A kind of simple atomic force microscopy (AFM) relocated technique, which takes advantage of homemade sample locator system, is used for investigating repeatedly imaging of some specific species on the whole substrate (over 1 x 1 cm(2)) with resolution about 400 nm. As applications of this sample locator system, single extended DNA molecules and plasmid DNA network are shown in different AFM operational modes: tapping mode and contact mode with different tips after the substrates have been moved.

Facilitated alkali metal ion (M+= Li+, Na+, K+, Rb+, and Cs+) transfers across the micro- and nano-water/1,2-dichloroethane (W/DCE) interfaces supported at the tips of micro- and nanopipets by dibenzo-18-crown-6 (DB18C6) have been investigated systematically using cyclic voltammetry. The theory developed by Matsuda et al. was applied to estimate the association constants of DB18C6 and M+ in the DCE phase based on the experimental voltammetric results. The kinetic measurements for alkali metal ion transfer across the W/DCE interface facilitated by DB18C6 were conducted using nanopipets or-submicropipets, and the standard rate constants (k(0)) were evaluated by analysis of the experimental voltammetric data. They increase in the following order: k(Cs+)(0) < k(Li+)(0) < k(Rb+)(0) < k(Na+)(0) < k(K+)(0), which is in accordance with their association constants except Cs+ and Li+.

To understand the systematic status of Larimichthys crocea in the Percoidei, we determined the complete mitochondrial (mt) genome sequence using 454 sequencing-by-synthesis technology. The complete mt genome is 16,466 bp in length including the typical structure of 22 tRNAs, 2 rRNAs, 13 protein-coding genes and the noncoding control region (CR). Further sequencing for the complete CR was performed using the primers Cyt b-F and 12S-R on six L crocea individuals and two L polyactis individuals. Interestingly, all seven CR sequences from L crocea were identical while the three sequences from L polyactis were distinct (including one from GenBank). Although the conserved blocks such as TAS and CSB-1, -2, and -3 are readily identifiable in the control regions of the two species, the typical central conserved blocks CSB-D, -E, and -F could not be detected, while they are found in Cynoscion acoupa of Sciaenidae and other Percoidei species. Phylogenetic analysis shows that L crocea is a relatively recently emerged species in Sciaenidae and this family is closely related to family Pomacanthidae within the Percoidei. L crocea, as the first species of Sciaenidae with complete mitochondrial genome available, will provide important information on the molecular evolution of the group. Moreover, the genus-specific pair of primers designed in this study for amplifying the complete mt control region will be very useful in studies on the population genetics and conservation biology of Larimichthys. Mitochondrial genome sequence and structure analysis has become a powerful tool for studying molecular evolution and phylogenetic relationships. To understand the systematic status of Trichiurus japonicus in suborder Scombroidei, we determined the complete mitochondrial genome (mitogenome) sequence using the long-polymerase chain reaction (long-PCR) and shotgun sequencing method. The entire mitogenome is 16,796 by in length and has three unusual features, including (1) the absence of tRNA(Pro) gene, (2) the possibly nonfunctional light-strand replication origin (O-L) showing a shorter loop in secondary structure and no conserved motif (5'-GCCGG-3'), (3) two sets of the tandem repeats at the 5' and 3' ends of the control region. The three features seem common for Trichiurus mitogenomes, as we have confirmed them in other three T. japonicus individuals and in T nanhaiensis. Phylogenetic analysis does not support the monophyly of Trichiuridae, which is against the morphological result. T. japonicus is most closely related to those species of family Scombridae; they in turn have a sister relationship with Perciformes members including suborders Acanthuroidei, Caproidei, Notothenioidei, Zoarcoidei, Trachinoidei, and some species of Labroidei, based on the current dataset of complete mitogenome. T japonicus together with T. brevis, T lepturus and Aphanopus carbo form a clade distinct from Lepidopus caudatus in terms of the complete Cyt b sequences. T. japonicus mitogenome, as the first discovered complete mitogenome of Trichiuridae, should provide important information on both genomics and phylogenetics of Trichiuridae. (C) 2009 Elsevier B.V. Molecular biotechnology of marine algae is referred to as the biotechnology on the identification, modification, production and utilization of marine algal molecules. It involves not only the manipulation of macromolecules such as DNA, RNA and proteins, but also deals with low molecular weight compounds such as secondary metabolites. In the last decade, molecular systematic researches to investigate the relationship and to examine the evolutionary divergence among Chinese marine algae have been carried out by Chinese scientists. For example, RAPD has been widely used in several laboratories to elucidate genetic variations of the reds, such as Porphyra, Gracilaria, Grateloupia and the greens such as Ulva and Enteromorpha. Some important data have been obtained. The study on molecular genetic markers for strain improvement is now in progress. In 1990s, genetic engineering of economic seaweeds such as Laminaria, Undaria, Porphyra, Gracilaria and Grateloupia has been studied in China. For Laminaria japonica, the successfully cultivated kelp in China, a model transformation system has been set up based on the application of plant genetic techniques and knowledge of the algal life history. Progress has been made recently in incorporating a vaccine gene into kelp genome. Evidence has been provided showing the expression of gene products as detectable vaccines. In the present paper, the progress of molecular biotechnological studies of marine algae in China, especially researches on elucidating and manipulating nucleic acids of marine algae, are reviewed.

Cyclic nucleotides (both cAMP and cGMP) play extremely important roles in cyanobacteria, such as regulating heterocyst formation, respiration, or gliding. Catalyzing the formation of cAMP and cGMP from ATP and GTP is a group of functionally important enzymes named adenylate cyclases and guanylate cyclases, respectively. To understand their evolutionary patterns, in this study, we presented a systematic analysis of all the cyclases in cyanobacterial genomes. We found that different cyanobacteria had various numbers of cyclases in view of their remarkable diversities in genome size and physiology. Most of these cyclases exhibited distinct domain architectures, which implies the versatile functions of cyanobacterial cyclases. Mapping the whole set of cyclase domain architectures from diverse prokaryotic organisms to their phylogenetic tree and detailed phylogenetic analysis of cyclase catalytic domains revealed that lineage-specific domain recruitment appeared to be the most prevailing pattern contributing to the great variability of cyanobacterial cyclase domain architectures. However, other scenarios, such as gene duplication, also occurred during the evolution of cyanobacterial cyclases. Sequence divergence seemed to contribute to the origin of putative guanylate cyclases which were found only in cyanobacteria. In conclusion, the comprehensive survey of cyclases in cyanobacteria provides novel insight into their potential evolutionary mechanisms and further functional implications.

Given the commercial and ecological importance of the Asian paddle crab, Charybdis japonica, there is a clearly need for genetic and molecular research on this species. Here, we present the complete mitochondrial genome sequence of C. japonica, determined by the long-polymerase chain reaction and primer walking sequencing method. The entire genome is 15,738 bp in length, encoding a standard set of 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNA genes, plus the putative control region, which is typical for metazoans. The total A+T content of the genome is 69.2%, lower than the other brachyuran crabs except for Callinectes sapidus. The gene order is identical to the published marine brachyurans and differs from the ancestral pancrustacean order by only the position of the tRNA (His) gene. Phylogenetic analyses using the concatenated nucleotide and amino acid sequences of 13 protein-coding genes strongly support the monophyly of Dendrobranchiata and Pleocyemata, which is consistent with the previous taxonomic classification. However, the systematic status of Charybdis within subfamily Thalamitinae of family Portunidae is not supported. C. japonica, as the first species of Charybdis with complete mitochondrial genome available, will provide important information on both genomics and molecular ecology of the group.

The region of Qingdao, China, experienced the world's largest green tide from May to July 2008. More than one million tons of fresh algal biomass of the green alga Ulva prolifera was harvested, while more was suspected to have sunk to the bottom. The original source of this seaweed was suspected to be from the south as revealed by satellite images. The floating biomass drifted with the water current northward and flourished in nearshore waters around Qingdao. However, direct biological evidence for "seed" source is lacking. It is still unclear whether this alga could survive the Qingdao local coastal environment and pose future danger of potential blooming. Systematic and seasonal sampling of waters in the intertidal zone at six collection sites along the Qingdao coast was conducted from December 2008 to April 2009. Forty-eight water samples were analyzed. From these, nine different morphotypes of Ulva were grown in the laboratory under standard temperature and light regimes. Growth of Ulva was observed in all water samples. However, molecular phylogenetic analyses revealed that the dominant U. prolifera strain of the 2008 bloom was absent in all the water-derived cultures during the sampling period. These results provide evidence that the dominant bloom-forming alga was unlikely able to survive the coastal waters (the minimal surface water temperature in February is 2A degrees C) in winter conditions in Qingdao, even though all the sampling locations were heavily covered by this alga in June 2008.

This paper reports the mega-, micro-sporogenesis and female-, male-gametogenesis of Swertia cincta for the first time, with the aim of discussing the systematic position of section Platynema and section Ophelia of Swertia. Anthers are tetrasporangiate. The development of anther walls conforms to the dicotyledonous type. The tapetum cells have dual origin and are similar to the glandular type. There are two middle layers. The endothecium and epidermis persist. Cytokinesis in the microsporrocyte meiosis is simultaneous type and the microscpore teads are tetrahedral. Pollen grains are 3-celled. The ovary is bicarpellum and unilocular. The placentation is of suparietal placentation with 12 series of ovules. The ovules. The ovule is unitegmic, tenuinucellar and ana-campylotropous, The embryo sac orignates from the single-archesporial cell. The one chalazal megaspore in lienar tetrad become the functional megasore. The development of embryo sac is of the polygonum type. Before fertilization, two polar nuclei fuse into one secondary nucleus. Three antipodal cells persisted and divided into 5-8 cells. A comparison between two sections indicates that section Plathnema is better treated as distinct section and is more advanced than section Ophelia according to the evolutionary trends of embryological characters.

There have been no detailed studies on reproductive biology of the Great Cormorant (Phalacrocorax carbo sinensis) in Qinghai-Tibet Plateau. We conducted such investigations during the breeding seasons of 1999 and 2000 in Qinghai-Lake Bird Isle, China. Great Cormorants began to migrate to Qinghai-Lake for reproduction from the middle of March and left from early October at the end of reproduction. Nesting periods were from early April to mid June and took 50 days. Egg-laying occurred during the three weeks from the end of April to 20 May. Females typically laid an egg every 1-2 days until clutch completion. Mean clutch size in the study area over two years was 3.3 (SE +/- 0.13, N = 68, range 1-5) and most (66.18%) fell within the range 3-4 eggs. Length of eggs averaged 61.01 mm and breadth averaged 34.13 mm. Fresh egg weight averaged 57.34 g (SE +/- 0.36, range 46.0-73.7 g, N = 179). Hatching success was 48.7% and fledging success was 64.9% over two years. Decline of available fish resources in Qinghai-Lake might be one of main causes of lower reproductive success. The causes of chick loss were possibly high altitude, high winds and prolonged rain.

A new species of Saussurea, S. erecta S. W Liu, J. T Pan A J. Q. Liu sp. nov., is described from Tibet. It resembles S. kingii but may be distinguished by having distinct stems and glabrous achenes. Saussurea kingii was placed in sect. Pseudoeriocoryne of subgen. Eriocoryne; this section was circumscribed by acaulescence and an inflorescence with congested capitula surrounded by a rosette of leaves. The discovery of S. erecta with distinct stems, cauline leaves and corymbose capitula blurred the delimitation of sect. Pseudoeriocoryne and suggested that the section may be polyphyletic. Both the close relationship and the significant difference between S. erecta and S. kingii were confirmed by analyses of nrDNA ITS sequences. The resulting phylogenies based on ITS data further suggest that Saussurea sect. Pseudoeriocoryne, as traditionally defined, does not constitute a monophyletic group. The rapid radiation and speciation of Saussurea in the Qinghai-Tibetan Plateau, as inferred from ITS phylogeny, are discussed. (c) 2005 The Linnean Society of London.

Pollen morphology of 40 species of Rheum, belonging to eight sections, was investigated under LM and SEM. Four new exine patterns were found in the species: a) microcchinate-foveolate, b) rugulate, c) verrucate-perforate, and d) verrucaterugulate ornamentation. In addition, two patterns, the Rheum-type pollens with microechinate-perforate and fine-reticulate, as previously described, were also confirmed in the present study. Based on above study the evolution trends of pollen morphology in the taxa involved were discussed phylogenetically as below. As microechinate-perforate exine pattern existed commonly, the pattern is, therefore, regarded as the most primitive among all the six types. The fine-reticulate type was thought as a derivative type, deriving from the basic micro echinate-foveolate-perforate pattern, and followed by the rugulate and verrucate-perforate ornamentation. The verrucate-rugulate ornamentation should be the most advanced. More than one pollen type often exist in most of the sections in Rheum. The pollen morphology of Rheum was strongly correlated with its geographical and ecological distribution. Three medicinally important species R. officinale, R. palmation and R. tanguticum can be palynologically distinguished by their ornamentations.
Karyomorphology of Biebersteinia Stephan (Geraniaceae) and its systematic and taxonomic significance

The systematic and taxonomic position of Biebersteinia Stephan has long been in dispute. The present paper describes for the first time the karyomorphology of two species in Biebersteinia Stephan. Both species commonly showed the interphase nuclei of the simple chromocenter type and the mitotic prophase chromosomes of the interstitial type. The karyotype formulae of both B. heterostemon and B. odora were 2n=10=2m(2sec)+8sm(2sec), belonging to the 3A type of Stebbins' classification. The karyotype of this genus is recorded for the first time. The basic chromosome numbers of four of the five known species of Biebersteinia have been recorded as x=5. The combination of resting nuclei of the simple chromocenter type, mitotic prophase chromosomes of the interstitial type, two pairs of chromosomes with four obvious secondary constrictions at the mitotic prophase and metaphase stages, and the peculiar 3A karyotype in Biebersteinia can be regarded as the karymorphological marker of this genus. The karyomorphological data presented here do not support the traditional grouping of this genus in Geraniaceae. The unique karyomorphology of Biebersteinia justifies its familiar or ordinal status, which is congruent with embryological, anatomical, chemical and molecular data. The systematic position of Biebersteinia needs further study.

The embryological characters of Crawfurdia delavayi Frabnch. are described and the systematic relationships of Crawfurdia discussed. Anthers are tetrasporangiate. The development of anther walls conforms to the Dicotyledonous type. The tapetum is of single origin. The development of the tapetum with uninucleate cells is of the glandular type. The tapetal cells on the connective side show radial elongation or periclinal division and intrude into the anther locule. The epidermis of anther walls persists and its cells become pillar and fibrous, and the endothecium degenerates. The ovary is bicarpellary and unilocular. The placentation is typically parietal with 8 rows of anatropous ovules. The development of embryo sac is of the polygonum type. Before fertilization, two polar nuclei fuse into a secondary nucleus. Three antipodal cells persist. Flowers are protandrous. Fertilization is porogamous. The development of the endosperm is of the nuclear type. The embryogeny corresponds to the solanad type physalis II variation. The embryological data indicate that it is better to separate Crawfurdia from Gentiana as an independent genus.

Karyomorphology of Sinadoxa corydalifolia and the karyotype of the diploid Adoxa moschatellina are investigated for the first time. Karyomorphological data indicate that Sinadoxa is directly akin to other two genera of the Adoxaceae, rather than related to Araliales; and three genera of the Adoxaceae should be a natural group. The karyomorphological combinations of the Adoxaceae delimit it from all taxa assumed to be related to it. The karyotype characteristics of all taxa of the Adoxaceae are compared and summarized. The mechanisms of speciation in the Adoxaceae are discussed.
