Organisaatio, prosessimalli ja menetelmät vaikuttavat toisiinsa sekä suorasti että prosessien ja tavoitteiden kautta epäsuorasti. Prosessimallit vaihtelevat eri organisaatioiden välillä, mutta työkalut ja menetelmät, erityisesti toteutusmenetelmät, saattavat vaihdella jopa eri projektien ja sovelluskehittäjien välillä. Toteutusmenetelmien yhtenäistämisellä tavoitellaan ohjelmistokehityksen tehokkuuden parantamista, ohjelmistojen laatutason nostamista ja työmotivaation kohottamista. Tämän diplomityön käytännön osuudessa selvitettiin ohjelmistokehitysorganisaation asenteita ja edellytyksiä toteutusmenetelmien yhtenäistämistä kohtaan. Diplomityön tuloksena laadittiin suositus siitä, kuinka parhaat käytännöt -dokumentti voidaan toteuttaa. Suosituksen mukaan kyseinen dokumentti tulisi jakaa kahdeksi dokumentiksi siten, että toinen dokumenteista kattaisi käytännöllisimmät toteutusmenetelmät, toinen sisältäisi suunnittelumenetelmät.

Tässä työssä esitetään tuloilmalaitteen virtausten laskennallinen tarkastelu huonetilassa virtauslaskentaohjelmiston avulla. Tarkastelu suoritetaan kolmiulotteisena sekä isotermisille että termisille tapauksille, jolloin huonetilan joihinkin pintoihin asetetaan lämpövuo ja epäpuhtauslähde. Työn tarkoituksena on arvioida CFX -virtauslaskentaohjelmiston soveltuvuutta huonetilavirtausten mallintamiseen ja tarkastella huonetilan ilmanvaihdon tehokkuutta eri tapauksissa. Arviointi suoritetaan vertaamalla laskentatuloksia mittaustuloksiin. Turbulenssimallien vaikutusta on arvioitu käyttämällä nollan yhtälön ja standardi k-e turbulenssimalleja. CFX:n k-e turbulenssimallilla lasketut nopeuden alenemat eri tilavuusvirroilla vastasivat paremmin mittaustuloksia kuin nollan yhtälön turbulenssimallilla, jolla erot olivat huomattavia. Huonetilan ilmanvaihdon tehokkuus vaihteli suuresti tuloilmalaitteen sijoituspaikan ja lämpövuon mukaan.

Tutkielmassa kartoitetaan tekijöitä, jotka vaikuttavat yrityksen kiinteistönhoidon make-or-buy –päätöksiin. Tutkielman päätavoitteena on selvittää case –yrityksen kiinteistönhoidon toimintoperusteiset kustannukset. Osatavoitteena testataan toimintoperusteisen kustannuslaskennan soveltuvuutta yrityksen tukitoimintojen kustannuslaskennassa. Tutkielma on tyypiltään laadullinen tutkimus ja tutkimusotteena on käytetty konstruktiivista tutkimusotetta. Tutkimusmetodiltaan työ on case-tutkimus. Tutkielmassa rakennetaan toimintoperusteinen kustannuslaskentamalli, jolla selvitetään case -yrityksen kiinteistönhoidon palveluiden tuottamisesta aiheutuvat kustannukset. Laskentatulosten pohjalta todetaan ne toiminnot, joihin kiinteistönhoidon järjestäminen case -yrityksessä sitoo rahaa ja joihin tulisi kohdistaa kustannusten leikkaamistoimenpiteet. Kiinteistönhoidon toimintoja analysoidaan yksitellen sekä toiminnon toimintoperusteisten kustannusten kautta että etsien tekijöitä, jotka vaikuttavat palvelun itse tekemiseen tai palvelun ostamiseen ulkopuoliselta toimittajalta. Tutkielmassa ei tehdä kannattavuusvertailuja itse tekemisen ja ostopalveluiden välillä.

Tämän tutkielman tavoitteena oli tarkastella tapahtumamarkkinoinnin suunnittelua osana yrityksen markkinointiviestintästrategiaa. Tutkielma jakautuu teoreettiseen ja empiiriseen osaan. Teoriaosassa muodostettavan tapahtumamarkkinoinnin suunnittelumallin avulla kuvataan empiriassa kohdeyrityksen tapahtumamarkkinoinnin suunnittelua osana yrityksen markkinointiviestintästrategiaa. Teoriaosan lähdeaineistona käytettiin keskeistä tapahtumamarkkinointia ja markkinointiviestintää käsittelevää kirjallisuutta. Tutkielman empiirinen aineisto kerättiin haastattelemalla kohdeyrityksen markkinointipäällikköä ja hyödyntämällä sekundääritietolähteitä. Tutkielman lähtökohtana oli ensin selvittää tapahtumamarkkinoinnin käsite sekä sen suhde markkinointiviestintään ennen varsinaisen suunnittelumallin muodostamista. Havaittiin, että tapahtumamarkkinointi käsitteenä on erittäin moniulotteinen, ja että tapahtumamarkkinointia käsitellään yrityksissä usein muusta markkinointiviestinnästä erillisenä kokonaisuutena. Verrattaessa teorian esittämää tapahtumamarkkinoinnin suunnittelumallia empirian todelliseen tilanteeseen huolellisen suunnittelun ja toteutuksen merkitys korostui tapahtumamarkkinoinnin onnistumisessa. Jotta tapahtumamarkkinointi toimisi entistä tehokkaammin markkinointiviestinnän osana, tulisi yritysten kiinnittää huomiota erityisesti tavoiteasetantaan ja seurantaan. Tutkielmassa korostuu eri markkinointiviestintäkeinojen integroimisen tärkeys, jotta yritys pystyy hyödyntämään tapahtumamarkkinointiaan parhaalla mahdollisella tavalla.

Tutkimuksen päätavoitteina on kehittää toimintolaskentamalli myyntiyhtiöön sekä rakentaa mittaristo myyntiyhtiön toimitusjohtajan suorituksen mittaukseen. Kohdeyrityksenä tutkimuksessa on UPM-Kymmene Oyj:n Painopaperitoimialojen Italian myyntiyhtiö. Tutkimusmetodologiana tutkimuksessa on käytetty konstruktiivista tutkimusotetta. Toimintolaskentamalliin tarvittavien toimintojen sekä ajurien määrittely sekä toimitusjohtajan suorituksen mittareiden kartoitus on tapahtunut pääosin lukuisiin haastatteluihin ja keskusteluihin perustuen. Mallin laskentaa varten tarvittavat kustannustiedot on kerätty Italian myyntiyhtiön vuoden 1999 puolivuotistilinpäätöksestä. Tutkimuksen tuloksena on todettu toimintolaskennan soveltuvan hyvin myyntiyhtiön kustannuslaskentamuodoksi. Toimitusjohtajan suorituksen mittaus resurssijohtamisen näkökulmasta on todettu hyvin vaikeaksi mittausalueeksi. Tärkeimmät johtamisen tehokkuuden mittarit on katsottu liittyvän henkilöstön kehittämisen ja osaamisen alueille.

Quartz Tuning Fork (QTF)-based Scanning Probe Microscopy (SPM) is an important field of research. A suitable model for the QTF is important to obtain quantitative measurements with these devices. Analytical models have the limitation of being based on the double cantilever configuration. In this paper, we present an electromechanical finite element model of the QTF electrically excited with two free prongs. The model goes beyond the state-of-the-art of numerical simulations currently found in the literature for this QTF configuration. We present the first numerical analysis of both the electrical and mechanical behavior of QTF devices. Experimental measurements obtained with 10 units of the same model of QTF validate the finite element model with a good agreement.

The urgent need of effective therapies for methicillin-resistant Staphylococcus aureus (MRSA) infective endocarditis (IE) is a cause of concern. We aimed to ascertain the in vitro and in vivo activity of the older antibiotic fosfomycin combined with different beta-lactams against MRSA and glycopeptide-intermediate-resistant S. aureus (GISA) strains. Time-kill tests with 10 isolates showed that fosfomycin plus imipenem (FOF+IPM) was the most active evaluated combination. In an aortic valve IE model with two strains (MRSA-277H and GISA-ATCC 700788), the following intravenous regimens were compared: fosfomycin (2 g every 8 h [q8h]) plus imipenem (1 g q6h) or ceftriaxone (2 g q12h) (FOF+CRO) and vancomycin at a standard dose (VAN-SD) (1 g q12h) and a high dose (VAN-HD) (1 g q6h). Whereas a significant reduction of MRSA-227H load in the vegetations (veg) was observed with FOF+IPM compared with VAN-SD (0 [interquartile range [IQR], 0 to 1] versus 2 [IQR, 0 to 5.1] log CFU/g veg; P = 0.01), no statistical differences were found with VAN-HD. In addition, FOF+IPM sterilized more vegetations than VAN-SD (11/15 [73%] versus 5/16 [31%]; P = 0.02). The GISA-ATCC 700788 load in the vegetations was significantly lower after FOF+IPM or FOF+CRO treatment than with VAN-SD (2 [IQR, 0 to 2] and 0 [IQR, 0 to 2] versus 6.5 [IQR, 2 to 6.9] log CFU/g veg; P < 0.01). The number of sterilized vegetations after treatment with FOF+CRO was higher than after treatment with VAN-SD or VAN-HD (8/15 [53%] versus 4/20 [20%] or 4/20 [20%]; P = 0.03). To assess the effect of FOF+IPM on penicillin binding protein (PBP) synthesis, molecular studies were performed, with results showing that FOF+IPM treatment significantly decreased PBP1, PBP2 (but not PBP2a), and PBP3 synthesis. These results allow clinicians to consider the use of FOF+IPM or FOF+CRO to treat MRSA or GISA IE.

There is an increasing need to develop improved systems for predicting the safety of xenobiotics. However, to move beyond hazard identification the available concentration of the test compounds needs to be incorporated. In this study cyclosporine A (CsA) was used as a model compound to assess the kinetic profiles in two rodent brain cell cultures after single and repeated exposures. CsA induced-cyclophilin B (Cyp-B) secretion was also determined as CsA-specific pharmacodynamic endpoint. Since CsA is a potent p-glycoprotein substrate, the ability of this compound to cross the blood-brain barrier (BBB) was also investigated using an in vitro bovine model with repeated exposures up to 14days. Finally, CsA uptake mechanisms were studied using a parallel artificial membrane assay (PAMPA) in combination with a Caco-2 model. Kinetic results indicate a low intracellular CsA uptake, with no marked bioaccumulation or biotransformation. In addition, only low CsA amounts crossed the BBB. PAMPA and Caco-2 experiments revealed that CsA is mostly trapped to lipophilic compartments and exits the cell apically via active transport. Thus, although CsA is unlikely to enter the brain at cytotoxic concentrations, it may cause alterations in electrical activity and is likely to increase the CNS concentration of other compounds by occupying the BBBs extrusion capacity. Such an integrated testing system, incorporating BBB, brain culture models and kinetics could be applied for assessing neurotoxicity potential of compounds.

Human chorionic gonadotropin (hCG) and luteinizing hormone (LH) are structurally and functionally similar glycoprotein hormones acting through the same luteinizing hormone chorionic gonadotropin receptor (LHCGR). The functions of LH in reproduction and hCG in pregnancy are well known. Recently, the expression of LHCGR has been found in many nongonadal tissues and cancers, and this has raised the question of whether LH/hCG could affect the function or tumorigenesis of these nongonadal tissues. We have also previously generated an hCG expressing mouse model presenting nongonadal phenotypes. Using this model it is possible to improve our understanding of nongonadal action of highly elevated LH/hCG. In the current study, we analyzed the effect of moderately and highly elevated hCG levels on male reproductive development and function. The main finding was the appearance of fetal Leydig cell (FLC) adenomas in prepubertal males. However, the development and differentiation of FLCs were not significantly affected. We also show that the function of hCG is different in FLCs and in adult Leydig cells (ALC), because in the latter cells hCG was not able to induce tumorigenesis. In FLCs, LHCGR is not desensitized or downregulated upon ligand binding. In this study, we found that the testicular expression of two G protein-coupled receptor kinases responsible for receptor desensitization or downregulation is increased in adult testis. Results suggest that the lack of LHCGR desensitization or downregulation in FLCs protect testosterone (Te) synthesis, but also predispose FLCs for LH/hCG induced adenomas. However, all the hCG induced nongonadal changes observed in male mice were possible to explain by the elevated Te level found in these males. Our findings indicate that the direct nongonadal effects of elevated LH/hCG in males are not pathophysiologically significant. In female mice, we showed that an elevated hCG level was able to induce gonadal tumorigenesis. hCG also induced the formation of pituitary adenomas (PA), but the mechanism was indirect. Furthermore, we found two new potential risk factors and a novel hormonally induced mechanism for PAs. Increased progesterone (P) levels in the presence of physiological estradiol (E2) levels induced the formation of PAs in female mice. E2 and P induced the expression and nuclear localization of a known cell-cycle regulator, cyclin D1. A calorie restricted diet was also able to prevent the formation of PAs, suggesting that obesity is able to promote the formation of PAs. Hormone replacement therapy after gonadectomy and hormone antagonist therapy showed that the nongonadal phenotypes observed in hCG expressing female mice were due to ovarian hyperstimulation. A slight adrenal phenotype was evident even after gonadectomy in hCG expressing females, but E2 and P replacement was able to induce a similar phenotype in WT females without elevated LH/hCG action. In conclusion, we showed that the direct effects of elevated hCG/LH action are limited only to the gonads of both sexes. The nongonadal phenotypes observed in hCG expressing mice were due to the indirect, gonadal hormone mediated effects of elevated hCG. Therefore, the gonads are the only physiologically significant direct targets of LHCGR signalling.

Epsilon toxin (Etx) from Clostridium perfringens is a pore-forming protein with a lethal effect on livestock, producing severe enterotoxemia characterized by general edema and neurological alterations. Site-specific mutations of the toxin are valuable tools to study the cellular and molecular mechanism of the toxin activity. In particular, mutants with paired cysteine substitutions that affect the membrane insertion domain behaved as dominant-negative inhibitors of toxin activity in MDCK cells. We produced similar mutants, together with a well-known non-toxic mutant (Etx-H106P), as green fluorescent protein (GFP) fusion proteins to perform in vivo studies in an acutely intoxicated mouse model. The mutant (GFP-Etx-I51C/A114C) had a lethal effect with generalized edema, and accumulated in the brain parenchyma due to its ability to cross the blood-brain barrier (BBB). In the renal system, this mutant had a cytotoxic effect on distal tubule epithelial cells. The other mutants studied (GFP-Etx-V56C/F118C and GFP-Etx-H106P) did not have a lethal effect or cross the BBB, and failed to induce a cytotoxic effect on renal epithelial cells. These data suggest a direct correlation between the lethal effect of the toxin, with its cytotoxic effect on the kidney distal tubule cells, and the ability to cross the BBB.

In the last two decades, cases of corruption have been unveiled in different countries, raising public awareness and reinforcing a trend in which society expects more from their leaders. Our objective in this paper is to examine the effects of corruption and seigniorage on inflation and growth rates. The model used in this article is an extension of the model used by Huang and Wei (2006). We find interesting results and one of them is that, under some conditions, corruption has a positive impact on the growth rate. JEL classification : D73, E52, E58, E62. Human activities have resulted in increased nutrient levels in many rivers all over Europe. Sustainable management of river basins demands an assessment of the causes and consequences of human alteration of nutrient flows, together with an evaluation of management options. In the context of an integrated and interdisciplinary environmental assessment (IEA) of nutrient flows, we present and discuss the application of the nutrient emission model MONERIS (MOdelling Nutrient Emissions into River Systems) to the Catalan river basin, La Tordera (north-east Spain), for the period 1996–2002. After a successful calibration and verification process (Nash-Sutcliffe efficiencies E=0.85 for phosphorus and E=0.86 for nitrogen), the application of the model MONERIS proved to be useful in estimating nutrient loads. Crucial for model calibration, in-stream retention was estimated to be about 50 % of nutrient emissions on an annual basis. Through this process, we identified the importance of point sources for phosphorus emissions (about 94% for 1996–2002), and diffuse sources, especially inputs via groundwater, for nitrogen emissions (about 31% for 1996–2002). Despite hurdles related to model structure, observed loads, and input data encountered during the modelling process, MONERIS provided a good representation of the major interannual and spatial patterns in nutrient emissions. An analysis of the model uncertainty and sensitivity to input data indicates that the model MONERIS, even in data-starved Mediterranean catchments, may be profitably used by water managers for evaluating quantitative nutrient emission scenarios for the purpose of managing river basins. As an example of scenario modelling, an analysis of the changes in nutrient emissions through two different future scenarios allowed the identification of a set of relevant measures to reduce nutrient loads.

Identification of order of an Autoregressive Moving Average Model (ARMA) by the usual graphical method is subjective. Hence, there is a need of developing a technique to identify the order without employing the graphical investigation of series autocorrelations. To avoid subjectivity, this thesis focuses on determining the order of the Autoregressive Moving Average Model using Reversible Jump Markov Chain Monte Carlo (RJMCMC). The RJMCMC selects the model from a set of the models suggested by better fitting, standard deviation errors and the frequency of accepted data. Together with deep analysis of the classical Box-Jenkins modeling methodology the integration with MCMC algorithms has been focused through parameter estimation and model fitting of ARMA models. This helps to verify how well the MCMC algorithms can treat the ARMA models, by comparing the results with graphical method. It has been seen that the MCMC produced better results than the classical time series approach.

SiO2/TiO2 nanostructured composites with three different ratios of Si:Ti were prepared using the sol-gel method. These materials were characterized using energy dispersive X-ray fluorescence, Fourier transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, high-resolution transmission electron microscopy, photoluminescence, Raman with Fourier transform infrared spectroscopy, and the specific surface area. The band gaps of materials were determined by diffuse reflectance spectra, and the values of 3.20 ± 0.01, 2.92 ± 0.02, and 2.85 ± 0.01 eV were obtained as a result of the proportional increases in the amount of Ti within the composite. The materials exhibit only the anatase (TiO2) crystalline phase and have crystalline domains ranging from 4 to 5 nm. The photodegradation process of methylene blue, royal blue GRL, and golden yellow GL dyes were studied with respect to their contact times, pH variations within the solution, and the variations in the dye concentration of the solution in response to only sunlight. The maximum amount of time for the mineralization of dyes was 90 min. The kinetics of the process follows an apparently first order model, in which the obtained rate constant values were 5.72 × 10-2 min-1 for methylene blue, 6.44 × 10-2min-1 for royal blue GRL, and 1.07 × 10-1min-1 for golden yellow.

The aim of this study is to analyse the content of the interdisciplinary conversations in Göttingen between 1949 and 1961. The task is to compare models for describing reality presented by quantum physicists and theologians. Descriptions of reality indifferent disciplines are conditioned by the development of the concept of reality in philosophy, physics and theology. Our basic problem is stated in the question: How is it possible for the intramental image to match the external object?Cartesian knowledge presupposes clear and distinct ideas in the mind prior to observation resulting in a true correspondence between the observed object and the cogitative observing subject. The Kantian synthesis between rationalism and empiricism emphasises an extended character of representation. The human mind is not a passive receiver of external information, but is actively construing intramental representations of external reality in the epistemological process. Heidegger's aim was to reach a more primordial mode of understanding reality than what is possible in the Cartesian Subject-Object distinction. In Heidegger's philosophy, ontology as being-in-the-world is prior to knowledge concerning being. Ontology can be grasped only in the totality of being (Dasein), not only as an object of reflection and perception. According to Bohr, quantum mechanics introduces an irreducible loss in representation, which classically understood is a deficiency in knowledge. The conflicting aspects (particle and wave pictures) in our comprehension of physical reality, cannot be completely accommodated into an entire and coherent model of reality. What Bohr rejects is not realism, but the classical Einsteinian version of it. By the use of complementary descriptions, Bohr tries to save a fundamentally realistic position. The fundamental question in Barthian theology is the problem of God as an object of theological discourse. Dialectics is Barth¿s way to express knowledge of God avoiding a speculative theology and a human-centred religious self-consciousness. In Barthian theology, the human capacity for knowledge, independently of revelation, is insufficient to comprehend the being of God. Our knowledge of God is real knowledge in revelation and our words are made to correspond with the divine reality in an analogy of faith. The point of the Bultmannian demythologising programme was to claim the real existence of God beyond our faculties. We cannot simply define God as a human ideal of existence or a focus of values. The theological programme of Bultmann emphasised the notion that we can talk meaningfully of God only insofar as we have existential experience of his intervention. Common to all these twentieth century philosophical, physical and theological positions, is a form of anti-Cartesianism. Consequently, in regard to their epistemology, they can be labelled antirealist. This common insight also made it possible to find a common meeting point between the different disciplines. In this study, the different standpoints from all three areas and the conversations in Göttingen are analysed in the frameworkof realism/antirealism. One of the first tasks in the Göttingen conversations was to analyse the nature of the likeness between the complementary structures inquantum physics introduced by Niels Bohr and the dialectical forms in the Barthian doctrine of God. The reaction against epistemological Cartesianism, metaphysics of substance and deterministic description of reality was the common point of departure for theologians and physicists in the Göttingen discussions. In his complementarity, Bohr anticipated the crossing of traditional epistemic boundaries and the generalisation of epistemological strategies by introducing interpretative procedures across various disciplines.
