This study evaluated the cytotoxicity of experimental adhesive systems (EASs) on odontoblast-like cells. Paper discs (n=132) were impregnated with 10 μL of each EAS-R1, R2, R3, R4, and R5 (in an ascending order of hydrophilicity), followed by photoactivation. R1 and R2 are nonsolvated hydrophobic blends, R3 represents a simplified etch-and-rinse adhesive system, and R4 and R5 represent simplified self-etch adhesive systems. Discs were immersed in Dulbecco's modified Eagle's medium for 24 h to obtain eluates applied on MDPC-23 cell cultures. No material was applied on discs used as control (R0). Cell viability [3-(4,5-dimethythiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay], total protein (TP) production, alkaline phosphatase (ALP) activity, type of cell death, and degree of monomer conversion Fourier transform infrared (%DC-FTIR) were evaluated. Data were analyzed by Kruskal-Wallis and Mann-Whitney tests (α=0.05). Considering R0 (control) as having 100% of cell viability, R1, R2, R3, R4, and R5 reduced the metabolic activity of cells by 36.4, 3.1, 0.2, 21.5, and 65.7%, respectively, but only R1 and R5 differed from R0. Comparing with R0, lower TP production was observed for R1, R4, and R5, while ALP activity decreased for R1 and R5. Necrotic cell death was predominant for all EASs, but only R1, R4, and R5 differed from R0. Only R5 presented a different apoptotic cell death ratio from R0. R1 presented the lowest %DC (ca. 37%), whereas R4 and R5 presented the highest (ca. 56%). In conclusion, R2 and R3 were not toxic to the MDPC-23 cells, suggesting that the degree of hydrophilicity or %DC of the EASs alone were not responsible for their cytopathic effects. © 2013 Wiley Periodicals, Inc.

Scope. To elucidate the morphological and biochemical in vitro effects exerted by caffeine, taurine, and guarana, alone or in combination, since they are major components in energy drinks (EDs). Methods and Results. On human neuronal SH-SY5Y cells, caffeine (0.125-2 mg/mL), taurine (1-16 mg/mL), and guarana (3.125-50 mg/mL) showed concentration-dependent nonenzymatic antioxidant potential, decreased the basal levels of free radical generation, and reduced both superoxide dismutase (SOD) and catalase (CAT) activities, especially when combined together. However, guarana-treated cells developed signs of neurite degeneration in the form of swellings at various segments in a beaded or pearl chain-like appearance and fragmentation of such neurites at concentrations ranging from 12.5 to 50 mg/mL. Swellings, but not neuritic fragmentation, were detected when cells were treated with 0.5 mg/mL (or higher doses) of caffeine, concentrations that are present in EDs. Cells treated with guarana also showed qualitative signs of apoptosis, including membrane blebbing, cell shrinkage, and cleaved caspase-3 positivity. Flow cytometric analysis confirmed that cells treated with 12.5-50 mg/mL of guarana and its combinations with caffeine and/or taurine underwent apoptosis. Conclusion. Excessive removal of intracellular reactive oxygen species, to nonphysiological levels (or antioxidative stress), could be a cause of in vitro toxicity induced by these drugs. © 2013 Fares Zeidán-Chuliá et al.

Nitensidine A is a guanidine alkaloid isolated from Pterogyne nitens, a common plant in South America. To gain insight into the biological activity of P. nitens-produced compounds, we examined herein their biological effects on osteoclasts, multinucleated giant cells that regulate bone metabolism by resorbing bone. Among four guanidine alkaloids (i.e., galegine, nitensidine A, pterogynidine, and pterogynine), nitensidine A and pterogynine exhibited anti-osteoclastic effects at 10 μM by reducing the number of osteoclasts on the culture plate whereas galegine and pterogynidine did not. The anti-osteoclastic activities of nitensidine A and pterogynine were exerted in a concentration-dependent manner, whereas nitensidine A exhibited an approximate threefold stronger effect than pterogynine (IC50 values: nitensidine A, 0.93 ± 0.024 μM; pterogynine, 2.7 ± 0.40 μM). In the present study, the anti-osteoclastic effects of two synthetic nitensidine A derivatives (nitensidine AT and AU) were also examined to gain insight into the structural features of nitensidine A that exert an anti-osteoclastic effect. The anti-osteoclastic effect of nitensidine A was greatly reduced by substituting the imino nitrogen atom in nitensidine A with sulfur or oxygen. According to the differences in chemical structures and anti-osteoclastic effects of the four guanidine alkaloids and the two synthetic nitensidine A derivatives, it is suggested that the number, binding site, and polymerization degree of isoprenyl moiety in the guanidine alkaloids and the imino nitrogen atom cooperatively contribute to their anti-osteoclastic effects. © 2013 Springer Science+Business Media Dordrecht.

This paper is devoted to study the 1D model of invasive avascular tumor growth, which takes into account cell division, death, and motility, proposed by Kolobov and collaborators in 2009. First, we examine the existence and uniqueness of the solution to this model. Second, we studied qualitatively and numerically the traveling wave solutions. Finally, we show some numerical simulations for the cell density and nutrient concentration. © 2013 NSP Natural Sciences Publishing Cor.

Image restoration is a research field that attempts to recover a blurred and noisy image. Since it can be modeled as a linear system, we propose in this paper to use the meta-heuristics optimization algorithm Harmony Search (HS) to find out near-optimal solutions in a Projections Onto Convex Sets-based formulation to solve this problem. The experiments using HS and four of its variants have shown that we can obtain near-optimal and faster restored images than other evolutionary optimization approach. © 2013 IEEE.

Research for acaricides with lower toxicity and impact on the environment has been intensified. An alternative would be the use of natural compounds or of synthetic products in lower concentrations than the ones sold commercially. Thus, this study describes the action of andiroba seed oil on the nuclei of the ovary and synganglion cells of Rhipicephalus sanguineus, and presents an analysis of the nuclear morphology of the nervous system cells of this tick species when exposed to permethrin. The results obtained showed that, although no changes have been observed in the genetic material of the ovary cells exposed to the andiroba oil, this compound, as well as permethrin, has neurotoxic action on the females of this species. The damages caused to the physiology of the synganglion, due to the loss of integrity of the genetic material, would result in the impairment of the metabolism of other systems of R. sanguineus ticks. © 2013 Elsevier Ltd.

A monitorização ambulatorial do eletrocardiograma (ECG) permite seguir as atividades cotidianas do paciente durante períodos de 24 horas (ou ainda maiores) possibilitando o estudo de casos que pudessem ter episódios arrítmicos fatais. Entretanto, o maior desafio tecnológico que este tipo de monitorização enfrenta é a perda de informação pela presença de ruídos e artefatos quando o paciente se move. A análise do intervalo QT de despolarização e repolarização ventricular do eletrocardiograma superficial é uma técnica não invasiva com um grande valor para a diagnose e prognósticos de cardiopatias e neuropatias, assim como para a predição da morte cardíaca súbita. A análise do desvio padrão do intervalo QT proporciona informação sobre a dispersão (temporal ou espacial) da repolarização ventricular, entretanto a influencia do ruído provoca erros na detecção do final da onda T que são apreciáveis devido ao fato dos valores pequenos do desvio padrão do QT tanto para sujeitos patológicos e quanto para os sãos. O objetivo geral desta tese é melhorar os métodos de processamento do sinal de ECG ambulatorial usando inteligência computacional, especificamente os métodos relacionados com a detecção do final da onda T, e os de reconhecimento morfológico de batimentos que invalidam a análise da variabilidade do intervalo QT. É proposto e validado (em termos de exatidão e precisão) um novo método e algoritmo para estimar o final da onda T baseado no calculo de áreas de trapézios, empregando sinais da base de dados QT da Physionet. O desempenho do método proposto foi testado e comparado com um dos métodos mais usados para detectar o final da onda T: o método baseado no limiar na primeira derivada. O método de inteligência computacional sugerido combina a extração de características usando o método de análise de componentes principais não lineares e a rede neural de tipo perceptron multicamada. O método de áreas de trapézios teve um bom desempenho em condições ruidosas e não depende de nenhum limiar empírico, sendo adequado para situações com níveis de elevados de ruído de banda larga. O método de reconhecimento morfológico de batimentos foi avaliado com sinais ambulatoriais com e sem artefatos pertencentes a bases de dados de prestigio internacional, e mostrou um bom desempenho.

Um dos problemas enfrentados pelos médicos na assistência a doentes com câncer refere-se aos dilemas relacionados à comunicação sobre a doença, especialmente pela associação ainda presente no imaginário social à idéia de morte e sofrimento, conferindo ao diagnóstico uma importante dimensão simbólica. Interpretado como uma construção social, o tema do diagnóstico do câncer é analisado com base na experiência de médicos que atuam no Hospital Ophir Loyola (PA), referência estadual no tratamento do câncer, compreendendo um total de 20 informantes, que concordaram em participar de entrevistas cujo enfoque era a comunicação sobre o diagnóstico, incluindo o enunciado inicial da doença e as informações dele derivadas. Por meio de uma abordagem sócio-antropológica, são analisadas diferentes variáveis, do doente, do médico, da família e da doença, incluindo algumas particularidades referentes aos contextos público e privado. Os dados sugerem que a relação do médico com o doente é influenciada pela condição de classe, com determinantes sócio-históricos e culturais, que tornam a comunicação um fenômeno complexo, agravado pelo limitado acesso aos serviços de saúde, contribuindo para que grande parte dos doentes já cheguem ao Hospital sem possibilidade de cura. Os resultados obtidos evidenciaram que, na realidade pesquisada, a influência da família no processo de decisão foi relevante para determinar os limites entre o dizer e o não dizer sobre a doença, incluindo o enunciado do diagnóstico e do prognóstico. A perda da esperança foi freqüentemente citada como um requisito para a não divulgação de informações, especialmente nos casos em que a doença está em uma fase avançada, havendo um maior silenciamento à medida que a doença progride.

Because an enriched environment (EE) enhances T-cell activity and T-lymphocytes contribute to immunopathogenesis during heterologous dengue virus (DENV) infections, we hypothesised that an EE increases dengue severity. To compare single serotype (SS) and antibody-enhanced disease (AED) infections regimens, serial intraperitoneal were performed with DENV3 (genotype III) infected brain homogenate or anti-DENV2 hyperimmune serum followed 24 h later by DENV3 (genotype III) infected brain homogenate. Compared AED for which significant differences were detected between the EE and impoverished environmental (IE) groups (Kaplan-Meyer log-rank test, p = 0.0025), no significant differences were detected between the SS experimental groups (Kaplan-Meyer log-rank test, p = 0.089). Survival curves from EE and IE animals infected with the AED regimen were extended after corticoid injection and this effect was greater in the EE than in the IE group (Kaplan-Meyer log-rank test, p = 0.0162). Under the AED regimen the EE group showed more intense clinical signs than the IE group. Dyspnoea, tremor, hunched posture, ruffled fur, immobility, pre-terminal paralysis, shock and death were associated with dominant T-lymphocytic hyperplasia and presence of viral antigens in the liver and lungs. We propose that the increased expansion of these memory T-cells and serotype cross-reactive antibodies facilitates the infection of these cells by DENV and that these events correlate with disease severity in an EE.

Cocaine is a widely used drug and its abuse is associated with physical, psychiatric and social problems. Abnormalities in newborns have been demonstrated to be due to the toxic effects of cocaine during fetal development. The mechanism by which cocaine causes neurological damage is complex and involves interactions of the drug with several neurotransmitter systems, such as the increase of extracellular levels of dopamine and free radicals, and modulation of transcription factors. The aim of this review was to evaluate the importance of the dopaminergic system and the participation of inflammatory signaling in cocaine neurotoxicity. Our study showed that cocaine activates the transcription factors NF-κB and CREB, which regulate genes involved in cellular death. GBR 12909 (an inhibitor of dopamine reuptake), lidocaine (a local anesthetic), and dopamine did not activate NF-κB in the same way as cocaine. However, the attenuation of NF-κB activity after the pretreatment of the cells with SCH 23390, a D1 receptor antagonist, suggests that the activation of NF-κB by cocaine is, at least partially, due to activation of D1 receptors. NF-κB seems to have a protective role in these cells because its inhibition increased cellular death caused by cocaine. The increase in BDNF (brain-derived neurotrophic factor) mRNA can also be related to the protective role of both CREB and NF-κB transcription factors. An understanding of the mechanisms by which cocaine induces cell death in the brain will contribute to the development of new therapies for drug abusers, which can help to slow down the progress of degenerative processes.
