A novel diagnosis method (by NGS) is being implemented in order to study a larger panel of genes for dyslipidaemia to improve patient diagnosis

O presente trabalho congrega, em si, o estabelecimento de uma ligação entre as áreas científica e artística, justificando o trabalho do actor com elementos inerentes à anatomia humana e aos progressos na ciência sobre o estudo do comportamento e cérebro humanos. A arte do actor é, neste estudo, vista como uma ciência de palco desenvolvida a partir do esqueleto e do corpo em vida, atentos aos impulsos psico-fisicos, prolongando a acção quotidiana na acção extra-quotidiana. Se na ciência a menor unidade, viva, do organismo humano é a célula, este estudo reclama o impulso como a unidade mínima do teatro e, por conseguinte, do trabalho de actor. Esta análise complementa-se, perspectivando a célula vivente como o núcleo da relação entre o invisível, como processo mental, e o visível como processo e manifestação física do trabalho do actor dentro e fora do palco. ABSTRACT; The present work congregates the creation of a connection between scientific and artistic areas, justifying the actor's work through characteristic elements of human anatomy and through the scientific advances on the study of the human brain and of human behaviour. ln this study, the actor's art, is viewed as a stage science based on the human skeleton and on the living body, both conscious of the psycho-physical impulses that extend everyday action to the extraordinary action. If in science the smallest living unit in the human body is the cell, then, this study argues that the impulse is the smallest unit in theater and therefore, of the actor's work. This analysis complements itself, envisaging the living cell as the core of the relationship between the invisible, as a mental process, and the visible, as a process of physical manifestation of the actor's work in(side) and out(side) of stage.

The aim of this study was to evaluate the level of awareness towards cholesterol levels in a Portuguese sample and to assess targets for interventions with gains in cardiovascular health.

This study aims to acknowledge the domain level and influence of the neuromarketing construct. This is done considering professionals at advertising agencies in Brazil. The presence of concepts related to this new approach is very little divulged, and there are little analysis performed on this area. Thus, the research is of qualitative and exploratory nature and used as primary fonts books, articles related to marketing, neuroscience, and psychology as well as secondary fonts. A profound interview was realized aiming the main advertising agencies in Brazil. The public was composed by managers responsible for planning. A content analysis was performed afterwards. The advances related to the brain science have permitted the development of technological innovation. These go primarily towards knowledge and unconscious experiences of consumers, which are responsible for the impulse of decision making and consumer behavior. These issues are related to Neuromarketing, that in turn, uses techniques such as FMRI, PET and FDOT. These scan the consumer s brain and produces imagines on the neuron s structures and functioning. This is seen while activities such as mental tasks for the visualization of brands, images or products, watching videos and commercials are performed. It is observed that the agencies are constantly in search of new technologies and are aware of the limitations of the current research instruments. On the other hand, they are not totally familiar with concepts related to neuromarketing. In relation to the neuroimage techniques it is pointed out by the research that there is full unawareness, but some agencies seem to visualize positive impacts with the use of these techniques for the evaluation of films and in ways that permit to know the consumer better. It is also seen that neuroimage is perceived as a technique amongst others, but its application is not real, there are some barriers in the market and in the agencies itself. These barriers as well as some questioning allied to the scarce knowledge of neuromarketing, make it not possible to be put into practice in the advertising market. It is also observed that even though there is greater use of neuromarketing; there would not be any meaningful changes in functioning and structuring of these agencies. The use of the neuro-image machines should be done in research institutes and centers of big companies. Results show that the level of domain of the neuromarketing construct in the Brazilian advertising agencies is only a theoretical one. Little is known of this subject and the neurological studies and absolutely nothing of neuroimage techniques

However, even though unrepaired UV-induced DNA damages are related to mutagenesis, cell death and tumorigenesis, they do not explain phenotypes such as neurodegeneration and internal tumors observed in patients with syndromes like Xeroderma Pigmentosum (XP) and Cockayne Syndrome (CS) that are associated with NER deficiency. Recent evidences point to a role of NER in the repair of 8-oxodG, a typical substrate of Base Excision Repair (BER). Since deficiencies in BER result in genomic instability, neurodegenerative diseases and cancer, it was investigated in this research the impact of XPC deficiency on BER functions in human cells. It was analyzed both the expression and the cellular localization of APE1, OGG1 e PARP-1, the mainly BER enzymes, in different NER-deficient human fibroblasts. The endogenous levels of these enzymes are reduced in XPC deficient cells. Surprisingly, XP-C fibroblasts were more resistant to oxidative agents than the other NER deficient fibroblasts, despite presenting the highest of 8-oxodG. Furthermore, subtle changes in the nuclear and mitochondrial localization of APE1 were detected in XP-C fibroblasts. To confirm the impact of XPC deficiency in the regulation of APE1 and OGG1 expression and activity, we constructed a XPC-complemented cell line. Although the XPC complementation was only partial, we found that XPC-complemented cells presented increased levels of OGG1 than XPC-deficient cells. The extracts from XPC-complemented cells also presented an elevated OGG1 enzimatic activity. However, it was not observed changes in APE1 expression and activity in the XPCcomplemented cells. In addition, we found that full-length APE1 (37 kDa) and OGG1- α are in the mitochondria of XPC-deficient fibroblasts and XPC-complemented fibroblasts before and after induction of oxidative stress. On the other hand, the expression of APE1 and PARP-1 are not altered in brain and liver of XPC knockout mice. However, XPC deficiency changed the APE1 localization in hypoccampus and hypothalamus. We also observed a physical interaction between XPC and APE1 proteins in human cells. In conclusion, the data suggest that XPC protein has a role in the regulation of OGG1 expression and activity in human cells and is involved mainly in the regulation of APE1 localization in mice. Aditionally, the response of NER deficient cells under oxidative stress may not be only associated to the NER deficiency per se, but it may include the new functions of NER enzymes in regulation of expression and cell localization of BER proteins

Neste trabalho foram analisados os parâmetros cinéticos e a atividade colinesterásica de duas espécies de peixes estuarinos: a corvina Micropogonias furnieri (Teleostei, Scianidae) e o bagre Cathorops spixii (Teleostei, Ariidae), buscando avaliar o uso destas espécies como bioindicadores da presença de compostos anticolinesterásicos no meio aquático. Os exemplares de corvina e bagre, respectivamente, foram coletados nos estados do RS (Lagoa dos Patos) e PR (Baía de Paranaguá) no inverno (corvina) e verão (corvina e bagre) em dois pontos de coleta: um local controle e outro poluído. Os peixes foram anestesiados (bezocaína, 200 ppm) e os cérebros dissecados, homogeneizados e centrifugados. Foram estimados os parâmetros cinéticos (Vmax e Kmap), utilizando iodeto de acetiltiocolina como substrato, nas seguintes concentrações: 0,025; 0,05; 0,2; 0,8; 1,6; 3,2 e 9 mM. Nos estudos de inibição enzimática foi utilizado o carbamato eserina em concentrações de 0,3 a 10 mM e de 1x10-4 a 1 µM, a fim de determinar os parâmetros cinéticos de inibição e a concentração de eserina que inibia 50% da atividade colinesterásica (CI50), respectivamente. Os resultados mostraram pouca variação nos valores de Kmap nas duas espécies, porém diferenças significativas nos valores de CI50 foram observadas, indicando que a ChE do cérebro da corvina M. furnieri é resistente à inibição por eserina. Nos estudos de cinética de inibição da ChE do cérebro do bagre C. spixii, foram encontradas diferenças entre alguns parâmetros, quando foram comparados os peixes coletados no local controle com aqueles coletados no local poluído. Houve uma maior atividade colinesterásica de bagres coletados no local poluído (p<0,05), sendo que o mesmo resultado foi observado em exemplares de corvinas M. furnieri coletados durante o inverno no local poluído. Os resultados obtidos in vitro demonstram que a ChE de M. furnieri possui pouca sensibilidade à eserina. 5 Porém, o fato de ter sido registrada inibição colinesterásica nos organismos coletados durante o verão região poluída sugere a possibilidade de alterações significativas na capacidade de bio-oxidação de pesticidas nesta espécie, ou a possibilidade de inibição por outros contaminantes que também afetam a atividade colinesterásica, como por exemplo, metais. No caso de C. spixii, a determinação dos parâmetros cinéticos e de atividade colinesterásica sugere que pode estar ocorrendo alterações bioquímicas nos peixes previamente expostos a contaminantes no ambiente, provavelmente devido a respostas adaptativas ao ambiente impactado. Desta forma, o presente estudo indica a importância de estudos cinéticos prévios, quando se utiliza ou pretende utilizar, a atividade colinesterásica como bioindicador da presença de compostos anticolinesterásicos no ambiente em espécies aquáticas.
