Objective: The purpose of this study was to examine the leukotoxin promoter types of Aggregatibacter actinomycetemcomitans clones in subjects with generalized aggressive periodontitis (GAgP) and in their family members (FM). Material and Methods: Thirty-five patients with GAgP (33.9+/-7.1 years), 33 of their FM (22.8+/-11.4 years), and 41 patients with chronic periodontitis (CP) (44.1+/-9.4 years) were clinically analyzed using the plaque index, gingival index, probing depth (PD), and clinical attachment level (CAL). Subgingival biofilm samples were collected from four interproximal periodontal sites (>PD and >CAL) of each patient. The presence of A. actinomycetemcomitans and its leukotoxic clone was confirmed by polymerase chain reaction (PCR). Results: A. actinomycetemcomitans was observed in 23 (51.1%) GAgP patients and 16 (30.1%) CP patients. Thirty-seven (94.8%) patients showed minimally leukotoxic strains and 2 (5.1%) showed highly leukotoxic strains. In the FM group, 10 (30.3%) had aggressive periodontitis (AgP), 12 (36.3%) had CP, 11 (33.3%) were periodontally healthy or had gingivitis, and 12.2% were A. actinomycetemcomitans positive. Greater full mouth PD and CAL were observed in GAgP patients positive for the bacteria than those negative for it (p<0.05), and the presence of A. actinomycetemcomitans positively correlated with GAgP (Odds ratio, 3.1; confidence interval, 1.4-7.0; p=0.009). Conclusions: The presence of A. actinomycetemcomitans was associated with the clinical condition of GAgP, with most patients exhibiting a generalized form of the disease and minimally leukotoxic clones. Most of the relatives of GAgP patients presented either CP or AgP.

Members of the genus Malassezia are lipophilic basidiomycetous yeasts, which are part of the normal cutaneous microbiota of humans and other warm-blooded animals. Currently, this genus consists of 14 species that have been characterized by phenetic and molecular methods. Although several molecular methods have been used to identify and/or differentiate Malassezia species, the sequencing of the rRNA genes and the chitin synthase-2 gene (CHS2) are the most widely employed. There is little information about the beta-tubulin gene in the genus Malassezia, a gene has been used for the analysis of complex species groups. The aim of the present study was to sequence a fragment of the beta-tubulin gene of Malassezia species and analyze their phylogenetic relationship using a multilocus sequence approach based on two rRNA genes (ITS including 5.8S rRNA and D1/D2 region of 26S rRNA) together with two protein encoding genes (CHS2 and beta-tubulin). The phylogenetic study of the partial beta-tubulin gene sequences indicated that this molecular marker can be used to assess diversity and identify new species. The multilocus sequence analysis of the four loci provides robust support to delineate species at the terminal nodes and could help to estimate divergence times for the origin and diversification of Malassezia species.

Inflammatory bowel disease (IBD) is a chronic and relapsing disease caused by exaggerated response of the immune system. It represents a significant health problem by limiting the quality of life and being the main risk factor for colorectal cancer. Despite of its importance, the high worldwide incidence and being the object of research for several decades, the etiology remains unknown. Studies indicates an interaction between genetic and environmental factors which together with the intestinal microbiota, leads to an uncontrolled immune response. One of the aggravating environmental factors often discussed is stress, as the daily life of the population in general is increasingly rushed. In order to demonstrate the influence of stress on IBD, this study aimed to standardize an experimental model of colitis induced by instillation of a trinitrobenzene sulfonic acid (TNBS) noninflammatory concentration plus exposure to stress that intensify the inflammation. Therefore, an experiment was done to determine what would be the noninflammatory concentration. In this step, four different concentrations of TNBS (1, 6, 12.5 or 40mg/ml) were tested and the lowest concentration capable of inducing a noninflammatory response in the gut was defined as 1 mg/ml. Then, a second experiment was performed which induced colitis and exposed the animals to restraint stress. The results, however, showed that this stimulus was not enough to exacerbate the damage caused by the 1 mg/ml concentration of TNBS in the colon. With some changes in the protocol, the third experiment associated cold and restraint, as well as changes on the day of euthanasia, which occurred immediately after the stress session. The results of myeloperoxidase activity measurement were unexpected due to the noninflammatory concentration of TNBS caused an intestinal inflammation similar to the concentration of 40 mg/ml. Petroleum and its subproducts are considered a treat for the environmental quality because of the many environmental accidents that may occur during exploitation, transport and storage. A common remediation technique used in the contaminated areas is based on the use of surfactants, mainly the chemical ones, because they have low production costs. In the other hand, some microorganisms have indicate capacities of producing surfactants that emulsify substances and as result, offer a bigger contact surface for the microbiota degradation. This biossurfactants stand out in comparison with the chemical surfactants because they present lower micelar concentration values, are more tolerant for temperature and pH variation, because they are biodegradable, have low toxicity, higher emulsification and hydrocarbon solubilization index. In this way, after the surfactant application, a toxicity evaluation have to be made to identify the treatment effects. In soil, the activity of some microbial enzymes can show the environmental behavior of the contaminant under different treatment conditions. Dehydrogenase is one example of those enzymes that can demonstrate indirectly the effect of the pollutant on the soil microorganisms. The aim of this paper was to evaluate the toxicity after the addition of a surfactant and/or Pseudomonas aeruginosa LBI in soil contaminated by a mineral automotive lubricant. The previous mentioned bacteria are a potential biossurfactant (rhamnolipid) producer. In order to evaluate the toxicity, the dehydrogenase test was run. In this test, trifeniltetrazolium compound (TTC) after utilized as an electron acceptor, turns into trifenil formazan (TPF), that can be indirectly quantified using the absorbance measured by the spectrophotometer UV-visible.
