Division of labor is a widely studied aspect of colony behavior of social insects. Division of labor models indicate how individuals distribute themselves in order to perform different tasks simultaneously. However, models that study division of labor from a dynamical system point of view cannot be found in the literature. In this paper, we define a division of labor model as a discrete-time dynamical system, in order to study the equilibrium points and their properties related to convergence and stability. By making use of this analytical model, an adaptive algorithm based on division of labor can be designed to satisfy dynamic criteria. In this way, we have designed and tested an algorithm that varies the response thresholds in order to modify the dynamic behavior of the system. This behavior modification allows the system to adapt to specific environmental and collective situations, making the algorithm a good candidate for distributed control applications. The variable threshold algorithm is based on specialization mechanisms. It is able to achieve an asymptotically stable behavior of the system in different environments and independently of the number of individuals. The algorithm has been successfully tested under several initial conditions and number of individuals.

Assessing video quality is a complex task. While most pixel-based metrics do not present enough correlation between objective and subjective results, algorithms need to correspond to human perception when analyzing quality in a video sequence. For analyzing the perceived quality derived from concrete video artifacts in determined region of interest we present a novel methodology for generating test sequences which allow the analysis of impact of each individual distortion. Through results obtained after subjective assessment it is possible to create psychovisual models based on weighting pixels belonging to different regions of interest distributed by color, position, motion or content. Interesting results are obtained in subjective assessment which demonstrates the necessity of new metrics adapted to human visual system.

La tesis se centra en el análisis de 356 números de la revista El Arquitecto Peruano publicados entre 1937 y 1977. La revista fue fundada en agosto de 1937 por Fernando Belaúnde Terry, arquitecto graduado en la University of Texas at Austin en 1935 quien la dirigió por 25 años hasta marzo de 1963. Desde abril de 1963 la dirige Miguel Cruchaga Belaunde. Durante esta última etapa la revista deja de ser regular, publicándose intermitentemente con varios años sin editarse, hasta desaparecer finalmente en 1977. La revista es la publicación periódica de arquitectura más importante del siglo XX en el Perú. La investigación ubica a la revista en el contexto nacional donde se desarrolló vinculando además los diversos eventos de la arquitectura y el urbanismo del panorama internacional sucedidos durante las tres décadas de estudio. El trabajo empieza por establecer una secuencia de periodos. Esta se basa en la identificación de una estructura fija existente en la revista conformada por las secciones permanentes. Una paulatina inserción de secciones temporales fue complementando y detallando nuevas temáticas y contenidos generando variantes de la estructura fija a lo largo del tiempo. Como resultado de este proceso se reconocen 5 periodos vinculados a una serie de temas vertebrales durante la dirección de Fernando Belaunde: Periodo 1 (Agosto de 1937 a marzo de 1942). Vivienda Unifamiliar. Periodo 2 (Abril de 1942 a mayo de 1944). Urbanismo moderno. Periodo 3 (Junio de 1944 a diciembre de 1949). Vivienda Colectiva. Periodo 4 (Enero-febrero de 1950 a enero-febrero de 1956). Proyectos y obras. Periodo 5 (Marzo-abril de 1956 a enero-febrero-marzo de 1963). Territorio. Los años finales bajo la dirección de Miguel Cruchaga conforman el último Periodo. En la década de los treinta, durante el primer periodo se analiza el rol que un grupo de revistas de arquitectura europeas y norteamericanas tuvieron en la construcción formal y temática de El Arquitecto Peruano. En sus inicios existen influencias de The Architectural Forum, The American Architect, Architecture, California Arts and Architecture y Architectural Digest . Y junto a estas, las revistas alemanas Moderne Bauformen e Innen Dekoration. A raíz de la Segunda Guerra Mundial, las referencias a las revistas alemanas desaparecen y las relaciones con Estados Unidos se intensifican por la posición del Perú dentro del área de influencia norteamericana. El canal de las revistas no fue el único medio de influencia, durante los años cuarenta hubo un marco de apertura hacia Latinoamérica que incluyó viajes de cooperación de arquitectos norteamericanos por el sur del continente. Como parte de estos vínculos, en 1945 Paul Lester Wiener de Town Planning Associates y Richard Neutra, - ambos miembros del CIAM y el segundo Presidente del CIAM USA- prestaron asistencia a la iniciativa más importante promovida por la revista peruana, un proyecto de vivienda popular para obreros y empleados llamado el Plan Peruano. La revista tuvo siempre una fuerte actitud de cambio de la realidad existente en el medio peruano. Para modelar su nueva versión de la realidad instrumentó una serie de campañas de prensa. En la más importante utilizó las ideas del urbanismo moderno que circulaban durante la guerra nacidas en los CIAM de entreguerras. El Arquitecto Peruano aprovecha la coyuntura de los primeros años de la posguerra para canalizar sus iniciativas, tomando como modelo de su propuesta la idea de la Neighborhood Unit, convirtiéndolo en proyecto y finalmente construyendo la Unidad Vecinal N°3, el proyecto de mayor escala asociado con la revista. A esta perspectiva favorable para las ideas vertidas por la publicación se une el hecho de que su Director, es elegido Diputado por Lima y es un miembro muy cercano al gobierno de turno que dio la cobertura legal y de gestión para cristalizar los proyectos desde 1945 hasta 1948. Durante este periodo se producen las propuestas de mayor alcance promovidas por Fernando Belaunde y difundidas por la revista, como el propio Plan Peruano de Vivienda, los Centros Climáticos de Invierno, el Plan Piloto de Lima y los Concursos de Vivienda popular. Existe un trabajo anterior sobre la revista, restringido al campo del urbanismo realizado en 1992 por el historiador Antonio Zapata y titulado El joven Belaunde: Historia de la revista El Arquitecto Peruano en donde se inclina el peso de la revista hacia la Vivienda Social y se señala a Walter Gropius como el arquitecto más influyente en la publicación. La presente tesis plantea que el Plan Peruano es la punta del iceberg, pero que todos estos proyectos relacionados con la revista, conforman un conjunto indisoluble y que el paquete completo tiene una directa influencia de proyectos, ideas y labor editorial de José Luis Sert desarrollada desde finales de los años veinte hasta los años cincuenta. Sus libros Can Our Cities Survive? , The Heart of the City y la revista del GATEPAC AC Documentos de Actividad Contemporánea tienen improntas claras en la revista peruana que atraviesan tres de los periodos propuestos. Durante la década de los cuarenta cuando Sert junto a Wiener fueron socios en Town Plannning Associates y tuvieron contacto directo con el director de la revista así como con los principales colaboradores, y durante la década posterior. A la luz de las evidencias analizadas, la figura de José Luis Sert emerge como la del arquitecto más influyente para la revista El Arquitecto Peruano a lo largo de todo su recorrido. ABSTRACT The thesis focuses on the analysis of 356 issues of the magazine El Arquitecto Peruano published between 1937 and 1977. The magazine was founded in august 1937 by Fernando Belaunde Terry, graduated from the University of Texas at Austin in 1935 who leaded it 25 years until March 1963. Since April 1963 was leaded by Miguel Cruchaga Belaunde. During this last stage the magazine ceases to be published on regular times, being published intermittently, unreleased several years, and finally disappear in 1977. El Arquitecto Peruano is the most important magazine of architecture of the twentieth century in Peru. The research located the magazine in the national context where was developed further by linking the various events of architecture and urbanism of the international scene occurred during the three decades of study. The research begins to establish a sequence of periods. This identification is based on a fixed structure existing in the journal formed by the permanent sections. A gradual insertion of temporary sections was complementing and detailing new themes and contents generating variants of the fixed structure over time. As a result of this process we recognize five periods linked to a series of central themes developed under the lead of Fernando Belaunde: Period 1 (September 1937 to March 1942): The Small House. Period 2 (April 1942 to May 1944). Modern urbanism. Period 3 (June 1944 to December 1949). Housing. Period 4 (January-February 1950 January-February 1956). Projects and Buildings. Period 5 (March-April 1956 January-February-March 1963). Territory. The final years under the direction of Miguel Cruchaga make the last Period. In the thirties, during the first period, is analyzed the role that a group of European and American magazines in architecture had in formal construction and thematic of the magazine. In the beginning there are influences of The Architectural Forum, The American Architect, Architecture, California Arts and Architecture and Architectural Digest. And along with these, the german magazine Moderne Bauformen and Innen Dekoration. Following World War II, the references to german magazines disappear and US relations are intensified by the position of Peru in the area of US influence. The magazines was not the only ways of influence, during the forties there was an open framework with Latin America that included cooperation trips of american architects on the south of the continent. As part of these links, in 1945 Paul Lester Wiener of Town Planning Associates and Richard Neutra, -both members of CIAM and the second President of the CIAM USAassisted the most important initiative promoted by the peruvian magazine, a popular housing project for workers and employees called El Plan Peruano. The magazine always has a strong attitude of change of the reality in the peruvian scene. To model the new version of reality created a series of media campaigns. In the biggest it used the ideas of modern urbanism that circulated during the war born in the CIAM of twenties and thirties. El Arquitecto Peruano take advantage of the situation of the early postwar years to direct their initiatives, modeled his proposal with the idea of Neighborhood Unit, making project and finally building the Unidad Vecinal N° 3, the larger scale project linked with the magazine. This favorable outlook for the ideas expressed by the publication joins the fact that its Director is elected deputy for Lima and is very close member to the current government who gave legal instruments and management to crystallize projects from 1945 to 1948. During this period of far-reaching proposals promoted by Fernando Belaunde and diffused by the magazine, as El Plan Peruano itself, the Centros Climaticos de Invierno, the Plan Piloto for Lima and the Concusos de Vivienda Popular. There is an earlier work on the magazine, restricted to the field of urbanism in 1992 by historian Antonio Zapata, entitled El Joven Belaunde: Historia de la revista El Arquitecto Peruano where the weight of the magazine leans Social Housing and notes Walter Gropius as the most influential architect in the publication. This thesis suggests that El Plan Peruano is the end of the iceberg, but these related projects to the magazine, form an inseparable whole and that the whole package has a direct influence of projects, ideas and editorial work of José Luis Sert developed from late twenties to the fifties. His book Can Our Cities Survive? , The Heart of the City and the magazine of GATEPAC AC Documentos de Actividad Contemporanea have clear signs in the peruvian magazine cross three of the proposed periods. During the forties when Sert and Wiener were partners in Town Plannning Associates and had direct contact with the director of the magazine as well as with key colaborators, and during the following decade. In light of the evidence examined, the shape of José Luis Sert emerges as the most influential architect to the peruvian magazine along its entire route.

The influence of training in labor risk prevention and the development of the resulting pre-emptive culture are analyzed within this paper. In order to achieve this, a quantitative analysis of the students of Building Degree in the Technic University of Madrid has been developed. This study has been made in all grades, valuating the previous knowledge acquired during compulsory education. It must be kept in mind that the students in 3rd and 4th grade have received general and specific compulsory training in prevention and safety in the building sector.

The 1,3–1,4-β-glucanase from Bacillus macerans (wtGLU) and the 1,4-β-xylanase from Bacillus subtilis (wtXYN) are both single-domain jellyroll proteins catalyzing similar enzymatic reactions. In the fusion protein GluXyn-1, the two proteins are joined by insertion of the entire XYN domain into a surface loop of cpMAC-57, a circularly permuted variant of wtGLU. GluXyn-1 was generated by protein engineering methods, produced in Escherichia coli and shown to fold spontaneously and have both enzymatic activities at wild-type level. The crystal structure of GluXyn-1 was determined at 2.1 Å resolution and refined to R = 17.7% and R(free) = 22.4%. It shows nearly ideal, native-like folding of both protein domains and a small, but significant hinge bending between the domains. The active sites are independent and accessible explaining the observed enzymatic activity. Because in GluXyn-1 the complete XYN domain is inserted into the compact folding unit of GLU, the wild-type-like activity and tertiary structure of the latter proves that the folding process of GLU does not depend on intramolecular interactions that are short-ranged in the sequence. Insertion fusions of the GluXyn-1 type may prove to be an easy route toward more stable bifunctional proteins in which the two parts are more closely associated than in linear end-to-end protein fusions.

Insights into the function of a gene can be gained in multiple ways, including loss-of-function phenotype, sequence similarity, expression pattern, and by the consequences of its misexpression. Analysis of the phenotypes produced by expression of a gene at an abnormal time, place, or level may provide clues to a gene’s function when other approaches are not illuminating. Here we report that an eye-specific, enhancer–promoter present in the P element expression vector pGMR is able to drive high level expression in the eye of genes near the site of P element insertion. Cell fate determination, differentiation, proliferation, and death are essential for normal eye development. Thus the ability to carry out eye-specific misexpression of a significant fraction of genes in the genome, given the dispensability of the eye for viability and fertility of the adult, should provide a powerful approach for identifying regulators of these processes. To test this idea we carried out two overexpression screens for genes that function to regulate cell death. We screened for insertion-dependent dominant phenotypes in a wild-type background, and for dominant modifiers of a reaper overexpression-induced small eye phenotype. Multiple chromosomal loci were identified, including an insertion 5′ to hid, a potent inducer of apoptosis, and insertions 5′ to DIAP1, a cell death suppressor. To facilitate the cloning of genes near the P element insertion new misexpression vectors were created. A screen with one of these vectors identified eagle as a suppressor of a rough eye phenotype associated with overexpression of an activated Ras1 gene.

The intracellular parasite Toxoplasma gondii resides within a specialized compartment, the parasitophorous vacuole (PV), that resists fusion with host cell endocytic and lysosomal compartments. The PV is extensively modified by secretion of parasite proteins, including the dense granule protein GRA5 that is specifically targeted to the delimiting membrane of the PV (PVM). We show here that GRA5 is present both in a soluble form and in hydrophobic aggregates. GRA5 is secreted as a soluble form into the PV after which it becomes stably associated with the PVM. Topological studies demonstrated that GRA5 was inserted into the PVM as a transmembrane protein with its N-terminal domain extending into the cytoplasm and its C terminus in the vacuole lumen. Deletion of 8 of the 18 hydrophobic amino acids of the single predicted transmembrane domain resulted in the failure of GRA5 to associate with the PVM; yet it remained correctly packaged in the dense granules and was secreted as a soluble protein into the PV. Collectively, these studies demonstrate that the secretory pathway in Toxoplasma is unusual in two regards; it allows soluble export of proteins containing typical transmembrane domains and provides a mechanism for their insertion into a host cell membrane after secretion from the parasite.

The membrane assembly of polytopic membrane proteins is a complicated process. Using Chinese hamster P-glycoprotein (Pgp) as a model protein, we investigated this process previously and found that Pgp expresses more than one topology. One of the variations occurs at the transmembrane (TM) domain including TM3 and TM4: TM4 inserts into membranes in an Nin-Cout rather than the predicted Nout-Cin orientation, and TM3 is in cytoplasm rather than the predicted Nin-Cout orientation in the membrane. It is possible that TM4 has a strong activity to initiate the Nin-Cout membrane insertion, leaving TM3 out of the membrane. Here, we tested this hypothesis by expressing TM3 and TM4 in isolated conditions. Our results show that TM3 of Pgp does not have de novo Nin-Cout membrane insertion activity whereas TM4 initiates the Nin-Cout membrane insertion regardless of the presence of TM3. In contrast, TM3 and TM4 of another polytopic membrane protein, cystic fibrosis transmembrane conductance regulator (CFTR), have a similar level of de novo Nin-Cout membrane insertion activity and TM4 of CFTR functions only as a stop-transfer sequence in the presence of TM3. Based on these findings, we propose that 1) the membrane insertion of TM3 and TM4 of Pgp does not follow the sequential model, which predicts that TM3 initiates Nin-Cout membrane insertion whereas TM4 stops the insertion event; and 2) “leaving one TM segment out of the membrane” may be an important folding mechanism for polytopic membrane proteins, and it is regulated by the Nin-Cout membrane insertion activities of the TM segments.

Contact of cultured mammary epithelial cells with the basement membrane protein laminin induces multiple responses, including cell shape changes, growth arrest, and, in the presence of prolactin, transcription of the milk protein β-casein. We sought to identify the specific laminin receptor(s) mediating the multiple cell responses to laminin. Using assays with clonal mammary epithelial cells, we reveal distinct functions for the α6β4 integrin, β1 integrins, and an E3 laminin receptor. Signals from laminin for β-casein expression were inhibited in the presence of function-blocking antibodies against both the α6 and β1 integrin subunits and by the laminin E3 fragment. The α6-blocking antibody perturbed signals mediated by the α6β4 integrin, and the β1-blocking antibody perturbed signals mediated by another integrin, the α subunit(s) of which remains to be determined. Neither α6- nor β1-blocking antibodies perturbed the cell shape changes resulting from cell exposure to laminin. However, the E3 laminin fragment and heparin both inhibited cell shape changes induced by laminin, thereby implicating an E3 laminin receptor in this function. These results elucidate the multiplicity of cell-extracellular matrix interactions required to integrate cell structure and signaling and ultimately permit normal cell function.

The Tim23 protein is an essential inner membrane (IM) component of the yeast mitochondrial protein import pathway. Tim23p does not carry an amino-terminal presequence; therefore, the targeting information resides within the mature protein. Tim23p is anchored in the IM via four transmembrane segments and has two positively charged loops facing the matrix. To identify the import signal for Tim23p, we have constructed several altered versions of the Tim23 protein and examined their function and import in yeast cells, as well as their import into isolated mitochondria. We replaced the positively charged amino acids in one or both loops with alanine residues and found that the positive charges are not required for import into mitochondria, but at least one positively charged loop is required for insertion into the IM. Furthermore, we find that the signal to target Tim23p to mitochondria is carried in at least two of the hydrophobic transmembrane segments. Our results suggest that Tim23p contains separate import signals: hydrophobic segments for targeting Tim23p to mitochondria, and positively charged loops for insertion into the IM. We therefore propose that Tim23p is imported into mitochondria in at least two distinct steps.

Efficient 3′-end processing of cell cycle-regulated mammalian histone premessenger RNAs (pre-mRNAs) requires an upstream stem–loop and a histone downstream element (HDE) that base pairs with the U7 small ribonuclearprotein. Insertions between these elements have two effects: the site of cleavage moves in concert with the HDE and processing efficiency declines. We used Xenopus oocytes to ask whether compensatory length insertions in the human U7 RNA could restore the fidelity and efficiency of processing of mouse histone insertion pre-mRNAs. An insertion of 5 nt into U7 RNA that extends its complementary to the HDE compensated for both defects in processing of a 5-nt insertion substrate; a noncomplementary insertion into U7 did not. Yet, the noncomplementary insertion mutant U7 was shown to be active on insertion substrates further mutated to allow base pairing. Our results suggest that the histone pre-mRNA becomes rigidified upstream of its HDE, allowing the bound U7 small ribonucleoprotein to measure from the HDE to the cleavage site. Such a mechanism may be common to other RNA measuring systems. To our knowledge, this is the first demonstration of length suppression in an RNA processing system.

Retroviral elements are found in abundance throughout the human genome but only rarely have alterations of endogenous genes by retroviral insertions been described. Herein we report that a human endogenous retrovirus (HERV) type C is inserted in the human growth factor gene pleiotrophin (PTN) between the 5′ untranslated and the coding region. This insert in the human genome expands the region relative to the murine gene. Studies with promoter-reporter constructs show that the HERV insert in the human PTN gene generates an additional promoter with trophoblast-specific activity. Due to this promoter function, fusion transcripts between HERV and the open reading frame of PTN (HERV-PTN) were detected in all normal human trophoblast cell cultures as early as 9 weeks after gestation (n = 7) and in all term placenta tissues (n = 5) but not in other normal adult tissues. Furthermore, only trophoblast-derived choriocarcinoma cell lines expressed HERV-PTN mRNA whereas tumor cell lines derived from the embryoblast (teratocarcinoma) or from other lineages failed to do so. We investigated the significance of HERV-PTN mRNA in a choriocarcinoma model by targeting this transcript with ribozymes and found that the depletion of HERV-PTN mRNA prevents human choriocarcinoma growth, invasion, and angiogenesis in mice. This suggests that the tissue-specific expression of PTN due to the HERV insertion in the human genome supports the highly aggressive growth of human choriocarcinoma and possibly of the human trophoblast.

The serpins are a family of proteinase inhibitors that play a central role in the control of proteolytic cascades. Their inhibitory mechanism depends on the intramolecular insertion of the reactive loop into β-sheet A after cleavage by the target proteinase. Point mutations within the protein can allow aberrant conformational transitions characterized by β-strand exchange between the reactive loop of one molecule and β-sheet A of another. These loop-sheet polymers result in diseases as varied as cirrhosis, emphysema, angio-oedema, and thrombosis, and we recently have shown that they underlie an early-onset dementia. We report here the biochemical characteristics and crystal structure of a naturally occurring variant (Leu-55–Pro) of the plasma serpin α1-antichymotrypsin trapped as an inactive intermediate. The structure demonstrates a serpin configuration with partial insertion of the reactive loop into β-sheet A. The lower part of the sheet is filled by the last turn of F-helix and the loop that links it to s3A. This conformation matches that of proposed intermediates on the pathway to complex and polymer formation in the serpins. In particular, this intermediate, along with the latent and polymerized conformations, explains the loss of activity of plasma α1-antichymotrypsin associated with chronic obstructive pulmonary disease in patients with the Leu-55–Pro mutation.

We report the isolation and expression of the Hox gene, Cnox-2, in Hydractinia symbiolongicarpus, a hydrozoan displaying division of labor. We found different patterns of aboral-to-oral Cnox-2 expression among polyp polymorphs, and we show that experimental conversion of one polyp type to another is accompanied by concordant alteration in Cnox-2 expression. Our results are consistent with the suggestion that polyp polymorphism, characteristic of hydractiniid hydroids, arose via evolutionary modification of proportioning of head to body column.

The specific formylation of initiator methionyl-tRNA by methionyl-tRNA formyltransferase (MTF; EC 2.1.2.9) is important for the initiation of protein synthesis in eubacteria and in eukaryotic organelles. The determinants for formylation in the tRNA are clustered mostly in the acceptor stem. As part of studies on the molecular mechanism of recognition of the initiator tRNA by MTF, we report here on the isolation and characterization of suppressor mutations in Escherichia coli MTF, which compensate for the formylation defect of a mutant initiator tRNA, lacking a critical determinant in the acceptor stem. We show that the suppressor mutant in MTF has a glycine-41 to arginine change within a 16-amino acid insertion found in MTF from many sources. A mutant with glycine-41 changed to lysine also acts as a suppressor, whereas mutants with changes to aspartic acid, glutamine, and leucine do not. The kinetic parameters of the purified wild-type and mutant Arg-41 and Lys-41 enzymes, determined by using the wild-type and mutant tRNAs as substrates, show that the Arg-41 and Lys-41 mutant enzymes compensate specifically for the strong negative effect of the acceptor stem mutation on formylation. These and other considerations suggest that the 16-amino acid insertion in MTF plays an important role in the specific recognition of the determinants for formylation in the acceptor stem of the initiator tRNA.
