O nosso trabalho consiste numa reflexão teórica sobre a aplicação prática da educação estética e das técnicas de teatro no ensino e aprendizagem das línguas e das literaturas. Pretendemos estabelecer relações entre a educação estética e o ensino, visando uma aproximação, em termos didáticos, entre a cultura, as artes e a educação literária e linguística, mas também conduzir a aprendizagem no sentido da observação atenta e refletida para o desenvolvimento do pensamento crítico e autónomo através da ação. A relação da imagem com o texto, tendo em vista a interpretação dramática, é uma forma de consolidar conhecimentos da obra literária inserida num contexto artístico mais amplo, e essa relação, para a realização de jogos dramáticos, serve a concretização em imagem viva, que é a do ator ou intérprete. Mas, acima de tudo, ela visa a formação integral e desenvolve a criatividade. A contemplação e o ato de criar só são possíveis se o aluno usufruir de uma educação estética, que fomenta em grande escala o gosto pelo texto inserido numa determinada cultura e na época em que se produziu. Nesta proposta, incluímos o exercício e a interiorização do conhecimento linguístico através das técnicas teatrais, enquanto práticas intrínsecas ao teatro, numa atitude comunicativa que implica interpretar e compreender, e que fomenta a autonomia do indivíduo. Pretendemos estimular o conhecimento integrado do aluno, associando a estética e as artes à aprendizagem das línguas, como meios impulsionadores da criatividade, da reinvenção e do fazer.

The development of human cell models that recapitulate hepatic functionality allows the study of metabolic pathways involved in toxicity and disease. The increased biological relevance, cost-effectiveness and high-throughput of cell models can contribute to increase the efficiency of drug development in the pharmaceutical industry. Recapitulation of liver functionality in vitro requires the development of advanced culture strategies to mimic in vivo complexity, such as 3D culture, co-cultures or biomaterials. However, complex 3D models are typically associated with poor robustness, limited scalability and compatibility with screening methods. In this work, several strategies were used to develop highly functional and reproducible spheroid-based in vitro models of human hepatocytes and HepaRG cells using stirred culture systems. In chapter 2, the isolation of human hepatocytes from resected liver tissue was implemented and a liver tissue perfusion method was optimized towards the improvement of hepatocyte isolation and aggregation efficiency, resulting in an isolation protocol compatible with 3D culture. In chapter 3, human hepatocytes were co-cultivated with mesenchymal stem cells (MSC) and the phenotype of both cell types was characterized, showing that MSC acquire a supportive stromal function and hepatocytes retain differentiated hepatic functions, stability of drug metabolism enzymes and higher viability in co-cultures. In chapter 4, a 3D alginate microencapsulation strategy for the differentiation of HepaRG cells was evaluated and compared with the standard 2D DMSO-dependent differentiation, yielding higher differentiation efficiency, comparable levels of drug metabolism activity and significantly improved biosynthetic activity. The work developed in this thesis provides novel strategies for 3D culture of human hepatic cell models, which are reproducible, scalable and compatible with screening platforms. The phenotypic and functional characterization of the in vitro systems performed contributes to the state of the art of human hepatic cell models and can be applied to the improvement of pre-clinical drug development efficiency of the process, model disease and ultimately, development of cell-based therapeutic strategies for liver failure.

Intitulado “O surto de Legionella de 2014 no Telejornal da RTP”, este Relatório analisa a emergência e ascensão de um tema de risco na agenda jornalística de um meio de comunicação, bem como o seu declínio e desaparecimento. Questionam-se os fatores que asseguraram o estatuto de matéria noticiável e todo o processo de construção social nas notícias. Através da análise de conteúdo das peças jornalísticas transmitidas no Telejornal e de uma breve revisão de literatura sobre o tema, concluímos que a comunicação exerce um papel fundamental na construção social de um risco. A cobertura do surto de Legionella desenvolveu-se ao ritmo com que se revelaram as suas consequências, da propaganda que foi feita, do interesse que despertou nas redações e do reflexo cultural que gerou na audiência. Tanto mais se verifica pelas circunstâncias inerentes a esta crise com a sua amplificação ou atenuação social nas notícias, tendo em conta a agenda jornalística e a saturação de um evento mediático. O interesse jornalístico pela estória foi estimulado pelo comportamento do principal departamento envolvido na crise – o Ministério da Saúde – e pela Direção-Geral de Saúde, e na origem do declínio da atenção estiveram eventos sociopolíticos - o aparecimento de um novo caso mediático.

INTRODUCTION: Some viruses of the Herpesviridae family are frequently the etiologic agents of oral lesions associated with HIV. The aim of this study was to identify the presence of herpes simplex virus types 1 and 2 (HSV-1, HSV-2), Varicella Zoster virus (VZV), Epstein-Barr virus (EBV), human cytomegalovirus (HCMV), human herpesvirus type 6, type 7 and type 8 (HHV-6, HHV-7 and HHV-8) in the oral cavity of HIV-infected children/adolescents and verify the association between viral subtypes and clinical factors. METHODS: The cells of oral mucosa were collected from 50 HIV infected children/adolescents, 3-13 years old (mean age 8.66). The majority (66%) of selected were girls, and they were all outpatients at the pediatric AIDS clinic of a public hospital in Rio de Janeiro. Nested-PCR was used to identify the viral types. RESULTS: Absence of immunosuppression was observed in 66% of the children. Highly active antiretroviral therapy (HAART) was used by 72.1% of selected and moderate viral load was observed in 56% of the children/adolescents. Viral types were found in 86% of the children and the subtypes were: HSV-1 (4%), HSV-2 (2%), VZV (4%), EBV (0%), HCMV (24%), HHV6 (18%), HHV-7 (68%), HHV8 (0%). CONCLUSIONS: The use of HAART has helped to reduce oral lesions, especially with herpes virus infections. The health professionals who work with these patients should be aware of such lesions because of their predictive value and the herpes virus can be found circulating in the oral cavity without causing lesions.

INTRODUCTION: Sequential antibiotic therapy (SAT) is safe and economical. However, the unnecessary use of intravenous (IV) administration usually occurs. The objective of this work was to get to know the effectiveness of an intervention to implement the SAT in a teaching hospital in Brazil. METHODS: This was a prospective and interventional study, historically controlled, and was conducted in the Hospital de Clínicas, Universidade Federal de Uberlândia, State of Minas Gerais, Brazil, a high complexity teaching hospital having 503 beds. In each of the periods, from 04/04/05 to 07/20/05 (pre-intervention) and from 09/24/07 to 12/20/07 (intervention), 117 patients were evaluated. After the pre-intervention period, guidelines were developed which were implemented during the intervention period along with educational measures and a reminder system added to the patients' prescription. RESULTS: In the pre-intervention and intervention periods, the IV antibiotics were used as treatment for a average time of 14.8 and 11.8 days, respectively. Ceftriaxone was the antibiotic most prescribed in both periods (23.4% and 21.6% respectively). Starting from the first prescription of antibiotics, the average length of hospitalization time was 21.8 and 17.5 days, respectively. The SAT occurred only in 4 and 5 courses of treatment, respectively, and 12.8% and 18.8% of the patients died in the respective periods. CONCLUSIONS: Under the presented conditions, the evaluated intervention strategy is ineffective in promoting the exchange of the antibiotic administration from IV to oral treatment (SAT).

The role of a set of gases relevant within the context of biomolecules and technologically relevant molecules under the interaction of low-energy electrons was studied in an effort to contribute to the understanding of the underlying processes yielding negative ion formation. The results are relevant within the context of damage to living material exposed to energetic radiation, to the role of dopants in the ion-molecule chemistry processes, to Electron Beam Induced Deposition (EBID) and Ion Beam Induced Deposition (IBID) techniques. The research described in this thesis addresses dissociative electron attachment (DEA) and electron transfer studies involving experimental setups from the University of Innsbruck, Austria and Universidade Nova de Lisboa, Portugal, respectively. This thesis presents DEA studies, obtained by a double focusing mass spectrometer, of dimethyl disulphide (C2H6S2), two isomers, enflurane and isoflurane (C3F5Cl5) and two chlorinated ethanes, pentachloroethane (C2HCl5) and hexachloroethane (C2Cl6), along with quantum chemical calculations providing information on the molecular orbitals as well as thermochemical thresholds of anion formation for enflurane, isoflurane, pentachloroethane and hexachloroethane. The experiments represent the most accurate DEA studies to these molecules, with significant differences from previous work reported in the literature. As far as electron transfer studies are concerned, negative ion formation in collisions of neutral potassium atoms with N1 and N3 methylated pyrimidine molecules were obtained by time-of-flight mass spectrometry (TOF). The results obtained allowed to propose concerted mechanisms for site and bond selective excision of bonds.

O presente relatório procura descrever o estágio desenvolvido na revista ELLE Portugal, entre os meses de setembro e dezembro de 2014, cumprindo os requerimentos necessários para conclusão do mestrado em Edição de Texto na Faculdade de Ciências Sociais e Humanas da Faculdade Nova de Lisboa. Além da descrição das atividades realizadas durante o mesmo, o documento apresenta reflexões sobre a Edição de Texto dentro das publicações periódicas internacionais pensadas para o público feminino, nomeadamente no tratamento da Tradução, da Adaptação e da Linguagem Feminina.

In the competitive landscape of the 21st century, effectively managing human capital in firms is considered to be a potential source of sustainable performance. Therefore, in this study, we tested the influence of high-performance work systems, as a talent management tool, on employees’ experience of developmental jobs. Then, we tested the mediating effect of such experiences on employees’ engagement, exhaustion, performance and turnover intention. With a sample of 254 employees of a diversity of companies and sectors of activity, our findings demonstrated that high-performance practices increase engagement, via the promotion of developmental experiences of fit, which improves performance and decreases turnover intention. Besides, those practices do not control for the pressure dimension of the developmental job experiences that increases exhaustion and turnover intention despite not worsening performance.

The purpose of this project is to understand if the brand Sagres is damaging the product Sagres Radler. The beer market in Portugal was studied and focus groups were used to perceive the impact of the brand in the product. The mother brand is bringing the beer association into a product designed to engage people that don’t like beer. With the insights, a new proposal was drawn and tested. Although it was not possible to prove that the new concept is better for the population, there are strong indications that the brand isn’t enabling the achievement of the proposed target.

Introduction We analyzed the vertical and monthly distributions of culicid species in the gallery forest of Brasília National Park, with an emphasis on the potential vectors of yellow fever (YF). Methods Between September 2010 and August 2011, mosquitoes were captured on the ground and in the canopy of the forest for five consecutive days per month, from nine to 15 hours. The mosquitoes were examined to verify natural infection with flaviviruses by isolation in Aedes albopictus Skuse, 1864 cells followed by indirect immunofluorescence. Results We identified 2,677 culicids distributed in 29 species. Most of the mosquitoes were captured at ground level (69%) during the rainy season (86%). The most abundant species were Sabethes (Sabethes) albiprivus Theobald, 1903; Limatus durhamii Theobald, 1901; Haemagogus (Conopostegus) leucocelaenus Dyar & Shannon, 1924; Haemagogus (Haemagogus) janthinomys Dyar, 1921; Aedes (Ochlerotatus) scapularis Rondani, 1848; Psorophora (Janthinosoma) ferox Von Humboldt, 1819; and Aedes (Ochlerotatus) serratus Theobald, 1901. Limatus durhamii, Limatus durhamii, Psorophora ferox, Aedes scapularis and Aedes serratus showed significant differences (p<0.05) in their habitat use. Limatus durhamii was found more often in the canopy, unlike the other species. During the rainy season, the most abundant species were Sa. albiprivus, Haemagogus leucocelaenus and Limatus durhamii. During the dry season, the potential YF vectors exhibited a very low frequency and abundance, except Aedes scapularis and Aedes serratus. No flavivirus was detected in the 2,677 examined mosquitoes. Conclusions We recommend continued and systematic entomological monitoring in areas vulnerable to the transmission of YF in the Federal District of Brazil.

This work analyses how the leverage ratio behaves through the cycle, vis-à-vis other capital ratios. For a sample of the largest Portuguese banks, the Basel III leverage ratio is indeed countercyclical. This result is relevant from a regulatory perspective, since the introduction of a limit on the leverage ratio will function as a restriction in the banks’ balance sheet size, reducing the economic costs associated with the excessive growth of leverage in periods of economic expansion followed by aggressive deleveraging in the downturn. However, one cannot exclude that restrictions on banks’ leverage incentivize its transference to less regulated intermediaries.

Introduction The incidence of canine leptospirosis in Brazil needs to be assessed Methods The same dogs in southern Brazil were sampled over two years to determine the prevalence, incidence and association of canine leptospirosis with various risk factors. Results In 2009, the prevalence was 33 (14.4%) of 228 dogs, with a predominance of serovar Canicola (33.4%). In 2010, 90 dogs were re-evaluated (the remaining dogs were lost to deaths, address changes and donations), and the prevalence was found to be 35 (38.9%) of 90, with the predominant serovar being Icterohaemorrhagiae (51.4%). Moreover, the incidence was 26 of 90 (28.9%), and the disease was statistically associated with age (2009) and street access (2010). Conclusions Our findings revealed instability in the dog population and age to be relevant risk factors for canine leptospirosis.

Os polímeros são materiais de grande interesse científico e tecnológico já que podem ser utilizados no desenvolvimento de dispositivos para aplicação em electrónica e fotónica. Tendo em conta a sua aplicação em dispositivos de conversão de energia solar em energia eléctrica, o estudo das propriedades eléctricas e ópticas de filmes destes materiais, quando irradiados por radiação ultravioleta e visível, é fundamental. Neste trabalho caracterizaram-se as propriedades eléctricas e ópticas de filmes finos obtidos pelas técnicas de derramamento e de camada-por-camada (Layer-by-Layer (LbL)) obtidos a partir do poli(cloreto de xilideno tetrahidrotiofeno) (PTHT) que após aquecimento é convertido em poli(p-fenileno vinileno) (PPV). Assim, e através da técnica de derramamento, foram preparados filmes sobre suportes sólidos com eléctrodos interdigitais de forma a proceder à medição da condutividade em modo de corrente alternada (AC), isto é, à medição da resistência e da capacidade em função da frequência. Verificou-se, portanto, que a presença de luz leva à formação de portadores de carga, ou seja, na presença de luminosidade os filmes apresentam uma menor capacitância e resistência, para além de degradar o material. Esta degradação foi verificada, também, a partir da espectrofotometria de Ultravioleta-Visível (UV-Vis) e de infravermelho (IV). Medições de condutividade de volume, isto é, colocando os filmes entre eléctrodos paralelos de funções de trabalho diferentes, permitiram traçar as curvas características e verificar que os dispositivos obtidos se comportam como díodos. Recorreu-se, também, a técnicas de espectrofotometria de ultravioleta do vácuo (VUV) e UV-Vis para caracterizar filmes LbL obtidos a partir do PTHT e de um polieletrólito comum o poli(estireno sulfonado) (PSS) antes e após aquecimento. Os espectros obtidos, após análise dos picos, mostram que a técnica de VUV é a técnica indicada para analisar a conversão do PTHT em PPV. Este trabalho permitiu concluir que as propriedades eléctricas e ópticas destes filmes, estão dependentes da presença de luminosidade, de oxigénio e da humidade relativa pelo que dispositivos obtidos a partir deste polímero devem ser, sempre, encapsulados.

Nesta dissertação é apresentada a metodologia adoptada no desenvolvimento de um sensor capaz de monitorizar a evolução da consolidação óssea proveniente de patologias tratadas com a utilização de aparelhos de fixação externa. O sensor tem como finalidade medir o deslocamento relativo entre os componentes do fixador externo que são proporcionais aos níveis de consolidação do osso. Surge como alternativa aos processos convencionais (raios-X, ultra-sons, etc.) que não possibilitam a detecção de imperfeições no processo de regeneração óssea numa fase precoce. Uma análise a vários materiais e geometrias revelaram que um sensor fabricado em latão e sob a forma de anel, após uma deformação de compressão de 4
