Purpose: To evaluate the clinical and radiologic response of patients with Graves' ophthalmopathy given low-dose orbital radiotherapy (RT) with a protracted fractionation.Methods and Materials: Eighteen patients (36 orbits) received orbital RT with a total dose of 10 Gy, fractionated in 1 Gy once a week over 10 weeks. of these, 9 patients received steroid therapy as well. Patients were evaluated clinically and radiologically at 6 months after treatment. Clinical response assessment was carried out using three criteria: by physical examination, by a modified clinical activity score, and by a verbal questionnaire considering the 10 most common signs and symptoms of the disease. Radiologic response was assessed by magnetic resonance imaging.Results: Improvement in ocular pain, palpebral edema, visual acuity, and ocular motility was observed in all patients. Significant decrease in symptoms such as tearing (p < 0.001) diplopia (p = 0.008), conjunctival hyperemia (p = 0.002), and ocular grittiness (p = 0.031) also occurred. Magnetic resonance imaging showed decrease in ocular muscle thickness and in the intensity of the T2 sequence signal in the majority of patients. Treatments were well tolerated, and to date no complications from treatment have been observed. There was no statistical difference in clinical and radiologic response between patients receiving RT alone and those receiving RT plus steroid therapy.Conclusion: RT delivered in at a low dose and in a protracted scheme should be considered as a useful therapeutic option for patients with Graves' ophthalmopathy. BACKGROUND: Despite numerous studies on endotracheal tube cuff pressure (CP) management, the literature has yet to establish a technique capable of adequately tilling the cuff with an appropriate volume of air while generating low CP in a less subjective way. the purpose of this prospective study was to evaluate and compare the CP levels and air volume required to fill the endotracheal tubes cuff using 2 different techniques (volume-time curve versus minimal occlusive volume) in the immediate postoperative period after coronary artery bypass grafting. METHODS: A total of 267 subjects were analyzed. After the surgery, the lungs were ventilated using pressure controlled continuous mandatory ventilation, and the same ventilatory parameters were adjusted. Upon arrival in the ICU, the cuff was completely deflated and re-inflated, and at this point the volume of air to fill the cuff was adjusted using one of 2 randomly selected techniques: volume-time curve and minimal occlusive volume. We measured the volume of air injected into the cuff, the CP, and the expired tidal volume of the mechanical ventilation after the application of each technique. RESULTS: the volume-time curve technique demonstrated a significantly lower CP and a lower volume of air injected into the cuff, compared to the minimal occlusive volume technique (P < .001). No significant difference was observed in the expired tidal volume between the 2 techniques (P = .052). However, when the subjects were submitted to the minimal occlusive volume technique, 17% (n = 47) experienced air leakage as observed by the volume-time graph. CONCLUSIONS: the volume-time curve technique was associated with a lower CP and a lower volume of air injected into the cuff, when compared to the minimal occlusive volume technique in the immediate postoperative period after coronary artery bypass grafting. Therefore, the volume-time curve may be a more reliable alternative for endotracheal tube cuff management.

AIM: To evaluate the suitability of reference genes in gastric tissue samples and cell lines.METHODS: the suitability of genes ACTB, B2M, GAPDH, RPL29, and 18S rRNA was assessed in 21 matched pairs of neoplastic and adjacent nonneoplastic gastric tissues from patients with gastric adenocarcinoma, 27 normal gastric tissues from patients without cancer, and 4 cell lines using reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR). the ranking of the best single and combination of reference genes was determined by NormFinder, geNorm (TM), BestKeeper, and DataAssist (TM). in addition, GenEx software was used to determine the optimal number of reference genes. To validate the results, the mRNA expression of a target gene, DNMT1, was quantified using the different reference gene combinations suggested by the various software packages for normalization.RESULTS: ACTB was the best reference gene for all gastric tissues, cell lines and all gastric tissues plus cell lines. GAPDH + B2M or ACTB + B2M was the best combination of reference genes for all the gastric tissues. On the other hand, ACTB + B2M was the best combination for all the cell lines tested and was also the best combination for analyses involving all the gastric tissues plus cell lines. According to the GenEx software, 2 or 3 genes were the optimal number of references genes for all the gastric tissues. the relative quantification of DNMT1 showed similar patterns when normalized by each combination of reference genes. the level of expression of DNMT1 in neoplastic, adjacent non-neoplastic and normal gastric tissues did not differ when these samples were normalized using GAPDH + B2M (P = 0.32), ACTB + B2M (P = 0.61), or GAPDH + B2M + ACTB (P = 0.44).CONCLUSION: GAPDH + B2M or ACTB + B2M is the best combination of reference gene for all the gastric tissues, and ACTB + B2M is the best combination for the cell lines tested. Objective: the aim of this study was to quantify mast cells at different time intervals after partial Achilles tendon rupture in rats treated with low-level laser therapy (LLLT). Background data: There is a high incidence of lesions and ruptures in the Achilles tendon that can take weeks and even months to heal completely. As the mast cells help in the healing repair phase, and LLLT has favorable effects on this tissue repair process, study of this modality on the quantity of mastocytes in the ruptured tendon is relevant. Methods: Sixty Wistar rats were subjected to partial Achilles' tendon rupture by direct trauma, randomized into 10 groups, and then divided into the group treated with 80mW aluminum gallium arsenide infrared laser diode, continuous wave, 2.8W/cm(2) power density, 40J/cm(2) energy density, and 1.12J total energy, and the simulation group. Both the groups were subdivided according to the histological assessment period of the sample, either 6h, 12h, 24h, 2 days, or 3 days after the rupture, to quantify the mastocytes in the Achilles' tendon. Results: the group subjected to LLLT presented a greater quantity of mastocytes in the periods of 6h, 12h, 24h, 2 days, and 3 days after rupture, compared with the simulation groups, but differences were detected between the sample assessment periods only in the simulation group. Conclusions: LLLT was shown to increase the quantity of mastocytes in the assessment periods compared with the simulation groups.

BACKGROUND: Previous investigation showed that the volume-time curve technique could be an alternative for endotracheal tube (ETT) cuff management. However, the clinical impact of the volume-time curve application has not been documented. the purpose of this study was to compare the occurrence and intensity of a sore throat, cough, thoracic pain, and pulmonary function between these 2 techniques for ETT cuff management: volume-time curve technique versus minimal occlusive volume (MOV) technique after coronary artery bypass grafting. METHODS: A total of 450 subjects were randomized into 2 groups for cuff management after intubation: MOV group (n = 222) and volume-time curve group (n = 228). We measured cuff pressure before extubation. We performed spirometry 24 h before and after surgery. We graded sore throat and cough according to a 4-point scale at 1, 24, 72, and 120 h after extubation and assessed thoracic pain at 24 h after extubation and quantified the level of pain by a 10-point scale. RESULTS: the volume-time curve group presented significantly lower cuff pressure (30.9 +/- 2.8 vs 37.7 +/- 3.4 cm H2O), less incidence and intensity of sore throat (1 h, 23.7 vs 51.4%; and 24 h, 18.9 vs 40.5%, P < .001), cough (1 h, 19.3 vs 48.6%; and 24 h, 18.4 vs 42.3%, P < .001), thoracic pain (5.2 +/- 1.8 vs 7.1 +/- 1.7), better preservation of FVC (49.5 +/- 9.9 vs 41.8 +/- 12.9%, P = .005), and FEV1, (46.6 +/- 1.8 vs 38.6 +/- 1.4%, P = .005) compared with the MOV group. CONCLUSIONS: the subjects who received the volume-time curve technique for ETT cuff management presented a significantly lower incidence and severity of sore throat and cough, less thoracic pain, and minimally impaired pulmonary function than those subjects who received the MOV technique during the first 24 h after coronary artery bypass grafting.

RESUMO: Este trabalho teve como objetivo comparar a severidade da mancha de Alternaria de genótipos de girassol em ambientes do Cerrado do Distrito Federal. Três experimentos foram avaliados, dois na Embrapa Cerrados (Planaltina, DF) e um localizado na Embrapa Produtos e Mercado (Recanto das Emas, DF). Foram feitas análises ao longo do ciclo da cultura com um intervalo de 15 dias, sendo a primeira aos 35 dias após emergência. Cada planta foi analisada em três partes: terço inferior, terço médio e terço superior. Ao final de cada experimento, a área abaixo da curva de progresso da doença (AACPD) foi calculada para cada genótipo. De acordo com os resultados observados, o ambiente de sequeiro na Embrapa Cerrados, que deteve a maior média da severidade da doença, entre os ambientes avaliados. Dentre os genótipos avaliados, MG 360 mostrou menor severidade da doença no sequeiro e AGUARA 06 no irrigado da Embrapa Cerrados. O genótipo BSG 42 na Embrapa Produtos e Mercado, se destacou pela menor severidade do fungo. Houve influência significativa do ambiente nos resultados de severidade da doença, que está estreitamente relacionada com os fatores climáticos como temperatura e umidade relativa do ar. A compreensão dessas condições favoráveis ao desenvolvimento do fungo é essencial quando se avalia a resistência em genótipos de girassol. ABSTRACT: This study had the purpose of comparing the severity of Alternaria leaf spot on genotypes of sunflower in the Brazilian Cerrado. Three field trials were established, two at Embrapa Cerrados (Planaltina, DF) and one at Embrapa Produtos e Mercados (Recanto das Emas, DF). Evaluations were made during the crop cycle every 15 days, the first one happening 35 days after crop emergency. Each plant was analyzed in three parts: lower, middle and upper thirds. At the end of each trial the average area under the disease progress curve (AUDPC), for each third, was calculated for all genotypes. According to the observed results, the dryland environment at Embrapa Cerrados had the highest mean disease severity, among the studied environments. Between the evaluated genotypes, MG360 showed less severity in dryland and AGUARA 06 in the irrigated environment at Embrapa Cerrados. At Embrapa Produtos e Mercado the genotype that presented the best performance was BRS G42. Environmental conditions deeply affected the trials, mainly temperature and relative humidity. Their influence on Alternaria leaf spot epidemics in the Cerrado region must be studied in more detail, to avoid misinterpreting data when evaluating sunflower genotypes for resistance to this important fungal disease.

RESUMO: O presente trabalho tem por objetivo caracterizar o comportamento de 16 genótipos de girassol em ambiente de sequeiro e ambiente irrigado do Cerrado do Distrito Federal visando aumentar disponibilidade de cultivares mais produtivas e adaptadas. Os ensaios foram conduzidos na área experimental da Embrapa Cerrados, Planaltina, DF. Os ensaios foram arranjados experimentalmente em blocos ao acaso, com quatro repetições. Os genótipos avaliados foram: CF 101, ADV 5504, BRS G42, BRS 323, HELIO 250, HELIO 251, SYN 045, SYN 3950HO, MG 305, MG 360, AGUARÁ 04, AGUARÁ 06, PARAÍSO 20, GNZ NEON, HLA 2012 e M734. Os caracteres avaliados foram rendimento estimado de grãos, tamanho do capítulo, peso de mil aquênios, altura de plantas, teor de óleo e dias para floração inicial. Houve diferenças significativas entre os genótipos para todas as características avaliadas. Dos 16 genótipos avaliados, os híbridos SYN 045 (3.786,2 kg ha-1) e MG 305 (4.987,2 kg ha-1) se sobressaíram quanto ao rendimento estimado de grãos. Quanto ao teor de óleo, os híbridos MG 306 (53,95%) e SYN 3950HO (49,49%) se destacaram. Foram identificados os genótipos mais promissores dentre os avaliados, podendo ser explorados em programas de melhoramento que visam o desenvolvimento de cultivares mais adaptadas.

RESUMO: O objetivo deste trabalho foi estimar caracteres genéticos, fenotípicos e ambientais relacionados aos componentes de produção e caracteres agromorfológicos de girassol em três núcleos rurais do Distrito Federal. Os experimentos foram conduzidos nas áreas experimentais da Embrapa Cerrados, Planaltina, DF, Embrapa Produtos e Mercado, Recanto das Emas, DF e na Fazenda Agua Limpa, da Universidade de Brasília. Através dos resultados obtidos, foram verificadas diferenças significativas entre os genótipos de girassol nos três núcleos rurais do Cerrado para todas as características agronômicas avaliadas. Ainda, baixos coeficientes de variação ambiental para quase todas as características, exceto para o tamanho de capitulo, indicaram boa precisão experimental e altos valores de herdabilidade, coeficientes de variação genéticos e acurácia evidenciaram condições favoráveis à seleção dos materiais para as características agronômicas avaliadas. ABSTRACT: The purpose of this study was to assess genetic, phenotypic and environmental characteristics related to agro-morphological traits of sunflower in three rural centers of Distrito Federal. The experiments were conducted at the experimental areas of Embrapa Cerrados, Planaltina, DF, Embrapa Produtos e Mercado, Recanto das Emas, DF e na Fazenda Agua Limpa, da Universidade de Brasília. Through the obtained results, were verified significate genotypic differences of sunflower at the three rural centers of Brazilian Savannah for all traits evaluated. In addition, low coefficients of environmental variation for almost all trait, except the head size, indicate good experimental precision and high values of heritability, genetic variation and accuracy showed favorable conditions to selecting materials for the agronomic traits evaluated.

RESUMO:Com o objetivo de avaliar o comportamento agronômico de genótipos de girassol no Cerrado do Distrito Federal, foram conduzidos ensaios na safrinha dos anos de 2014 e 2015, na estação experimental da Embrapa Cerrados, Planaltina, DF. O delineamento experimental foi de blocos ao acaso com quatro repetições, e foram avaliados 12 genótipos de girassol: HLA 2015, NTC 90, SYN 065, M734, BRS G44, HLA 2014, BRS G45, BRS G43, HLA 2013, HLA 2017, BRS G46, HLA 2016. As características avaliadas foram rendimento de grãos, tamanho do capitulo, peso de mil aquênios, altura de plantas e dias de floração inicial. Diferenças significativas foram encontradas para as características avaliadas. Os genótipos que se destacaram em relação ao rendimento de grãos foram HLA 2014 (3.161 kg ha-1) e a testemunha M743 (3.212 kg ha-1). Além disso, o ensaio do ano 2014 apresentou uma média de rendimento maior (2.829 kg ha-1) e mais precoces (63,10 dias) em relação a 2015. O trabalho permitiu a identificação de materiais promissores para exploração em programas de melhoramento genético. ABSTRACT: Aiming the evaluation on agronomic behavior of sunflower genotypes in the Brazilian savannah, experiments were carried on in the second crop of 2014 and 2015 at Centro de Pesquisa Agropecuária dos Cerrados (Embrapa), Planaltina, DF. A complete randomized block design was used with four replications and 12 genotypes of sunflower were analyzed: HLA 2015, NTC 90, SYN 065, M734, BRS G44, HLA 2014, BRS G45, BRS G43, HLA 2013, HLA 2017, BRS G46, HLA 2016.The evaluated characteristics were grain yield, head, weight thousand achenes, plant height, and flowering time. Significant differences were found in all evaluated characteristics. The genotypes that stood out in seed yield were HLA 2014 (3161 kg ha-1) and M743 (3212 kg ha-1). Besides, the 2014 experiment presented a seed yield average higher (2829 kg ha-1), and earlier flowering (63,10 days) when compared to 2015 experiment. This study allowed the identification of promising materials to explore in breeding programs.

The creative industries sector faces a constantly changing context characterised by the speed of the development and deployment of digital information systems and Information Communications Technologies (ICT) on a global scale. This continuous digital disruption has had significant impact on the whole value chain of the sector: creation and production; discovery and distribution; and consumption of cultural goods and services. As a result, creative enterprises must evolve business and operational models and practices to be sustainable. Enterprises of all scales, type, and operational model are affected, and all sectors face ongoing digital disruption. Management consultancy practitioners and business strategy academics have called for new strategy development frameworks and toolkits, fit for a continuously changing world. This thesis investigates a novel approach to organisational change appropriate to the digital age, in the context of the creative sector in Scotland. A set of concepts, methods, tools, and processes to generate theoretical learning and practical knowing was created to support enterprises to digitally adapt through undertaking journeys of change and organisational development. The framework is called The AmbITion Approach. It was developed by blending participatory action research (PAR) methods and modern management consultancy, design, and creative practices. Empirical work also introduced to the framework Coghlan and Rashford’s change categories. These enabled the definition and description of the extent to which organisations developed: whether they experienced first order (change), second order (adaptation) or third order (transformation) change. Digital research tools for inquiry were tested by a pilot study, and then embedded in a longitudinal study over two years of twentyone participant organisations from Scotland’s creative sector. The author applied and investigated the novel approach in a national digital development programme for Scotland’s creative industries. The programme was designed and delivered by the author and ran nationally between 2012-14. Detailed grounded thematic analysis of the data corpus was undertaken, along with analysis of rich media case studies produced by the organisations about their change journeys. The results of studies on participants, and validation criteria applied to the results, demonstrated that the framework triggers second (adaptation) and third order change (transformation) in creative industry enterprises. The AmbITion Approach framework is suitable for the continuing landscape of digital disruption within the creative sector. The thesis contributes to practice the concepts, methods, tools, and processes of The AmbITion Approach, which have been empirically tested in the field, and validated as a new framework for business transformation in a digital age. The thesis contributes to knowledge a theoretical and conceptual framework with a specific set of constructs and criteria that define first, second, and third order change in creative enterprises, and a robust research and action framework for the analysis of the quality, validity and change achieved by action research based development programmes. The thesis additionally contributes to the practice of research, adding to our understanding of the value of PAR and design thinking approaches and creative practices as methods for change.
