Acute promyelocytic leukemia is characterized by gene rearrangements that always involve the retinoic acid receptor alpha on chromosome 15. In the majority of patients t(15;17) is detected, which generates the promyelocytic leukemia gene/retinoic acid receptor alpha rearrangement. This rearrangement interacts with several proteins, including the native promyelocytic leukemia gene, thus causing its delocalization from the nuclear bodies, impairing its function. The immunofluorescence staining technique using the anti-PML antibody may be used to provide a rapid diagnosis and to immediately start therapy using all-trans retinoic acid. The experience of the International Consortium on Acute Promyelocytic Leukemia has demonstrated that early mortality was significantly reduced by adopting the immunofluorescence technique. All-trans retinoic acid combined with chemotherapy is the standard therapy; this promotes complete remission rates greater than 90% and cure rates of nearly 80%. However, early mortality is still an important limitation and hematologists must be aware of the importance of treating newly diagnosed acute promyelocytic leukemia as a medical emergency.

UVA light (320–400 nm) represents approximately 95% of the total solar UV radiation that reaches the Earth’s surface. UVA light induces oxidative stress and the formation of DNA photoproducts in skin cells. These photoproducts such as pyrimidine dimers (cyclobutane pyrimidine dimers, CPDs, and pyrimidine (6-4) pyrimidone photoproducts, 6-4PPs) are removed by nucleotide excision repair (NER). In this repair pathway, the XPA protein is recruited to the damage removal site; therefore, cells deficient in this protein are unable to repair the photoproducts. The aim of this study was to investigate the involvement of oxidative stress and the formation of DNA photoproducts in UVA-induced cell death. In fact, similar levels of oxidative stress and oxidised bases were detected in XP-A and NER-proficient cells exposed to UVA light. Interestingly, CPDs were detected in both cell lines; however, 6-4PPs were detected only in DNA repairdeficient cells. XP-A cells were also observed to be significantly more sensitive to UVA light compared to NER-proficient cells, with an increased induction of apoptosis, while necrosis was similarly observed in both cell lines. The induction of apoptosis and necrosis in XP-A cells using adenovirus-mediated transduction of specific photolyases was investigated and we confirm that both types of photoproducts are the primary lesions responsible for inducing cell death in XP-A cells and may trigger the skin-damaging effects of UVA light, particularly skin ageing and carcinogenesis.

The relation between the intercepted light and orchard productivity was considered linear, although this dependence seems to be more subordinate to planting system rather than light intensity. At whole plant level not always the increase of irradiance determines productivity improvement. One of the reasons can be the plant intrinsic un-efficiency in using energy. Generally in full light only the 5 – 10% of the total incoming energy is allocated to net photosynthesis. Therefore preserving or improving this efficiency becomes pivotal for scientist and fruit growers. Even tough a conspicuous energy amount is reflected or transmitted, plants can not avoid to absorb photons in excess. The chlorophyll over-excitation promotes the reactive species production increasing the photoinhibition risks. The dangerous consequences of photoinhibition forced plants to evolve a complex and multilevel machine able to dissipate the energy excess quenching heat (Non Photochemical Quenching), moving electrons (water-water cycle , cyclic transport around PSI, glutathione-ascorbate cycle and photorespiration) and scavenging the generated reactive species. The price plants must pay for this equipment is the use of CO2 and reducing power with a consequent decrease of the photosynthetic efficiency, both because some photons are not used for carboxylation and an effective CO2 and reducing power loss occurs. Net photosynthesis increases with light until the saturation point, additional PPFD doesn’t improve carboxylation but it rises the efficiency of the alternative pathways in energy dissipation but also ROS production and photoinhibition risks. The wide photo-protective apparatus, although is not able to cope with the excessive incoming energy, therefore photodamage occurs. Each event increasing the photon pressure and/or decreasing the efficiency of the described photo-protective mechanisms (i.e. thermal stress, water and nutritional deficiency) can emphasize the photoinhibition. Likely in nature a small amount of not damaged photosystems is found because of the effective, efficient and energy consuming recovery system. Since the damaged PSII is quickly repaired with energy expense, it would be interesting to investigate how much PSII recovery costs to plant productivity. This PhD. dissertation purposes to improve the knowledge about the several strategies accomplished for managing the incoming energy and the light excess implication on photo-damage in peach. The thesis is organized in three scientific units. In the first section a new rapid, non-intrusive, whole tissue and universal technique for functional PSII determination was implemented and validated on different kinds of plants as C3 and C4 species, woody and herbaceous plants, wild type and Chlorophyll b-less mutant and monocot and dicot plants. In the second unit, using a “singular” experimental orchard named “Asymmetric orchard”, the relation between light environment and photosynthetic performance, water use and photoinhibition was investigated in peach at whole plant level, furthermore the effect of photon pressure variation on energy management was considered on single leaf. In the third section the quenching analysis method suggested by Kornyeyev and Hendrickson (2007) was validate on peach. Afterwards it was applied in the field where the influence of moderate light and water reduction on peach photosynthetic performances, water requirements, energy management and photoinhibition was studied. Using solar energy as fuel for life plant is intrinsically suicidal since the high constant photodamage risk. This dissertation would try to highlight the complex relation existing between plant, in particular peach, and light analysing the principal strategies plants developed to manage the incoming light for deriving the maximal benefits as possible minimizing the risks. In the first instance the new method proposed for functional PSII determination based on P700 redox kinetics seems to be a valid, non intrusive, universal and field-applicable technique, even because it is able to measure in deep the whole leaf tissue rather than the first leaf layers as fluorescence. Fluorescence Fv/Fm parameter gives a good estimate of functional PSII but only when data obtained by ad-axial and ab-axial leaf surface are averaged. In addition to this method the energy quenching analysis proposed by Kornyeyev and Hendrickson (2007), combined with the photosynthesis model proposed by von Caemmerer (2000) is a forceful tool to analyse and study, even in the field, the relation between plant and environmental factors such as water, temperature but first of all light. “Asymmetric” training system is a good way to study light energy, photosynthetic performance and water use relations in the field. At whole plant level net carboxylation increases with PPFD reaching a saturating point. Light excess rather than improve photosynthesis may emphasize water and thermal stress leading to stomatal limitation. Furthermore too much light does not promote net carboxylation improvement but PSII damage, in fact in the most light exposed plants about 50-60% of the total PSII is inactivated. At single leaf level, net carboxylation increases till saturation point (1000 – 1200 μmolm-2s-1) and light excess is dissipated by non photochemical quenching and non net carboxylative transports. The latter follows a quite similar pattern of Pn/PPFD curve reaching the saturation point at almost the same photon flux density. At middle-low irradiance NPQ seems to be lumen pH limited because the incoming photon pressure is not enough to generate the optimum lumen pH for violaxanthin de-epoxidase (VDE) full activation. Peach leaves try to cope with the light excess increasing the non net carboxylative transports. While PPFD rises the xanthophyll cycle is more and more activated and the rate of non net carboxylative transports is reduced. Some of these alternative transports, such as the water-water cycle, the cyclic transport around the PSI and the glutathione-ascorbate cycle are able to generate additional H+ in lumen in order to support the VDE activation when light can be limiting. Moreover the alternative transports seems to be involved as an important dissipative way when high temperature and sub-optimal conductance emphasize the photoinhibition risks. In peach, a moderate water and light reduction does not determine net carboxylation decrease but, diminishing the incoming light and the environmental evapo-transpiration request, stomatal conductance decreases, improving water use efficiency. Therefore lowering light intensity till not limiting levels, water could be saved not compromising net photosynthesis. The quenching analysis is able to partition absorbed energy in the several utilization, photoprotection and photo-oxidation pathways. When recovery is permitted only few PSII remained un-repaired, although more net PSII damage is recorded in plants placed in full light. Even in this experiment, in over saturating light the main dissipation pathway is the non photochemical quenching; at middle-low irradiance it seems to be pH limited and other transports, such as photorespiration and alternative transports, are used to support photoprotection and to contribute for creating the optimal trans-thylakoidal ΔpH for violaxanthin de-epoxidase. These alternative pathways become the main quenching mechanisms at very low light environment. Another aspect pointed out by this study is the role of NPQ as dissipative pathway when conductance becomes severely limiting. The evidence that in nature a small amount of damaged PSII is seen indicates the presence of an effective and efficient recovery mechanism that masks the real photodamage occurring during the day. At single leaf level, when repair is not allowed leaves in full light are two fold more photoinhibited than the shaded ones. Therefore light in excess of the photosynthetic optima does not promote net carboxylation but increases water loss and PSII damage. The more is photoinhibition the more must be the photosystems to be repaired and consequently the energy and dry matter to allocate in this essential activity. Since above the saturation point net photosynthesis is constant while photoinhibition increases it would be interesting to investigate how photodamage costs in terms of tree productivity. An other aspect of pivotal importance to be further widened is the combined influence of light and other environmental parameters, like water status, temperature and nutrition on peach light, water and phtosyntate management.

The structural peculiarities of a protein are related to its biological function. In the fatty acid elongation cycle, one small carrier protein shuttles and delivers the acyl intermediates from one enzyme to the other. The carrier has to recognize several enzymatic counterparts, specifically interact with each of them, and finally transiently deliver the carried substrate to the active site. Carry out such a complex game requires the players to be flexible and efficiently adapt their structure to the interacting protein or substrate. In a drug discovery effort, the structure-function relationships of a target system should be taken into account to optimistically interfere with its biological function. In this doctoral work, the essential role of structural plasticity in key steps of fatty acid biosynthesis in Plasmodium falciparum is investigated by means of molecular simulations. The key steps considered include the delivery of acyl substrates and the structural rearrangements of catalytic pockets upon ligand binding. The ground-level bases for carrier/enzyme recognition and interaction are also put forward. The structural features of the target have driven the selection of proper drug discovery tools, which captured the dynamics of biological processes and could allow the rational design of novel inhibitors. The model may be perspectively used for the identification of novel pathway-based antimalarial compounds.

The hydrogen production in the green microalga Chlamydomonas reinhardtii was evaluated by means of a detailed physiological and biotechnological study. First, a wide screening of the hydrogen productivity was done on 22 strains of C. reinhardtii, most of which mutated at the level of the D1 protein. The screening revealed for the first time that mutations upon the D1 protein may result on an increased hydrogen production. Indeed, productions ranged between 0 and more than 500 mL hydrogen per liter of culture (Torzillo, Scoma et al., 2007a), the highest producer (L159I-N230Y) being up to 5 times more performant than the strain cc124 widely adopted in literature (Torzillo, Scoma, et al., 2007b). Improved productivities by D1 protein mutants were generally a result of high photosynthetic capabilities counteracted by high respiration rates. Optimization of culture conditions were addressed according to the results of the physiological study of selected strains. In a first step, the photobioreactor (PBR) was provided with a multiple-impeller stirring system designed, developed and tested by us, using the strain cc124. It was found that the impeller system was effectively able to induce regular and turbulent mixing, which led to improved photosynthetic yields by means of light/dark cycles. Moreover, improved mixing regime sustained higher respiration rates, compared to what obtained with the commonly used stir bar mixing system. As far as the results of the initial screening phase are considered, both these factors are relevant to the hydrogen production. Indeed, very high energy conversion efficiencies (light to hydrogen) were obtained with the impeller device, prooving that our PBR was a good tool to both improve and study photosynthetic processes (Giannelli, Scoma et al., 2009). In the second part of the optimization, an accurate analysis of all the positive features of the high performance strain L159I-N230Y pointed out, respect to the WT, it has: (1) a larger chlorophyll optical cross-section; (2) a higher electron transfer rate by PSII; (3) a higher respiration rate; (4) a higher efficiency of utilization of the hydrogenase; (5) a higher starch synthesis capability; (6) a higher per cell D1 protein amount; (7) a higher zeaxanthin synthesis capability (Torzillo, Scoma et al., 2009). These information were gathered with those obtained with the impeller mixing device to find out the best culture conditions to optimize productivity with strain L159I-N230Y. The main aim was to sustain as long as possible the direct PSII contribution, which leads to hydrogen production without net CO2 release. Finally, an outstanding maximum rate of 11.1 ± 1.0 mL/L/h was reached and maintained for 21.8 ± 7.7 hours, when the effective photochemical efficiency of PSII (ΔF/F'm) underwent a last drop to zero. If expressed in terms of chl (24.0 ± 2.2 µmoles/mg chl/h), these rates of production are 4 times higher than what reported in literature to date (Scoma et al., 2010a submitted). DCMU addition experiments confirmed the key role played by PSII in sustaining such rates. On the other hand, experiments carried out in similar conditions with the control strain cc124 showed an improved final productivity, but no constant PSII direct contribution. These results showed that, aside from fermentation processes, if proper conditions are supplied to selected strains, hydrogen production can be substantially enhanced by means of biophotolysis. A last study on the physiology of the process was carried out with the mutant IL. Although able to express and very efficiently utilize the hydrogenase enzyme, this strain was unable to produce hydrogen when sulfur deprived. However, in a specific set of experiments this goal was finally reached, pointing out that other than (1) a state 1-2 transition of the photosynthetic apparatus, (2) starch storage and (3) anaerobiosis establishment, a timely transition to the hydrogen production is also needed in sulfur deprivation to induce the process before energy reserves are driven towards other processes necessary for the survival of the cell. This information turned out to be crucial when moving outdoor for the hydrogen production in a tubular horizontal 50-liter PBR under sunlight radiation. First attempts with laboratory grown cultures showed that no hydrogen production under sulfur starvation can be induced if a previous adaptation of the culture is not pursued outdoor. Indeed, in these conditions the hydrogen production under direct sunlight radiation with C. reinhardtii was finally achieved for the first time in literature (Scoma et al., 2010b submitted). Experiments were also made to optimize productivity in outdoor conditions, with respect to the light dilution within the culture layers. Finally, a brief study of the anaerobic metabolism of C. reinhardtii during hydrogen oxidation has been carried out. This study represents a good integration to the understanding of the complex interplay of pathways that operate concomitantly in this microalga.

Die Arbeit beschreibt Untersuchungen zum nichtphoto- chemischen Lochbrennen, das bei 1.4 Kelvin in Form von rein lichtinduzierten Frequenzsprüngen einzelner in p-Terphenyleingebetteter Terrylenmoleküle beobachtet werden kann. Dabei zeigen alle Chromophore aus der X1-Einbaulage ein exzellent reproduzierbares Verhalten, sowohl im bistabilen primären Photozyklus wie auch in dem daran angegliederten sekundärenPhotozyklus, welcher aus drei weiteren spektralen Positionen besteht. Aus den Ergebnissen der nach der genauen Charakterisierung dieser Eigenschaft des Systems durchgeführten Experimente - Fluoreszenzspektroskopie der Photoprodukte, Stark-Effekt-Messungen und Polarisationsmodulation - wird ein Modell für die den lichtinduzierten Änderungen der Absorptionsfrequenzzugrundeliegenden Konformationsänderungender Wirt/Gast- Struktur abgeleitet und diskutiert. Die mittlerweile verfügbaren Ergebnisse von diesbezüglichen molekular- dynamischen Simulationen einer Theoriegruppe ausBordeaux, die alle grundlegenden Annahmen dieses Modellsbestätigen und eine noch genauere mikroskopische Beschreibung des Systems liefern, werden zur Abrundung der Darstellung ebenfalls vorgestellt. Außerdem geht die Dissertation auf die durchgeführten Einzelmolekül- untersuchungen an Terrylen in p-Terphenyl bei Raumtemperatur ein und stellt das im Rahmen der Arbeit aufgebaute temperaturvariable laserscannende Konfokalmikroskop im Detail vor.

Die supramolekularen Organisationen von 5-n-Alkoxyisophthaläuren und 2,5-Di-n-alkoxyterephthalsäuren bilden durch sekundäre Wechselwirkungen in Form van der Waals-Kräften und Wasserstoffbrückenbindungen hochgeordnete lamellare Strukturen, die sowohl mittels Röntgen-kristallgraphie als auch durch Rastertunnelmikroskopie (STM) visualisierbar sind. Diese Art der Aggregationsmuster ermöglicht den potentiell reaktiven Gruppen (Diacetylene, Zimtsäuren) in der Alkoxykette, einen optimalen Abstand und geometrische Orientierungen im Kristall einzunehmen, um lichtinduzierte Reaktionen im Festkörper durchzuführen. Einführung einer Amidgruppe an die Alkoxykette erweitert dieses Konzept, durch Erhalt einer zusätzlichen Wasserstoffbrückenbindung im hydrophoben Bereich der Organisation.Die Stabilität solche supramolekularer Aggregate kann durch Einführung semifluorierter Alkoxyketten stark beeinflußt werden, da die Inkompatibilität der pefluorierten Gruppen durch viele organische Verbindungen das Gleichgewicht nicht-kovalenter Wechselwirkung drastisch verändert. Diese Eigenschaft der semifluorierten 5-n-Alkoxyisopthalsäuren und 2,5-Di-n-alkoxyterephthalsäuren zeigt sich erstaunlicherweise jedoch nur bei einem großen Anteil des perfluorierten Alkylteils als der des nicht-perfluorierten. Da sich dann die perfluorierten Alkylteile untereinander organisieren können, erscheint die Änderung der Assoziationsstruktur von interdigitiert-lamellar zu nicht-interdigitiert-lamellar plausibel.Das gezielte Design eines neuen Organisationsmusters der 5-n-Alkoxyisophthalsäuren gelang durch Brechung der Symmetrie dieses Bausteins. Die Einführung zweier Alkoxyketten ließ die zweidimensionale Anordnung der wasserstoffbrückengebundenen Aromate unverändert, resultierte aber mit Ausbruch einer Alkoxykette aus dieser Ebene.

Die Analyse der CML-Zellinie K562 mittels Fluoreszenz in situ Hybridisierung (FISH), Multiplex-FISH (M-FISH) und comparativer genomischer Hybridisierung (CGH) ergab einen hypotriploiden Karyotyp mit 67 Chromosomen und 21 verschiedenen Marker-Chromosomen. Das bei über 90% der CML-Patienten nachgewiesene Ph-Chromosom entsteht durch die reziproke Translokation t(9;22)(q34;q11). Bei 5 - 10% der Patienten resultiert das Ph-Chromosom aus varianten Translokation. Anhand der Untersuchung dreier varianter Translokation mittels Bruchpunkt-übergreifender FISH-Proben für die BCR- und ABL-Gene werden drei verschiedene Mechanismen der Entstehung komplexer Translokationen dargestellt. Das Auftreten sekundärer Aberrationen wurde in 15 CML-Blastenkrisen untersucht. Zudem wurde anhand der CGH-Analyse von CD34-positiven Zellen, Monozyten, Granulozyten und T-Zellen die Zellinienspezifität sekundärer Aberrationen untersucht. In einem Fall wurde eine sekundäre Aberration in allen vier Fraktionen gefunden. In zwei Fällen traten sekundäre Aberrationen in allen untersuchten Fraktionen mit Ausnahme der T-Zellen auf. Aufgrund dieser Ergebnisse lassen sich zwei alternative Modelle der Tumor-Progression der CML ableiten: 1. Sekundäre Mutatonen treten vor der Differenzierung der hämatopoetischen Stammzelle auf. 2. Sekundäre Mutationen treten in einer hämatopoetischen Vorläuferzelle nach der T-Zell-Differenzierung auf.

Zusammenfassung In der vorliegenden Arbeit wurden 74736 bp genomischer DNA-Sequenzder Hämoglobingen-Gruppe D aus der Chironomiden Art Chironomus tentansentschlüsselt und analysiert. Durch Datenbankrecherchen undSequenz-Vergleiche wurden 29 vollständige Hämoglobin-Geneidentifiziert und klassifiziert. Es zeigt sich, daß alle derzeitbekannten Hämoglobin-Gene der Chironomiden auch in Chironomus tentansvorhanden sind. Zusätzlich konnten in Chironomus tentans sechs neueHämoglobin-Varianten identifiziert werden, die bislang weder aufProtein- noch auf Gen-Ebene in anderen Spezies nachgewiesenwurden. Die Hämoglobin-Gene liegen in dichter Abfolge innerhalbdes Clusters, wobei durchschnittlich etwa alle 2 kb ein Gen zufinden ist. Die Abfolge der Hämoglobin-Gene innerhalb derGengruppe wird nur an einer Stelle durch ein interspergiertes Genaus der Familie der Glukosetransporter unterbrochen. Desweiterenkonnten zwei retrotransponierbare Elemente der SINE-Klasse (CP1)innerhalb des Hämoglobingen-Clusters identifiziert werden. AlleGene besitzen die für ihre Expression erforderlichenSignalsequenzen, so daß es sich höchstwahrscheinlich um aktiveGene handelt. Die abgeleiteten Aminosäure-Sequenzen weisen alleCharakteristika sauerstofftransportierender Moleküle auf. Da es sich bei den Hämoglobinen um eine sehr alte Genfamiliehandelt, kann die vergleichende Analyse derHämoglobin-Genstruktur bei Vertebraten, Invertebraten, Pflanzenund Protozoen zur Rekonstruktion der Intron-Evolution genutztwerden. Die Konservierung von Intronpositionen in homologen Genenverschiedener Taxa gilt dabei als Maß für das relativestammesgeschichtliche Alter der Introns. Eine Vielzahl derHämoglobin-Gene von Invertebraten weisen ein Intron im zentralenGenbereich auf. Auch bei einigen Chironomiden-Arten konntendiese 'zentralen Introns' nachgewiesen werden. DieHämoglobin-Gene von Chironomus tentans galten hingegen bislang als intronlos.Die vorliegende Untersuchung zeigt, daß auch zweiHämoglobin-Gene dieser Spezies je ein kurzes Intron aufweisen.Der Vergleich der Intronverteilung in den Hämoglobin-Genen derChironomiden führt zu dem Ergebnis, daß alle vorhandenen Intronsam sparsamsten (im Sinne des 'maximum parsimony'-Prinzips) durchunabhängige Insertionen in ein intronlosesVorläufer-Hämoglobin-Gen erklärt werden können. Alle bislangin Chironomiden beschriebenen Introns sind mit großerWahrscheinlichkeit nicht ortholog (Hankeln et al., 1997; dieseArbeit). Das Vorläufer-Hämoglobin-Gen in Chironomiden besaßdaher vermutlich kein 'zentrales Intron'. Die in Chironomidengefundenen Verhältnisse stellen somit die von Go (1981)formulierte Hypothese der Ursprünglichkeit des 'zentralenIntrons' in Hämoglobin-Genen in Frage. Die in Invertebraten undPflanzen beschriebenen 'zentralen Introns' sind vermutlich nichthomolog und dementsprechend auch nicht auf ein Intron imanzestralen Globin zurückzuführen. Vielmehr implizieren die inhohem Maße variablen Positionen der 'zentralen Introns' beiPflanzen und Invertebraten ihre unabhängige Insertion in diejeweiligen Globin-Gene nach der Aufspaltung der Taxa. Grundsätzlich können zwei Klassen von Hämoglobin-Genen inChironomiden unterschieden werden. Die überwiegende Mehrzahl derHämoglobine wird von Genen kodiert, die nur in einer Kopie imGenom vorliegen. Sie werden dementsprechend als 'single copy'Varianten bezeichnet. Für andere Hämoglobin-Varianten konntehingegen eine Vielzahl leicht unterschiedlicher Gene beschriebenwerden. Diese bilden sogenannte Gen-Subfamilien. In Chironomus tentans konntegezeigt werden, daß neben den 7B-Genen auch die 7A-Gene eineeigene Subfamilie bilden. Die 'single copy' Varianten zeichnensich im Interspezies-Vergleich durch ihre konservierteNukleotid-Sequenz aus: Sie unterliegen während ihrer Evolutionoffenbar einer stabilisierenden Selektion, d.h. Veränderungenihrer Protein-Sequenzen werden nur in geringem Maße toleriert.Auch ihre räumliche Anordnung innerhalb der Gengruppe istzwischenartlich konserviert. Der Vergleich der 'single copy'Varianten innerhalb einer Art zeigt, daß diese sehr deutlicheSequenz-Unterschiede zueinander aufweisen. Sie bilden somit einkonserviertes Sortiment an Hämoglobin-Genen, das weitgehend vorder Radiation der Arten entstanden ist und eine über dieArtgrenzen hinweg unveränderte 'Hämoglobin-Grundausstattung'gewährleistet. Im Gegensatz hierzu zeichnen sich die Mitglieder vonHämoglobin-Gen-Sub-familien durch eine hohe Variabilität aus:Nukleotid-Sequenz, Anzahl und Organisation der Gene innerhalb derGenfamilie weisen im zwischenartlichen Vergleich zahlreicheUnterschiede auf. Es ist daher nur selten möglich allein aufGrundlage der Nukleotid-Sequenzen orthologe Genpaare zuidentifizieren. Die orthologen Gene der 7B-Subfamilie aus Chironomus tentansund chth konnten ausschließlich anhand korrespondierenderIntergen-Sequenzen einander zugeordnet werden. Somit sind dieGen-Subfamilien präferenziell an der Entstehung einesspeziesspezifischen Gen-Repertoires beteiligt. Variationen derNukleotid-Sequenz, Gen-Anzahl und Gen-Organisation innerhalb derSubfamilie werden im Gegensatz zu den 'single copy' Varianten ineinem hohen Maße toleriert. Aufgrund der hohen Sequenz-Übereinstimmungen zwischen denMitgliedern der Gen-Subfamilien unterliegen diese einer Vielzahlvon Rearrangements, die in Gen-Duplikationen, Deletionen undSequenz-Homogenisierungen resultieren. So führten beispielsweiseGenduplikationen durch ungleiches, homologes Crossing-over mitgroßer Wahrscheinlichkeit zur Entstehung und Expansion der7A-Subfamilie. Auch die Gene Cte12-1 und Cte 12-2 sind vermutlichdas Ergebnis eines rezenten Duplika-tions-Ereignisses. DerMechanismus der Retrotransposition, der zu einer Duplika-tioneines 3`-untranslatierten Bereichs innerhalb der 7A-Subfamilieführte, scheint für die Entstehung derHämoglobin-Multiplizität in Chironomiden hingegen wenigerbedeutsam zu sein. Innerhalb der 7A-Subfamilie ist eineAngleichung der Gene durch konzertierte Sequenz-Evolution zubeobachten. Der nukleotidweise Vergleich von Gen-Sequenzen zeigtam Beispiel der Gene 7A7 und 7A8, daß die konzertierte Evolutiondieser Gen-Varianten auf dem Mechanismus der Genkonversionberuht. Auch die Gen-Subfamilie 7B unterliegt offenbar in hohemMaße einer solchen Sequenz-Homogenisierung. Im Sinne einer molekularen Uhr sollten synonyme Basenaustauscheweitgehend neutral sein und sich proportional zur Zeit in denGenen anhäufen. Der Vergleich der Hämoglobin-Gen-Sequenzenzeigt, daß große Unterschiede in der Anzahl der synonymenBasenaustausche zwischen orthologen Genen nicht zwangsläufig dasErgebnis einer frühen Trennung dieser Gene sind. Die Übertragungvon Sequenzen zwischen paralogen Genen kann die Anzahl dersynonymen Basenaustausche orthologer Gene in kürzester Zeitverändern und den tatsächlichen Zeitpunkt der Trennung zweierorthologen Gene überdecken. Werden Genkonversionen nichterkannt, weil beispielsweise nicht alle Gene der Gruppevollständig erfaßt werden konnten, führt der Vergleichorthologer Gen-Sequenzen zwangsläufig zu falschen evolutionärenGendistanzen. Da die Mitglieder der Hämoglobin-Genfamiliebesonders häufig Rekombinations-Prozessen unterliegen, sind siedaher möglicherweise weniger nützliche Kanditaten für dieErmittlung evolutionärer Distanzen zwischen den verschiedenenChironomiden-Arten. Insgesamt zeigen die Ergebnisse dieser Arbeit, daß anhanddetaillierter phylogenetischer Analysen sich die Evolution derHämoglobin-Multigenfamilie von Chironomiden umfassendbeschreiben läßt. Ob einzelne, besonders gut konservierteGen-Varianten (wie z. B. die Gene Cte 8 und Cte W) einespezifische physiologische Funktion erfüllen oder ob dieGen-Subfamilien, die ein speziesspezifisches Genrepertoirebilden, an der Einnischung der verschiedenen Arten beteiligtsind, sollte durch weiterführende Untersuchungen (z. B. derGenexpression sowie der physiologischen Eigenschaften einzelnerVarianten) ermittelt werden können.

Es wurden neue funktionalisierte Carbazole und anellierte Benzo[a]carbazole als potentielle pharmakologische Therapeutika durch 1,6-pi-Elektrocyclisierung auf photo-chemischem, thermischem und sonochemischem Weg synthetisiert und die Synthesemethoden der 1,6-pi-Elektrocyclisierung sowie der 2,3-Divinylindole und der 2-Aryl-3-vinylindole als entsprechende Ausgangsprodukte validiert und evaluiert. Es gelang weder das nach den Woodward-Hoffmann-Regeln erwartete primäre Cyclisierungsprodukt mit Indolochinodimethanstruktur noch die Existenz des in einer photochemischen Abfangreaktion daraus resultierenden Cycloprodukts NMR-spektroskopisch nachzuweisen, um den stereochemischen Verlauf der Cyclisierung vorherzusagen. Ergebnisse der quantenchemischen Berechnungen der Eduktmoleküle (AO-Koeffizienten der MO's, HOMO/LUMO-Energien) sowie der Übergangszustandsgeometrien der Cyclisierungen decken sich mit den experimentellen Daten. Divinyl- und 2-Aryl-3-vinylindole sind als Systeme mit Hexatriensymmetrie aufzufassen, deren Cyclisierungsverhalten sich mit den Woodward-Hoffmann-Regeln beschreiben läßt. Im Vergleich der verschiedenen 1,6-pi-Elektrocyclisierungsmethoden zeigte sich, daß die photochemische Variante eine elegante Synthesemethode darstellt, um funktionalisierte Carbazole und Benzo[a]carbazole mit unterschiedlichen pharmakologischen Aktivitäten unter schonenden Reaktionsbedingungen mit den vergleichbar höchsten Ausbeuten zu erhalten. Demgegenüber lieferten die Ultraschallreaktionen keine nachweisbaren Cyclisierungsprodukte. Die thermische Cyclisierung führte zur Gruppe der 1,2-Dihydrocarbazole. Sie bildeten sich in einer Folgereaktion durch [1,5s]-H-Verschiebung aus dem primär entstandenen Woodward-Hoffmann-Cyclisierungsprodukt. In abschließenden DNA-Bindestudien mit verschiedenen Testsystemen zeigte keine der synthetisierten Testsubstanzen DNA-Bindungsaktivität.

Photochemie von ungesättigten Sternverbindungen.Wirt-Gast-Systeme bei stilbenoiden Dendrimeren. Diese Arbeit beschäftigt sich mit der Synthese und denphotochemischen Eigenschaften stilbenoider und chalkonoiderSternverbindungen.Die Synthese der stilbenoiden Dendrimere gelingt über einekonvergente in einzelnen Generationen gekoppelteSynthesestrategie durch Wittig-Horner-Olefinierung inKombination mit einer Schutzgruppentechnik.Experimente mit den stibenoiden Dendrimeren 1. Generationhaben gezeigt, dass geänderte Belichtungsbedingungen(monochromatisches Licht in Gegenwart einesTriplett-Sensibilisators) die photochemische[2+2]Cycloaddition bzw. CC-Verknüpfung unterdrücken. Dabeiverläuft selektiv eine cis/trans-Isomerisierung, die füroptische Schaltung genutzt werden könnte.Mit Triplett-Sensibilisatoren wie Diacetyl oder Acetonkonnten auch Wirt-Gast-Systeme mit stilbenoiden Dendrimerengebildet werden. Das zeigt die Möglichkeit der Einlagerungvon Gastmolekülen in freie Kavitäten der Dendrimere.Die Synthese der chalkonoiden Sternverbindungen gelingt übereine dreifache Aldolkondensation des 1,3,5-Triacetylbenzolsmit unsubstituierten oder 3,4,5-trialkoxysubstituiertenBenzaldehyden. Mit einer geeigneten Verzweigungseinheitwurden auch Armbauteile zur Generationenerweiterungsynthetisiert.Die photochemischen Eigenschaften der chalkonoidenSternverbindungen in Lösung und im Festzustand wurdenstudiert und die entstandenen Produkte charakterisiert. BeiBelichtung im Festzustand läuft bei topochemischer Kontrolleeine Photodimerisierung unter Bildung eines Cyclobutanringsdurch Cycloaddition olefinischer Doppelbindungen ab, wobeiein Kopf-Schwanz-anti-Dimer erhalten wird. BeiPhotodimerisierung in Lösung entsteht einKopf-Kopf-anti-Dimer als Hauptprodukt. Das kann zur Syntheseder sonst schwer zugänglichen [4.4.4](1,3,5)Cyclophanegenutzt werden.

Comparative fluorescence in situ hybridization (FISH) mapping revealed four large DNA segments which have been conserved in their entirety between human chromosome 3 and Bornean orangutan chromosome 2 as well as three evolutionary breakpoints which distinguish between the human and Bornean orangutan chromosome forms. Examination of the structural and functional features of evolutionary breakpoints provides new insights into the possible effects of evolutionary rearrangements on genome function and the relationship between human chromosome pathology and evolution. FISH of human BAC clones which were assesssed in human genomic sequence to primate chromosomes, combined with precise breakpoint localizations by polymerase chain reaction (PCR) analysis of flow-sorted chromosomes and in silico analysis, were used to characterize the evolutionary breakpoints. The aim of this Ph.D. project has been the photophysical and photochemical characterization of new photo- and redox-active supramolecular systems. In particular we studied two different classes of compounds: metal complexes and dendrimers. Two different families of bis-cyclometalated neutral Ir(III) complexes are presented and their photophysical properties are discussed. The first family of complexes contains two 2-phenylpyridyl (ppy) or 2-(4,6-difluorophenyl)pyridyl (F2ppy) cyclometalated ligands and an ancillary ligand constituted by a phenol-oxazoline (phox), which can be substituted in the third position with a fluorine group (Fphox). In the second part of this study, we present another family of bis-cyclometalated Ir(III) complexes in which the ancillary ligand could be a chiral or an achiral bis-oxazoline (box). We report on their structural, electrochemical, photophysical, and photochemical properties. Complexes containing phox and Fphox ancillary ligands show blue luminescence with very high quantum yield, while complexes with box ligands do not show particularly interesting photophysical properties. Surprisingly these complexes give an unexpected photoreaction when irradiated with UV light in presence of dioxygen. This photoreaction originates a stable, strong blue emitting and particularly interesting photoproduct. Three successive generations of a family of polyethyleneglycol (PEG)-coated Pd(II) tetrabenzoporphyrin (PdTBP)-based dendritic nanoprobes are presented, and their ability to sensitize singlet oxygen and inflict cellular photodamage are discussed. It was found that the size of the dendrimer has practically no effect on the singlet oxygen sensitization efficiency, that approximate the unity, in spite of the strong attenuation of the triplet quenching rate with an increase in the dendrimer generation. Nevertheless, when compared against a commonly used singlet oxygen sensitizer, as Photofrin, the phosphorescent probes were found to be non-phototoxic. The lack of phototoxicity is presumably due to the inability of PEGylated probes to associate with cell surfaces and/or penetrate cellular membranes. The results suggest that protected phosphorescent probes can be safely used for oxygen measurements in biological systems in vivo. A new family of two photoswitchable (G0(Azo) and G1(Azo)) dendrimers with an azobenzene core, two cyclam units as coordination sites for metal ions, and luminescent naphthalene units at the periphery have been characterized and their coordination abilities have been studied. Because of their proximity, the various functional groups of the dendrimer may interact, so that the properties of the dendrimers are different from those exhibited by the separated functional units. Both the naphthalene fluorescence and the azobenzene photoisomerization can be observed in the dendrimer, but it has been shown that (i) the fluorescent excited state of the naphthalene units is substantially quenched by excimer and exciplex formation and by energy transfer to the azobenzene units, and (ii) in the latter case the fluorescence quenching is accompanied by the photosensitized isomerization of the trans → cis, and, with higher efficiency, the cis → trans reaction. Complexation of these dendrimers, both trans and cis isomers, with Zn(II) ions shows that complexes of 1:1 and 2:1 metal per dendrimer stoichiometry are formed showing different photophysical and photochemical properties compared to the corresponding free ligands. Practically unitary efficiency of the sensitized isomerization of trans → cis and cis → trans reaction is observed, as well as a slight increase in the naphthalene monomer emission. These results are consistent with the coordination of the cyclam amine units with Zn(II), which prevents exciplex formation. No indication of a concomitant coordination of both cyclam to a single metal ion has been obtained both for trans and cis isomer.

Carbonyl compounds are very important volatil organic compounds receiving increasing attention due to their critical role in tropospheric organic chemistry and to their effects on human health. They are first photo-oxidation stage of almost all organic compounds and originators of free radicals (OH, O3...). Therfore, continuous determination of their atmospheric concentrations is essential. Currently, field measurements are very limited mainly because of the analytical challenges posed by trace concentrations and interferences arising from atmospheric co-pollutants. Additionally, hydroxyl radical (OH) plays a central role in atmospheric chemistry processes. The measurement of atmospheric OH concentrations is essential for the comprehension of atmospheric chemical and photochemical processes. In this context, objectives of this work were: To perform a continuous survey of atmospheric concentrations of carbonyl compounds on different sites around the area of Bologna. For this purpose, an automatic instrument for the continuous measurement (one hour of time resolution) was developed and used. Additionally, the development of a complete and original analytical method was realised. To develop an original OH generation system in order to evaluate the effect of OH and the reactivity of solid and/or liquid materials of interest. This included the validation of an analytical method for the quantification of OH concentrations generated. Effect of OH on ceramic materials and reactivity of snow surface samples, involved as sink of OH in atmospheric chemistry, were evaluated.

A numerical model for studying the influences of deep convective cloud systems on photochemistry was developed based on a non-hydrostatic meteorological model and chemistry from a global chemistry transport model. The transport of trace gases, the scavenging of soluble trace gases, and the influences of lightning produced nitrogen oxides (NOx=NO+NO2) on the local ozone-related photochemistry were investigated in a multi-day case study for an oceanic region located in the tropical western Pacific. Model runs considering influences of large scale flows, previously neglected in multi-day cloud resolving and single column model studies of tracer transport, yielded that the influence of the mesoscale subsidence (between clouds) on trace gas transport was considerably overestimated in these studies. The simulated vertical transport and scavenging of highly soluble tracers were found to depend on the initial profiles, reconciling contrasting results from two previous studies. Influences of the modeled uptake of trace gases by hydrometeors in the liquid and the ice phase were studied in some detail for a small number of atmospheric trace gases and novel aspects concerning the role of the retention coefficient (i.e. the fraction of a dissolved trace gas that is retained in the ice phase upon freezing) on the vertical transport of highly soluble gases were illuminated. Including lightning NOx production inside a 500 km 2-D model domain was found to be important for the NOx budget and caused small to moderate changes in the domain averaged ozone concentrations. A number of sensitivity studies yielded that the fraction of lightning associated NOx which was lost through photochemical reactions in the vicinity of the lightning source was considerable, but strongly depended on assumptions about the magnitude and the altitude of the lightning NOx source. In contrast to a suggestion from an earlier study, it was argued that the near zero upper tropospheric ozone mixing ratios which were observed close to the study region were most probably not caused by the formation of NO associated with lightning. Instead, it was argued in agreement with suggestions from other studies that the deep convective transport of ozone-poor air masses from the relatively unpolluted marine boundary layer, which have most likely been advected horizontally over relatively large distances (both before and after encountering deep convection) probably played a role. In particular, it was suggested that the ozone profiles observed during CEPEX (Central Equatorial Pacific Experiment) were strongly influenced by the deep convection and the larger scale flow which are associated with the intra-seasonal oscillation.
