The Mediterranean region is characterised by a variable climate with most of the rain falling during the winter and frequent summer droughts. Such warm, dry periods are ideal for the growth of large algal blooms that often consist of potentially toxic Cyanobacteria. This makes the management of water for human use particularly challenging in such a climate and it is important to understand how such blooms can be avoided or at least be reduced in size. PROTECH (Phytoplankton RespOnses To Environmental CHange) is a model that simulates the dynamics of different species of phytoplankton populations in lakes and reservoirs. Its distinct advantage over similar models is its ability to simulate the relative composition of the algal flora, allowing both quantitative and qualitative conclusions to be drawn e.g. whether Cyanobacteria could be a potential problem. PROTECH has been applied primarily to lakes and reservoirs in northern Europe. Recently, however, the model has been applied to water bodies in lower latitudes, including Australia to a water supply reservoir in the south of Spain, El Gergal. El Gergal is the last in a chain of reservoirs that supply water to the city of Seville. It was brought into service in April 1979 and has a maximum storage volume of 35 000 000 m3. This article summarises the application of PROTECH in order to simulate the following problems: • the effect of a large influx of Ceratium biomass into El Gergal from another reservoir • the effect of using alternative water sources instead of the Guadalquivir River (used occasionally to raise water levels in El Gergal) • the effect of installing tertiary sewage treatment on the Cala River • the effect of simulated drought conditions on phytoplankton in the reservoir.

A bacia hidrográfica do rio São Domingos constitui uma das sub-bacias do rio Muriaé pertencente ao sistema Paraíba do Sul e, tem seus limites coincidentes com os limites do município de São José de Ubá, sendo este o segundo maior produtor de tomate do estado do Rio de Janeiro, com sua principal fonte econômica baseada na agropecuária. Este tipo de atividade resulta em utilização de produtos químicos nas lavouras e juntamente a ocupação inadequada resulta em modificação das paisagens e da mata nativa, resultando em diversos tipos de impactos no ambiente. Neste estudo foram abordados os impactos relacionados a concentração de metais e sua proveniência através das assinaturas isotópicas Pb/Pb, utilizando para tal o procedimento analítico de lixiviação dos sedimentos de corrente e abertura total de rochas para a obtenção de razões isotópicas e concentrações de metais por ICP-MS. Os resultados mostraram que as contribuições nos sedimentos de corrente se dão a partir de cinco fontes compreendidos no intervalo de assinatura isotópica 206Pb/207Pb 1,1229 e 1,1949, representadas em intervalos bem definidos. Por correlação com as respectivas concentrações se observa que, preferencialmente, existe maior influência das rochas do embasamento, seguido de atividade antrópicas como a urbanização e disposição de lixo doméstico. As maiores concentrações estão associadas ao cobre, chumbo, estrôncio, níquel e zinco. Contudo todas as concentrações de metais obtidas se encontram abaixo da legislação vigente. Desta forma a contaminação antrópica é limitada a regiões de maior densidade populacional e as influências naturais predominam na área da bacia.

A utilização de testes de biocompatibilidade de materiais odontológicos é necessária para avaliar a segurança dos mesmos. Listerine é um enxaguatório comercial usado para a prevenção e tratamento da gengivite. O objetivo do estudo foi avaliar os efeitos citotóxico e genotóxico do Listerine em culturas de Escherichia coli e plasmídios. Na avaliação da citotoxicidade, culturas de E. coli AB1157 e BW9091 foram incubadas com Listerine (10, 50 e 100%) e o crescimento acompanhado pela densidade óptica (DO) em 600nm por 7 horas(h). Para avaliar a sobrevivência, culturas de E. coli AB1157, em fase exponencial, foram centrifugadas, ressuspensas em solução salina (NaCl 0,9%) e incubadas (1h, 37C) com Listerine (10, 50, 100%, 1h, 37 C). Alíquotas foram semeadas em placas de Petri contendo meio nutritivo nos tempos 0, 30 e 60 minutos e armazenadas em estufa bacteriológica (18h, 37 C). As unidades formadoras de colônias contadas e as frações de sobrevivência (FS) calculadas. Como controles, culturas tratadas salina ou etanol (21,6%). Para genotoxicidade, plasmídios pBSK foram incubados com Listerine (10, 50 e 100%) e com etanol (2,16%, 10,8% e 21,6%), associados ou não ao SnCl2(200g/mL, 30 minutos, temperatura ambiente), realizada eletroforese em gel de agarose (0,8%, 8V/cm), observados por transiluminação UV e obtido o percentual da forma superespiralada (%SE). Os resultados indicam que o enxaguatório Listerine foi capaz de inibir o crescimento bacteriano de culturas de E. coli na maior concentração utilizada. O enxaguatório, na maior concentração, diminuiu a sobrevivência das culturas bacterianas testadas. Listerine não modificou o perfil eletroforético do plasmídios, indicando ausência de efeito genotóxico e também foi capaz de proteger os plamídios da ação do SnCl2. Além disso, o etanol, na mesma concentração presente no Listerine, não alterou o perfil eletroforético dos plasmídios, sendo capaz de protegê-lo da ação do SnCl2. Os resultados indicaram que o Listerine apresentou efeito citotóxico em culturas de E. coli e ausência de potencial genotóxico em plamídios, sendo capaz de protegê-los, bem como o etanol, dos efeitos genotóxicos do SnCl2.

The disposal of sewage is the most important item in public sanitation. It is the most important present day problem in every city whether large or small. The direct cause of the majority of epidemics is the contamination of the water supply of the city by the excreta of man or animal. Public health varies directly as public sanitation, and if the public sanitation be good, the liability of sickness caused by contamination of the water supply is greatly lessened. When a city outgrows its sewerage system the public health becomes endangered. There are two causes for the increased amount of sewerage, increase in population and increase in industrial and manufacturing wastes. The main problem in this connection is the ultimate disposal of the matter which reaches the sewers.

Ten year comparison of fish survey's with respect to diversity evenness and composition of fish communities. The upper Patuxent River was divided into Piedmont Plateau and Coastal Plain regions, not only for geographical purposes, but also because of the clustering of sewage treatment plants in the Coastal Plain region. In the Piedmont Plateau region, the fish species diversity changed very little from 1966 to 1977 ( Little Patuxent -- 2.82 to 2.66; Middle Patuxent -- 2.86 to 2.83; and main stem -- 2.46 to 2.63), except in a section of Little Patuxent River at and below the City of Columbia where the species diversity index showed a significant reduction from 2.97 to 1.99, and in a section of the main stem Patuxent River immediately downstream from the Brighton Dam of the Triadelphia Reservoir where the index increased significantly from 1.66 to 3.20. In the Coastal Plain region, a significant reduction in the fish species diversity index occurred between 1966 and 1977 below the two sewage treatment plant outfalls : Savage -- 2.69 to 0 and Patuxent-Crofton -- 3.06 to 1.33. Also, the substantial reduction in the species diversity index which had already occurred in 1966 below the six other plant: outfalls of Fort Meade No. I, Fort Meade No. 2, Maryland House of Correction, Maryland City , Parkway and Bowie, remained depressed in 1977. On the other hand, below the Horsepen Sewage Treatment Plant (a tertiary plant practicing dechlorination) the species diversity index increased from 1.91 to 2.8. (PDF contains 48 pages)

[ES]Este proyecto pretende plantear una solución viable y que, por tanto, resulte económicamente rentable, para la gestión de los lodos de depuradora generados en una estación de tratamiento de aguas residuales. Así, este documento se centrará, por una parte, en realizar un acercamiento al mundo de la depuración de aguas residuales y en estudiar los distintos procesos y tratamientos que se llevan a cabo en una depuradora, y por otra, planteará la valorización energética de los lodos mediante su combustión en una planta de generación de energía eléctrica como la solución al problema de gestión que suponen, no sin antes haber valorado las distintas alternativas posibles en cuanto a la salida que se les podría dar a estos fangos. De esta forma, la base del proyecto, una vez se haya optado por la alternativa de la valorización energética como la opción a desarrollar, consistirá en plantear los principios y fundamentos necesarios para el diseño de la planta de generación de energía eléctrica mediante lodos de depuradora que se pretenderá instalar. Se realizará, por tanto, una descripción de la planta generadora de electricidad detallándose el proceso que se llevará a cabo en la misma. Asimismo, se realizará el dimensionamiento de los diferentes equipos que tomarán parte en dicho proceso. Para finalizar, se planteará el estudio económico y de rentabilidad del proyecto que constituirá la creación de una planta de generación eléctrica mediante lodos, con el fin de comprobar el grado de viabilidad del mismo, y se analizarán los posibles riesgos a los que puede exponerse un proyecto de este tipo.

After artificial activation or fertilization of non-nucleate fragments or eggs of the sea urchin, the mitochondria actively synthesize RNA. The RNA made in non-nucleate fragments is shown to be mostly single stranded and to be associated primarily with the low speed pellet of centrifuged cellular homogenates.

Protein synthesis is observed in non-nucleate fragments in the presence or absence of the mitochondrial RNA synthesis: it is found to be qualitatively similar but quantitatively less in the absence of the RNA synthesis. The continued syntheses of proteins in the non-nucleate fragments in the absence of mitochondrial RNA synthesis provides additional evidence for the presence of a stable messenger RNA component in the unfertilized sea urchin egg.

Since the uptake or actinomycin D was found to be inhibited by the presence of a fertilization membrane, ethidium bromide, at 10 μgs/ml, is used as an effective inhibitor of RNA synthesis in non-nucleate fragments and in early cleavage stage embryos. However, this same concentration of ethidium bromide is found to be only partially effective in blocking RNA synthesis at the mesenchyme blastula stage of development.

Low concentrations of ethidium bromide (2 and 5 μgs/ml) are found not to be lethal but to be capable of producing moderate developmental defects. In the presence of concentrations of ethidium bromide adequate to inhibit all the mitochondrial RNA synthesis (10 μgs/ml of ethidium bromide), from fertilization on, the embryos do not cleave beyond the 4-8 cell stages. When similar concentrations of ethidium bromide are added at an early mesenchyme blastula stage, the embryos do not gastrulate but continue to swim for more than 24 additional hours (adequate for control embryos to develop to a late prism stage). These results lead to the conclusion that mitochondrial RNA synthesis may be very essential for normal development to occur.

DNA is synthesized in the non-nucleate fragments of sea urchin eggs. None of the newly synthesized DNA is found in the closed circular form. When phenol extracted directly from the fragments, the DNA is found to sediment at approximately 38 and 27s in sucrose gradients but neither of these size classes could be found associated with the isolated mitochondria. The template for the synthesis of DNA in non-nucleate fragments remains unknown.

O presente trabalho tem por objetivo disponibilizar metodologia para o projeto de tratamento de esgoto sanitário através de disposição oceânica com utilização de emissário submarino, para localidades de pequeno porte. Também apresenta metodologia simplificada para obtenção de alguns dados oceanográficos necessários, na perspectiva de atender aos administradores dessas pequenas prefeituras envolvidas nas questões de atendimento de sua população quanto aos serviços de tratamento dos esgotos sanitários. Esses municípios, por serem pequenos e em país em desenvolvimento, carecem de recursos financeiros para as soluções convencionais de tratamento de esgotos sanitários. O trabalho contém uma revisão da bibliografia técnica relativa ao processo objeto deste estudo, não só referente ao projeto hidráulico como também à estabilidade física da tubulação do emissário assentada no leito do mar. Julgou-se necessário a realização, deste trabalho depois que se verificou que a implantação de emissários submarinos com diâmetros até da ordem de 300 mm, em geral tem seus custos inferiores aos dos sistemas convencionais.

An article discussing changes observed in phytoplankton of the Lake District. An overview is given of previous phytoplankton studies undertaken in the area, detailing some changes found in various waterbodies. Water quality changes in Lake Windermere are mentioned, including the gradual increase of dissolved organic matter (DOM), believed to be caused by the increase of sewage to the lake. The lakes in the Alps are given as an example of a similar anthropogenic pollution scenario. The treatment of a Lake District tarn with bone meal is described. The article goes on to discuss the composition of plankton throughout the year under a variety of climatic conditions. A figure shows seasonal variation in the consistuents of phytoplankton in Windermere (north basin).

Theoretical and experimental studies were made on two classes of buoyant jet problems, namely:

1) an inclined, round buoyant yet in a stagnant environment with linear density-stratification;

2) a round buoyant jet in a uniform cross stream of homogenous density.

Using the integral technique of analysis, assuming similarity, predictions can be made for jet trajectory, widths, and dilution ratios, in a density-stratified or flowing environment. Such information is of great importance in the design of disposal systems for sewage effluent into the ocean or waste gases into the atmosphere.

The present study of a buoyant jet in a stagnant environment has extended the Morton type of analysis to cover the effect of the initial angle of discharge. Numerical solutions have been presented for a range of initial conditions. Laboratory experiments were conducted for photographic observations of the trajectories of dyed jets. In general the observed jet forms agreed well with the calculated trajectories and nominal half widths when the value of the entrainment coefficient was taken to be α = 0.082, as previously suggested by Morton.

The problem of a buoyant jet in a uniform cross stream was analyzed by assuming an entrainment mechanism based upon the vector difference between the characteristic jet velocity and the ambient velocity. The effect of the unbalanced pressure field on the sides of the jet flow was approximated by a gross drag term. Laboratory flume experiments with sinking jets which are directly analogous to buoyant jets were performed. Salt solutions were injected into fresh water at the free surface in a flume. The jet trajectories, dilution ratios and jet half widths were determined by conductivity measurements. The entrainment coefficient, α, and drag coefficient, Cd, were found from the observed jet trajectories and dilution ratios. In the ten cases studied where jet Froude number ranged from 10 to 80 and velocity ratio (jet: current) K from 4 to 16, α varied from 0.4 to 0.5 and Cd from 1.7 to 0.1. The jet mixing motion for distance within 250D was found to be dominated by the self-generated turbulence, rather than the free-stream turbulence. Similarity of concentration profiles has also been discussed.

Although its shores lack sizeable settlements, its main inflow receives sewage effluent from a major tourist centre (Keswick) and is subject to episodic floods. These influences, the growing development of leisure activities at the lake (e.g. sailing, time-share units), and recent road-construction, have led to past appraisals of ecological impacts and lake management. The lake has not been the subject of intense and long-term ecological study, but much scattered information exists that is relevant to future management decisions. In the present Report, commissioned by North West Water, such information - published and unpublished - is surveyed. Especial attention is given to evidence bearing on susceptibility to change, affecting the lake environment and its biota or species of conservation interest. Extensive use has been made of the results of a recent (1986-7) seasonal survey by the FBA.

The word ”Broads” is used to describe a series of relatively shallow lakes resulting from the flooding of medieval peat diggings. Broadland is essentially freshwater, but because the rivers have such low gradients the lower reaches are brackish. The influence of tide is particularly apparent on the River Yare; in Norwich 40 km from the sea there is a vertical movement of half a metre at spring tide. This study examines the problems that the broadlands are facing. The problems are basically the progressive loss of aquatic plants, in particular the macro- phytes, animal life, outbreaks of avian botulism, occasional fish kills due to a toxin produced by the blue-green alga Prymesium parvum and the emergence of very heavy algal blooms. The main factor for the deteriation of the Broaslands is the eutrophication resulting from enhanced nutrient inputs, in particular of nitrates and phosphates, from a variety of sources. Esthwaite Water is the most productive or eutrophic lake in the English Lake District. Since 1945 its water quality has been determined from weekly or biweekly measurements of temperature, oxygen, plant nutrients and phytoplankton abundance. The lake receives phosphorus from its largely lowland-pasture catchment, sewage effluent from the villages of Hawkshead and Near Sawrey, and from a cage-culture fish farm. From 1986 phosphorus has been removed from the sewage effluent of Hawkshead which was considered to contribute between 47% and 67% of the total phosphorus loading to the lake. At the commencement of phosphorus removal regular measurements of phosphorus in the superficial 0-4 cm layer of lake sediment were made from cores collected at random sites. Since 1986 the mean annual concentration of alkali-extractable sediment phosphorus has decreased by 23%. This change is not significant at the 5% level but nearly so. There has been no marked change in water quality over this period. Summer dominance of blue-green algae which arose in the early 1980s after decline of the previous summer forms, Ceratium spp., has been maintained. Improvement in water quality is unlikely to be achieved at the present phosphorus loading.

Recent advances in our knowledge of the genetic structure of human caliciviruses (HuCVs) and small round-structured viruses (SRSVs) have led to the development of polymerase chain reaction (PCR)-based molecular tests specific for these viruses. These methods have been developed to detect a number of human pathogenic viruses in environmental samples including water, sewage and shellfish. HuCVs and SRSVs are not culturable, and no animal model is currently available. Therefore there is no convenient method of preparing viruses for study or for reagent production. One problem facing those attempting to use PCR-based methods for the detection of HuCVs and SRSVs is the lack of a suitable positive control substrate. This is particularly important when screening complex samples in which the levels of inhibitors present may significantly interfere with amplificiation. Regions within the RNA polymerase regions of two genetically distinct human caliciviruses have been amplified and used to produce recombinant baculoviruses which express RNA corresponding to the calicivirus polymerase. This RNA is being investigated as a positive control substrate for PCR testing, using current diagnostic primer sets. Recombinant baculovirus technology will enable efficient and cost-effective production of large quantities of positive control RNA with a specific known genotype. We consider the development of these systems as essential for successful screening and monitoring applications.
