Proper dorsal–ventral patterning in the developing central nervous system requires signals from both the dorsal and ventral portions of the neural tube. Data from multiple studies have demonstrated that bone morphogenetic proteins (BMPs) and Sonic hedgehog protein are secreted factors that regulate dorsal and ventral specification, respectively, within the caudal neural tube. In the developing rostral central nervous system Sonic hedgehog protein also participates in ventral regionalization; however, the roles of BMPs in the developing brain are less clear. We hypothesized that BMPs also play a role in dorsal specification of the vertebrate forebrain. To test our hypothesis we implanted beads soaked in recombinant BMP5 or BMP4 into the neural tube of the chicken forebrain. Experimental embryos showed a loss of the basal telencephalon that resulted in holoprosencephaly (a single cerebral hemisphere), cyclopia (a single midline eye), and loss of ventral midline structures. In situ hybridization using a panel of probes to genes expressed in the dorsal and ventral forebrain revealed the loss of ventral markers with the maintenance of dorsal markers. Furthermore, we found that the loss of the basal telencephalon was the result of excessive cell death and not a change in cell fates. These data provide evidence that BMP signaling participates in dorsal–ventral patterning of the developing brain in vivo, and disturbances in dorsal–ventral signaling result in specific malformations of the forebrain.

Understanding the mechanism of thermodynamic stability of an RNA structure has significant implications for the function and design of RNA. We investigated the equilibrium folding of a thermophilic ribozyme and its mesophilic homologue by using hydroxyl radical protection, small-angle x-ray scattering, and circular dichroism. Both RNAs require Mg2+ to fold to their native structures that are very similar. The stability is measured as a function of Mg2+ and urea concentrations at different temperatures. The enhanced stability of the thermophilic ribozyme primarily is derived from a tremendous increase in the amount of structure formed in the ultimate folding transition. This increase in structure formation and cooperativity arises because the penultimate and the ultimate folding transitions in the mesophilic ribozyme become linked into a single transition in the folding of the thermophilic ribozyme. Therefore, the starting point, or reference state, for the transition to the native, functional thermophilic ribozyme is significantly less structured. The shift in the reference state, and the resulting increase in folding cooperativity, is likely due to the stabilization of selected native interactions that only form in the ultimate transition. This mechanism of using a less structured intermediate and increased cooperativity to achieve higher functional stability for tertiary RNAs is fundamentally different from that commonly proposed to explain the increased stability of thermophilic proteins.

Experiments were performed to confirm that the aldimine bond formation is a spontaneous reaction, because attempts to find an enzyme catalyzing the last decisive step in betaxanthin biosynthesis, the aldimine formation, failed. Feeding different amino acids to betalain-forming hairy root cultures of yellow beet (Beta vulgaris L. subsp. vulgaris “Golden Beet”) showed that all amino acids (S- and R-forms) led to the corresponding betaxanthins. We observed neither an amino acid specificity nor a stereoselectivity in this process. In addition, increasing the endogenous phenylalanine (Phe) level by feeding the Phe ammonia-lyase inhibitor 2-aminoindan 2-phosphonic acid yielded the Phe-derived betaxanthin. Feeding amino acids or 2-aminoindan 2-phosphonic acid to hypocotyls of fodder beet (B. vulgaris L. subsp. vulgaris “Altamo”) plants led to the same results. Furthermore, feeding cyclo-3-(3,4-dihydroxyphenyl)-alanine (cyclo-Dopa) to these hypocotyls resulted in betanidin formation, indicating that the decisive step in betacyanin formation proceeds spontaneously. Finally, feeding betalamic acid to broad bean (Vicia faba L.) seedlings, which are known to accumulate high levels of Dopa but do not synthesize betaxanthins, resulted in the formation of dopaxanthin. These results indicate that the condensation of betalamic acid with amino acids (possibly including cyclo-Dopa or amines) in planta is a spontaneous, not an enzyme-catalyzed reaction.

An allele of the 1-aminocyclopropane-1-carboxylic acid (ACC) synthase gene (Md-ACS1), the transcript and translated product of which have been identified in ripening apples (Malus domestica), was isolated from a genomic library of the apple cultivar, Golden Delicious. The predicted coding region of this allele (ACS1-2) showed that seven nucleotide substitutions in the corresponding region of ACS1-1 resulted in just one amino acid transition. A 162-bp sequence characterized as a short interspersed repetitive element retrotransposon was inserted in the 5′-flanking region of ACS1-2 corresponding to position −781 in ACS1-1. The XhoI site located near the 3′ end of the predicted coding region of ACS1-2 was absent from the reverse transcriptase-polymerase chain reaction product, revealing that exclusive transcription from ACS1-1 occurs during ripening of cv Golden Delicious fruit. DNA gel-blot and polymerase chain reaction analyses of genomic DNAs showed clearly that apple cultivars were either heterozygous for ACS1-1 and ACS1-2 or homozygous for each type. RNA gel-blot analysis of the ACS1-2 homozygous Fuji apple, which produces little ethylene and has a long storage life, demonstrated that the level of transcription from ACS1-2 during the ripening stage was very low.

To demonstrate that crystallographic methods can be applied to index and interpret diffraction patterns from well-ordered quasicrystals that display non-crystallographic 5-fold symmetry, we have characterized the properties of a series of periodic two-dimensional lattices built from pentagons, called Fibonacci pentilings, which resemble aperiodic Penrose tilings. The computed diffraction patterns from periodic pentilings with moderate size unit cells show decagonal symmetry and are virtually indistinguishable from that of the infinite aperiodic pentiling. We identify the vertices and centers of the pentagons forming the pentiling with the positions of transition metal atoms projected on the plane perpendicular to the decagonal axis of quasicrystals whose structure is related to crystalline η phase alloys. The characteristic length scale of the pentiling lattices, evident from the Patterson (autocorrelation) function, is ∼τ2 times the pentagon edge length, where τ is the golden ratio. Within this distance there are a finite number of local atomic motifs whose structure can be crystallographically refined against the experimentally measured diffraction data.

The dose effect of pure daidzin on the suppression of ethanol intake in Syrian golden hamsters was compared with that of crude daidzin contained in a methanol extract of Radix puerariae (RP). EC50 values estimated from the graded dose-response curves for pure daidzin and RP extract daidzin are 23 and 2.3 mg per hamster per day, respectively. Apparently the antidipsotropic activity of the RP extract cannot be accounted for solely by its daidzin content (22 mg/g). In addition to daidzin, six other isoflavones were identified in the RP extract and quantified--namely, puerarin (160 mg per g of extract), genistin (3.7 mg/g), daidzein (2.6 mg/g), daidzein-4',7-diglucoside (1.2 mg/g), genistein (0.2 mg/g), and formononetin (0.16 mg/g). None of these, administered either alone or combined, contributes in any significant way to the antidipsotropic activity of the extract. Plasma daidzin concentration-time curves determined in hamsters administered various doses of pure daidzin or RP extract by i.p.injection indicate that the crude extract daidzin has approximately 10 times greater bioavailability than the pure compound. Reconstruction of the dose-response effects for pure and crude daidzin using bioavailable daidzin rather than administered dose gives a single curve. Synthetic daidzin added to the RP extract acquires the bioavailability of the endogenous daidzin that exists naturally in the extract. These results show that (i) daidzin is the major active principle in methanol extracts of RP, and (ii) additional constituents in the methanol extract of RP assist uptake of daidzin in golden hamsters.

To examine the role of complement components as regulators of the expression of endothelial adhesive molecules in response to immune complexes (ICs), we determined whether ICs stimulate both endothelial adhesiveness for leukocytes and expression of E-selectin and intercellular and vascular cell adhesion molecules 1 (ICAM-1 and VCAM-1). We found that ICs [bovine serum albumin (BSA)-anti-BSA] stimulated endothelial cell adhesiveness for added leukocytes in the presence of complement-sufficient normal human serum (NHS) but not in the presence of heat-inactivated serum (HIS) or in tissue culture medium alone. Depletion of complement component C3 or C8 from serum did not prevent enhanced endothelial adhesiveness stimulated by ICs. In contrast, depletion of complement component C1q markedly inhibited IC-stimulated endothelial adhesiveness for leukocytes. When the heat-labile complement component C1q was added to HIS, the capacity of ICs to stimulate endothelial adhesiveness for leukocytes was completely restored. Further evidence for the possible role of C1q in mediating the effect of ICs on endothelial cells was the discovery of the presence of the 100- to 126-kDa C1q-binding protein on the surface of endothelial cells (by cytofluorography) and of message for the 33-kDa C1q receptor in resting endothelial cells (by reverse transcription-PCR). Inhibition of protein synthesis by cycloheximide blocked endothelial adhesiveness for leukocytes stimulated by either interleukin 1 or ICs in the presence of NHS. After stimulation with ICs in the presence of NHS, endothelial cells expressed increased numbers of adhesion molecules (E-selectin, ICAM-1, and VCAM-1). Endothelial expression of adhesion molecules mediated, at least in part, endothelial adhesiveness for leukocytes, since leukocyte adhesion was blocked by monoclonal antibodies directed against E-selectin. These studies show that ICs stimulate endothelial cells to express adhesive proteins for leukocytes in the presence of a heat-labile serum factor. That factor appears to be C1q.

Replication-incompetent retroviral vectors encoding histochemical reporter genes have been used for studying lineal relationships in a variety of species. A crucial element in the interpretation of data generated by this method is the identification of sibling relationships, or clonal boundaries. The use of a library of viruses in which each member is unique can greatly facilitate this aspect of the analysis. A previously reported murine retroviral library containing about 80 members demonstrated the utility of the library approach. However, the relatively low number of tags in the murine library necessitated using low infection rates in order to give confidence in clonal assignments. To obviate the need for low infection rates, a far more complex library was created and characterized. The CHAPOL library was constructed such that each member encodes a histochemical reporter gene and has a DNA tag derived from a degenerate oligonucleotide pool synthesized to have a complexity of > 1 x 10(7). The library was tested after infection of cells in vitro or in vivo. The DNA tag from each histochemically labeled cell or clone of cells was recovered by PCR and sequenced for unambiguous identification. Three hundred and twenty tags have been identified after infection, and so far no tag has been seen to result from more than one independent infection. Thus, an equal distribution of inserts is suggested, and Monte Carlo analysis predicts a complexity of > 10(4) members.

El Acceso Abierto en la comunicación académica se viene desarrollando por dos grandes vías: la ruta verde y la ruta dorada. Algunos estudios analizan la extensión y la magnitud de este fenómeno, pero pocos muestran los efectos en términos de impacto y visibilidad. Nuestro objetivo es analizar estos efectos desde la perspectiva del modelo económico de las revistas incluidas en la base de datos Scopus, en Latinoamérica. Los resultados muestran que tanto a nivel internacional como temático, la presencia de las revistas que contribuyen al desarrollo de la "ruta verde" sobrepasan el porcentaje de las revistas de la ruta dorada. Mientras en las regiones periféricas y emergentes, estas últimas revistas predominan ocupando las últimas posiciones de las distribuciones de impacto

This perspective article presents an overview of the Open Access movement in Argentina, from a global and regional (Latin American) context. The article describes the evolution and current state of initiatives by examining two principal approaches to Open Access in Argentina: golden and green roads. The article will then turn its attention to: the support that Open Access receives from governmental sources; collaboration with international projects; and the perspective of Argentine authors regarding Open Access and self-archiving. It concludes with a reflection on the outlook, the main barriers and opportunities for Open Access in Argentina

O Transtorno do Espectro do Autismo (TEA) inclui um conjunto de sintomas, tais como dificuldade para sustentar contato visual direto e comprometimento da linguagem. Apesar da Terapia Assistida Por Animais (TAA) com cães ser considerada uma modalidade terapêutica eficaz para promover o desenvolvimento de pessoas com TEA, ainda não são se sabe quais características dos cães possibilitam alcançar sucesso na terapia. Esta análise quantitativa tem como objetivo verificar o impacto de abordagens laterais e frontais de cães e humanos nas expressões emocionais de alegria e rejeição de crianças com TEA. Através da análise de vídeos de TAA, foram mensuradas duração e frequência das abordagens laterais e frontais de cães e humanos dirigidas às crianças para comparar possíveis diferenças entre ambos e também para verificar se a abordagem escolhida afetava o tipo de expressão emocional exibida pela criança. Os participantes deste projeto foram 11 crianças, 8 do sexo masculino e 3 do sexo feminino, entre 5 e 11 anos. Seis crianças foram atendidas por uma psicóloga, uma condutora e um Border Collie. O segundo grupo era composto pela mesma psicóloga, uma condutora e uma Golden Retriever. Escalas de avaliação foram aplicadas para confirmar o diagnóstico de TEA. Os cães foram previamente avaliados e treinados por uma instituição que atua na área de TAA. Cinco minutos de 8 sessões foram analisadas: um bloco de seis sessões com o cão, uma sessão anterior e uma sessão posterior a este bloco. Para verificar possíveis diferenças temperamentais entre cães, o C-barq (Canine Behavioral Assesment & Research Questionnaire) foi aplicado para analisar o temperamento de ambos. Embora esta análise tenha demonstrado diferenças em relação às categorias busca de atenção e nível de energia dos cães, não foram verificadas diferenças estatísticas entre os cães, em relação às variáveis analisadas neste estudo. Na comparação entre cães e humanos, os cães foram mais efetivos para conseguir expressões de alegria independentemente do tipo de abordagem escolhida. Comparando-se o tempo de abordagem de cães e humanos até obterem expressão emocional das crianças, observou-se uma importante diferença estatística. Os resultados sugerem que os cães exibiram menor latência que humanos para todas expressões emocionais analisadas: alegria (2= 7,312, p=0,007), de rejeição (2= 11,277, p-0,001) e neutras (2=9,097, p=0,043). Além disso, os resultados sugerem que, no contexto da TAA, não há relação entre abordagem lateral ou frontal e expressões de alegria, rejeição ou neutras de crianças com TEA. As expressões de alegria foram mais frequentes diante das abordagens laterais dos cães do que das abordagens frontais, no entanto não foi verificada significância estatística. Em relação aos humanos também não foi verificada preferência por uma abordagem especifica. Assim, os resultados sugerem que a latência para a exibição de uma expressão emocional das crianças depende mais de quem aborda do que do posicionamento lateral ou frontal quando a abordagem é realizada

Esta tese analisa a poesia de Joaquim de Sousa Andrade (1832-1902), ou Sousândrade, e tem por objetivo discutir o imaginário político republicano como elo de ligação entre os poemas O Guesa, especialmente o Canto Décimo (1877), Novo Éden: Poemeto da adolescência, 1888-1889 (1893), Harpa de Ouro (189?) e a continuação O Guesa, o Zac (1902). A cronologia dessas obras indica que é a partir dos anos vividos por Sousândrade nos Estados Unidos que as figurações da República passam a permear os seus escritos. No Canto Décimo, gestado em Nova York, Sousândrade traça um panorama da sociedade estadunidense na década de 1870 e, embora haja críticas à certas práticas econômicas nesse canto, conforme discutiremos com vagar, destaca-se a emulação do poeta em exaltar aquele modelo sociopolítico. Essa atitude do escritor maranhense afinava-se com a visão de brasileiros imigrados nos Estados Unidos naquele mesmo período, os quais formavam uma rede de pensadores liberais comprometidos com a modernização do Brasil. Esse diálogo transatlântico que começa a ser traçado entre Estados Unidos e Brasil caracteriza uma alternativa ao modelo de civilização europeu até então em voga, tema ainda pouco explorado pela historiografia literária. De volta a pátria, Sousândrade dá prosseguimento ao seu fazer poético e, além do seu engajamento na nova conjuntura política do país, dedica-se à construção do mito fundador da República brasileira nos poemas concebidos desde 1893. A trajetória do poeta republicano foi marcada pelo seu anseio de progresso e modernização do Brasil, tarefa na qual ele empenhou-se política e literariamente.

La enfermedad cardiovascular sigue siendo la principal causa de morbilidad y mortalidad a nivel mundial en países desarrollados, fundamentalmente en pacientes con DM tipo 2, en algunas poblaciones puede representar el 50% o más de las muertes por diabetes (Joseph and Golden, 2014). Esto se debe en gran medida a factores ya conocidos como la predisposición genética, la aterogénesis acelerada, la inflamación crónica, la isquemia silente y la presencia de patologías co-existentes como la hipertensión o la dislipidemia. La diabetes es sin duda alguna, uno de los problemas de salud más graves del siglo XXI y actualmente en España, la prevalencia nacional es de 10,83% (FDI 2013). Las células progenitoras endoteliales juegan un papel clave en los procesos de reparación endotelial. En los pacientes con DM tipo 2 y enfermedad cardiovascular, se sabe que la funcionalidad de las EPCs es deficiente, aunque el mecanismo exacto de disfunción aún es incierto. Además, está bien descrito que en la evolución natural de los pacientes con DM tipo 2 presentan un mayor número de complicaciones y con mayor frecuencia estos pacientes estarán abocados a procedimientos de revascularización. Múltiples estudios (Sidhu and Boden, 2015; Verma et al., 2013) que han señalado la importancia de una adecuada terapia de reparación endotelial (terapia con EPCs), que ayudaría a disminuir las alteraciones en los procesos de reendotelización en los pacientes con DM tipo 2 y enfermedad cardiovascular, y por consiguiente disminuiría la aparición de la enfermedad cardiovascular (ECV)...

El inicio del siglo XXI supone una nueva época dorada para el retrato fotográfico, puesto que aparece de forma masiva entre los usuarios, asentándose como un hecho social muy arraigado entre la sociedad. En el estudio analizamos algunas de las funciones culturales que cumple entre las que encontramos la de reconstruir nuestra memoria personal a través de imágenes de nuestra vida, servir como medio de comunicación y publicidad a través del cual nos relacionamos con los demás gracias a las redes sociales e internet, y por último su faceta artística en la que camina por senderos de total libertad permeabilizando sus fronteras con otros géneros fotográficos, y sirviendo de base para recoger diversas temáticas ajenas –en principio- al propio retrato.
