The microbiological flora of 108 water samples was explored to evaluate the role of recreational waters as a possible source of human diseases in Araraquara, S.P., Brazil. These waters included six swimming pools and three lakes with beaches. The number of total and fecal coliforms, Escherichia coli, fecal streptococci, Pseudomonas aeruginosa, Staphylococcus aureus, Candida albicans, and heterotrophic organisms was determined. As was the occurrence of Salmonella, Shigella, Yersinia, enteropathogenic E. coli (EPEC), enteroinvasive E. coli (EIEC), Mycobacteria, yeasts in general and dermatophytes. Shigella, Yersinia, EIEC and dermatophytes were not isolated. Other organisms or groups of microorganisms were found in variable proportions. From this study it is concluded that recreational waters used by the population of Araraquara, may be contaminated with potentially pathogenic microorganism and this may serve as a source of human diseases.

The objectives of the present study were: to evaluate the variation in total number of microorganisms in laying hen fecal samples and to identify the pathogenic microorganisms possibly present such as Staphylococcus aureus, Salmonella sp., Clostridium botulinum, C chauvoei, Campylobacter sp., Escherichia coli, and Corynebacterium sp. The experiment was conducted at the School of Agrarian and Veterinarian Sciences of Jaboticabal, - UNESP. Fecal samples were collected from the aviary. A fully randomized experimental design was used with 7 treatments and 4 replications, for a total of 28 samples. A decrease in total number of microorganisms was detected when treatment T0 (zero days of storage) was compared with treatment T6 (42 days of storage), with oscillations in numbers being observed during this period of time. Pathogenic bacteria such as Clostridium noyvi, C perfringens, C sordelli, C septicum and C noyvi type B were detected, as well as non-pathogenic bacteria such as Bacillus sp. and Staphylococcus epidermidis.

To evaluate the nutritional value of rations supplemented with different protein sources, an in vivo digestibility trial was conducted to determine the coefficients of digestibility for dry matter (DMDC) and crude protein (CPDC), nitrogen balance (NB) and the fecal non-protein nitrogen (NPN). Twenty - four 11-months - old lambs weighing 30.4 +/- 3.0 kg were blocked on the basis of their body weight and randomly allocated to three treatments groups; suplemented with cottonseed meal (CSM), starea (ST) or urea (UR). The DMDC was statistically superior for the Starea treatment (67.7%) over the others (64.3% and 64.1% for CSM and UR). The CSM, UR and ST rations had no influence on CPDC (63.9; 66.9 and 69.4%, respectively) and on fecal NPN (1,3; 1,2 e 1,3 g/day, respectively). The NB results were similar also among treatments (13,4; 9,0 e 10,5 g/day to CSM, UR and ST, respectively) although the ST treatment lambs excreted larger amounts of nitrogen in the urine (7.7 g/day) in relation to CSM (4.8g/day) and similar to UR treatment (6.5 g/day). Starea supplement in ruminant diets increased dry matter digestibility when compared to cottonseed meal and urea, but did not improve the efficiency of the nitrogen utilization by lambs.

Intestinal parasitic infections are currently a source of concern for Public Health agencies in developing and developed countries. Since three ovum-and-parasite stool examinations have been demonstrated to provide sensitive results, we designed a practical and economical kit (TF-Test) that is now commercially available (Immunoassay Com. Ind. Ltda., S (a) over tildeo Paulo, Brazil). This kit allows the separate collection of three fecal specimens into a preservative solution. The specimens are then pooled, double-filtered, and concentrated by a single rapid centrifugation process. The TF-Test was evaluated in four different laboratories in a study using 1,102 outpatients and individuals living in an endemic area for enteroparasitosis. The overall sensitivity found using the TF-Test (86.2-97.8%) was significantly higher (P<0.01) than the sensitivity of conventional techniques such as the Coprotest (NIL Comercio Exterior Ltda, São Paulo, Brazil) and the combination of Lutz/Hoffman, Faust, and Rugai techniques (De Carli, Diagnostico Laboratorial das Parasitoses Humanas. Metodos e Tecnicas, 1994), which ranged from 48.3% to 75.9%. When the above combined three specimen technique was repeated with three specimens collected on different days, its sensitivity became similar (P > 0.01) to that of the TF-Test. The kappa index values of agreement for the TF-Test were consistent (P < 0.01), being higher and ranking in a better position than conventional techniques. The high sensitivity, cost/benefit ratio, and practical aspects demonstrate that the TF-Test is suitable for individual diagnosis, epidemiological inquiries, or evaluation of chemotherapy in treated communities. Dry matter intake (DMI) of coast-cross grazing by crossbred Holstein-Zebu and Zebu lactating cows was calculated using in vitro dry matter digestibility from extrusa (four esophageal fistulated cows) and fecal output estimate with mordent chromium. Pasture was rotationally grazed with three days grazing period and 27 days testing period, adopting a stocking rate of 1.6 and 3.2 cows/ha, during the dry and rainy season respectively. Voluntary DMI was estimated from degradation characteristics using different equations. Predicted coast-cross DMI varied with models. The prediction of tropical forages dry matter intake from equations based in ruminal degradation parameters needs farther investigation before being employed in practice.

Toxocara vitulorum, a parasite of the small intestine of cattle and water buffaloes, is mainly acquired by calves via the colostrum/milk from infected cows. To understand the development of immune responses in calves, antibody levels to a soluble extract antigen (Ex) from T. vitulorum infective larvae were measured by an indirect ELISA with sera of 15 buffalo calves, which were sampled every 15 days for the first 180 days after birth and 9 buffalo cows during the perinatal period. From all serum samples examined during the first 180 days, antibody level was lowest and highest in calves at 1 day of age before and after suckling colostrum, respectively, suggesting that the origin of antibodies was the colostrum. Immediately after birth, antibody levels in suckled calves remained at high levels until day 15, began to decrease to lower levels between 15 and 30 days and remained relatively stable until 120 days. By comparing the immune responses of these animals with their parasitological status it was considered possible to determine if passively acquired or actively produced antibodies provided protection against the infection. High numbers of T. vitulorum eggs in the feces between 30 and 60 days indicated that passively acquired antibodies did not provide protection against the infection, at least during these first days, and the maximum fecal egg counts during 30-45 days were coincident with decreased antibody levels. Between 60 and 120 days, when serum antibodies were detected at reduced, but stable levels, adult nematodes were expelled from the intestines and no more T. vitulorum eggs were found, suggesting development of acquired resistance. However, the potential and functional protective role of the antibodies against T. vitulorum infection and the process of self-cure requires further investigation. (C) 2001 Elsevier B.V. B.V. A total of 24 male and female equines of mixed breed, 10-20 months of age and naturally infected with internal parasites was utilized in a controlled test to evaluate the efficacy of a moxidectin 2% gel formulation at the dosage of 0.4 mg moxidectin per kg of live weight and an ivermectin 1.87% commercial paste formulation at the dosage 0.2 mg ivermectin per kg applied orally Animals were allocated into three groups of eight horses each based on pre-treatment eggs per gram (EPG) counts and treatments were randomized among the groups. One group was kept as untreated controls. One animal in the moxidectin-treated group died before the end of the trial from a cause unrelated to treatment leaving a total of seven animals in this group. Fecal egg counts were performed three times post-treatment and the number of parasites remaining in each animal was determined. Statistical analyses using geometric means were performed at the 1% level of significance. Both moxidectin and ivermectin preparations reduced initial EPG from a mean of 1600 to 0 on Days 5, 7 and at the end of the trial on Day 14. Efficacy percentages of moxidectin and ivermectin against immature and adult nematodes were as follows: Trichostrongylus axel, Parascaris equorum, Strongylus edentatus, S. vulgaris, Triodontophorus spp. and Gyalocephalus capitatus, 100% for both products; Habronema muscae 99.5 and 99.6%, respectively, Strongyloides westeri, 100 and 99.2%, respectively; Oxyuris equi, 99.6 and 100%, respectively; small strongyles, 99.7% for both products. of the latter, the most numerous were: Cylicocyclus insigne, Cylicostephanus longibursatus and Cyathostomum catinatum. No Gasterophilus nasalis were found in horses from either treated group, while two of eight control horses had infections with this parasite. Moxidectin showed greater efficacy (84.9%) than ivermectin (67.8%) against Strongylus vulgaris larvae found in the mesenteric artery aneurisms, but the difference was not statistically significant. Total parasite counts for both treated groups were significantly lower (p<0.01) than in the non-treated group. No significant differences were noted between moxidectin and ivermectin. Efficacy against the 30 nematode species found in this study was very evident for both products. As expected, neither moxidectin nor ivermectin was effective in controlling the tapeworm Anoplocephala perfoliata. No adverse reactions were observed during the experimental period. Two groups of Holstein-Friesian and Nelore calves, five animals each, about nine months old, received, by oral route, 1,000 infective larvae (L-3) per kg of body weight of Haemonchus placei. Blood samples were collected by venipuncture, at weekly intervals, from one week before, to eight weeks after infection. Hematological studies comprised the hematocrit, differential leukocyte counts, hemoglobin, fibrinogen and plasma protein determinations. Parasitological examinations covered weekly fecal egg counts (EPG) and worm burden counts at necropsy. Samples of the abomasal mucosa were submitted to gross examination and histopathological studies. Both groups had increasing EPG after the fifth week, with Holstein calves showing higher counts than the Nelore. Holstein calves had anemia and hipoproteinemia from the third week post-infection to the end of the experiment, whereas Nelore calves showed no significant differences in those, parameters. Holstein calves had significantly larger worm counts than the Nelore. The gross and histopathological lesions in the abomasum at necropsy were very similar, although macroscopically they look more apparent in the Holstein group. These results showed that Holstein calves are more susceptible to the infection and pathogenic effects of H. placei than Nelore calves.

Between January and December 1996, the food habits of a relict population of jaguars were studied in 220 km(2) Linhares Forest Preserve, which comprises much of the remaining old-growth Atlantic Forest of Espirito Santo, Brazil. Fecal analysis indicated opportunistic feeding on 24 prey species (N = 101 scats). Mammals represented 87 percent of the total items, followed by reptiles (9.8%) and birds (2.8%). Considering prey weight, 23.4 percent of the items weighed 1-3 kg, 40.5 percent were 3-10 kg, and 27.7 percent weighed more than 10 kg. Analysis of relative prey frequency and biomass indicated that the diet was concentrated in two prey types: long-nosed armadillo and white-lipped peccary. Literature data suggest that forest jaguars rely on the same mammal prey over their entire geographic range.

A total of 145 capybara (Hydrochoerus hydrochaeris) fecal samples from the state of Sdo Paulo, Brazil, were screened for Cryptosporidium spp. oocysts using the malachite green method. Eight samples (5.52%) showed positive results and were further submitted to nested PCR reaction for amplification of fragments of 18S rRNA gene and 60-kDa glycoprotein gene for determination of species, alleles and subtypes of Cryptosporidium. Sequencing of the PCR products of the 18S rRNA gene fragments and 60-kDa glycoprotein gene fragments showed that for both genes all Cryptosporidium isolates from capybara were respectively 100% genetically similar to a bovine isolate of C. parvum and to C parvum subtype IIaA15G2R1. To the best of our knowledge this is the first report of Cryptosporidium infection in this rodent. The finding of zoonotic C parvum infection in a semi-aquatic mammal that inhabits anthroponotic habitats raises the concern that human water supplies may be contaminated with zoonotic Cryptosporidium oocysts from wildlife. This experiment was carried out with six half-bred Bretao-Campolinacolts with ileum fistulated to evaluate the difference in the ileum dry matter flow estimated by chromic oxide, cellulose, NDF, lignin and fecal lignin through the collection of ileum samples digesta at 28 h intervals, totalizing six samples per animal, starting at 10:00 a.m. The animals were fed ad libitum with the following diets: R1: clephantgrass, R2: elephantgrass plus ground corn, and R3: elephantgrass plus ground corn plus soybean meal. The data was statistically described, based on the coefficient of variation. The values of dry matter prececal digestion coefficients were, respectively, for diets 1, 2 and 3, at six schedules, for cellulose (-16.4; 21.4 and 6.6%), NDF(-34.7; 28.8 and -12.8%), to lignin (-51.5; -5.1 and -25.7%), in two schedules for cellulose (-13.4; 25.6 and 21.5%), fecal lignin (-37.1, 16.6 and -6.4%) and chromic oxide (-219.3, 36.4 and 9.5%). The coefficients of variation were, respectively, for the diets i, 2 and 3, at six schedules, for cellulose (148.3; 107.5 and 522.7%), NDF (95.4; 80.9 and 314.3%), lignin (210.2; 752.3 and 209.6%), at two schedules for cellulose(148.5; 80.7 and 70.0%), fecal lignin, (262.4; 177.9 and 723.5%) and chromic oxide (141.1; 158.9 and 473.4%). In diet i, the ileum dry matter now were over estimated for all markers, showing that chosen collection lime to estimate the flows were not adequate. Based on the coefficient of variation of the diets 2 and 3, the cellulose at two schedules was the most marker indicator to determine the ileum dry matter flow.
