Objective: In Southern European countries up to one-third of the patients with hereditary hemochromatosis (HH) do not present the common HFE risk genotype. In order to investigate the molecular basis of these cases we have designed a gene panel for rapid and simultaneous analysis of 6 HH-related genes (HFE, TFR2, HJV, HAMP, SLC40A1 and FTL) by next-generation sequencing (NGS). Materials and Methods: Eighty-eight iron overload Portuguese patients, negative for the common HFE mutations, were analysed. A TruSeq Custom Amplicon kit (TSCA, by Illumina) was designed in order to generate 97 amplicons covering exons, intron/exon junctions and UTRs of the mentioned genes with a cumulative target sequence of 12115bp. Amplicons were sequenced in the MiSeq instrument (IIlumina) using 250bp paired-end reads. Sequences were aligned against human genome reference hg19 using alignment and variant caller algorithms in the MiSeq reporter software. Novel variants were validated by Sanger sequencing and their pathogenic significance were assessed by in silico studies. Results: We found a total of 55 different genetic variants. These include novel pathogenic missense and splicing variants (in HFE and TFR2), a very rare variant in IRE of FTL, a variant that originates a novel translation initiation codon in the HAMP gene, among others. Conclusion: The merging of TSCA methodology and NGS technology appears to be an appropriate tool for simultaneous and fast analysis of HH-related genes in a large number of samples. However, establishing the clinical relevance of NGS-detected variants for HH development remains a hard-working task, requiring further functional studies.

In the 2000’s, the Internet became the preferred mean for the citizens to communicate. The YouTube, Twitter, Facebook, LinkedIn, i.e., the social networks in general appeared together with the Web 2.0, which allows an extraordinary interaction between citizens and the democratic institutions. The trade unions constantly fight governments’ decisions, especially in periods of crisis like the one that the world, Europe and, in particular, Portugal are facing. In this regard, the use of e-participation platforms is expected to strengthen the relationship between trade unions and the education community. This paper reports the research about the planning and driving of a series of experiments of online public consultation, launched by teachers’ trade unions. These experiments are compared with those of other countries, such as Australia, United Kingdom and United States of America. A quantitative analysis of the results regarding hits, subscriptions, and response rates is presented, and it is compared with the 90-9-1 rule, the ASCU model and data from government agencies. The experiments performed used the Liberopinion, an online platform that supports bidirectional asynchronous communication. A better understanding of the benefits of these collaborative environments is expected by promoting quality of interaction between actors.

The insoluble residues of samples from ODP Sites 626 and 627 can be subdivided into four groups: (1) illite, 7 A minerals, quartz and feldspar; (2) smectite and zeolite (clinoptilolite); (3) palygorskite and in places sepiolite; and (4) glauconite and pyrite. Whereas group 1 is clearly terrigenous and group 4 authigenic, group 2 is most probably authigenic, as indicated by its abundance in samples with small insoluble residues and its appearance in SEM photographs. Group 3 is authigenic in Albian peritidal dolomite and possibly terrigenous in middle Miocene slumps and debris flows. Smectite crystallinity increases with age. This increase, however, is less pronounced in the Bahamian carbonate-rich samples than in the carbonate-poor silts south of Guatemala (DSDP Leg 84, Sites 569 and 570, the only comparable investigation) as far as can be judged from such a small number of samples.

Woodlark Basin, an area of continental extension, is an ideal location to study the evolution of permeability and the development of overpressures within an active rift basin. In this investigation, we measured sediment permeabilities of cores from Woodlark Basin and used numerical modeling to determine if pore fluid overpressures are likely at the base of the rift basin. Constant-rate flow tests were conducted on cores from Site 1108, located in the rift basin, and Sites 1115 and 1118, located on the northern margin of the basin. Results of the laboratory tests indicated permeabilities that range from 1.5 x 10**-18 to 1 x 10**-16 m**2. Results of the numerical modeling of Site 1108 suggest that overpressures due to sedimentation are unlikely.

The development of the ecosystem approach and models for the management of ocean marine resources requires easy access to standard validated datasets of historical catch data for the main exploited species. They are used to measure the impact of biomass removal by fisheries and to evaluate the models skills, while the use of standard dataset facilitates models inter-comparison. North Atlantic albacore tuna is exploited all year round by longline and in summer and autumn by surface fisheries and fishery statistics compiled by the International Commission for the Conservation of Atlantic Tunas (ICCAT). Catch and effort with geographical coordinates at monthly spatial resolution of 1° or 5° squares were extracted for this species with a careful definition of fisheries and data screening. In total, thirteen fisheries were defined for the period 1956-2010, with fishing gears longline, troll, mid-water trawl and bait fishing. However, the spatialized catch effort data available in ICCAT database represent a fraction of the entire total catch. Length frequencies of catch were also extracted according to the definition of fisheries above for the period 1956-2010 with a quarterly temporal resolution and spatial resolutions varying from 1°x 1° to 10°x 20°. The resolution used to measure the fish also varies with size-bins of 1, 2 or 5 cm (Fork Length). The screening of data allowed detecting inconsistencies with a relatively large number of samples larger than 150 cm while all studies on the growth of albacore suggest that fish rarely grow up over 130 cm. Therefore, a threshold value of 130 cm has been arbitrarily fixed and all length frequency data above this value removed from the original data set.
