligem não só os trabalhadores da iniciativa privada, pois são igualmente encontradas no segmento estatal, contrariando a expectativa de que o aparato governamental eliminaria as condições insalubres e criaria outras melhores nas quais prevalecesse à promoção de saúde. Diante desse panorama questionou-se porque, uma vez que, pelo menos do ponto de vista da sociedade leiga, esses servidores estão submetidos a condições privilegiadas de trabalho. O presente estudo objetivou identificar e descrever possíveis relações entre o clima organizacional e o burnout em servidores públicos de uma instituição federal de ensino. Objetivou-se ainda descrever o clima organizacional predominante. A pesquisa realizada teve cunho quantitativo, tipo estudo de caso e exploratória. A coleta de dados deu-se por meio das escalas ECO (escala de clima organizacional), ECB (escala de caracterização do burnout) e um questionário sociodemográfico, todos os instrumentos autoaplicáveis eletronicamente disponíveis à instituição. Participaram do estudo 201 servidores públicos federais, com idade média de 37 anos, majoritariamente de nível superior e casados. Os resultados revelaram que cerca de um quarto dos participantes raramente experimentaram burnout, no entanto outra quarta parte deles frequentemente experimentaram altos níveis de burnout, resultado bastante expressivo. Os servidores perceberam clima organizacional mediano, destacando-se a boa coesão entre os colegas de trabalho e a percepção de baixa recompensa. Merece destaque a grande dispersão entre as percepções de clima, o que permite inferir haver subclimas não identificados nesta investigação, possivelmente ocasionados por uma força de clima fraca e pela participação dos servidores de unidades de ensino geograficamente distintas, geridas por gestores locais com relativa autonomia. Os resultados dos cálculos de correlação revelaram que, quanto menos os participantes percebem apoio da chefia e da organização, coesão entre colegas, e mais controle/pressão, mais exaustos se sentem, mais desumanizam as pessoas com quem tratam e mais se decepcionam no trabalho e vice-versa. Conforto físico menor está associado a maior desumanização e a mais decepção no trabalho e vice-versa; e que controle/pressão, relaciona-se positiva e fracamente com desumanização e vice-versa. Desta forma, a hipótese de que existe associação entre burnout e clima organizacional foi confirmada. Os resultados também revelaram que os servidores com burnout, perceberam pior clima organizacional que os seus pares sem burnout, confirmando a segunda hipótese. Esses servidores também se mostraram neutros quanto à percepção de apoio da chefia e conforto físico; não percebem controle pressão, nem recompensa; todavia percebem coesão entre os colegas. Esses resultados sugerem que os participantes têm se apoiado nessas relações para suportar a indiferença e ausência de estímulos experimentados no trabalho. Os resultados obtidos nesse estudo permitiram concluir que o clima organizacional é fraco, provavelmente influenciado por uma cultura organizacional fraca, explicando a heterogeneidade da percepção do clima organizacional pelos servidores. Além disso, embora haja burnout entre poucos participantes, há que se atentar que cerca de um quarto deles, encontra-se acometido desta síndrome e isto poderá contagiar os demais.

Correlates of virus load and characteristics of virus-producing cells in tonsillar tissue were investigated. Our results suggest that when less than 1:100 tonsillar CD4+ T cells from individuals infected with HIV type-1 (HIV-1) contain replication competent provirus, the level of CD4+ T cells in tonsils is comparable to that observed in uninfected individuals. Virus load at or above this level was associated with low CD4 cell numbers in tonsillar tissue. Only a few percent of all infected T cells in tonsillar tissue were active virus producers, with minor differences observed between individuals. Plasma viremia was found to correlate with infectious virus load in tonsillar tissue. With less than 1:1,000 of CD4 cells in lymphoid tissues being involved in active virus production, direct cytopathic effect by HIV-1 on infected CD4 cells is unlikely to fully explain the immunodeficiency seen in AIDS.

A filamentary model of “metallic” conduction in layered high temperature superconductive cuprates explains the concurrence of normal state resistivities (Hall mobilities) linear in T (T−2) with optimized superconductivity. The model predicts the lowest temperature T0 for which linearity holds and it also predicts the maximum superconductive transition temperature Tc. The theory abandons the effective medium approximation that includes Fermi liquid as well as all other nonpercolative models in favor of countable smart basis states.

This paper describes the NMR observation of 15N—15N and 1H—15N scalar couplings across the hydrogen bonds in Watson–Crick base pairs in a DNA duplex, hJNN and hJHN. These couplings represent new parameters of interest for both structural studies of DNA and theoretical investigations into the nature of the hydrogen bonds. Two dimensional [15N,1H]-transverse relaxation-optimized spectroscopy (TROSY) with a 15N-labeled 14-mer DNA duplex was used to measure hJNN, which is in the range 6–7 Hz, and the two-dimensional hJNN-correlation-[15N,1H]-TROSY experiment was used to correlate the chemical shifts of pairs of hydrogen bond-related 15N spins and to observe, for the first time, hJHN scalar couplings, with values in the range 2–3.6 Hz. TROSY-based studies of scalar couplings across hydrogen bonds should be applicable for large molecular sizes, including protein-bound nucleic acids.

A method for the study of the control of the attainment of thermal acclimation has been applied to the crabs, Cancer pagurus and Carcinus maenas. Crabs were heterothermally acclimated by using an anterior–posterior partition between two compartments, one at 8°C and the other at 22°C. One compartment held a three-quarter section of the crab including the central nervous system (CNS), eye stalks, and ipsilateral legs; the other held a quarter section including the contralateral legs. Criteria used to assess the acclimation responses were comparisons of muscle plasma membrane fatty acid composition and “fluidity.” In both species, the major fatty acids of phosphatidylcholine were 16:0, 18:1, 20:5, and 22:6, whereas phosphatidylethanolamine contained significantly less 16:0 but more 18:0; these fatty acids comprised 80% of the total. Differences in fatty acid composition were demonstrated between fractions obtained from the ipsilateral and contralateral legs from the same heterothermally acclimated individual. In all acclimation states (except 22CNS, phosphatidylcholine fraction), membrane lipid saturation was significantly increased with acclimation at 22° as compared with 8°C. Membrane fluidity was determined by using 1,3-diphenyl-1,3,5 hexatriene (DPH) fluorescence polarization. In both species, membranes from legs held at 8° were more fluid than from legs held at 22°C irrespective of the acclimation temperature of the CNS. Heterothermal acclimation demonstrated that leg muscle membrane composition and fluidity respond primarily to local temperature and were not predominately under central direction. The responses between 8°C- and 22°C-acclimated legs were more pronounced when the CNS was cold-acclimated, so a central influence cannot be excluded.

A mechanism of ion transport across membranes is reported. Microbial transport of Fe3+ generally delivers iron, a growth-limiting nutrient, to cells via highly specific siderophore-mediated transport systems. In contrast, iron transport in the fresh water bacterium Aeromonas hydrophila is found to occur by means of an indiscriminant siderophore transport system composed of a single multifunctional receptor. It is shown that (i) the siderophore and Fe3+ enter the bacterium together, (ii) a ligand exchange step occurs in the course of the transport, and (iii) a redox process is not involved in iron exchange. To the best of our knowledge, there have been no other reports of a ligand exchange mechanism in bacterial iron transport. The ligand exchange step occurs at the cell surface and involves the exchange of iron from a ferric siderophore to an iron-free siderophore already bound to the receptor. This ligand exchange mechanism is also found in Escherichia coli and seems likely to be widely distributed among microorganisms.

The structure of the catalytically inactive mutant (C215S) of the human protein-tyrosine phosphatase 1B (PTP1B) has been solved to high resolution in two complexes. In the first, crystals were grown in the presence of bis-(para-phosphophenyl) methane (BPPM), a synthetic high-affinity low-molecular weight nonpeptidic substrate (Km = 16 μM), and the structure was refined to an R-factor of 18.2% at 1.9 Å resolution. In the second, crystals were grown in a saturating concentration of phosphotyrosine (pTyr), and the structure was refined to an R-factor of 18.1% at 1.85 Å. Difference Fourier maps showed that BPPM binds PTP1B in two mutually exclusive modes, one in which it occupies the canonical pTyr-binding site (the active site), and another in which a phosphophenyl moiety interacts with a set of residues not previously observed to bind aryl phosphates. The identification of a second pTyr molecule at the same site in the PTP1B/C215S–pTyr complex confirms that these residues constitute a low-affinity noncatalytic aryl phosphate-binding site. Identification of a second aryl phosphate binding site adjacent to the active site provides a paradigm for the design of tight-binding, highly specific PTP1B inhibitors that can span both the active site and the adjacent noncatalytic site. This design can be achieved by tethering together two small ligands that are individually targeted to the active site and the proximal noncatalytic site.

The 2H,13C,15N-labeled, 148-residue integral membrane protein OmpX from Escherichia coli was reconstituted with dihexanoyl phosphatidylcholine (DHPC) in mixed micelles of molecular mass of about 60 kDa. Transverse relaxation-optimized spectroscopy (TROSY)-type triple resonance NMR experiments and TROSY-type nuclear Overhauser enhancement spectra were recorded in 2 mM aqueous solutions of these mixed micelles at pH 6.8 and 30°C. Complete sequence-specific NMR assignments for the polypeptide backbone thus have been obtained. The 13C chemical shifts and the nuclear Overhauser effect data then resulted in the identification of the regular secondary structure elements of OmpX/DHPC in solution and in the collection of an input of conformational constraints for the computation of the global fold of the protein. The same type of polypeptide backbone fold is observed in the presently determined solution structure and the previously reported crystal structure of OmpX determined in the presence of the detergent n-octyltetraoxyethylene. Further structure refinement will have to rely on the additional resonance assignment of partially or fully protonated amino acid side chains, but the present data already demonstrate that relaxation-optimized NMR techniques open novel avenues for studies of structure and function of integral membrane proteins.

The feasibility of using carbohydrate-based vaccines for the immunotherapy of cancer is being actively explored at the present time. Although a number of clinical trials have already been conducted with glycoconjugate vaccines, the optimal design and composition of the vaccines has yet to be determined. Among the candidate antigens being examined is Lewisy (Ley), a blood group-related antigen that is overexpressed on the majority of human carcinomas. Using Ley as a model for specificity, we have examined the role of epitope clustering, carrier structure, and adjuvant on the immunogenicity of Ley conjugates in mice. A glycolipopeptide containing a cluster of three contiguous Ley-serine epitopes and the Pam3Cys immunostimulating moiety was found to be superior to a similar construct containing only one Ley-serine epitope in eliciting antitumor cell antibodies. Because only IgM antibodies were produced by this vaccine, the effect on immunogenicity of coupling the glycopeptide to keyhole limpet hemocyanin was examined; although both IgM and IgG antibodies were formed, the antibodies reacted only with the immunizing structure. Reexamination of the clustered Ley-serine Pam3Cys conjugate with the adjuvant QS-21 resulted in the identification of both IgG and IgM antibodies reacting with tumor cells, thus demonstrating the feasibility of an entirely synthetic carbohydrate-based anticancer vaccine in an animal model.
