Os objetivos deste trabalho foram definir as faixas críticas de concentração de nutrientes e avaliar o estado nutricional das lavouras cafeeiras de alta, média e baixa produtividade em quatro regiões do Estado de Minas Gerais. Foram coletadas folhas em 312 unidades amostrais de 168 lavouras, tanto no ano de alta produtividade quanto no de baixa produtividade, conforme o ciclo bienal do cafeeiro. Nas folhas foram determinadas as concentrações de macronutrientes e dos micronutrientes Cu, Fe, Zn, Mn e boro. Com os resultados das análises foliares, estabeleceram-se as faixas críticas das concentrações dos nutrientes no ano de baixa e alta produtividade, em cada região estudada, utilizando como referência lavouras de alta produtividade (mais de 1.800 kg/ha de café beneficiado na média do biênio). Realizou-se ainda o diagnóstico do estado nutricional das lavouras nas regiões em estudo determinando-se as freqüências de nutrientes em níveis deficiente, adequado e excessivo. As faixas críticas não diferiram entre os anos amostrados, tampouco entre as regiões em estudo. Nas regiões amostradas predominam problemas com micronutrientes.

Background: Various patterns of HIV-1 disease progression are described in clinical practice and in research. There is a need to assess the specificity of commonly used definitions of long term non-progressor (LTNP) elite controllers (LTNP-EC), viremic controllers (LTNP-VC), and viremic non controllers (LTNP-NC), as well as of chronic progressors (P) and rapid progressors (RP). Methodology and Principal Findings: We re-evaluated the HIV-1 clinical definitions, summarized in Table 1, using the information provided by a selected number of host genetic markers and viral factors. There is a continuous decrease of protective factors and an accumulation of risk factors from LTNP-EC to RP. Statistical differences in frequency of protective HLA-B alleles (p-0.01), HLA-C rs9264942 (p-0.06), and protective CCR5/CCR2 haplotypes (p-0.02) across groups, and the presence of viruses with an ancestral genotype in the "viral dating" (i.e., nucleotide sequences with low viral divergence from the most recent common ancestor) support the differences among principal clinical groups of HIV-1 infected individuals. Conclusions: A combination of host genetic and viral factors supports current clinical definitions that discriminate among patterns of HIV-1 progression. The study also emphasizes the need to apply a standardized and accepted set of clinical definitions for the purpose of disease stratification and research.

The objective of this study was to obtain genetic marker information in the Gyr breed by analyzing bGH and Pit-1 gene polymorphisms and to verify their association with milk production traits. One sample including 40 Gyr bulls were genotyped at two bGH gene restriction sites (bGH- AluI and bGH-MspI) and at one restriction site in the Pit-1 gene (Pit-1 HinfI). The bGH-MspI(-) allele was favorable for fat milk percentage. The heterozigous Pit-1 HinfI (+/-) bulls were superior for fat milk production, in relation to homozigous Pit-1 HinfI (+/+). The Pit-1 and bGH genes are strong candidates in the dairy cattle QTL search, and zebuine populations are promising samples for this purpose.

The performance of Pseudomonas biocontrol agents may be improved by applying mixtures of strains which are complementary in their capacity to suppress plant diseases. Here, we have chosen the combination of Pseudomonas fluorescens CHA0 with another well-characterized biocontrol agent, P. fluorescens Q2-87, as a model to study how these strains affect each other's expression of a biocontrol trait. In both strains, production of the antimicrobial compound 2,4-diacetylphloroglucinol (DAPG) is a crucial factor contributing to the suppression of root diseases. DAPG acts as a signaling compound inducing the expression of its own biosynthetic genes. Experimental setups were developed to investigate whether, when combining strains CHA0 and Q2-87, DAPG excreted by one strain may influence expression of DAPG-biosynthetic genes in the other strain in vitro and on the roots of wheat. DAPG production was monitored by observing the expression of lacZ fused to the biosynthetic gene phlA of the respective strain. Dual-culture assays in which the two strains were grown in liquid medium physically separated by a membrane revealed that Q2-87 but not its DAPG-negative mutant Q2-87::Tn5-1 strongly induced phlA expression in a DeltaphlA mutant of strain CHA0. In the same way, phlA expression in a Q2-87 background was induced by DAPG produced by CHA0. When coinoculated onto the roots of wheat seedlings grown under gnotobiotic conditions, strains Q2-87 and CHA0, but not their respective DAPG-negative mutants, were able to enhance phlA expression in each other. In summary, we have established that two nonrelated pseudomonads may stimulate each other in the expression of an antimicrobial compound important for biocontrol. This interpopulation communication occurs in the rhizosphere, i.e., at the site of pathogen inhibition, and is mediated by the antimicrobial compound itself acting as a signal exchanged between the two pseudomonads.

O objetivo deste trabalho foi desenvolver um método de produção de mudas de tomateiro a partir de estacas laterais enraizadas em hidroponia. Foram realizados três experimentos. Nos experimentos 1 e 2, as estacas foram enraizadas em espuma fenólica e em solução nutritiva, respectivamente, e os tratamentos representaram as posições de retirada das estacas das axilas das folhas de 1 a 5 da base para o ápice. No experimento 3, estacas enraizadas nos experimentos 1 e 2, foram transplantadas para vasos contendo solução nutritiva e cultivadas até a produção do primeiro cacho. Com relação à massa de matéria fresca das raízes, pontos de interseção da raiz, massa de matéria seca dos caules e das folhas, comprimento de raízes, área foliar, número de folhas e diâmetro do caule, nos experimentos 1 e 2, não foi observada diferença estatística significativa entre os tratamentos 1, 2, 3 e 4. No experimento 3, apenas o diâmetro dos frutos apresentou diferença estatística entre os tratamentos. O tratamento 3 foi superior ao 5, e os tratamentos 1, 2, 3 e 4 foram semelhantes entre si. Em sistema hidropônico, podem ser produzidas mudas de tomate a partir de estacas, utilizando-se espuma fenólica ou solução nutritiva como meio de enraizamento.

The performance of magnetic nanoparticles is intimately entwined with their structure, mean size and magnetic anisotropy. Besides, ensembles offer a unique way of engineering the magnetic response by modifying the strength of the dipolar interactions between particles. Here we report on an experimental and theoretical analysis of magnetic hyperthermia, a rapidly developing technique in medical research and oncology. Experimentally, we demonstrate that single-domain cubic iron oxide particles resembling bacterial magnetosomes have superior magnetic heating efficiency compared to spherical particles of similar sizes. Monte Carlo simulations at the atomic level corroborate the larger anisotropy of the cubic particles in comparison with the spherical ones, thus evidencing the beneficial role of surface anisotropy in the improved heating power. Moreover we establish a quantitative link between the particle assembling, the interactions and the heating properties. This knowledge opens new perspectives for improved hyperthermia, an alternative to conventional cancer therapies.

BACKGROUND: HIV-infected individuals have an increased risk of myocardial infarction. Antiretroviral therapy (ART) is regarded as a major determinant of dyslipidemia in HIV-infected individuals. Previous genetic studies have been limited by the validity of the single-nucleotide polymorphisms (SNPs) interrogated and by cross-sectional design. Recent genome-wide association studies have reliably associated common SNPs to dyslipidemia in the general population. METHODS AND RESULTS: We validated the contribution of 42 SNPs (33 identified in genome-wide association studies and 9 previously reported SNPs not included in genome-wide association study chips) and of longitudinally measured key nongenetic variables (ART, underlying conditions, sex, age, ethnicity, and HIV disease parameters) to dyslipidemia in 745 HIV-infected study participants (n=34 565 lipid measurements; median follow-up, 7.6 years). The relative impact of SNPs and ART to lipid variation in the study population and their cumulative influence on sustained dyslipidemia at the level of the individual were calculated. SNPs were associated with lipid changes consistent with genome-wide association study estimates. SNPs explained up to 7.6% (non-high-density lipoprotein cholesterol), 6.2% (high-density lipoprotein cholesterol), and 6.8% (triglycerides) of lipid variation; ART explained 3.9% (non-high-density lipoprotein cholesterol), 1.5% (high-density lipoprotein cholesterol), and 6.2% (triglycerides). An individual with the most dyslipidemic antiretroviral and genetic background had an approximately 3- to 5-fold increased risk of sustained dyslipidemia compared with an individual with the least dyslipidemic therapy and genetic background. CONCLUSIONS: In the HIV-infected population treated with ART, the weight of the contribution of common SNPs and ART to dyslipidemia was similar. When selecting an ART regimen, genetic information should be considered in addition to the dyslipidemic effects of ART agents.

Métodos para avaliar a atividade microbiana no solo são fundamentais no monitoramento ambiental de áreas degradadas. O objetivo deste trabalho foi investigar a atividade microbiana de solo do semi-árido cultivado com Atriplex nummularia Lindl. em áreas que receberam rejeito salino durante um e três anos, em comparação com um solo nativo, sem cultivo e não irrigada. O solo cultivado por três anos e que recebeu rejeito salino apresentou, no período seco, valores de pH, CE e atividade de hidrólise do diacetato de fluoresceína (FDA) superiores aos das demais áreas. No entanto, foi observada correlação negativa entre o carbono microbiano e os valores do quociente metabólico (qCO2). A biomassa microbiana e a fosfatase alcalina também foram superiores no solo cultivado por três anos e que recebeu rejeito salino em relação ao solo nativo sem irrigação, confirmando o desempenho de plantas halófitas na melhoria da qualidade do solo sob condições de estresse salino. O cultivo de A. nummularia constitui uma das alternativas para utilização de rejeito salino proveniente da dessalinização por osmose reversa.

BACKGROUND/AIMS: Bacillus Calmette Guerin (BCG) infection causes hepatic injury following granuloma formation and secretion of cytokines which render mice highly sensitive to endotoxin-mediated hepatotoxicity. This work investigates the role of inducible nitric oxide synthase (iNOS) in liver damage induced by BCG and endotoxins in BCG-infected mice. METHODS: Liver injury and cytokine activation induced by BCG and by LPS upon BCG infection (BCG/LPS) were compared in wild-type and iNOS-/- mice. RESULTS: iNOS-/- mice infected with living BCG are protected from hepatic injury when compared to wild-type mice which express iNOS protein in macrophages forming hepatic granulomas. In addition, iNOS-/- mice show a decrease in BCG-induced IFN-gamma serum levels. LPS challenge in BCG-infected mice strongly activates iNOS in the liver and spleen of wild-type mice which show important liver damage associated with a dramatic increase in TNF and IL-6 and also Th1 type cytokines. In contrast, iNOS-/- mice are protected from liver injury after BCG/LPS challenge and their TNF, IL-6 and Th1 type cytokine serum levels raise moderately. CONCLUSIONS: These results demonstrate that nitric oxide (NO) from iNOS is involved in hepatotoxicity induced by both mycobacterial infection and endotoxin effects upon BCG infection and that inhibition of NO from iNOS protects from liver injuries.

O objetivo do trabalho foi avaliar a inclusão de silagem de grãos úmidos de milho, em substituição ao milho seco, em rações para frangos de corte criados nos sistemas convencional e alternativo. Foram utilizados 720 pintos machos Cobb, com um dia de idade, distribuídos em blocos ao acaso, em esquema fatorial 2x3 (dois sistemas de criação: convencional e alternativo; três níveis de silagem de grãos úmidos, em substituição ao milho seco: 0%, 30% e 60%), com quatro repetições cada. Foram obtidas médias de peso, ganho de peso, consumo de ração, conversão alimentar, fator de produção e mortalidade aos 21, 42 e 49 dias de idades. Aos 49 dias, foram abatidas 28 aves, por tratamento, para determinação do rendimento de carcaça, cortes e gordura abdominal. A inclusão de até 60% de silagem de grãos úmidos, em substituição ao milho seco, reduziu o custo da ração, sem alterar o desempenho das aves aos 21, 42 e 49 dias, nem o rendimento de carcaça aos 49 dias. No sistema de criação alternativo, independentemente do nível de inclusão de silagem de grãos úmidos, o desempenho dos frangos foi inferior, porém o retorno econômico foi maior.

The work presented here is part of a larger study to identify novel technologies and biomarkers for early Alzheimer disease (AD) detection and it focuses on evaluating the suitability of a new approach for early AD diagnosis by non-invasive methods. The purpose is to examine in a pilot study the potential of applying intelligent algorithms to speech features obtained from suspected patients in order to contribute to the improvement of diagnosis of AD and its degree of severity. In this sense, Artificial Neural Networks (ANN) have been used for the automatic classification of the two classes (AD and control subjects). Two human issues have been analyzed for feature selection: Spontaneous Speech and Emotional Response. Not only linear features but also non-linear ones, such as Fractal Dimension, have been explored. The approach is non invasive, low cost and without any side effects. Obtained experimental results were very satisfactory and promising for early diagnosis and classification of AD patients.

BACKGROUND: Physiological changes associated with pregnancy may alter antiretroviral plasma concentrations and might jeopardize prevention of mother-to-child HIV transmission. Lopinavir is one of the protease inhibitors more frequently prescribed during pregnancy in Europe. We described the free and total pharmacokinetics of lopinavir in HIV-infected pregnant and non-pregnant women, and evaluated whether significant alterations in its disposition and protein binding warrant systematic dosage adjustment. METHODS: Plasma samples were collected at first, second and third trimester of pregnancy, at delivery, in umbilical cord and postpartum. Lopinavir free and total plasma concentrations were measured by HPLC-MS/MS. Bayesian calculations were used to extrapolate total concentrations to trough (Cmin). RESULTS: A total of 42 HIV-positive pregnant women and 37 non-pregnant women on lopinavir/ritonavir were included in the study. Compared to postpartum and control values, total lopinavir Cmin was decreased moderately (31-39%) during pregnancy, and free Cmin minimally, showing significant alteration only at delivery (-35%). However, total and free Cmin remained in all patients above the target concentrations for wild-type virus of 1,000 ng/ml, and above the unbound IC50(WT) of 0.64-0.77 ng/ml of lopinavir, respectively. Lopinavir free fractions remained higher during pregnancy compared to postpartum and controls, and were influenced by α-1-acid-glycoprotein and albumin decrease. Free cord-to-mother ratio (0.43) was 2.7-fold higher than total cord-to-mother ratio (0.16), suggesting higher fetal exposure. CONCLUSIONS: The moderate decrease of total lopinavir concentrations during pregnancy is not associated with proportional decrease in free concentrations. Both reach a nadir at delivery, albeit not to an extent that would put treatment-naive women at risk of insufficient exposure to the free, pharmacologically active concentrations of lopinavir. No dosage adjustment is therefore needed during pregnancy as it is unlikely to further enhance treatment efficacy but could potentially increase the risk of maternal and fetal toxicity. Nonetheless, in case of viral resistance in treatment-experienced pregnant women, loss of virological control or questionable adherence, it is justified to consider lopinavir dosage adjustment based on total plasma concentration measurement.

Alzheimer's disease is the most prevalent form of progressive degenerative dementia; it has a high socio-economic impact in Western countries. Therefore it is one of the most active research areas today. Alzheimer's is sometimes diagnosed by excluding other dementias, and definitive confirmation is only obtained through a post-mortem study of the brain tissue of the patient. The work presented here is part of a larger study that aims to identify novel technologies and biomarkers for early Alzheimer's disease detection, and it focuses on evaluating the suitability of a new approach for early diagnosis of Alzheimer’s disease by non-invasive methods. The purpose is to examine, in a pilot study, the potential of applying Machine Learning algorithms to speech features obtained from suspected Alzheimer sufferers in order help diagnose this disease and determine its degree of severity. Two human capabilities relevant in communication have been analyzed for feature selection: Spontaneous Speech and Emotional Response. The experimental results obtained were very satisfactory and promising for the early diagnosis and classification of Alzheimer’s disease patients.

Abacus, és una eina de reforç al càlcul mental.Aquesta eina té dos propòsits, enfocats a dues situacions prou diferents: per una banda ajudar als nens que comencen a estudiar càlcul, per a qui aquesta eina servirà per a millorar la seva facilitat en el càlcul; per una altra banda gent a qui han detectat alguna malaltia degenerativa, a qui l¿exercici mental podria ajudar.
