The research described in this PhD thesis focuses on proteomics approaches to study the effect of oxidation on the modification status and protein-protein interactions of PTEN, a redox-sensitive phosphatase involved in a number of cellular processes including metabolism, apoptosis, cell proliferation, and survival. While direct evidence of a redox regulation of PTEN and its downstream signaling has been reported, the effect of cellular oxidative stress or direct PTEN oxidation on PTEN structure and interactome is still poorly defined. In a first study, GST-tagged PTEN was directly oxidized over a range of hypochlorous acid (HOCl) concentration, assayed for phosphatase activity, and oxidative post-translational modifications (oxPTMs) were quantified using LC-MS/MS-based label-free methods. In a second study, GSTtagged PTEN was prepared in a reduced and reversibly H2O2-oxidized form, immobilized on a resin support and incubated with HCT116 cell lysate to capture PTEN interacting proteins, which were analyzed by LC-MS/MS and comparatively quantified using label-free methods. In parallel experiments, HCT116 cells transfected with a GFP-tagged PTEN were treated with H2O2 and PTENinteracting proteins immunoprecipitated using standard methods. Several high abundance HOCl-induced oxPTMs were mapped, including those taking place at amino acids known to be important for PTEN phosphatase activity and protein-protein interactions, such as Met35, Tyr155, Tyr240 and Tyr315. A PTEN redox interactome was also characterized, which identified a number of PTEN-interacting proteins that vary with the reversible inactivation of PTEN caused by H2O2 oxidation. These included new PTEN interactors as well as the redox proteins peroxiredoxin-1 (Prdx1) and thioredoxin (Trx), which are known to be involved in the recycling of PTEN active site following H2O2-induced reversible inactivation. The results suggest that the oxidative modification of PTEN causes functional alterations in PTEN structure and interactome, with fundamental implications for the PTEN signaling role in many cellular processes, such as those involved in the pathophysiology of disease and ageing.

The increased occurrence of cyanobacteria (blue-green algae) blooms and the production of associated cyanotoxins have presented a threat to drinking water sources. Among the most common types of cyanotoxins found in potable water are microcystins (MCs), a family of cyclic heptapeptides containing substrates. MCs are strongly hepatotoxic and known to initiate tumor promoting activity. The presence of sub-lethal doses of MCs in drinking water is implicated as one of the key risk factors for an unusually high occurrence of primary liver cancer. ^ A variety of traditional water treatment methods have been attempted for the removal of cyanotoxins, but with limited success. Advanced Oxidation Technologies (AOTs) are attractive alternatives to traditional water treatments. We have demonstrated ultrasonic irradiation and UV/H2O2 lead to the degradation of cyanotoxins in drinking water. These studies demonstrate AOTs can effectively degrade MCs and their associated toxicity is dramatically reduced. We have conducted detailed studies of different degradation pathways of MCs and conclude that the hydroxyl radical is responsible for a significant fraction of the observed degradation. Results indicate preliminary products of the sonolysis of MCs are due to the hydroxyl radical attack on the benzene ring and substitution and cleavage of the diene of the Adda peptide residue. AOTs are attractive methods for treatment of cyanotoxins in potable water supplies. ^ The photochemical transformation of MCs is important in the environmental degradation of MCs. Previous studies implicated singlet oxygen as a primary oxidant in the photochemical transformation of MCs. Our results indicate that singlet oxygen predominantly leads to degradation of the phycocyanin, pigments of blue green algae, hence reducing the degradation of MCs. The predominant process involves isomerization of the diene (6E to 6Z) in the Adda side chain via photosensitized isomerization involving the photoexcited phycocyanin. Our results indicate that photosensitized processes play a key role in the environmental fate and elimination of MCs in the natural waters. ^

Advancements in the micro-and nano-scale fabrication techniques have opened up new avenues for the development of portable, scalable and easier-to-use biosensors. Over the last few years, electrodes made of carbon have been widely used as sensing units in biosensors due to their attractive physiochemical properties. The aim of this research is to investigate different strategies to develop functionalized high surface carbon micro/nano-structures for electrochemical and biosensing devices. High aspect ratio three-dimensional carbon microarrays were fabricated via carbon microelectromechanical systems (C-MEMS) technique, which is based on pyrolyzing pre-patterned organic photoresist polymers. To further increase the surface area of the carbon microstructures, surface porosity was introduced by two strategies, i.e. (i) using F127 as porogen and (ii) oxygen reactive ion etch (RIE) treatment. Electrochemical characterization showed that porous carbon thin film electrodes prepared by using F127 as porogen had an effective surface area (Aeff 185%) compared to the conventional carbon electrode. To achieve enhanced electrochemical sensitivity for C-MEMS based functional devices, graphene was conformally coated onto high aspect ratio three-dimensional (3D) carbon micropillar arrays using electrostatic spray deposition (ESD) technique. The amperometric response of graphene/carbon micropillar electrode arrays exhibited higher electrochemical activity, improved charge transfer and a linear response towards H2O2 detection between 250&mgr;M to 5.5mM. Furthermore, carbon structures with dimensions from 50 nano-to micrometer level have been fabricated by pyrolyzing photo-nanoimprint lithography patterned organic resist polymer. Microstructure, elemental composition and resistivity characterization of the carbon nanostructures produced by this process were very similar to conventional photoresist derived carbon. Surface functionalization of the carbon nanostructures was performed using direct amination technique. Considering the need for requisite functional groups to covalently attach bioreceptors on the carbon surface for biomolecule detection, different oxidation techniques were compared to study the types of carbon-oxygen groups formed on the surface and their percentages with respect to different oxidation pretreatment times. Finally, a label-free detection strategy using signaling aptamer/protein binding complex for platelet-derived growth factor oncoprotein detection on functionalized three-dimensional carbon microarrays platform was demonstrated. The sensor showed near linear relationship between the relative fluorescence difference and protein concentration even in the sub-nanomolar range with an excellent detection limit of 5 pmol.

Although group 14 organometallic compounds (Si, Sn) have been well developed as transmetallation reagents in cross-coupling reactions, the application of organogermanium compounds as cross-coupling reagents is still a relatively new area with few papers published. This study aimed to develop methods for the synthesis of new classes of vinyl germane and vinyl silane compounds, mainly Z and E tris(trimethylsilyl)germanes and silanes, which were then applied to Pd-catalyzed cross-couplings with aryl and alkenyl halides. The stereoselective radical-mediated desulfonylation of vinyl sulfones with tris(trimethyl)germanium or silane hydrides provided access to the synthesis of trans vinyl germanes or silanes. Alternatively hydrogermylation or hydrosilylation of terminal alkynes gave cis vinyl germanes or silanes. The application of these new classes of organometallic compounds in cross-coupling reactions with various aryl and alkenyl halides under aqueous [NaOH/H2O2/Pd(PPh 3)4] and anhydrous [KH/t-BuOOH/Pd(PPh 3)4] oxidative conditions were investigated. ^ It was found that the vinyl tris(trimethylsilyl)germanes successfully underwent Pd-catalyzed cross-couplings with aryl and alkenyl halides and aryl triflates under aqueous and anhydrous oxidative conditions. These procedures provided examples of "ligand-free" Pd-catalyzed coupling of organogermanes with aryl and alkenyl halides. Interestingly, couplings with fluorinated vinyl germanes appeared to occur more easily than with the corresponding (α-fluoro)vinyl stannanes and silanes since neither addition of an extra ligand nor activation with fluoride was necessary. The vinyl tris(trimethyl)silanes were found to be alternative substrates for the Hiyama reaction. The coupling of TTMS-silanes with various aryl, heteroaryl as well as alkenyl halides proceeded smoothly upon treatment with hydrogen peroxide in the presence of sodium hydroxide and fluoride ion. ^

A abordagem metagenômica tem permitido o acesso ao material genético de microrganismos não cultivados e tem sido usada para identificação de novos genes. Apesar da importância dos mecanismos de reparo de DNA para a manutenção da integridade genômica nosso conhecimento sobre mecanismos de reparo de DNA é baseado em organismos modelo como E. coli e pouco é conhecido sobre os organismos de vida livre e não cultivados. Neste trabalho, a abordagem metagenômica foi aplicada para descobrir novos genes envolvidos com a manutenção da integridade genômica. Um clone positivo foi identificado por replicar a biblioteca metagenômica em meio seletivo contendo H2O2. O clone metagenômico foi capaz de complementar parcialmente a deficiência em reparo de DNA de cepas simples e duplo-mutantes de E.coli (recA e xthA nfo, respectivamente) submetidas ao estresse gerado por H2O2 e MMS.A análise de sequência mostrou uma ORF codificando para uma proteína hipotética membro da superfamília Exo_Endo_Phos (PF03372) e, a filogenia indicou que a mesma não está inclusa em nenhuma das subfamílias EEP. Assim, uma nova nuclease foi identificada e experimentalmente caracterizada in vivo e in vitro. Ensaios específicos utilizando a nuclease purificada e oligonucleotideos fluorescentemente marcados revelaram sua atividade 3´-5´exonuclease, em substratos simples e dupla-fita, dependente de Magnésio e sensível a EDTA. Uma vez que este é o primeiro relato e caracterização de uma enzima obtida a partir de abordagem metagenômica mostrando uma atividade exonuclease, foi nomeada EXOMEG1

With the increasing environmental awareness, maximizing biodegradability and minimizing ecotoxicity is the main driving force for new technological developments. Thus, can be developed new biodegradable lubricants for use in environmentally sensitive areas. The aim of this study was to obtain new bio-lubricants from passion fruit (Passiflora edulis Sims f. flavicarpa Degener) and moringa (Moringa oleifera Lamarck) epoxidized oils and develop a new additive package using experimental design for their use as a hydraulic fluid. In the first stage of this work was performed the optimization of the epoxidation process of the oils using fractional experimental design 24-1 , varying the temperature, reaction time, ratio of formic acid and hydrogen peroxide. In the second step was investigated the selectivity, thermodynamics and kinetics of the reaction for obtaining the two epoxides at 30, 50 and 70 °C. The result of the experimental design confirmed that the epoxidation of passion fruit oil requires 2 hours of reaction, 50 °C and a ratio H2O2/C=C/HCOOH (1:1:1). For moringa oil were required 2 hours reaction, 50 °C and a ratio of H2O2/C=C/HCOOH (1:1:1.5). The results of the final conversions were equal to 83.09% (± 0.3) for passion fruit oil epoxide and 91.02 (±0,4) for moringa oil epoxide. Following was made the 23 factorial design to evaluate which are the best concentrations of corrosion inhibitor and anti-wear (IC), antioxidant (BHA) and extreme pressure (EP) additives. The bio-lubricants obtained in this step were characterized according to DIN 51524 (Part 2 HLP) and DIN 51517 (Part 3 CLP) standards. The epoxidation process of the oils was able to improve the oxidative stability and reduce the total acid number, when compared to the in natura oils. Moreover, the epoxidized oils best solubilized additives, resulting in increased performance as a lubricant. In terms of physicochemical performance, the best lubricant fluid was the epoxidized moringa oil with additives (EMO-ADI), followed by the epoxidized passion fruit oil with additives (EPF-ADI) and, finally, the passion fruit in natura oil without additives (PFO). Lastly, was made the investigation of the tribological behavior under conditions of boundary lubrication for these lubricants. The tribological performance of the developed lubricants was analyzed on a HFRR equipment (High Frequency Reciprocating Rig) and the coefficient of friction, which occurs during the contact and the formation of the lubricating film, was measured. The wear was evaluated through optical microscopy and scanning electron microscopy (SEM). The results showed that the addition of extreme pressure (EP) and anti-wear and corrosion inhibitor (CI) additives significantly improve the tribological properties of the fluids. In all assays, was formed a lubricating film that is responsible for reducing the coefficient of metal-to-metal wear. It was observed that the addition of EP and IC additives in the in natura vegetable oils of passion fruit and moringa did not favor a significant reduction in wear. The bio-lubricants developed from passion fruit and moringa oils modified via epoxidation presented satisfactory tribological properties and shown to be potential lubricants for replacement of commercial mineral-based fluids.

The correlation between the type 1 diabetes mellitus and oxidative stress have been described in several studies, however its underlying mechanisms are not fully elucidated. The present work aimed to evaluate the effects of four weeks of streptozootocin-induced (STZ) diabetes in the redox homeostasis of rat hepatocytes. Thus, the liver of male Wistar rats from control and diabetic groups were collected and the activity and expression of antioxidant enzymes, as well the main markers of oxidative stress and content of H2O2 in these tissues were measured. The diabetes induced the activity of superoxide dismutase (SOD) and the gene expression of its mitochondrial isoform, SOD2. However, the expression of SOD1, the cytoplasmic isoform, was reduced by this disease. The activity and expression of catalase (CAT), as well the expression of glutathione peroxidase 1 (GPX1) and peroxiredoxin 4 (PRX4) were drastically reduced in the hepatocytes of diabetics rats. Even with this debility in the peroxidases mRNA expression, the content of H2O2 was reduced in the liver of diabetics rats when compared to the control group. The diabetes caused an increase of lipid peroxidation and a decrease of protein thiol content, showing that this disease causes distinct oxidative effects in different cell biomolecules. Our results indicate that four week of diabetes induced by STZ is already enough to compromise the enzymatic antioxidant systems of the hepatocytes.

Advanced Oxidation Processes (AOP) are techniques involving the formation of hydroxyl radical (HO•) with high organic matter oxidation rate. These processes application in industry have been increasing due to their capacity of degrading recalcitrant substances that cannot be completely removed by traditional processes of effluent treatment. In the present work, phenol degrading by photo-Fenton process based on addition of H2O2, Fe2+ and luminous radiation was studied. An experimental design was developed to analyze the effect of phenol, H2O2 and Fe2+ concentration on the fraction of total organic carbon (TOC) degraded. The experiments were performed in a batch photochemical parabolic reactor with 1.5 L of capacity. Samples of the reactional medium were collected at different reaction times and analyzed in a TOC measurement instrument from Shimadzu (TOC-VWP). The results showed a negative effect of phenol concentration and a positive effect of the two other variables in the TOC degraded fraction. A statistical analysis of the experimental design showed that the hydrogen peroxide concentration was the most influent variable in the TOC degraded fraction at 45 minutes and generated a model with R² = 0.82, which predicted the experimental data with low precision. The Visual Basic for Application (VBA) tool was used to generate a neural networks model and a photochemical database. The aforementioned model presented R² = 0.96 and precisely predicted the response data used for testing. The results found indicate the possible application of the developed tool for industry, mainly for its simplicity, low cost and easy access to the program.

O aumento da esperança média de vida tem elevado a prevalência de doenças neurodegenerativas, como é o caso da doença de Parkinson. Nos últimos anos a procura de novas soluções terapêuticas, assim como a minimização dos efeitos dos tratamentos atualmente utilizados tem promovido a procura de novas soluções. Deste modo, o objetivo deste trabalho consistiu no estudo dos mecanismos moleculares de neurotoxicidade induzidos pela dopamina (DA) e 6-hidroxidopamina (6-OHDA) num modelo celular do neuroblastoma humano (SH-SY5Y), bem como na avaliação do potencial neuroprotetor de extratos de algas com elevada capacidade antioxidante. O efeito neurotóxico da DA e 6-OHDA, assim como o efeito protetor dos extratos das algas com maior atividade antioxidante (Sargassum muticum, Saccorhiza polyschides, Padina pavonica, Codium tomentosum, Ulva compressa) foi avaliado através da viabilidade celular das células SH-SY5Y utilizando o método de MTT. De modo a compreender os efeitos induzidos na viabilidade celular pela DA e 6-OHDA procedeu-se ao estudo da atividade da caspase-3, alterações do potencial mitocondrial e quantificação de H2O2. Os resultados demonstraram um claro efeito dependente da concentração da DA (30-3000μM) e 6-OHDA (10-1000μM) na viabilidade celular das células SH-SY5Y, bem como do tempo de exposição (6-48h). No que diz respeito a prevenção do efeito neurotóxico da DA (1000μM (56,41±5,05% de células viáveis); 24h) e 6-OHDA (100μM (66,76±3,24% de células viáveis);24h) pelos extratos das algas (1mg/mL; 24h) verificou-se que os extratos que apresentaram um efeito preventivo mais marcado pertencem as algas Sargassum muticum (82,37±6,41% de células viáveis e 115,8±8,53% de células viáveis, após tratamento com DA e 6-OHDA, respetivamente), Saccorhiza polyschides (89,26±8,62% de células viáveis e 106,51±4,26% de células viáveis, após tratamento com DA e 6-OHDA, respetivamente) e Codium tomentosum (81,28±3,68% de células viáveis e 103,17±7,25% de células viáveis, após tratamento com DA e 6-OHDA, respetivamente). A morte celular induzida pela DA e pela 6-OHDA foi acompanhada pelo aumento da atividade da caspase-3 quando comparado com o controlo (DA - 66,46±1,49fluorescência (u.a)/mg de proteína/minuto; 6-OHDA - 22,56±1,71fluorescência (u.a)/mg de proteína/minuto; controlo – 4,8 ±0,48fluorescência (u.a)/mg de proteína/minuto), pela presença de elevadas quantidades de peróxido de hidrogénio (H2O2) (363,81±28,58 % do controlo e 214,26 ± 8,46 % do controlo, após tratamento com DA e 6-OHDA, respetivamente) e pela despolarização da membrana mitocondrial (162,3±2,34 % do controlo e 144,7±2,87 % do controlo, após tratamento com DA e 6-OHDA, respetivamente). Por sua vez, durante o tratamento com extratos das algas (1mg/mL) na presença de DA e 6-OHDA verificou-se uma inibição da atividade da caspase-3 induzida pelas algas Sargassum muticum (2,53±2,49fluorescência (u.a)/mg de proteína/minuto e 4,52±1,36 fluorescência (u.a)/mg de proteína/minuto, após tratamento com DA e 6-OHDA, respetivamente), Saccorhiza polyschides (4,71±0,70fluorescência (u.a.)/mg de proteína/minuto e 2,73±1,11 fluorescência (u.a.)/mg de proteína/minuto, após tratamento com DA e 6-OHDA, respetivamente) e Codium tomentosum (17,05±1,72fluorescência (u.a.)/mg de proteína/minuto e 2,58±1,77fluorescência (u.a)/mg de proteína/minuto, após tratamento com DA e 6-OHDA, respetivamente). De igual modo verificou-se uma diminuição da produção de H2O2 pelas células SH-SY5Y na presença dos extratos das algas Sargassum muticum (132,58 ± 10,68% controlo), Saccorhiza polyschides (150,54 ± 23,54% controlo) e Codium tomentosum (54,074 ± 6,66% do controlo), quando expostas a 6-OHDA, contudo não se verificou o mesmo efeito na presença de DA. Relativamente ao potencial mitocondrial observou-se uma inibição da despolarização mitocondrial induzida pela DA e 6-OHDA nas células SH-SY5Y pela presença dos extratos das algas Sargassum muticum (135,7±2,97% controlo e 49,3±1,17% controlo, após tratamento com DA e 6-OHDA, respetivamente), Saccorhiza polyschides (126,7±5,46% controlo e 94,3±1,70% controlo, após tratamento com DA e 6-OHDA, respetivamente). Os resultados obtidos demonstraram o potencial citoprotetor dos extratos de algas sobre efeitos neurotóxicos induzidos pela DA e 6-OHDA no modelo celular SH-SY5Y. O efeito protetor é mediado pela diminuição da condição de stress oxidativo, com redução da produção de H2O2, diminuição da atividade da caspase-3 e prevenção da alteração do potencial mitocondrial induzido pela DA e 6-OHDA. Conclui-se que os extratos de algas produzem moléculas bioativas com elevado potencial antioxidante, podendo ser uma fonte promissora de novos compostos neuroprotetores com aplicação terapêutica para doenças neurodegenerativas como a doença de Parkinson.

No âmbito da saúde e farmacologia a procura de compostos para a prevenção e tratamento de doenças é constante, tendo sido valorizada atualmente a medicina tradicional aliada à existência de diversos compostos naturais provenientes de plantas, fungos e bactérias com ação anti-inflamatória e antimicrobiana. Logo é valorizada a investigação e estudos específicos das propriedades terapêuticas de compostos, dos mecanismos de ação e compreensão dos seus efeitos, úteis ao avanço da medicina. Posto isto, aliada à relevância do stresse oxidativo, associado a danos oxidativos e envolvimento em várias doenças humanas e no processo de envelhecimento, surge a necessidade de construção de uma ferramenta que permita estudar o efeito de drogas ao nível do stresse oxidativo, de modo a avaliar o potencial terapêutico, clarificar a respetiva função e abranger a aplicação de compostos que têm sido cada vez mais valorizados. Assim, foi delineado como objetivo o melhoramento genético de estripes Saccharomyces cerevisiae, de modo a promover a acumulação de drogas e, consequentemente promover as condições que despoletam a ação dos ativadores Yap1 e Msn2 ao nível do stresse oxidativo. Neste contexto, foi otimizada uma metodologia para a construção de novas estirpes haploides de S. cerevisiae com seis cruzamentos distintos que combinam a deleção independente de três transportadores diferentes (TxΔ, TyΔ e TzΔ) e, em simultâneo, cada um dos ativadores Yap1 e Msn2 em fusão com a Gfp (A-GFP). Estas estirpes haploides foram selecionadas por análises fenotípicas, confirmadas ao nível do genótipo pretendido por PCR e ensaios de microscopia de fluorescência quando sujeitas previamente ao tratamento com H2O2, que não só confirmou o genótipo A-GFP, como também permitiu avaliar qualitativamente a sensibilidade das estirpes ao H2O2 pela observação da localização nuclear da Gfp. No sentido de obter uma ferramenta simples de fácil manipulação e baixo custo que facilite a exposição de compostos e permanência no interior das células, de modo a potenciar o seu efeito a baixas concentrações e permitir uma análise mais assertiva, criou-se uma ferramenta que permitirá a caracterização de propriedades e efeitos pró-oxidantes e/ou antioxidantes de drogas ao nível do stresse oxidativo em levedura

We measured light absorption in 42 marine snow, sea ice, seawater, brine, and frost flower samples collected during the OASIS field campaign between February 27 and April 15, 2009. Samples represented multiple sites between landfast ice and open pack ice in coastal areas approximately 5 km west of Barrow, Alaska. The chromophores that are most commonly measured in snow, H2O2, NO3-, and NO2-, on average account for less than 1% of sunlight absorption in our samples. Instead, light absorption is dominated by unidentified "residual" species, likely organic compounds. Light absorption coefficients for the frost flowers on first-year sea ice are, on average, 40 times larger than values for terrestrial snow samples at Barrow, suggesting very large rates of photochemical reactions in frost flowers. For our marine samples the calculated rates of sunlight absorption and OH production from known chromophores are (0.1-1.4) x 10**14 (photons/cm**3/s) and (5-70) x 10**-12 (mol/L/s), respectively. Our residual spectra are similar to spectra of marine chromophoric dissolved organic matter (CDOM), suggesting that CDOM is the dominant chromophore in our samples. Based on our light absorption measurements we estimate dissolved organic carbon (DOC) concentrations in Barrow seawater and frost flowers as approximately 130 and 360 µM C, respectively. We expect that CDOM is a major source of OH in our marine samples, and it is likely to have other significant photochemistry as well.

The mineralogy and geochemistry of a suite of nine manganese nodules from the South Atlantic have been determined. The Ce/La ratios of the nodules were investigated to see if they could be used as redox indicators to trace the oxygen content of the ambient water mass and the flow path of the Antarctic Bottom Water as has previously been successfully carried out in the Pacific Ocean. The Ce/La ratios of the nodules decrease in the sequence Lazarev Sea, Weddell Sea (10.4 and 9.7)>East Georgia Basin (6.5 and 7.1)>Argentine Basin (5.0), but then increase in the Brazil Basin (6.2) and Angola Basin (9.8 and 15.1). A further decrease was observed in the Cape Basin (7.6). An extremely high Ce/La ratio of 24.4 had already been determined for nodules sampled north of the Nares Abyssal Plain in the western North Atlantic. These data reflect the more complicated pattern of bottom water flow in the South Atlantic than in the South Pacific. The penetration of more oxygenated North Atlantic Deep Water into the South Atlantic accounts for the higher Ce/La ratios in the nodules from the Angola and Brazil basins. Based on this study, the flow path of the Antarctic Bottom Water could only be traced as far north as the Argentine Basin. The unique geochemistry of nodules from the central Angola Basin (high Mn/Fe and Ce/La ratios, high contents of Ni, Cu, Zn and Mo) appears to be a function of the nature of the overlying water mass and of the multiple diagenetic sources of metals to the nodules.
