The venom of Crotalus durissus terrificus snakes presents various substances, including a serine protease with thrombin-like activity, called gyroxin, that clots plasmatic fibrinogen and promote the fibrin formation. The aim of this study was to purify and structurally characterize the gyroxin enzyme from Crotalus durissus terrificus venom. For isolation and purification, the following methods were employed: gel filtration on Sephadex G75 column and affinity chromatography on benzamidine Sepharose 6B; 12% SDS-PAGE under reducing conditions; N-terminal sequence analysis; cDNA cloning and expression through RT-PCR and crystallization tests. Theoretical molecular modeling was performed using bioinformatics tools based on comparative analysis of other serine proteases deposited in the NCBI (National Center for Biotechnology Information) database. Protein N-terminal sequencing produced a single chain with a molecular mass of similar to 30 kDa while its full-length cDNA had 714 bp which encoded a mature protein containing 238 amino acids. Crystals were obtained from the solutions 2 and 5 of the Crystal Screen Kit (R), two and one respectively, that reveal the protein constitution of the sample. For multiple sequence alignments of gyroxin-like B2.1 with six other serine proteases obtained from snake venoms (SVSPs), the preservation of cysteine residues and their main structural elements (alpha-helices, beta-barrel and loops) was indicated. The localization of the catalytic triad in His57, Asp102 and Ser198 as well as S1 and S2 specific activity sites in Thr193 and Gli215 amino acids was pointed. The area of recognition and cleavage of fibrinogen in SVSPs for modeling gyroxin B2.1 sequence was located at Arg60, Arg72, Gln75, Arg81, Arg82, Lis85, Glu86 and Lis87 residues. Theoretical modeling of gyroxin fraction generated a classical structure consisting of two alpha-helices, two beta-barrel structures, five disulfide bridges and loops in positions 37, 60, 70, 99, 148, 174 and 218. These results provided information about the functional structure of gyroxin allowing its application in the design of new drugs.

On tomato production in Brazil, around two/thirties is designated to consume "in nature". Thus, the skin aspect and its quality became an important factor on buying decision. Tomato presents high mass of water, and its quality depends on temperature and humidity variation. Water loss brings weight and fruit aspect loss, affecting the quality. This work aimed to verify the influence of harvest season on the fruit quality grown in alternative, organic and biodynamic systems. It was evaluated tomato quality characteristics, including mass loss, texture, pH, titratable acidity, soluble solids. The cycle stage of the tomato plant interferes on fruit quality. The third, fourth and fifth harvests were those which showed the highest values to quality parameters, with more fresh mass fruit. The sixth harvest showed fruits with high total soluble solids and sugar contents. on the evaluation of mass loss on the three harvest seasons, it was observed that on the first harvest there was a smaller loss. The period of harvest cycle interferes on the time of storage, and fruitd harvested on the first have more conservation time in relation to the other ones.

Alternaria alternata, the causal agent of Alternaria brown spot (ABS), causes necrosis on leaves, twigs, and fruit, reducing the productivity and quality of fruits. Tangerines and their hybrids are highly susceptible to the disease. Species, hybrids, and cultivars of Citrus from the germplasm bank of the Estacao Experimental de Citricultura de Bebedouro, São Paulo, Brazil, were evaluated in 2004 and 2005 with respect to their resistance to A. alternata, both through natural infection and by inoculation. Detached leaves were also used to demonstrate susceptibility or resistance to the disease. Ten cultivars of Satsumas (Citrus unshiu), and 14 cultivars of Clementine mandarin (C. clementina) did not show any symptoms of the disease in their leaves, either through natural infection or when inoculated in the field. The Burguess SRA-412, Wallent SRA-438, Carvalhais, Ampefy SRA-459, Ananas SRA, and Macaque SRA-426 mandarin hybrids (C. reticulata) did not show symptoms of the disease under natural or artificial infection in the field. Some cultivars of C. deliciosa, C. tangerina, C. erythrosa, and C. temple showed symptoms of the disease, even though no previous record of their susceptibility to Alternaria brown spot had been previously reported. The hybrids Fairchild, Nova, Page, Fortune, and Sunburst were susceptible to the disease. However, Fremont mandarin (a crossing between C. clementina and C. reticulata), Encore (C. nobilis x C. deliciosa), and Fallglo (C. reticulata x C. paradisi) did not show symptoms in field, and few symptoms were verified in detached leaves. These materials are promising for the cultivation of tangerines, and will enable genetic improvement for the development of cultivars resistant to Alternaria brown spot. (c) 2007 Elsevier B.V. Biofertilizers generated from the anaerobic and aerobic digestion of cattle manure, known as Bio1 and Bio2, respectively, were studied with regard to their microbial composition and effect on the mycelial growth of Phyllosticta citricarpa, the causal agent of citrus black spot (CBS). Two field experiments were conducted to determine the biofertilizer's potential (Bio1) in controlling CBS (2001/2002 and 2002/2003 crops). It was observed that the greatest number of microorganisms was found in the aerobically produced biofertilizer. Bio2 did not inhibit the mycelial growth of P. citricarpa. Mycelial growth of P. citricarpa was inversely proportional to the Bio1 biofertilizer concentration. In the 2001/2002 cropping season, the Biol effect in controlling CBS was directly proportional to its concentration, at the rate of 0 (healthy fruit), with R-2 = 0.88. Biol had a significant effect in controlling CBS, at a concentration of 10%, during the 2001/2002 cropping season, with DI values of 0.246 and 0.229 for the. 10 and 20% doses, respectively, compared to DI of 0.329 for the control. A directly proportional effect of the biofertilizer concentration on the percentage of fruits with a rating of zero was. also observed in the 2002/2003 cropping season, with R-2 = 0.48. However, even at doses higher than in the preceding cropping season, the biofertilizer was less effective, possibly due to a higher occurrence of the disease. Copper oxychloride and combined applications of copper oxychloride and carbendazim plus mancozeb controlled the disease. The possibility of using the biofertilizer as a protective biofungicide to replace copper oxychloride, especially in organic agriculture, should be explored. (c) 2005 Elsevier Ltd. O objetivo deste trabalho foi estudar o efeito do diâmetro dos botões florais de algodoeiro na massa corporal de adultos do bicudo-do-algodoeiro, Anthonomus grandis Boheman (Coleoptera: Curculionidae). O experimento foi realizado em condições de campo e laboratório, em Jaboticabal (SP), Brasil. Foram realizadas seis amostragens, coletando-se ao acaso dez plantas nas cultivares Coodetec 405 e Fibermax 986, avaliando-se o número e o diâmetro de botões florais. Os botões florais com orifícios de oviposição foram individualizados em frascos e observados diariamente para a visualização da emergência dos adultos. Botões florais com maiores diâmetros proporcionam maior massa corporal de adultos de A. grandis recém-emergidos nas duas cultivares.

O objetivo do presente trabalho foi determinar o acúmulo e exportação de macronutrientes pela cultura da melancia, híbrido Tide. O experimento foi conduzido no município de Borborema - SP. As amostragens de plantas foram realizadas aos 15, 30, 45, 60 e 75 dias após transplantio (DAT), para determinação da massa seca e do acúmulo e exportação dos nutrientes (N, P, K, Ca, Mg e S). O acúmulo de massa seca foi lento até os 30 DAT, intensificando-se a partir deste período e atingindo, no final do ciclo, a contribuição média da parte vegetativa de 31% e dos frutos de 69%. Até 30 DAT, o acúmulo de nutrientes também foi pequeno, não ultrapassando 2% do total. Com a frutificação, houve forte incremento na quantidade de nutrientes acumulados. O período de maior demanda para N, Ca e Mg foi de 45 a 60 DAT e para P, K e S de 60 a 75 DAT. do total acumulado, a parte vegetativa foi responsável por 29,6% e os frutos com 70,4%. A ordem decrescente dos nutrientes acumulados pela cultura foi: K>N>Ca>Mg>P>S. Para a produtividade alcançada de 40 t ha-1 observou-se a seguinte exportação de nutrientes pelos frutos: 106,4 kg ha-1 de N, 11,1 kg ha-1 de P, 118,0 kg ha-1 de K, 4,3 kg ha-1 de Ca, 6,8 kg ha-1 de Mg e 6,0 kg ha-1 de S.

O objetivo desta pesquisa foi estudar o processo de maturação de sementes de sabiá (Mimosa caesalpiniifolia Benth.). Foram realizadas dezesseis colheitas semanais de frutos e sementes de árvores, no município de Areia - Paraiba, no período de agosto a novembro de 2001. As colheitas iniciaram-se aos 105 dias e estenderam-se até os 210 dias após a antese. Foram avaliadas as dimensões, o teor de água e a massa fresca e seca de frutos e sementes, bem como a germinação e o vigor (primeira contagem de germinação, comprimento e massa fresca e seca das plântulas). Concluiu-se que a maturidade fisiológica das sementes de sabiá ocorre entre 154 e168 dias após a antese, podendo a colheita ser realizada até 189 dias após a antese, uma vez que a partir deste período ocorre elevada perda de frutos e sementes devido à dispersão natural.
