Copper and iron metabolism intersect in mammals. Copper deficiency simultaneously leads to decreased iron levels in some tissues and iron deficiency anemia, whereas it results in iron overload in other tissues such as the intestine and liver. The copper requirement of the multicopper ferroxidases hephaestin and ceruloplasmin likely explains this link between copper and iron homeostasis in mammals. We investigated the effect of in vivo and in vitro copper deficiency on hephaestin (Heph) expression and activity. C57BL/6J mice were separated into 2 groups on the day of parturition. One group was fed a copper-deficient diet and another was fed a control diet for 6 wk. Copper-deficient mice had significantly lower hephaestin and ceruloplasmin (~50% of controls) ferroxidase activity. Liver hepcidin expression was significantly downregulated by copper deficiency (~60% of controls), and enterocyte mRNA and protein levels of ferroportin1 were increased to 2.5 and 10 times, respectively, relative to controls, by copper deficiency, indicating a systemic iron deficiency in the copper-deficient mice. Interestingly, hephaestin protein levels were significantly decreased to ~40% of control, suggesting that decreased enterocyte copper content leads to decreased hephaestin synthesis and/or stability. We also examined the effect of copper deficiency on hephaestin in vitro in the HT29 cell line and found dramatically decreased hephaestin synthesis and activity. Both in vivo and in vitro studies indicate that copper is required for the proper processing and/or stability of hephaestin.

In oligotrophic waters the light spectrum is mostly blue, and therefore the physiological and biochemical responses to blue light occurring in the coral tissue and in the symbiotic algae are important. Examination of the wavelength dependence of two free radical scavenger enzyme activity revealed an increase in activity in the blue light range (440-480 nm) compared to the red (640680 nm) in the full visible light (400-700 nm) range. These data show for the first time the relationship between the action spectra of photosynthesis and the activity of two main antioxidant enzymes in the symbiotic coral Favia favus. It was found that in the animal (host) the enzyme response to the spectral distribution of light was higher than that of the zooxanthellae, probably due to accumulation of free radicals within the host tissue. Furthermore, we found that the activity of these enzymes is affected in nature by the length of the day and night, and in the laboratory, by the duration of the illumination. Changes in the pigment concentrations were also observed in response to growth under the blue region and the whole PAR spectrum, while fluorescence measurements with the fast repetition rate fluorometer (FRRF) showed a decrease in the sigma cross section and a decrease in the quantum yield also in the blue part of the spectrum. These changes of scavenger enzymes activity, pigment concentration and fluorescence yield at different light spectra are vital in acclimatization and survival of corals in shallow water environments with high light radiation. Variation in the rate at which parasitic gnathiid isopod juveniles emerged from the benthos at Lizard Island, Great Barrier Reef, Australia, was examined (I) every 4 or 8 h throughout the day and night over a 24 h period, (2) over a 12 h period during the day or night, and (3) during different lunar phases (weeks). The number of gnathiids sampled per 4 or 8 h was low, with only 30% of the traps containing gnathiids and the abundance ranging from 0 to 3 gnathiids m(-2). The number of gnathiids that emerged over 12 h, in contrast, ranged from 0 to 36 m(-2). During the third and fifth weeks sampled, more gnathiids emerged during the day than at night. This coincided with the full moon and new moon. Most gnathiids that emerged from the reef during the day (98 %) had not fed, in contrast to those sampled at night (71%). Of the gnathiids with no engorged gut, most (97 %) of those collected during the day were small (II. mm) compared to those collected at night (19%), the latter being mostly >1 mm. Of the gnathiids with an engorged gut, most were sampled at night (83 %) and 97 % were >1 mm in size. These percentages suggest differences in the emergence behaviour among Life stages or species of gnathiids. This study, which shows that gnathiids do emerge during the day and supports other studies showing that gnathiids also attack fishes during the day, has important implications for understanding the role of cleaner fish and their main food source, gnathiids, as it shows there is a constant source of gnathiids emerging from the reef during the day and night in search of hosts.

(U–Th)/He dating of goethite, when combined with quantification of diffusive 4He loss by the 4He/3He methodology, provides reliable corrected ages for minerals precipitated in weathering profiles. We have combined (U–Th)/He dating of supergene goethite with 40Ar/39Ar dating of supergene manganese oxides to study the weathering history and landscape evolution in the Hamersley Province, northwestern Australia. Incremental heating 40Ar/39Ar analysis of 187 grains of Mn oxides from 65 samples (44 hand specimens) collected from weathering profiles at seven field sites across the Hamersley Province yield precipitation ages ranging from 63.4 ± 0.9 to 1.5 ± 0.2 Ma. These results, combined with previous results of 40Ar/39Ar dating of Mn oxides (Vasconcelos, 1998 Vasconcelos, P.V., 1998. Unpub. report, pp. 1–278.Vasconcelos, 1998 and Cochrane, 2003), reveal a protracted and episodic history of weathering and landscape evolution, which was already ongoing in Late Cretaceous and spans the Palaeogene and Neogene. Seventy-three grains of goethite from 39 samples extracted from 21 hand specimens, collected from the same field sites where the Mn oxides originated, were dated by the (U–Th)/He method. Internally consistent (U–Th)/He ages, which range from 84.3 ± 12.2 to 3.3 ± 0.5 Ma, have been obtained for most samples when corrections are applied for 10% helium diffusive loss. The geochronological results obtained show remarkable similarity in the distribution of ages associated with supergene mineral precipitation. The widespread occurrence of iron oxides such as goethite in soils and weathering profiles and the successful application of (U–Th)/He dating of goethite offers great opportunities for extracting the wealth of palaeoclimatic and palaeoenvironmental information recorded by these profiles on the surface of terrestrial planets such as Earth and Mars.

O trabalho que tem por título: O Direito é torcido à Porta porque a Justiça se encontra deitada por terra é um exercício exegético que tem por objeto a perícope Am 5,10-13. Na análise semântica dos seus principais vocábulos evidenciou-se uma realidade social, política e econômica paradoxal em Israel, sob o comando de Jeroboão II (787-747 a.C.), como resultado de uma expansão territorial e comercial, de vitórias militares e da organização de um Estado tributarista. Esse modelo de sociedade gerou um antagonismo social entre uma elite abastada que esbanjava luxo e ostentação, à custa do suor e da fome de uma população empobrecida, especialmente a classe camponesa, que trabalhava para sustentar as benesses do mundo urbano. É de dentro dessa realidade que ecoa o grito de Amós como denúncia a esse estado de coisas, como palavra de desgraça e condenação a toda sorte de desmandos praticados em Israel. Entre esses a falência do sistema judiciário, pela prática da exploração e corrupção por parte dos magistrados, de ricos comerciantes e latifundiários, desviando o pobre do seu direito de recorrer em sua defesa perante o tribunal. Em razão disso, Amós anuncia a ruína de Israel, com o Dia de Javé, que será um anti-Êxodo, e aponta uma exigência ético-religiosa como forma de reverter esse não futuro para Israel, que se traduz no compromisso de estabelecer à Porta o Direito e a Justiça.

Ulrico Zuínglio, que iniciou a Reforma protestante na Suíça, rompeu com uma concepção antiga dos sacramentos, não os considerando meios de graça. Os sacramentos do batismo e da Ceia do Senhor, mantidos pelo reformador, recebem novos enfoques, em particular a Ceia, ao ser considerada apenas uma recordação do sacrifício de Cristo na cruz. A ruptura deu-se também na dissociação da Ceia do Senhor do Dia do Senhor. João Calvino, reformador de Genebra e considerado o principal teólogo da chamada Tradição Reformada, manteve a interpretação dos sacramentos como meios de graça e defendeu, embora sem sucesso, a celebração dominical da Ceia do Senhor. A tese demonstra a presença dominante da concepção de Zuínglio nas denominações presbiterianas brasileiras e suas implicações litúrgicas, a despeito de essas igrejas considerarem-se calvinistas. Inclui um estudo da mediação norte-americana na teologia dos sacramentos, tendo em vista que o presbiterianismo implantou-se definitivamente no Brasil, no século XIX, através de missionários procedentes dos Estados Unidos. Esses missionários consideraram a população católica brasileira como principal alvo de evangelização, praticando o rebatismo de conversos, prática esta que se tornaria usual no presbiterianismo brasileiro. Aspectos da liturgia de igrejas presbiterianas brasileiras, como a celebração da Ceia do Senhor de modo informal e mesmo, por vezes, com omissão da Oração Eucarística ou das palavras da instituição do sacramento, a presença ou não de elementos clássicos do culto, como o Credo Apostólico ou Niceno e a Oração do Senhor, constituíram-se objeto da presente pesquisa in loco. Algumas tentativas de recuperação de elementos da tradição litúrgica encontram resistências no presbiterianismo brasileiro, seja em razão de uma identidade negativa , o anticatolicismo, seja pelo atual contexto cultural de relativização de valores em detrimento de marcos históricos de identidade cristã. A pesquisa constata que a herança teológica zuingliana, em cujo desenvolvimento intervieram novos fatores, relaciona-se a dificuldades de inserção das principais igrejas presbiterianas brasileiras no movimento ecumênico do século XX.(AU)

Destacar a importância da reportagem investigativa, e mostrar o valor que deve ser observado no empenho da atuação profissional ética de um repórter de televisão, capaz de mudar o rumo da história política recente do Brasil, é o que se propõe o presente trabalho. A prática do jornalismo autêntico demonstra ser transformadora. É o que acontece com a atuação do repórter Caco Barcellos. Ele se propôs a perseguir uma história por mais de um ano e, ao final, mostrar uma verdade escondida durante 30 anos pelo poder público. Com tal reportagem, possibilitou a reparação de uma injustiça social cometida contra uma jovem estudante brasileira. A força da reportagem apresentada num telejornal é incontestável, em razão do potencial que representa a televisão na penetração em todas as camadas da sociedade como veículo de informação. Sendo a reportagem apresentada em série de três dias consecutivos pelo Jornal Nacional, eleva seu peso significativamente, devido à liderança de audiência do telejornal da Rede Globo. O investimento da emissora numa reportagem como a apresentada neste trabalho depende fundamentalmente da presença de um repórter profundamente identificado com a luta contra a injustiça social. E é este o perfil de Caco Barcellos. Ele cresceu na periferia pobre de uma metrópole, e desde muito cedo se revoltou contra a ação policial que discrimina o cidadão comum e privilegia os poderosos. Conseguiu transformar a raiva acumulada em ações efetivas através da reportagem investigativa reveladora da injustiça social cometida diariamente. Quando o espaço era pequeno para o tamanho do que tinha para reportar, Caco lançou livros, ganhou prêmios e enfrentou processos judiciais, movidos por policiais interessados em destruir a vida profissional do repórter que ousou trabalhar sério e encontrar os documentos que provavam os desmandos praticados contra inocentes, friamente assassinados. O resultado é uma atuação exemplar, com um método de trabalho digno de ser seguido, refletindo-se na série de reportagens que desmontou uma farsa montada pelo exército, e que pode ser considerada como uma verdadeira aula de jornalismo.

Este estudo, inserido no contexto do professor da educação profissional no Brasil, traz à tona uma realidade pouco discutida no meio acadêmico. Discussão esta que para muitos educadores não faz sentido por acreditarem que os cursos técnicos não fazem parte da educação, mas sim de uma formação do indivíduo para o trabalho. Contudo, se é formação, deve existir alguém a formar este profissional, então, quem é este professor? Será mesmo um docente ou apenas um mero reprodutor de técnicas e conhecimentos científicos capaz de levar seu aluno a aprender apertar um parafuso ? Ou será alguém de uma criatividade e conhecimentos inquestionáveis, mas cuja práxis em sala de aula deixa a desejar por lhe faltar a metodologia para o processo ensino-aprendizagem? Para tentar entender um pouco mais deste universo esta análise será feita por meio de entrevistas com professores Engenheiros, Tecnólogos e Administradores de Empresa licenciados e não licenciados atuando na formação profissional, além de uma análise sobre os cursos de licenciatura no Brasil, conhecidos por Programas Especiais de Formação Pedagógica de Docentes, voltados a estes profissionais e o que trazem da contribuição em suas práticas diárias.(AU)

In this paper the performance of opening and closing returns, for the components of the FT-30 will be studied. It will be shown that for these stocks opening returns have higher volatility and a greater tendency towards negative serial correlation than closing returns. Unlike previous studies this contrasting performance cannot solely be attributed to differences in the trading mechanism across the trading day. All the stocks used in our sample trade thought the day using a uniform trading mechanism. In this paper, we suggest that it is differences in the speed that closing and opening returns adjust to new information that causes differences in return performance. By estimating the Amihud and Mendelson (1987) [Amihud, Yakov, & Mendelson, Haim (1987). Trading mechanisms and stock returns: An empirical investigation, Journal of Finance, 62 533-553.] partial adjustment model with noise, we show that opening returns have a tendency towards over-reaction, while closing returns have a tendency towards under-reaction. We suggest that it is these differences that cause a substantial proportion (although not all) of the asymmetric return patterns associated with opening and closing returns. © 2005 Elsevier Inc. Purpose - One of the principal organizational developments in the last decade has been the pervasive influence of computer mediated communication (CMC) tools. The purpose of this paper is to closely interrogate the day-to-day role of e-mail in explicating, influencing and shaping social and information interactions within an organization. Design/methodology/approach - A series of in-depth interviews (n = 29) were undertaken to elicit employee opinions on their e-mail adaptation, experiences and practices. Findings - The paper provides insights into the polymorphic role of e-mail, particularly the way in which it is adapted by individuals within the organization. Specifically, it shows how this tool interacts within day-to-day work activities and tasks. Research limitations/implications - This paper investigates only one CMC tool, e-mail, although it is envisaged that this initial work will be used to raise a new understanding of the socially skilled adaptation of other CMC tools by employees as well as leaders. Practical implications- Previously unreported insights into employee opinion are delineated in order to provide a focus from which organizations can train and develop their employees and leaders to maximise knowledge creation within the organization. Originality/value - This study assesses CMC from an under-researched "real-life" perspective in which everyday interactions are used to understand employee reactions to e-mail communication and hence foster an atmosphere in which these interactions assist organizational development.

W. G. Sebald’s special interest in marginal authors shaped his critical perception of Austrian postwar writers. Focusing on his critical writings on Herbert Achternbusch and the schizophrenic poet Ernst Herbeck—considered by Sebald as paradigms of a disregarded, “minor” form of literature—this essay demonstrates how on the one hand Sebald placed their texts within an anthropological context of an “untamed thinking” (pensée sauvage) and how on the other he sought to reverse the standard practice of evaluating the literary value of “minor” writers from the perspective of established authors. As this critical approach indirectly suggests, literary criticism needs to adopt a “minor” stance to be truly able to unlock the meaning of normally marginalised texts.

Contact lenses have become a popular method of vision correction for millions of people globally. As with all devices designed for use within the body, interactions occur between the implanted material and the surrounding biological fluid. A common complaint of lens wearers is that they often experience symptoms of dry eye whilst wearing lenses. This sensation is often heightened towards the end of the day. Through the course of this study, various analytical techniques have been utilised including one dimensional electrophoresis and Western Blotting to study the protein profiles of tear samples. By studying the tears of non-contact lens wearers, it was possible to analyse what could be considered normal, healthy, individuals. A clinical study was also undertaken which followed a population of individuals from the neophyte stage to one whereby they were accustomed lens wearers. Tears were monitored at regular intervals throughout the course of this study and worn contact lenses were also analysed for proteins that had been deposited both on and within the lens. Contact lenses disrupt the tear film in a physical manner by their very presence. They are also thought to cause the normal protein profile to deviate from what would be considered normal. The tear film deposits proteins and lipids onto and within the lens. The lens may therefore be depriving the tear film of certain necessary components. The ultimate aim of this thesis was to discover how, and to what extent, lenses affected tear proteins and if there were any proteins in the tear fluid that had the potential to be used as biochemical markers. Should this be achievable it may be possible to identify those individuals who were more likely to become intolerant lens wearers. This study followed the changes taking place to the tear film as an effect of wearing contact lenses. Twenty-eight patients wore two different types of silicone hydrogel lenses in both a daily wear and a continuous wear regime. The tear protein profiles of the lens-wearers were compared with a control group of non-lens wearing individuals. The considerable amount of data that was generated enabled the clearly observable changes to the four main tear proteins to be monitored.

The election on 6 May 2007 of Nicolas Sarkozy as President of the Republic ushered in the promise of a new era in France. Sarkozy’s presidency follows those of the Socialist François Mitterrand (1981-95) and the neo-Gaullist Jacques Chirac (1995-2007), who together occupied France’s highest political office for more than a quarter-century. From the outset, Sarkozy’s presidential campaign was predicated on the need for change in France, for a “rupture” with the past; and his emphatic victory against the Socialist Ségolène Royal gave him a mandate to effect this. The legislative elections of June 2007, by assuring a strong majority in the National Assembly for Sarkozy’s centre-right Union pour un Mouvement Populaire (UMP), left the way clear for implementing the new President’s reform agenda over the next five years. This article examines the political context within which Sarkozy was elected to power, the main proposals of his presidential program, the challenges he faces, and his prospects for bringing real change to France.

Post-operative infections resulting from total hip arthroplasty are caused by bacteria such as Staphylococcus aureus and Pseudomonas aeruginosa entering the wound perioperatively or by haemetogenous spread from distant loci of infection. They can endanger patient health and require expensive surgical revision procedures. Gentamicin impregnated poly (methyl methacrylate) bone cement is traditionally used for treatment but is often removed due to harbouring bacterial growth, while bacterial resistance to gentamicin is increasing. The aim of this work was to encapsulate the antibiotics vancomycin, ciprofloxacin and rifampicin within sustained release microspheres composed of the biodegradable polymer poly (dl-lactide-co-glycolide) [PLCG] 75:25. Topical administration to the wound in hydroxypropylmethylcellulose gel should achieve high local antibiotic concentrations while the two week in vivo half life of PLCG 75:25 removes the need for expensive surgical retrieval operations. Unloaded and 20% w/w antibiotic loaded PLCG 75:25 microspheres were fabricated using a Water in Oil emulsification with solvent evaporation technique. Microspheres were spherical in shape with a honeycomb-like internal matrix and showed reproducible physical properties. The kinetics of in vitro antibiotic release into newborn calf serum (NCS) and Hank's balanced salt solution (HBSS) at 37°C were measured using a radial diffusion assay. Generally, the day to day concentration of each antibiotic released into NCS over a 30 day period was in excess of that required to kill St. aureus and Ps. auruginosa. Only limited microsphere biodegradation had occurred after 30 days of in vitro incubation in NCS and HBSS at 37°C. The moderate in vitro cytotoxicity of 20% w/w antibiotic loaded microspheres to cultured 3T3-L1 cells was antibiotic induced. In conclusion, generated data indicate the potential for 20% w/w antibiotic loaded microspheres to improve the present treatment regimens for infections occurring after total hip arthroplasty such that future work should focus on gaining industrial collaboration for commercial exploitation.

The thesis examines Kuhn's (1962, 1970) concept of paradigm, assesses how it is employed for mapping intellectual terrain in the social sciences, and evaluates it's use in research based on multiple theory positions. In so doing it rejects both the theses of total paradigm 'incommensurability' (Kuhn, 1962), and also of liberal 'translation' (Popper, 1970), in favour of a middle ground through the 'language-game of everyday life' (Wittgenstein, 1953). The thesis ultimately argues for the possibility of being 'trained-into' new paradigms, given the premise that 'unorganised experience cannot order perception' (Phillips, 1977). In conducting multiple paradigm research the analysis uses the Burrell and Morgan (1979) model for examining the work organisation of a large provincial fire Service. This analysis accounts for firstly, a 'functionalist' assessment of work design, demonstrating inter alia the decrease in reported motivation with length of service; secondly, an 'interpretive' portrayal of the daily accomplishment of task routines, highlighting the discretionary and negotiated nature of the day's events; thirdly, a 'radical humanist' analysis of workplace ideology, demonstrating the hegemonic role of officer training practices; and finally, a 'radical structuralist' description of the labour process, focusing on the establishment of a 'normal working day'. Although the argument is made for the possibility of conducting multiple paradigm research, the conclusion stresses the many institutional pressures serving to offset development.
