The aim of the study was to verify whether 8 weeks of resistance training employing maximal isokinetic eccentric (IERT) knee extensor actions would reduce the acute force loss observed after high-intensity treadmill running exercise. It was hypothesized that specific IERT would induce protective effects against muscle fatigue and ultrastructural damages, preventing or reducing the loss in mechanical muscle function after running. Subjects were tested before and after IERT protocol for maximal isometric, concentric and eccentric isokinetic knee extensor strength (60 and 180 s-1). In a second session, subjects performed treadmill running (~35 min) and the previously mentioned measurements were repeated immediately after running. Subsequently, subjects were randomized to training (n = 12) consisting of 24 sessions of maximal IERT knee extensors actions at 180 s-1, or served as controls (n = 8). The effects of acute running-induced fatigue and training on isokinetic and isometric peak torque, and rate of force development (RFD) were investigated. Before IERT, running-induced eccentric torque loss at 180 s-1 was -8 %, and RFD loss was -11 %. Longitudinal IERT led to reduced or absent acute running-induced losses in maximal IERT torque at 180 s-1 (+2 %), being significantly reduced compared to before IERT (p < 0.05), however, RFD loss remained at -11 % (p > 0.05). In conclusion, IERT yields a reduced strength loss after high-intensity running workouts, which may suggest a protective effect against fatigue and/or morphological damages. However, IERT may not avoid reductions in explosive muscle actions. In turn, this may allow more intense training sessions to be performed, facilitating the adaptive response to running training. © 2013 Springer-Verlag Berlin Heidelberg.

Background: The present study aims to evaluate the effects of orthodontic movement (OM) on the periodontal tissues of rats with ligature-induced periodontal disease. Methods: Eighty-eight rats were divided into four groups: 1) negative control (sham operated); 2) periodontal disease; 3) OM; and 4) periodontal disease followed by OM (OMP). Rats were sacrificed 3 hours or 1, 3, or 7 days after OM commencement. Bone volume fraction (BVF) and bone mineral density (BMD) were assessed in hemimaxillae by microcomputed tomography analysis. Expression of the proinflammatory cytokines interleukin (IL)-1β and tumor necrosis factor (TNF)-α were evaluated in gingival samples by quantitative polymerase chain reaction and enzyme-linked immunosorbent assay, and in the furcation region by immunohistochemistry analysis (IHC). Results: The OMP group had lower BVF and BMD levels compared to the other groups at day 7 (P <0.05). Maximum messenger ribonucleic acid expression of both cytokines was observed in the OMP group at day 1 (P <0.05). In the same period, all proteins were expressed in high levels for all test groups compared to the control group. The number of cells positive for IL-1β and TNF-α by IHC was highest in the OMP group at day 1, with progressive reduction thereafter. Conclusion: The results suggest that OM acts synergistically with periodontal disease in periodontal breakdown through upregulation of proinflammatory cytokines.

BackgroundEvidence is emerging that highlights the far-reaching consequences of a high-fat diet (HFD) on kidney morphology and function disorders.MethodsThe present study was performed on 3-, 5-, 7- and 9-week-old HFD female rats compared with the appropriate gender and age-matched animals. We evaluated the kidney expression of angiotensin type II receptor and fibrotic and epithelial-to-mesenchymal transition (EMT) markers, by immunoblotting and immunohistochemical and histological techniques, in parallel with kidney function.ResultsIn the current study, the time-course HFD-treated group showed, by immunoblotting and immunohistochemical analysis, an early time-course increase in the expression of transforming growth factor β-1 (TGFβ-1) in the entire kidney of HFD-treated rats, compared with that observed in the control group. Simultaneously, the study shows a transient increase in the expression of ZEB2 in the HFD whole kidney accompanied by a fall in the E-cadherin expression and increased collagen and fibronectin deposition. A pronounced decrease in fractional urinary sodium excretion was also demonstrated in the long-term HFD-treated rats. The decreased FENa + was accompanied by a fall in FEPNa + and FEPP Na +, which occurred in association with significantly decreased CCr and, certainly on the sodium-filtered load. The reduction in the glomerular filtration rate (GFR) occurred in parallel to proteinuria and glomerular desmin overexpression.ConclusionsThe results of the current study suggest that podocyte injury in parallel with observed proteinuria and evidence of EMT transformation are associated with long-term loss of kidney function and renal sodium and water retention. © 2013 The Author. A ligação entre as zonas urbanas e as questões ambientais ficam mais próximas na medida em que cresce a conscientização global de conservar, melhorar e valorizar os serviços ambientais prestados pela natureza para a sustentabilidade da vida, dentro e fora da cidade. Cobertura vegetal (ou cobertura verde) está dentre as principais fontes de tais serviços. Uma vez que o processo de urbanização se mostra irreversível e os problemas ambientais urbanos se alastram em tamanho e extensão, a presença do verde está diretamente relacionada aos indicadores de qualidade de vida urbana. Como reflexo do processo de urbanização, a cidade de Belém perdeu uma grande porcentagem de seus ecossistemas naturais, de modo que este trabalho se concentrou em analisar alguns serviços ecossistêmicos—qualidade do ar, poluição do ar e regulação do clima - fornecidos pela qualidade e pela quantidade de cobertura vegetal local, considerando as alterações na distribuição espaço-temporal, em três distritos administrativos. Um marco teórico foi construído e analisado; a cobertura vegetal foi calculada, utilizando-se NDVI e Cobertura Vegetal Fracional em imagens do LANDSAT 5, ao longo de um período de 23 anos. A partir de uma proposta de escala mais detalhada de NDVI, análises quantitativas e qualitativas da cobertura verde evidenciaram perda significativa de cobertura muito densa, densa, moderada e aumento de áreas de pouca ou nenhuma vegetação. Ademais, lesão das áreas verdes sinalizou tendências de aumento da poluição do ar, da poluição sonora e da temperatura. A carência de dados relacionados ao meio ambiente não deixa dúvida sobre a urgência de investimento nos serviços ambientais provenientes da cobertura vegetal, para a sustentabilidade urbana em Belém, cujos cenários previstos são de drásticas perdas de área verde. Mais pesquisas e iniciativas de instituições públicas e privadas são necessárias para a contribuição aos serviços ambientais em Belém e, consequentemente, ao bem-estar público.

OBJETIVO: Avaliar o efeito do resveratrol sobre a via de sinalização da insulina e melhora do quadro inflamatório no miocárdio de ratos Wistar obesos induzidos por dieta.MÉTODOS: Ratos Wistar foram divididos em grupos: controle (dieta padrão para roedores), obeso (dieta hiperlipídica) e obeso suplementado com resveratrol (20 mg/kg/dia), por 8 semanas (n=10). Ao final do período experimental, realizou-se o teste de tolerância à insulina, nos tempos 0 (sem insulina), 5, 10, 15, 20, 25 e 30 minutos após injeção intraperitoneal de insulina (2 U/kg). O peso corporal e o tecido adiposo epididimal foram mensurados. Fragmentos do miocárdio foram extraídos para análises da via da insulina e moléculas pró-inflamatórias através de Western blot.RESULTADOS: Os resultados indicam que a intervenção com resveratrol aumenta a constante de decaimento da glicose, fosforilação do receptor de insulina, substrato do receptor de insulina e da proteína quinase B. A suplementação de resveratrol também reduziu os níveis proteicos do fator de necrose tumoral alfa e de moléculas envolvidas com a transdução do sinal pró-inflamatório (quinase indutora do kappa B e fator nuclear kappa B). Os resultados ainda sugerem que a melhora na sensibilidade à insulina e a redução das moléculas pró-inflamatórias ocorreram independentemente da perda de peso corporal e da redução do tecido adiposo epididimal.CONCLUSÃO: A suplementação de resveratrol aumenta a sensibilidade à insulina, o que está relacionado à redução de fatores inflamatórios no miocárdio.

An experiment employing three hundred and twenty 81-week-old Lohmann LSL commercial-breed hens was conducted to compare alternative induced-molting methods with the conventional method (fasting). Induced molting lasted 28 days at most, production and quality being monitored for four periods of 28 days thereafter. A completely randomized experimental design with five treatments, eight replicates of eight birds each per plot was adopted. The following experimental treatments were applied until a loss of 26% of body weight was reached: T1 - fasting, T2 - wheat bran ad libitum, T3 - rice bran ad libitum, T4 - cracked rice ad libitum, T5 - ground alfalfa ad libitum. Birds were then fed production diet ad libitum, except for those on treatment T1 (fasting) which received 30, 60 and 100 g/bird/day and then feed ad libitum. During induced molting the birds were exposed to a natural photoperiod and at day 28 that period was increased by 30 minutes/week until reaching 16 hours of light/day. The characteristics evaluated during induced molting were: feed intake, body weight changes and laying percentage. In the post-molt period, performance (feed intake, laying percentage, egg weight, egg mass, feed conversion ratio per dozen and per egg mass and percentage of broken eggs) and egg quality (specific gravity, eggshell breaking strength, percentages of eggshell, yolk, and albumen, eggshell thickness, yolk color and Haugh unit) were evaluated. Every 28 days one egg was collected from each repetition for three consecutive days for quality assessment. The use of rice bran and wheat bran is viable as molting inducers since the birds given those treatments display performance and egg quality similar to those fasted during the induced molting and also because these ingredients promote easier handling, eliminates the need for grinding and feed-mixing equipment and, being less aggressive, provide greater bird welfare.

Objective: Local invasion of bone is a frequent complication of oral squamous cell carcinoma (OSCC). Development of these osteolytic lesions is mediated by osteoclasts. Receptor activation of NF-kappa B ligand (RANKL) signaling, counteracted by osteoprotegerin (OPG), regulates osteoclastogenesis. Previous studies in rodent models have demonstrated that inhibition of RANKL decreases tumor growth and lesions within bone. However, the contributory role of OSCC cells to this disease process has yet to be defined.Methods: RANKL expression was assessed in a panel of OSCC cell lines by qPCR, flow cytometry, and ELISA. Induction of osteoclastogenesis was assessed by co-culture with macrophages or with OSCC-derived conditioned medium. In an animal model of bone invasion, nude mice were injected intratibially with UMSCC-11B cells expressing a RANKL luciferase promoter to detect tumor-derived RANKL activity. Osteolytic lesions were analyzed by X-ray, micro-CT, and histological methods. RANKL expression was assessed in human OSCC tissues by immunohistochemistry.Results: We demonstrated that OSCCs express varied levels of all RANKL isoforms, both membrane-bound and soluble RANKL. Both co-culture and treatment with OSCC-conditioned media induced osteoclastogenesis. In mice, we demonstrated human RANKL promoter activity during bone invasion. Over the course of the experiment, animals suffered osteolytic lesions as RANKL-driven luciferase expression increased with time. After 8 weeks, human-derived RANKL was detected in areas of bone resorption by immunohistochemistry. Similar epithelial RANKL expression was detected in human OSCC tissues.Conclusion: These data demonstrate the ability of OSCCs to produce RANKL, directly altering the tumor microenvironment to increase osteoclastogenesis and mediate local bone invasion. Background: The aim of this study is to characterize and evaluate the host response caused by three different models of experimental periodontitis in mice.Methods: C57BL/6 wild-type female mice were distributed into six experimental groups and sacrificed at 7, 15, and 30 days after the induction of periodontal disease: 1) group C: no treatment control group; 2) group L: periodontal disease induced by ligature; 3) group G-Pg: oral gavage with Porphyromonas gingivalis (Pg); 4) group G-PgFn: oral gavage with Fusobacterium nucleatum + Pg; 5) group I-Pg: heat-killed Pg injected into the palatal mucosa between the molars; and 6) group I-V: phosphatebuffered saline injected into the palatal mucosa. The samples were used to analyze the immune-inflammatory process in the gingival tissue via descriptive histologic and real-time polymerase chain reaction analyses. The alveolar bone loss was evaluated using microcomputed tomography. The data were analyzed using the Kruskal-Wallis test, followed by a post hoc Dunn test and analysis of variance, followed by a Tukey test using a 5% significance level.Results: Only the ligature model displayed significant alveolar bone loss in the initial period (7 days), which was maintained with time. The group injected with heat-killed Pg displayed significant alveolar bone loss starting from day 15, which continued to progress with time (P < 0.05). A significant increase (P < 0.05) in the gene expression of proinflammatory cytokines (interleukin-6 and -1b) and proteins involved in osteoclastogenesis (receptor activator of nuclear factor-kB ligand and osteoprotegerin) was observed in the ligature group on day 7.Conclusion: The ligature and injection of heat-killed Pg models were the most representative of periodontal disease in humans, whereas the oral gavage models were not effective at inducing the disease under the experimental conditions.

Esta pesquisa analisou as diversas soluções para atenuar os níveis de ruído dos tratores e propor um projeto de redução do ruído na sua fonte. Para tal, se propôs projetar, executar e testar alterações em componentes mecânicos causadores de ruído nos tratores, fazendo com que esses níveis se enquadrem em valores considerados salubres. Todas alterações e ensaios foram executados num trator Massey Ferguson MF 290. Para a redução do ruído foram atacados as 3 fontes principais de ruído do trator : o sistema de exaustão de gases, a hélice do sistema de arrefecimento e o sistema de admissão de ar. Como conclusão final pode-se afirmar que é possível atenuar o ruído de tratores agrícolas até níveis salubres (que não causem perda de audição a seus operadores), atuando-se exclusivamente nas fontes de ruído (motor e seus acessórios).
