Despite the resistance developed by the Mycobacterium tuberculosis (MTb) strains, isoniazid (INK) has been recognized as one of the best drug for treatment of Tuberculosis (Tb). The coordination of INH to ruthenium metal centers was investigated as a strategy to enhance the activity of this drug against the sensitive and resistant strains of MTb. The complexes trans-[Ru(NH3)(4)(L)(INH)](2+) (L = SO2 or NH3) were isolated and their chemical and antituberculosis properties studied. The minimal inhibitory concentration (MIC) data show that [Ru(NH3)(5)(INH)](2+) was active in both resistant and sensitive strains, whereas free INK (non-coordinated) showed to be active only against the sensitive strain. The coordination of INH to the metal center in both [Ru(NH3)(5)(INH)](2+) and trans-[Ru(NH3)(4)(SO2)(INH)](2+) complexes led to a shift in the INH oxidation potential to less positive values compared to free INH. Despite, the ease of oxidation of INH did not lead to an increase in the in vitro INH activity against MTb, it might have provided sensitivity toward resistant strains. Furthermore, ruthenium complexes with chemical structures analogous to those described above were synthesized using the oxidation products of INK as ligands (namely, isonicotinic acid and isonicotinamide). These last compounds were not active against any strains of MTb. Moreover, according to DFT calculations the formation of the acyl radical, a proposed intermediate in the INH oxidation, is favored in the [Ru(NH3)(5)(INH)](2+) complex by 50.7 kcal mol(-1) with respect to the free INH. This result suggests that the stabilization of the acyl radical promoted by the metal center would be a more important feature than the oxidation potential of the INH for the antituberculosis activity against resistant strains. The compounds [NiX 2(PPh 3) 2] (where X is Cl -, Br -, I -, NO - 3, NCS -; and PPh 3 is triphenylphosphine) were prepared and characterized by infrared and atomic absorption spectroscopies and by carbon and hydrogen analyses. Simultaneous thermogravimetric (TG) and derivative thermogravimetric (DTG) curves of these complexes were recorded in air. The decrease in mass observed indicates conversion of the complexes to oxides. The thermal decomposition of the halogen and nitrate complexes occurred in a number of steps; the thiocyanate complex decomposed in a single step. © 1994.

Cu(II)/neocuproine (2,9-dimethyl-1,10-phenanthroline) complexes were utilized for spectrophotometric determination of total polyphenol content in beers. This procedure is based on the reduction of Cu(II) by polyphenols in hydroethanolic medium (pH 7.0) in the presence of neocuproine, yielding Cu(I)/complexes with maximum absorption at 454 nm. The sensitivity of the proposed method was compared with the AOAC method using tannic and gallic acid as standards. The average apparent molar absorptivity, in L cm(-1) mol(-1), of tannic acid (3.50 +/- 0.20) x 10(5) and gallic acid (5.12 +/- 0.21) x 10(4), was twice as high for the proposed method. A lower limit of detection (LOD) (2.9 x 10(-1) mg L-1) was found when tannic acid was used in the proposed method. Additionally, less interference from the most common additives in beers was noticed. Total content of polyphenols was analyzed in 17 Brazilian samples. Results ranged from 35.5 to 556 mg L-1 of tannic acid, with higher values for recovery rates (45.4-118%, mean 85.0%) than for gallic acid. Although tannic acid is a mixture of polygalloyl glucoses, the total polyphenol content found in the samples suggests that tannic acid should be used as a standard. Im Rahmen dieser Arbeit wurden zweikernige Modellkomplexe zur Untersuchung der Radikal-Metallwechselwirkung innerhalb des wasseroxidierenden Zentrums des Photo¬systems II synthetisiert und eine magneto-strukturelle Korrelation dieser Komplexe erstellt. Als Liganden wurden diverse sechs- bis siebenzähnige Chelatliganden verwendet, welche über zwei Koordinationstaschen und eine verbrückende Phenolatgruppe verfügen. Zwei daran gebundene Manganionen liegen in einer wohl definierten Umgebung nicht koordinativ gesättigt vor. An die freien Koordinationsstellen können weitere ein bis zwei Brückenliganden binden, bei denen es sich in dieser Arbeit hauptsächlich um Carboxylate handelt. Durch die Verwendung eines diamagnetischen Brückenliganden konnte die magnetische Spin-Spin-Austauschwechselwirkung zwischen den spintragenden Manganionen über die verbrücken¬de Phenolatgruppe bestimmt werden. Komplexe, welche über Manganionen in den gleichen Oxidationsstufen, aber über unterschiedliche Carboxylatbrückenliganden verfügen, weisen ähnliche magnetische Austauschwechselwirkungen zwischen den Metallzentren auf. Diese Beobachtung konnte durch eine strukturelle Ähnlichkeit dieser Komplexe erklärt werden. Mittels Aufsummieren der Bindungslängen der verbrückenden Phenolateinheit zu beiden Zentralionen kann innerhalb dieser Komplexe jeweils die Länge des Wechselwirkungspfades erhalten werden, welcher die magnetische Austauschwechselwirkung maßgeblich beein¬flusst. Je länger der Wechselwirkungspfad ist, desto kleiner ist die Austausch¬wechsel¬wirkung. Durch Austausch der diamagnetischen Carboxylate durch paramagnetische benzoat¬substituierte Nitronyl Nitroxid Radikale wurden den Komplexen ein bis zwei weitere Spinzentren hinzugefügt, welche mit den Spins der Zentralionen wechselwirken können. Simulationen der magnetischen Suszeptibilitätsmessungen liefern Werte für die magneti¬schen Austausch¬wechselwirkungen zwischen den Nitronyl Nitroxid Radikalen und den Manganionen, die in allen Fällen schwach ferromagnetisch zwischen 0 und 4,7 cm-1 sind. In einer Auftragung dieser Austauschwechselwirkungen gegen die Mangan-Carboxylat-Bindungs¬längen von strukturell charakterisierten äquivalenten acetatverbrückten Komplexen, kann eine lineare Abhängigkeit gezeigt werden.

During the Equamarge II cruise (February 4 to March 21, 1988), on board the R. V. "Jean Charcot", 12.500 kms of continuous geophysical profiling have been recorded along three sectors of the Equatorial Atlantic. Two segments ofthe West African transform margin have been intensively surveyed off Guinea and off Ivory Coast and Ghana. The active Romanche fracture zone has been surveyed in details on a distance of about 100 kms. These data (multibeam bathymetry, continuous seismic profiling, magnetism and gravity) have been supplemented by 16 geological stations (dredging and coring). This report gives a synthetic review of the onboard analysis and allows to better understand the geological structures of the three surveyed areas.

TFIIH is a multifunctional RNA polymerase II transcription factor that possesses DNA-dependent ATPase, DNA helicase, and protein kinase activities. Previous studies have established that TFIIH enters the preinitiation complex and fulfills a critical role in initiation by catalyzing ATP-dependent formation of the open complex prior to synthesis of the first phosphodiester bond of nascent transcripts. In this report, we present direct evidence that TFIIH also controls RNA polymerase II activity at a postinitiation stage of transcription, by preventing premature arrest by very early elongation complexes just prior to their transition to stably elongating complexes. Unexpectedly, we observe that TFIIH is capable of entering the transcription cycle not only during assembly of the preinitiation complex but also after initiation and synthesis of as many as four to six phosphodiester bonds. These findings shed new light on the role of TFIIH in initiation and promoter escape and reveal an unanticipated flexibility in the ability of TFIIH to interact with RNA polymerase II transcription intermediates prior to, during, and immediately after initiation.

By combining two previously generated null mutations, Ii° and M°, we produced mice lacking the invariant chain and H-2M complexes, both required for normal cell-surface expression of major histocompatibility complex class II molecules loaded with the usual diverse array of peptides. As expected, the maturation and transport of class II molecules, their expression at the cell surface, and their capacity to present antigens were quite similar for cells from Ii°M° double-mutant mice and from animals carrying just the Ii° mutation. More surprising were certain features of the CD4+ T cell repertoire selected in Ii°M° mice: many fewer cells were selected than in Ii+M° animals, and these had been purged of self-reactive specificities, unlike their counterparts in Ii+M° animals. These findings suggest (i) that the peptides carried by class II molecules on stromal cells lacking H-2M complexes may almost all derive from invariant chain and (ii) that H-2M complexes edit the peptide array displayed on thymic stromal cells in the absence of invariant chain, showing that it can edit, in vivo, peptides other than CLIP.

A hyperphosphorylated form of the largest subunit of RNA polymerase II (pol IIo) is associated with the pre-mRNA splicing process. Pol IIo was detected in association with a subset of small nuclear ribonucleoprotein particle and Ser-Arg protein splicing factors and also with pre-mRNA splicing complexes assembled in vitro. A subpopulation of pol IIo was localized to nuclear "speckle" domains enriched in splicing factors, indicating that it may also be associated with RNA processing in vivo. Moreover, pol IIo was retained in a similar pattern following in situ extraction of cells and was quantitatively recovered in the nuclear matrix fraction. The results implicate nuclear matrix-associated hyperphosphorylated pol IIo as a possible link in the coordination of transcription and splicing processes.

Electron spin echo electron-nuclear double resonance (ESE-ENDOR) experiments performed on a broad radical electron paramagnetic resonance (EPR) signal observed in photosystem II particles depleted of Ca2+ indicate that this signal arises from the redox-active tyrosine YZ. The tyrosine EPR signal width is increased relative to that observed in a manganese-depleted preparation due to a magnetic interaction between the photosystem II manganese cluster and the tyrosine radical. The manganese cluster is located asymmetrically with respect to the symmetry-related tyrosines YZ and YD. The distance between the YZ tyrosine and the manganese cluster is estimated to be approximately 4.5 A. Due to this close proximity of the Mn cluster and the redox-active tyrosine YZ, we propose that this tyrosine abstracts protons from substrate water bound to the Mn cluster.
