P>According to the hygiene hypothesis, the increased incidence of allergic and autoimmune diseases in developed countries is mainly explained by the decreased contact between the human population and certain environmental agents as lactobacillus, mycobacteria and helminths. In this study, we evaluated the effect of multiple infections with Strongyloides venezuelensis on the development of experimental autoimmune encephalomyelitis (EAE) in Lewis rats. Multiple infections before EAE induction were not able to change the evolution of the disease. No alterations were observed in weight loss, clinical score and inflammation intensity at the central nervous system. The presence of significant levels of parasite-specific IgG1 but not IgG2b suggested a Th2 polarization. However, the percentage and absolute number of CD4+CD25+Foxp3+ T cells were not changed, being their levels in the spleen and lymph nodes of infected rats comparable to the ones found in normal animals. These results suggest that a Th2-polarized response without concomitant expansion of Foxp3+ regulatory T cells was not able to modify EAE progression. Even though these results do not threaten the hygiene hypothesis, they suggest that this paradigm might be an oversimplification. They also emphasize the need of a study to compare the immunoregulatory ability associated with different helminth spp.

This study aimed to compare Petrifilm Aerobic Count (AC) plates and the conventional pour plate methodology using the de Man-Rogosa-Sharpe (MRS) agar for the enumeration of lactic acid bacteria (LAB) in fermented milks (FMs), with different starter cultures added. FM samples (n = 66) were collected and plated on both methodologies, with incubation under anaerobic conditions at 35C for 48 h. The count results were compared by analysis of variance (P <= 0.05) and regression analysis. No differences between the mean counts obtained by both methodologies were observed, even when distinct FMs were compared. Considering all samples, a high correlation level was obtained between Petrifilm AC and MRS agar (r = 0.92), but these indexes were lower in FMs with Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus (r = 0.90) and Lactobacillus fortis (r = 0.81). Despite some slight interferences, Petrifilm AC has proven to be a convenient methodology on enumerating LAB in FM.

Aim: In the Amazon region of Brazil, the fruits of Caesalpinia ferrea Martius (Brazilian ironwood) are widely used as an antimicrobial and healing medicine in many situations including oral infections. This study aimed to evaluate the antimicrobial activity of Caesalpinia ferrea Martius fruit extract against oral pathogens. Materials and methods: Polyphenols estimation and spectral analysis ((1)H NMR) of the methanol extract were carried out. The microorganisms Candida albicans, Streptococcus mutans, Streptococcus salivarius, Streptococcus oralis and Lactobacillus casei were tested using the microdilution method for planktonic cells (MIC) and a multispecies biofilm model. Chlorhexidine was used as positive control. Results: Polyphenols in the extract were estimated at 7.3% and (1)H NMR analysis revealed hydroxy phenols and methoxilated compounds. MIC values for Candida albicans, Streptococcus mutans, Streptococcus salivarius, Streptococcus oralis and Lactobacillus casei were 25.0, 40.0, 66.0, 100.0, 66.0 mu g/mL, respectively. For the biofilm assay, chlorhexidine and plant extract showed no growth at 10(-4) and 10(-5) microbial dilution, respectively. At 10-4 and 10-5 the growth values (mean +/- SD) of the negative controls (DMSO and saline solution) for Streptococcus mutans, Streptococcus sp. and Candida albicans were 8.1 +/- 0.7, 7.0 +/- 0.6 and 5.9 +/- 0.9 x 10(6) CFU, respectively. Conclusion: Caesalpinia ferrea fruit extract can inhibit in vitro growth of oral pathogens in planktonic and biofilm models supporting its use for oral infections. (C) 2009 Elsevier Ireland Ltd. This study evaluated the caries risk of asthmatic patients on the basis of mutans streptococci (MS) and lactobacilli levels in saliva samples as well as the index of oral hygiene and dental caries (DMFT index). The study population was composed of 80 asthmatic children, aged 3-15 years, who use specific medication, and 80 matched, healthy control children. The parents were interviewed about oral health-related factors. The World Health Organization criteria were used for dental examinations. The Kohler and Bratthal methodology was used to detect salivary MS levels and dilutions of saliva were done for lactobacilli counting. No differences between asthma and control groups were observed for caries prevalence in children aged 3-6 and 7-10 years, except in severe cases in the younger group. However, higher caries prevalence for permanent dentition was observed in 11- to 15-year-old asthmatic children. An increased dental biofilm was observed in the asthma group, as well as salivary levels of MS. No differences were observed in levels of lactobacilli. No statistical correlations were found between medication, frequency of treatment, method of consumption and caries experience, dental biofilm and salivary levels of MS or lactobacilli. However, there was a correlation between MS levels and treatment duration. The logistic regression revealed that MS level is an important risk factor for increased caries experience. Asthma should be evaluated as a risk factor for caries experience because it can increase the levels of MS and the dental biofilm. Copyright (C) 2011 S. Karger AG, Basel

Aims : To study the effects of amylomaize starch and modified (carboxymethylated and acetylated) amylomaize starches on the composition of colonic bacteria and the production of volatile fatty acids, in mice. Methods and Results : Balb/c mice were fed with experimental diets containing various amount of amylomaize and modified amylomaize starches. Colonic bacterial populations and short-chain fatty acids were monitored. Results showed that the increases in indigenous bifidobacteria were detected in mice fed all starches tested; however, the highest numbers were observed in the group fed with 40% unmodified amylomaize starch. The starch type influenced the populations of indigenous Lactobacillus , Bacteroides and coliforms. High Lactobacillus numbers were achieved in the colon of mice fed with high concentration of amylomaize starch. Acetylated amylomaize starch significantly reduced the population of coliforms. In addition, orally dosed amylomaize utilizing bifidobacteria reached their highest levels when fed together with amylomaize or carboxymethylated amylomaize starch and in both cases butyrate levels were markedly increased. Conclusions: These results indicate that different amylomaize starches could generate desirable variation in gut microflora and that particular starches may be used to selectively modify gut function. Significance and Impact of Study: Amylomaize starch appeared to enhance the desirable composition of colonic bacteria in mice, and suggested it possessed the potential prebiotic properties.MTherefore, resistant starch and its chemical derivatives may exert beneficial impacts to the human colon.

The health benefits provided by probiotic bacteria have led to their increasing use in fermented and other dairy products. However, their viability in these products is low. Encapsulation has been investigated to protect the bacteria in the product's environment and improve their survival. There are two common encapsulation techniques, namely extrusion and emulsion, to encapsulate the probiotics for their use in the fermented and other dairy products. This review evaluates the merits and limitations of these two techniques, and also discusses the supporting materials and special treatments used in encapsulation processes. Resumo: A cárie dentária, um problema que tem atingido populações em grande parte do mundo, é a doença de maior prevalência da cavidade oral, gerando graves consequências económicas e sociais. Esta doença tem sido estudada ao longo do tempo em diferentes países com o emprego de diversos índices, geralmente para o estudo da sua prevalência, a avaliação de medidas preventivas e o adequado planeamento das acções e serviços de saúde oral. O objectivo deste Projecto foi determinar se a escovagem quando realizada na escola com pasta fluoretada, e supervisionada pelos professores, duas vezes por dia, seria ou não eficaz na diminuição das populações microbianas de Streptococcus mutans e Lactobacillus e na consequente diminuição da incidência de cárie dentária. Material e métodos: Foram seleccionadas todas as crianças (universo = 178), com idades compreendidas entre os 5, 6 e 7 anos, residentes no Concelho de Aljustrel e a frequentar o pré-escolar e o primeiro ciclo do ensino básico oficial. Foi realizada a escovagem bi-diária com pasta fluoretada a 500 ppm F-, na escola, segundo o método de Bass modificado, supervisionado pelos professores titulares de turma, que tiveram formação da técnica de escovagem utilizada. Durante os 3 anos de estudo foram realizadas 6 observações dentárias e recolhas salivares para contagem de Streptococcus mutans e Lactobacillus e avaliação da capacidade tampão da saliva. Resultados: O grupo de estudo no início da intervenção apresentava valores dos índices de cárie dentária mais elevados do que os do grupo de controlo (mais 0,109 no CPO-S, 0,0749 no CPO-D, 1,505 no cpo-s e 0,831 no cpo-d), porém sem diferenças de significância estatística. A análise estatística dos resultados não veio confirmar este pressuposto uma vez que o grupo de estudo apresentou um aumento percentual ligeiramente maior do índice CPO-D (12,5%) do que o grupo de controlo (11,6%). Para além deste aspecto, ao contrário do que seria de esperar, não foi possível detectar nenhuma diferença estatisticamente significativa em nenhum dos índices de cárie dentária (cpo-s, cpo-d, CPO-S e CPO-D) entre o grupo de estudo e grupo de controlo entre a 1ª e última observação.Ainda que os resultados do estudo aqui apresentado tenham ficado aquém do esperado, deveria ser efectuada a escovagem diária na escola, uma vez por dia, com pasta fluoretada a 1000 ppm F-, atendendo a que esta medida contribui para a promoção da saúde e prevenção da doença e é facilitadora da construção de estilos de vida saudáveis.-------ABSTRACT: Dental caries, a problem that has affected populations worldwide, is one of the most prevalent diseases of the oral cavity, causing severe economic and social consequences. This disease has been studied over time in different countries with the use of various indices, usually for the knowledge of its prevalence, evaluation of preventive measures and appropriate planning of actions and oral services. The aim of this study was to determine whether toothbrushing when performed in schools, with fluoride toothpaste, and supervised by teachers twice a day, was effective in reducing microbial populations of Streptococcus mutans and Lactobacillus with consequent reduction in the incidence of dental caries. Material and Methods: All children aged 5, 6, and 7 years, from Aljustrel County, attending official pre-school and first cycle of basic educatio were selected. Toothbrushing was performed twice a day with toothpaste with 500 ppm F-,in the school, according to the modified Bass method, supervised by professors in the class, who were trained in the brushing technique used. During the study period were performed 6 observations of the dental status, and were also collected saliva for the count of Streptococcus mutans and Lactobacillus, and assessment of buffering capacity of saliva. Results: The study group at the beginning of the intervention had higher values of dental caries than the control group (more than 0,109 in DMF-S, 0, 0749 in DMF, dmf-s 1,505 and 0,831 in dmf-t) although without statistical significance. The expected results were not confirmed, since the study group had a slightly higher percentage increase of the DMF-T (12,5%) than the control group (11,6%). Apart from that, contrary to what one would expect, we could not detect any statistical significant difference in any of the indices of dental caries (dmf-s, dmf-t and DMF-S, DMF-T) between the study and the control group in all study periods. Although the study results were not has expected, toothbrushing should be performed daily at school, once a day with fluoride toothpaste with 1000 ppm F-, since this measure contributes to health promotion and disease prevention and encourages healthy lifestyles.

Avaliar a microbiota intestinal de indivíduos que sofreram acidente ocupacional com materiais biológicos e receberam anti-retrovirais foi o objetivo deste estudo. O grupo de estudo constou de 23 indivíduos com idade entre 18-45 anos, sendo 13 doadores de sangue e 10 que sofreram acidente ocupacional. Foram avaliados a microbiota intestinal, antropometria e exames laboratoriais pré, pós e 30 dias após o término da medicação. Zidovudina mais lamivudina foi utilizada em 70% dos indivíduos associado ao nelfinavir, 20% ao efavirenz e 10% ao ritonavir. As alterações nutricionais e dietéticas-laboratoriais e de microbiota intestinal foram analisadas em três momentos. M1: até dois dias do início da profilaxia; M2: no último dia da profilaxia e M3: 30 dias após o término da profilaxia. Náuseas, vômitos e diarréia estiveram presentes em 50% no segundo momento do estudo. Sobrepeso em 70%, desnutrição e eutrofia em 10%, dos indivíduos, não se modificaram durante o estudo. Transaminases, triglicérides, LDL-colesterol se elevaram no segundo momento e normalizaram 30 dias após término da medicação. Houve redução significativa dos Lactobacillus, Bifidobacterium e Bacteróides nos três momentos. Uso de anti-retrovirais provocou impacto significativo na microbiota intestinal dos indivíduos, sem recuperação em 30 dias.

Avaliou-se in vitro a atividade antimicrobiana, antifúngica e antiaderente da aroeira-do-sertão, malva e goiabeira sobre microrganismos do biofilme dental e candidose oral. Os extratos mostraram-se eficazes, inibindo o crescimento das bactérias do biofilme dental e fungos da candidose oral, sugerindo a utilização dessas plantas como meio alternativo na terapêutica odontológica.

É do conhecimento geral que a população portuguesa está cada vez mais preocupada com a sua saúde e qualidade de vida, bem como em mudar alguns dos seus hábitos alimentares. Consequentemente, a procura por alimentos que proporcionam, para além de boas propriedades organolépticas e nutritivas, benefícios à saúde do consumidor, aumentou significativamente nas últimas décadas, onde os produtos lácteos fermentados surgem em grande destaque. Vários são os microrganismos empregues no desenvolvimento de produtos lácteos fermentados. Na produção de iogurtes, são utilizadas culturas lácteas tradicionais (Lactobacillus delbrueckii subsp. bulgaricus e Streptococcus thermophilus), também denominadas por flora específica do iogurte, e/ou bactérias probióticas, como as bifidobactérias, devido às suas boas características tecnológicas, terapêuticas e sensoriais. O trabalho descrito nesta dissertação de mestrado foca-se na validação e implementação de metodologias que permitem a contagem da flora específica do iogurte e de bifidobactérias, em iogurtes, na empresa SGS Portugal, S.A. A enumeração da flora específica do iogurte teve como base a norma ISO 7889:2003, por meio da técnica de sementeira por incorporação no meio de cultura apropriado e posterior contagem em placa dos microrganismos presentes. Foram analisadas 20 amostras de iogurtes, obtendo-se valores de contagens da flora específica do iogurte na ordem de 108 UFC/g de produto, indo ao encontro dos valores estabelecidos pela legislação portuguesa em vigor. A enumeração de bifidobactérias teve como base a norma ISO 29981:2010, por meio da técnica de sementeira por incorporação no meio de cultura apropriado e posterior contagem em placa dos microrganismos presentes. Das 20 amostras de iogurtes probióticos analisadas, apenas 15 obtiveram resultados positivos, apresentando valores de contagens de bifidobactérias superiores a 106 UFC/g de produto. Estas amostras, para além de apresentarem valores de acordo com os valores estabelecidos pela legislação portuguesa em vigor, são terapeuticamente eficazes, durante todo o prazo de validade do produto, uma vez que apresentam valores de contagem de bactérias probióticas superiores a 106 UFC/g, sendo este o padrão mínimo necessário para que um iogurte seja considerado como probiótico. Com base nos resultados obtidos e no cumprimento dos parâmetros preconizados nos procedimentos de validação de métodos microbiológicos, foi possível a validação destas metodologias e as suas implementações na empresa SGS Portugal, S.A.

OBJECTIVE: To assess the effect of a new feed soy product fermented by Enterococcus faecium and Lactobacillus jugurti on the serum lipid levels of rabbits with induced hypercholesterolemia. METHODS: Thirty-two rabbits were divided into 4 groups as follows: 1) control (C); 2) hypercholesterolemic (H); 3) hypercholesterolemic + fermented product (HPF); and 4) control + fermented product (CPF). The H and HPF groups were fed with a diet with 0.15% (p/p) cholesterol in the first 15 days. C and CPF groups received regular food preparation. The HPF and CPF groups received 10 mL daily of the fermented 30 days. Blood samples were drawn at the beginning of the study and at the 15th and 30th days. Concentrations of total cholesterol, HDL-cholesterol, and triglycerides were analyzed. RESULTS: After 15 days, the HPF group showed a total cholesterol concentration lower (18.4%) than that of the H group (p=0.05), but this difference disappeared after 30 days. No change was observed in total cholesterol levels of C and CPF groups. After 15 days, the HDL-cholesterol was higher (17.8%) in the HPF group, but the triglyceride levels remained unchanged in all groups during the same period of time. CONCLUSION: The soy fermented product caused an 18.4% reduction in total cholesterol and a 17.8% increase in the HDL-fraction. It may, therefore, be a possible coadjutor in the treatment of hypercholesterolemia.

The microbiological assay method of Snell and Wright for niacine was studied and some modifications of the basal medium were proposed. A maximal growth of the "Lactobacillus arabinosus" was obtained by the addition to the basal medium of 25 mg % asparagine and increasing the percentages of glucose and sodium acetate. Liver and yeast extracts were assayed satisfactory and the niacine added was recovered quantitatively.

Invasive candidiasis (IC) is a relatively common syndrome in neonates and children and is associated with significant morbidity and mortality. These guidelines provide recommendations for the prevention and treatment of IC in neonates and children. Appropriate agents for the prevention of IC in neonates at high risk include fluconazole (A-I), nystatin (B-II) or lactoferrin ± Lactobacillus (B-II). The treatment of IC in neonates is complicated by the high likelihood of disseminated disease, including the possibility of infection within the central nervous system. Amphotericin B deoxycholate (B-II), liposomal amphotericin B (B-II), amphotericin B lipid complex (ABLC) (C-II), fluconazole (B-II), micafungin (B-II) and caspofungin (C-II) can all be potentially used. Recommendations for the prevention of IC in children are largely extrapolated from studies performed in adults with concomitant pharmacokinetic data and models in children. For allogeneic HSCT recipients, fluconazole (A-I), voriconazole (A-I), micafungin (A-I), itraconazole (B-II) and posaconazole (B-II) can all be used. Similar recommendations are made for the prevention of IC in children in other risk groups. With several exceptions, recommendations for the treatment of IC in children are extrapolated from adult studies, with concomitant pharmacokinetic studies. Amphotericin B deoxycholate (C-I), liposomal amphotericin B (A-I), ABLC (B-II), micafungin (A-I), caspofungin (A-I), anidulafungin (B-II), fluconazole (B-I) and voriconazole (B-I) can all be used.
