植物在长期的进化过程中,已经对其生存环境具备了各种适应对策。放牧影响下草原植物的生态适应策略,决定了其是否能够忍耐或者适应放牧生境从而维持自身的生存和种群的延续。研究植食性动物对植物的影响有助于制定合理的放牧制度和草地利用方式,从而为防止草原退化和恢复退化草地提供重要的理论依据;同时研究植物对放牧的响应策略,对于草原生物多样性的保护和草原生物资源的合理利用具有重要的理论和实践意义。   本文以中科院内蒙古草原生态系统定位研究站放牧综合试验样地中的小叶锦鸡儿为主要研究对象。通过长期不同放牧强度的放牧试验和唾液涂抹等模拟试验,从形态学、有性生殖、种子萌发和遗传多样性等方面,探讨了小叶锦鸡儿对放牧家畜(绵羊)采食的生物学响应。本研究得到以下主要结论:   1. 通过小叶锦鸡儿形态和有性生殖的实验,可以看到放牧改变了该种植物的形态和生殖特性,不同放牧强度对其影响的程度是不相同的。在啃食压力下,小叶锦鸡儿的营养、生殖和防御之间存在消长关系。随着放牧强度的增加,小叶锦鸡儿对营养器官和有性生殖器官的投资均减少,而对防御器官的投资有增加的趋势,主要体现在:个体的小型化(植株高度、叶轴长度、小叶大小)和果荚数目及成熟种子数都随着放牧强度的增加而明显减少;物理性防御器官――刺,其密度和长度都明显的增加。同时,放牧也对植物花粉的品质产生了消极影响。   2. 放牧不仅影响了植物体本身的生物学特性,而且影响了子代的生物学特性。不同放牧强度下的植株产生的种子,其萌发速率明显不同。同时放牧强度和沙埋深度对小叶锦鸡儿的出苗率均具有显著影响,随着沙埋深度增加,出苗率明显降低,0~2cm是其适宜出苗的沙埋深度;浅层沙埋处理下,轻度放牧和重度放牧的出苗率差异显著。与轻度放牧相比,重牧条件下同一沙埋深度的种子出苗时间明显推迟;在相同放牧压力下,沙埋深度也影响了出苗时间。   3. 采食活动对植物本身的形态、生殖以及子代的萌发特性都产生了影响;通过AFLP实验证明了小叶锦鸡儿在长期的放牧历史活动中已经发生了遗传多样性的变化,重度放牧强度下的植株与轻度放牧条件下的植株具有相对较远的遗传距离,也就是说,小叶锦鸡儿种群的分化与放牧强度具有密切的关系。   4. 三种不同生活型植物(灌木-小叶锦鸡儿、半灌木-冷蒿和草本-羊草)对绵羊唾液涂抹的响应不同,刈割和涂抹绵羊唾液能够增加植物的净地上生物量,并促进植株增加地上部分的光合产物投资。同时表明,简单的机械剪除不能够真正反应放牧家畜采食所产生的生物学效应。

青藏高原是世界上最高的高原,被称为“地球第三极”,环境条件相当严酷。本研究通过ISSR标记的方法研究青藏高原两种相对克隆构型的克隆植物紫花针茅(Stipa purpurea Griseb.)和青藏苔草(Carex moorcroftii Falc. ex Boott)不同种群的遗传多样性及克隆格局,在分子水平上揭示两种植物对极端环境适应的机理。主要结果如下: 与其它针茅属植物相比,紫花针茅各种群平均遗传多样性相对较低,其遗传变异主要存在于种群之内;与其它苔草属植物相比,青藏苔草各种群平均遗传多样性也较低,而总的遗传多样性较高,其遗传变异主要存在于种群之间。紫花针茅各种群平均遗传多样性高于青藏苔草各种群平均遗传多样性,尽管差异不显著。紫花针茅和青藏苔草各种群间具有较高的遗传分化。 紫花针茅具有与其它分蘖型克隆植物相当的克隆多样性,说明有性生殖在其生活史中起着重要作用。青藏苔草具有与其它克隆植物相当的克隆多样性,而与北极地区苔草属植物相比,其克隆多样性则较低。 Pearson相关显示,紫花针茅遗传多样性与纬度、经度均成显著负相关,而与海拔高度相关关系不显著;青藏苔草克隆多样性及遗传多样性与纬度、经度、海拔高度均不相关,而克隆多样性与遗传多样性的比值则与纬度显著相关。唐古拉山对紫花针茅遗传格局未产生显著影响,山南北地区遗传多样性无显著差别;唐古拉山对青藏苔草遗传格局产生了重要影响,唐古拉山南北地区基因交流有显著隔离,唐古拉山以北地区种群遗传多样性显著高于以南地区,推测其种群自北向南迁移过程中的瓶颈效应起着重要作用。青藏苔草种群的克隆多样性与遗传多样性成显著正相关。Mantel test检验显示,紫花针茅和青藏苔草的地理距离与遗传距离间的关系均不显著。 紫花针茅以分蘖生长的方式由源株向外扩展,有明确、致密的边界,野外观察和室内检测的结果显示其环上其它基株也可进行生长成活,可形成一种克隆环镶嵌格局,克隆环或其边界可能是幼苗定居有利的生境;空间自相关也显示在较小的尺度内紫花针茅遗传相关性强,推测克隆繁殖在其中也起重要作用。青藏苔草种群不同尺度的取样其克隆多样性不同,小面积(1 × 1 m2)的克隆多样性均小于大面积的(10 × 10 m2),空间自相关也显示克隆生长在其中起重要作用;尽管青藏苔草有较强的克隆生长能力,在环境条件较好时其呈密集分布,表现出较强的可塑性。 总之,尽管青藏高原环境非常严酷,紫花针茅与青藏苔草仍有较高水平的克隆多样性和一定的遗传多样性;高原严酷的环境造成了两种植物较大的遗传隔离。紫花针茅环状结构对其有性更新有重要作用,青藏苔草克隆生长具有较大可塑性,这些都是植物对高原严酷环境的适应形式。

构树(Broussonetia papyrifera L.)为桑科(Moraceae)构树属(Broussonetia)落叶乔木,广泛分布于亚洲东部及太平洋岛屿。它是我国重要的经济林木,具有重要的经济价值,其树皮纤维品质优良,自古就是造纸的优良原料;叶片可用作饲料;果实具有重要的药用价值;环境适应性强,是迅速绿化荒山、荒滩和盐碱地的理想树种。因此对构树这些特性的深入研究和开发利用具有非常重要的实际应用价值。本研究以日本和国内构树主要分布区域的的10种生态型及杂交构树共23份材料,摸索并改进了构树DNA的提取方法,建立了稳定的SRAP分子标记体系,以杂交构树组培苗为材料从120对引物组合中筛选出条带信息较高的17对SRAP引物,以这些引物对23份构树材料进行PCR扩增和标记分析。 本研究取得的主要结果如下: (1)本实验首次将SRAP技术应用于构树的研究中,建立起构树稳定的SRAP-PCR反应体系;实验中对影响扩增的5个主要因素进行了优化,确定20 μL PCR反应体系中各因素最适浓度:模板DNA浓度80 ng/(20 μL),Mg2+浓度2.0 mM,dNTP浓度0.6 mM,引物浓度0.8 mM,Taq 酶浓度1.5 U/(20 μL)。 (2)从120对引物组合中筛选出来的17对SRAP引物对21份不同生态型构树样本(21份材料指的是除两份杂交构树材料外的其它生态型构树,以下同)进行PCR扩增,共扩增出439条带,平均每对引物25.5条,其大小介于100~1,000 bp之间,其中多态性条带319条,占总数的72.67%。 (3)用Popgene1.32软件进行分析,计算出Shannon信息指数(I)值为0.2275(0.2042),物种水平的Nei基因多样性(H)值为0.1336(0.1436),表明各生态型构树之间的平均遗传多态性不高,中国大陆各生态型构树Shannon信息指数(I)值仅为0.1675(0.2271),物种水平的Nei基因多样性(H)值为0.1039(0.1540),群体遗传多样性较低,构树的遗传分化主要存在于中国和日本之间。 (4)用NTSYS-2.10e软件进行聚类分析,发现不同生态型构树按距离关系远近及分布区域可划分为不同类群。在遗传相似系数0.57附近,对21份材料进行聚类分析发现其可分为两个群体,一类为日本生态型,另一类为中国生态型,表明日本构树与中国野生种构树种源遗传相似性较小。中国各生态型构树在遗传相似系数0.91处可分为5类,总体而言,中国各地区之间的构树遗传相似度较高。对杂交构树分析表明,其亲缘关系与日本构树(母本)更接近。 (5)日本及杂交构树的SCAR标记。本研究找到两条日本及杂交构树的特异性条带,回收、测序,再根据序列往里重新设计引物,转变成稳定性更好,更直观的SCAR标记,这为挑选性状优良的日本及杂交构树提供重要的参考,对其育种有一定的指导意义。其中一条片段经与NCBI数据库比对发现与拟南芥磺基转移酶家族基因具有较高的同源性。

长鳞红景天(Rhodiola gelida)属景天科红景天属,多年生草本。主要分布在我国新疆中部天山。在天山一号冰川观测试验站附近的高寒草甸上,从海拔3850m~4010m均有长鳞红景天分布。长鳞红景天是一种重要的药用植物,但目前尚未见到有关其遗传多样性方面的研究报道。本文利用相关序列扩增多态性(sequence-related amplified polymorphism,SRAP)分子标记技术,对新疆天山不同海拔高度3个长鳞红景天种群的遗传多样性,种群内及种群间的遗传变异进行研究,为有效保护和合理利用长鳞红景天物种资源提供理论依据。主要结果如下: (1)基于CTAB法,建立了适合长鳞红景天的DNA提取方法,获得的DNA经琼脂糖电泳和紫外分析仪检测,浓度和质量均较高。该法有利于去除长鳞红景天植物材料中多糖、多酚类等物质,获得的高质量基因组DNA能够满足后续的PCR实验要求。 (2)采用正交实验对影响长鳞红景天SRAP-PCR反应的4个主要因素(Taq酶用量、Mg2+浓度、dNTP浓度、引物浓度)进行优化试验,选出这4个因素的最佳水平组合,建立了长鳞红景天SRAP-PCR最佳反应体系。同时,根据最佳体系筛选出扩增稳定、多态性丰富的SRAP引物。通过梯度PCR确定引物的最佳退火温度,得到了适合长鳞红景天SRAP-PCR反应的最佳扩增程序。 (3)对20对SRAP引物的扩增结果进行统计分析显示:长鳞红景天物种水平的多态位点百分率(P)为96.00 %,Shannon信息指数(I)为0.4484、基因多样性指数(H)为0.2922,表现出较高的遗传多样性水平;在海拔梯度上,长鳞红景天各种群间的遗传多样性水平有一定差异,但没有呈现规律性的变化;AMOVA分子变异分析显示,在总遗传变异中24.76 % 的变异存在于种群间,75.24 % 的变异存在于种群内,种群间的基因分化系数(Gst)为0.1749,表明3个长鳞红景天种群的变异主要存在于种群内,但种群间也出现了一定程度的遗传分化;长鳞红景天3个种群间的基因流为2.3595,遗传距离平均值为0.0981(变化范围:0.0768 ~0.1196),遗传距离较近,说明海拔高度对长鳞红景天的地理隔离较小;相关性分析表明长鳞红景天种群的遗传多样性与各生态因子的相关性均不显著。

The CGIAR Research Program on Aquatic Agricultural Systems (AAS) seeks to reduce poverty and improve food security for many small-scale fishers and farmers who are dependent on aquatic agriculture systems by partnering with local, national and international partners to achieve large-scale development impact. This study on promising practices in food security and nutrition assistance to vulnerable households in the Tonle Sap region forms part of the preliminary research that informs AAS work in the highly productive Mekong Delta and Tonle Sap Lake floodplain. The study aims to identify and learn from promising practices that have had a positive impact on the food security and nutrition of vulnerable households in the Tonle Sap region.

采用水平式淀粉胶电泳技术 ,对云南龙陵黄山羊、宁蒗黑头山羊、马关无角山羊和路南圭山羊等 4个保种山羊的 1 2 0个个体共 3 9个基因座位的基因多态性进行了研究。结果显示 ,云南 4个保种山羊品种在AKP、CES - 1、ESD、GOI、LAP、MDH、ME和NP基因座位出现多态。多态座位基因在不同保种山羊中分布不同。多态基因座位百分比 (P)在 4个保种山羊中分别为 0 .2 0 51 ,0 .1 53 8,0 .1 2 82和 0 .1 53 8。平均杂合度 (H)分别为 0 .0 95,0 .0 61 4 ,0 .0 4 67和 0 .0 662。用UPGMA法对由基因频率计算得到的Nei氏标准遗传距离进行聚类分析 ,结果表明云南保种山羊具地理分布及品种特点 ,龙陵黄山羊和其它 3个品种的遗传距离最远。

文采用蛋白电泳技术研究滇南小耳猪血液蛋白多态性。共分析了滇南小耳猪32个遗传位 点,其中AKP、CAT、ES、PA、6PGD和TF等6个位点检测到多态性,多态位点百分比P=0.1875,平均 杂合度H=0.0712。结果表明滇南小耳猪的血液蛋白多态程度较高,反映在蛋白质水平上的遗传多样 性较为丰富。

mtDNA genotypes of six domestic horses (three adult short horses whose heights are under 1 m and three common domestic horses) from a small region of 15 km(2) in Malipo county of Yunnan province of China were investigated by the technique of restriction fragment length polymorphism (RFLP) with restriction endonucleases which recognize 6-bp sequences. An average of fragments for an individual was obtained. Unlike other domestic animals, this population of horses exhibits high mtDNA genetic diversity. Each of the six horses has a specific mtDNA genotype showing a pattern of multiple maternal origins, as suggested by fossil and literature records. We think the population of horses is an amazing seed-resource pool of horses and hence deserves to be paid more attention from the view of conservation genetics. However it is also remarkable that we did not find any typical mtDNA genetic markers which would discriminate between short horses and common domestic horses.

The genetic diversity and phylogeny of 26 isolates of Bursaphelenchus xlophilus from China, Japan, Portugal and North America were investigated based on the D2/3 domain of 28S rDNA, nuclear ribosomal Internal Transcribed Spacer (ITS) sequences, and random amplified polymorphic DNA (RAPD) analysis. The genetic diversity analysis showed that the D2/3 domain of 28S rDNA of isolates of B. xlophilus from China, Portugal, Japan and the US were identical and differed at one to three nucleotides compared to those from Canada. ITS sequences of isolates from China and Portugal were the same; they differed at one or two nucleotides compared to those of Japanese isolates and at four and 23 nucleotides compared to those front the US and Canada, respectively. The phylogenetic analysis indicated that Chinese isolates share a common ancestor with one of the two Japanese clades and that the Canadian isolates form a sister group of the clade comprised of isolates from China, Portugal,Japan, and the US. The relationship between Japanese isolates and those from China was closer than with the American isolates. The Canadian isolates were the basal group of B. xylophilus. This suggests that B. xlophilus originated in North America and that the B. xylphilus that occurs in China could have been first introduced from Japan. Further analysis based on RAPD analysis revealed that the relationship among isolates from Guangdong, Zhejiang, Shandong, Anhui provinces and Nanjing was the closest, which suggests that pine wilt disease in these Chinese locales was probably dispersed from Nanjing, where this disease first occurred in China.

对6个牛微卫星座位的遗传变异及多态性分析,以期了解BMY牛和婆罗门牛的群体遗传结构与育成情况.6个微卫星座位的多态信息含量为在0.524~0.752间,BMY牛、婆罗门牛两个群体的平均期望杂合度和观察杂合度值接近,分别为0.737 6和0.739 6,0 641 2和0.653 7,进入横交固定第二世代的BMY牛期望杂合度值较高,这与我们的育种进展相符.红安格斯的期望杂合度较低(0.460 9)接近日本和牛0.471,暗示红安格斯牛的同质性较高.

A fragment of mitochondrial DNA (mtDNA) control region (similar to700 bp) was sequenced in 104 individuals from 20 breeds (three Chinese domestic breeds, five recently derived breeds and 12 introduced breeds) of domestic rabbits, Oryctolagus cuniculus . Nineteen sites were polymorphic, with 18 transitions and one insertion/deletion, and eight haplotypes (A1, A2, A3, A4, A5, A6, A7 and A8) were identified. Haplotype A1 was the most common and occurred in 89 individuals. In the 25 Chinese rabbits, only haplotype A1 was observed, while four haplotypes (A1, A3, A5 and A6) were found in 26 recently derived individuals. Haplotype A2 was shared by seven individuals among three introduced strains. The other six haplotypes accounted for 0. 96-1. 92% of the animals. Combined with the published sequences of European rabbits, a reduced median-joining network was constructed. The Chinese rabbit mtDNAs were scattered into two clusters of European rabbits. These results suggest that the (so-called) Chinese rabbits were introduced from Europe. Genetic diversity in Chinese rabbits was very low.

Previous study using protein electrophoresis shows no polymorphism in 44 nuclear loci of Sichuan golden monkey (Rhinopithecus roxellana), which limits our understandings of its population genetic patterns in the nuclear genome. In order to obtain sufficient information, we scanned 14 microsatellite loci in a sample of 32 individuals from its three major habitats (Minshan, Qinling and Shennongiia). A considerable amount of polymorphisms were detected. The average heterozygosities in the local populations were all above 0.5. The differentiations among local populations were significant. There was evidence of geneflow among subpopullations, but geneflow between Qinling and Shennongjia local populations was the weakest. Minshan and Qinling populations might have gone through recent bottlenecks. The estimation of the ratio of the effective population sizes among local populations was close to that from census sizes. Comparisons to available mitochondria data suggested that R. roxellana's social structures played an important role in shaping its population genetic patterns. Our study showed that the polymorphism level of R. roxellana was no higher than other endangered species; therefore, measures should be taken to preserve genetic diversity of this species.

Background: Human skeletal system has evolved rapidly since the dispersal of modern humans from Africa, potentially driven by selection and adaptation. Osteogenin (BMP3) plays an important role in skeletal development and bone osteogenesis as an antagonist of the osteogenic bone morphogenetic proteins, and negatively regulates bone mineral density. Methodology/Principal Findings: Here, we resequenced the BMP3 gene from individuals in four geographically separated modern human populations. Features supportive of positive selection in the BMP3 gene were found including the presence of an excess of nonsynonymous mutations in modern humans, and a significantly lower genetic diversity that deviates from neutrality. The prevalent haplotypes of the first exon region in Europeans demonstrated features of long-range haplotype homogeneity. In contrast with findings in European, the derived allele SNP Arg192Gln shows higher extended haplotype homozygosity in East Asian. The worldwide allele frequency distribution of SNP shows not only a high-derived allele frequency in Asians, but also in Americans, which is suggestive of functional adaptation. Conclusions/Significance: In conclusion, we provide evidence for recent positive selection operating upon a crucial gene in skeletal development, which may provide new insight into the evolution of the skeletal system and bone development.
