RESUMO - (Indigofera L. (Leguminosae, Papilionoideae) no estado de São Paulo, Brasil). Em um levantamento realizado no estado de São Paulo, com exsicatas de vários herbários e material coletado no campo, foram encontradas nove espécies de Indigofera L.: I. asperifolia Bong. ex Benth., I. bongardiana (Kuntze) Burk var. bongardiana, I. guaranitica Hassl., I. hirsuta L., I. lespedezioides Kunth, I. spicata Forssk., I. suffruticosa Mill. subsp. suffruticosa, I. trita subsp. scabra (Roth) De Kort & Thijsse e I. truxillensis Kunth. Quatro destas espécies são citadas pela primeira vez para o estado, sendo que duas, I. spicata e I. trita subsp. scabra, são também novas ocorrências para o Brasil. Além de uma chave para a identificação dos taxa, são fornecidas descrições e ilustrações dos mesmos.

Neste trabalho foram analisados os grãos de pólen de seis táxons do gênero Brunfelsia: B. bonodora (Vell.) J.F. Macbr., B. brasiliensis (Spreng.) L.B. Sm. & Downs var. brasiliensis, B. brasiliensis subsp. macrocalyx (Dusén) Plowman, B. hydrangeiformis subsp. capitata (Benth.) Plowman, B. latifolia (Pohl) Benth. e B. uniflora D. Don. Os grãos de pólen foram tratados com ACLAC 60% (exceto os de B. latifolia, que sofreram o processo da acetólise). Os grãos de pólen foram, posteriormente, mensurados, descritos, foto e eletromicrografados. A análise sob MEV foi utilizada visando a elucidação de dúvidas sobre a ornamentação da exina. Constatou-se que os grãos de pólen são médios ou grandes; isopolares; suboblatos ou oblato-esferoidais; âmbito subcircular, quadrangular a pentagonal; área polar de muito pequena a grande; 3-5-colporados; sexina variavelmente rugulada porém, melhor visualizada sob MEV.

Novos táxons infraespecíficos e combinações em Pilocarpus são apresentados em antecipação à completa publicação da revisão do gênero: P. grandiflorus Engl. var. recurvus Skorupa; P. pauciflorus A. St.-Hil. subsp. clavatus Skorupa; P. racemosus Vahl subsp. goudotianus (Tul.) Skorupa; P. racemosus Vahl subsp. goudotianus (Tul.) Skorupa var. mollis (Cuatrec.) Skorupa; P. racemosus Vahl subsp. goudotianus (Tul.) Skorupa var. heterochromus (Kaastra) Skorupa. Informações sobre a distribuição geográfica, hábitat e fenologia são fornecidos para estes táxons.

Floristic and phytosociological surveys were carried out for 12 months in the Embrapa-SPSB, Petrolina, Pernambuco, Brazil. A transect was laid on starting at the river bank extending for 790 m away from the river and divided into 140 10 × 10 m contiguous plots. In each plot, all standing plants, alive or dead, with stem diameter at soil level > 3 cm and total height > 1 m were sampled. Along this transect, an elevation range of 9.40 m was registered and five topographical environments were identified: riverside (MR), dike (D), floodable depression (DI), boundary terrace (TL) - all of them belonging to the fluvial terrace with Fluvic Neosol and Haplic Cambisol both silty textured eutrophic soils - and the inlander tableland (TS), with medium sandy-textured Red-Yellow Argisols. Fourty-eight species/morphospecies, distributed into 39 genera and 21 families, were identified. Four phytogeoenvironments (MR, D + TL, DI + TL, and TS) were registered based on environmental variations and floristic similarities among plots using cluster analyses. The MR environment showed the largest total density, total basal area, maximum and medium heights and maximum diameter. Moreover, it had 8.1% of plants with heights above 8 m against 0.6% for D + TL, 0.2% for DI + TL, and 0% for TS. The species with the largest importance value were Inga vera subsp. affinis (DC.) T.D. Pennington in MR, Mimosa bimucronata Kunth in D + TL and DI + TL and M. tenuiflora (Willd.) Poir. in TS.

The main goal of this thesis is to increase understanding on evolutionary and ecological factors that have contributed to differences in parasite numbers in insects. Furthermore, the thesis addresses the effects of parasites on their hosts. The most important findings were: The Northern damselfly’s (Coenagrion hastulatum) immune response to artificial pathogen increased with increasing parasite numbers (Article I). Marginal, more isolated C. hastulatum populations on the edge of distribution have fewer parasites when compared to distribution’s core populations (Article II). The Banded damselfly Calopteryx splendens individuals with higher homozygosity have more parasites, however, the rate of homozygosity did not differ between populations (Article III). Parasite prevalence was affected by whether the host species occurred in allopatric or sympatric population: sympatric C. splendens populations with sister species the Beautiful damselfly Calopteryx virgo harbored more parasites (Article IV). Parasites were associated with the wing spot size, an ornament under sexual selection, and thus may play an important role in character displacement, i.e. the size of the wing spot (Article V). To conclude with, this thesis brings about new information on the parasite infection patterns in insects, proposing several factors to contribute to these patters, as well as it addresses the effects of parasites on their hosts, from individual to population level.

The protease ZapA, secreted by Proteus mirabilis, has been considered to be a virulence factor of this opportunistic bacterium. The control of its expression requires the use of an appropriate methodology, which until now has not been developed. The present study focused on the replacement of azocasein with fluorogenic substrates, and on the definition of enzyme specificity. Eight fluorogenic substrates were tested, and the peptide Abz-Ala-Phe-Arg-Ser-Ala-Ala-Gln-EDDnp was found to be the most convenient for use as an operational substrate for ZapA. A single peptide bond (Arg-Ser) was cleaved with a Km of 4.6 µM, a k cat of 1.73 s-1, and a catalytic efficiency of 376 (mM s)-1. Another good substrate for ZapA was peptide 6 (Abz-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg-Gln-EDDnp) which was cleaved at a single bond (Phe-Ser) with a Km of 13.6 µM, a k cat of 3.96 s-1 and a catalytic efficiency of 291 (mM s)-1. The properties of the amino acids flanking the scissile bonds were also evaluated, and no clear requirement for the amino acid residue at P1 was found, although the enzyme seems to have a preference for a hydrophobic residue at P2.

The opportunistic bacterium Proteus mirabilis secretes a metalloprotease, ZapA, considered to be one of its virulence factors due to its IgA-degrading activity. However, the substrate specificity of this enzyme has not yet been fully characterized. In the present study we used fluorescent peptides derived from bioactive peptides and the oxidized ß-chain of insulin to determine the enzyme specificity. The bradykinin- and dynorphin-derived peptides were cleaved at the single bonds Phe-Ser and Phe-Leu, with catalytic efficiencies of 291 and 13 mM/s, respectively. Besides confirming already published cleavage sites, a novel cleavage site was determined for the ß-chain of insulin (Val-Asn). Both the natural and the recombinant enzyme displayed the same broad specificity, demonstrated by the presence of hydrophobic, hydrophilic, charged and uncharged amino acid residues at the scissile bonds. Native IgA, however, was resistant to hydrolysis by ZapA.

An alternative device for the immobilization of the hind limb of the rat was developed to study the effects of chronic disuse on the soleus and tibialis anterior muscles, maintained for 3 weeks in the shortening and the stretching positions, respectively. The proposed device is made of steel mesh and cotton materials, and has some advantages when compared to cast or plaster cast: it is cheaper, lighter (12 g or 4% of the body weight of the rat) and the same unit can be easily adjusted and used several times in the same animal or in animals of similar size. Immobilization is also useful to restrain the movements of the hip, knee, and ankle joints. Male rats (291 ± 35 g and aged 14 ± 2 weeks) were used to develop and test the model. The soleus muscle of 18 rats was maintained in a shortened position for 21 consecutive days and lost 19 ± 7% of its length (P = 0.008) and 44 ± 6% of its weight (P = 0.002) compared to the contralateral intact muscle. No difference (P = 0.67) was found in the stretched tibialis anterior of the same hind limb when compared to the contralateral muscle. No ulcer, sore or foot swelling was observed in the animals. Immobilization was effective in producing chronic muscle disuse in the hind limbs of rats and is an acceptable alternative to the traditional methods of immobilization such as cast or plaster cast.
