Increasing attention is being paid to the possible development of non-invasive tests for the assessment of the quality of Fruits. We propose a novel non-destructive method for the measurement of the internal optical properties of fruits and vegetables by means of lime-resolved reflectance spectroscopy in the visible and NIR range. A Fully automated instrumentation for time-resolved reflectance measurements was developed. It is based on mode-locked laser sources and electronics for time-correlated single photon counting, and provides a time-resolution of 120-160 ps. The system was used to probe the optical properties of several species and varieties of Fruits and vegetables in the red and NIR range (650-1000 nm). In most Fruits, the absorption line shape is dominated by the absorption peak of water, centred around 970 nm. Generally, the absorption spectra also show the spectral features typical of chlorophyll, with maximum at 675 nm. In particular, for what concerns apples, variations in peak intensity are observed depending on the variety, the degree of ripeness as well as the position on the apple. For all the species and varieties considered, the transport scattering coefficient decreases progressively upon increasing the wavelength.

A presente dissertação teve como objetivos contextualizar e analisar as perspectivas da educação salesiana a partir do documento das Linhas Orientadoras da Missão Educativa do Instituto das Filhas de Maria Auxiliadora, tendo em vista aprofundar o contexto histórico e sociocultural da fundação do Instituto das Filhas de Maria Auxiliadora e do desenvolvimento da missão educativa salesiana, com a finalidade de atuar no campo da educação, indicar os fundamentos interdisciplinares que compreendem os fundamentos teóricos para a prática educativa em relação aos aspectos sociológicos, antropológicos, teológicos e pedagógicos, além de algumas experiências práticas da educação salesiana. A pesquisa considera a contextualização da sua prática educativa a partir de diversas contribuições teóricas realizadas em torno da história do Instituto e dos conhecimentos interdisciplinares que integram seus propósitos e seus projetos educativos. Com a perspectiva de analisar as orientações e conteúdos do documento é utilizada a técnica da análise documental que permite interpretar o seu significado, analisar e indicar algumas perspectivas de educação salesiana presentes no documento e que envolvem os seus princípios, propósitos e as suas ações em relação à educação. Por meio da observação participante alguns aspectos práticos das perspectivas da educação salesiana foram demonstrados. Portanto, a investigação permite o mapeamento das perspectivas fundamentais da educação salesiana a partir das Linhas Orientadoras e a possibilidade de aprofundar a significatividade das perspectivas de referência para a prática educativa conforme são indicadas no documento: as dimensões cultural, evangelizadora, social e comunicativa da educação, que são os elementos e princípios fundamentais do Sistema Educativo Salesiano. A concepção de educação salesiana que se constrói a partir de uma visão interdisciplinar, complexa e dimensional, com uma perspectiva de uma estratégia projetual. Os seus princípios integram alguns valores que são considerados como fundamentais para a realização de uma educação integral: a comunidade educativa, o trabalho educativo realizado em equipe, a espiritualidade juvenil, a preventividade e a pedagogia do ambiente. Sobretudo, os núcleos centrais indicados pelo documento orientam as comunidades para a realização de um pensamento projetual, considerando as diversas dimensões da educação, com uma especial abertura para uma contínua projeção das suas intenções e da sua prática educativa. Enfim, encontramos algumas contribuições para a educação a partir do pensamento pedagógico e prático da educação salesiana, especialmente em relação à proposta de pensar a educação de forma complexa e multidimensional.

Toxoplasma gondii is a coccidian parasite with a global distribution. The definitive host is the cat (and other felids). All warm-blooded animals can act as intermediate hosts, including humans. Sexual reproduction (gametogony) takes place in the final host and oocysts are released in the environment, where they then sporulate to become infective. In intermediate hosts the cycle is extra-intestinal and results in the formation of tachyzoites and bradyzoites. Tachyzoites represent the invasive and proliferative stage and on entering a cell it multiplies asexually by endodyogeny. Bradyzoites within tissue cysts are the latent form. T. gondii is a food-borne parasite causing toxoplasmosis, which can occur in both animals and humans. Infection in humans is asymptomatic in more than 80% of cases in Europe and North-America. In the remaining cases patients present fever, cervical lymphadenopathy and other non-specific clinical signs. Nevertheless, toxoplasmosis is life threatening if it occurs in immunocompromised subjects. The main organs involved are brain (toxoplasmic encephalitis), heart (myocarditis), lungs (pulmonary toxoplasmosis), eyes, pancreas and parasite can be isolated from these tissues. Another aspect is congenital toxoplasmosis that may occur in pregnant women and the severity of the consequences depends on the stage of pregnancy when maternal infection occurs. Acute toxoplasmosis in developing foetuses may result in blindness, deformation, mental retardation or even death. The European Food Safety Authority (EFSA), in recent reports on zoonoses, highlighted that an increasing numbers of animals resulted infected with T. gondii in EU (reported by the European Member States for pigs, sheep, goats, hunted wild boar and hunted deer, in 2011 and 2012). In addition, high prevalence values have been detected in cats, cattle and dogs, as well as several other animal species, indicating the wide distribution of the parasite among different animal and wildlife species. The main route of transmission is consumption of food and water contaminated with sporulated oocysts. However, infection through the ingestion of meat contaminated with tissue cysts is frequent. Finally, although less frequent, other food products contaminated with tachyzoites such as milk, may also pose a risk. The importance of this parasite as a risk for human health was recently highlighted by EFSA’s opinion on modernization of meat inspection, where Toxoplasma gondii was identified as a relevant hazard to be addressed in revised meat inspection systems for pigs, sheep, goats, farmed wild boar and farmed deer (Call for proposals -GP/EFSA/BIOHAZ/2013/01). The risk of infection is more highly associated to animals reared outside, also in free-range or organic farms, where biohazard measure are less strict than in large scale, industrial farms. Here, animals are kept under strict biosecurity measures, including barriers, which inhibit access by cats, thus making soil contamination by oocysts nearly impossible. A growing demand by the consumer for organic products, coming from free-range livestock, in respect of animal-welfare, and the desire for the best quality of derived products, have all led to an increase in the farming of free-range animals. The risk of Toxoplasma gondii infection increases when animals have access to environment and the absence of data in Italy, together with need for in depth study of both the prevalence and genotypes of Toxoplasma gondii present in our country were the main reasons for the development of this thesis project. A total of 152 animals have been analyzed, including 21 free-range pigs (Suino Nero race), 24 transhumant Cornigliese sheep, 77 free-range chickens and 21 wild animals. Serology (on meat juice) and identification of T. gondii DNA through PCR was performed on all samples, except for wild animals (no serology). An in-vitro test was also applied with the aim to find an alternative and valid method to bioassay, actually the gold standard. Meat samples were digested and seeded onto Vero cells, checked every day and a RT-PCR protocol was used to determine an eventual increase in the amount of DNA, demonstrating the viability of the parasite. Several samples were alos genetically characterized using a PCR-RFLP protocol to define the major genotypes diffused in the geographical area studied. Within the context of a project promoted by Istituto Zooprofilattico of Pavia and Brescia (Italy), experimentally infected pigs were also analyzed. One of the aims was to verify if the production process of cured “Prosciutto di Parma” is able to kill the parasite. Our contribution included the digestion and seeding of homogenates on Vero cells and applying the Elisa test on meat juice. This thesis project has highlighted widespread diffusion of T. gondii in the geographical area taken into account. Pigs, sheep, chickens and wild animals showed high prevalence of infection. The data obtained with serology were 95.2%, 70.8%, 36.4%, respectively, indicating the spread of the parasite among numerous animal species. For wild animals, the average value of parasite infection determined through PCR was 44.8%. Meat juice serology appears to be a very useful, rapid and sensitive method for screening carcasses at slaughterhouse and for marketing “Toxo-free” meat. The results obtained on fresh pork meat (derived from experimentally infected pigs) before (on serum) and after (on meat juice) slaughter showed a good concordance. The free-range farming put in evidence a marked risk for meat-producing animals and as a consequence also for the consumer. Genotyping revealed the diffusion of Type-II and in a lower percentage of Type-III. In pigs is predominant the Type-II profile, while in wildlife is more diffused a Type-III and mixed profiles (mainly Type-II/III). The mixed genotypes (Type-II/III) could be explained by the presence of mixed infections. Free-range farming and the contact with wildlife could facilitate the spread of the parasite and the generation of new and atypical strains, with unknown consequences on human health. The curing process employed in this study appears to produce hams that do not pose a serious concern to human health and therefore could be marketed and consumed without significant health risk. Little is known about the diffusion and genotypes of T. gondii in wild animals; further studies on the way in which new and mixed genotypes may be introduced into the domestic cycle should be very interesting, also with the use of NGS techniques, more rapid and sensitive than PCR-RFLP. Furthermore wildlife can become a valuable indicator of environmental contamination with T. gondii oocysts. Other future perspectives regarding pigs include the expansion of the number of free-range animals and farms and for Cornigliese sheep the evaluation of other food products as raw milk and cheeses. It should be interesting to proceed with the validation of an ELISA test for infection in chickens, using both serum and meat juice on a larger number of animals and the same should be done also for wildlife (at the moment no ELISA tests are available and MAT is the reference method for them). Results related to Parma ham do not suggest a concerning risk for consumers. However, further studies are needed to complete the risk assessment and the analysis of other products cured using technological processes other than those investigated in the present study. For example, it could be interesting to analyze products such as salami, produced with pig meat all over the Italian country, with very different recipes, also in domestic and rural contexts, characterized by a very short period of curing (1 to 6 months). Toxoplasma gondii is one of the most diffuse food-borne parasites globally. Public health safety, improved animal production and protection of endangered livestock species are all important goals of research into reliable diagnostic tools for this infection. Future studies into the epidemiology, parasite survival and genotypes of T. gondii in meat producing animals should continue to be a research priority.

The efficiency of a Laue lens for X and Gamma ray focusing in the energy range 60 ÷ 600 keV is closely linked to the diffraction efficiency of the single crystals composing the lens. A powerful focusing system is crucial for applications like medical imaging and X ray astronomy where wide beams must be focused. Mosaic crystals with a high density, such as Cu or Au, and bent crystals with curved diffracting planes (CDP) are considered for the realization of a focusing system for X rays, owing to their high diffraction efficiency. In this work, a comparison of the efficiency of CDP crystals and mosaic crystals was performed on the basis of the theory of X-ray diffraction. Si, GaAs and Ge CDP crystals with optimized thicknesses and moderate radii of curvature of several tens of metres demonstrate comparable or superior performance with respect to the higher atomic number mosaic crystals generally used. A simplified approach for calculating the integrated reflectivity of the crystals is applied. A bending technique used during this work to realize CDP crystals consists in a controlled surface damaging induced by a mechanical lapping process. A compressive strained layer of few micrometres in thickness is generated and causes the convex curvature of the damaged side of the crystal. Another new bending technique is developed and the main results are shown. The process consists on a film deposition of a selected bi-component epoxy resin on one side of crystal, made uniform in thickness by mean of a spin-coater. Choosing the speed of spin-coating, so changing the thickness of the film, a control of radius of curvature can be obtained. Moreover the possibility to combine the two bending technique to obtain CDP crystal with a stronger curvature in rather thick crystals was demonstrated. Detailed characterization of Si, and GaAs CDP crystals at low and high x-ray energies are performed on flat and bent crystals obtained with the damaging and the resin deposition technique. As expected an increase of diffraction efficiency in asymmetrical diffraction geometry in CDP crystals with respect to the flat ones is observed. On the other hand an unexpected increase of the integrated intensity in symmetrical geometry, not predicted by the theory, is observed in all the measurements performed with different set up. The experimental trend of the integrated reflectivity as a function of the radius of curvature is in a good agreement with that predicted by the theory of bent perfect crystals, so it is possible to conclude that the surface damage has a limited effect on the crystal reflectivity. A study of the integrated reflectivity in the energy range of interest (100÷350 keV) in CDP crystals realized with damaging and resin deposition technique at symmetrical and asymmetrical geometries was performed at ILL Institute. Also at these energies the diffraction efficiency of bent crystals was much larger (a 12 time increase is observed for bent crystals in asymmetrical 111 geometry) than that measured in flat crystals. The diffraction efficiency of CDP crystals realized with both techniques tends to coincide with that of flat crystals at very high energies (> 200 keV). This suggesting that also real flat perfect crystals can be considered as strongly bent or mosaic crystals at very high X ray energies.

La ricerca punta a ricostruire il complesso e travagliato processo decisionale che porta alla realizzazione del quarto centro siderurgico di Taranto. Il grande stabilimento viene progettato nel 1955, approda sui tavoli del comitato di Presidenza della Finsider nel settembre del 1956 e dopo quasi quattro anni di dibattiti e dietrofront ottiene il via libera dal consiglio dei Ministri il 9 giugno del 1959. La prima pietra del centro verrà posta il 10 luglio del 1960, il primo settore (un tubificio) inaugurato nel 1961, mentre il resto del complesso entrerà in funzione nel 1964. Ispiratore del centro è Pasquale Saraceno e gli uomini dell'associazione Svimez, convinti sostenitori dell'industrializzazione dell'Italia Meridionale. Il progetto sostenuto apertamente dalla Democrazia Cristiana a partire dal 1956 e in seguito da tutte le forze politiche, viene convintamente osteggiato dall'Impresa Pubblica, Finsider in testa, che vede messa a repentaglio l'autonomia imprenditoriale del gruppo e i propri equilibri economici. Il centro meridionale è considerato strategicamente penalizzante ed economicamente sconveniente, dato che i maggiori centri industriali italiani del tempo si trovano nell'Italia Settentrionale (quindi una localizzazione meridionale comporterebbe un aggravio di costi per le operazioni di trasporto dell'acciaio). L'Impresa pubblica opterebbe su una politica imprenditoriale più prudente, incentrata sul potenziamento graduale dei centri già esistenti. Non così la Politica decisa ad attuare una svolta in grado di realizzare evidenti progressi nell'economia meridionale. La nostra ricerca ha cercato di ricostruire l'impegno e le manovre dei principali partiti politici e parallelamente le strategie e le ragioni dell'Impresa pubblica tenendo anche conto del grande dibattito sorto sulla stampa nazionale tra favorevoli e contrari al progetto e al coinvolgimento dell'opinione pubblica meridionale in particolare pugliese. La ricerca si divide in tre parti (dedicate alle origini del progetto (1954-1955), gli sviluppi della vicenda (1957-1958), il via libera al centro (1959-1960) e si articola in sei capitoli (Meridione e industrializzazione: verso il quarto centro siderurgico; La siderurgia italiana verso il boom: il progetto del IV centro a ciclo integrale; La polemica Iri – Dc: uno scontro istituzionale; Il Pci e il centro siderurgico di Taranto; La vicenda Vado Ligure e la posizione dell'impresa privata; L'approvazione del progetto). La tesi cerca di mettere a fuoco la posizione della Politica (Dc e Pci su tutti), della Tecnica (Iri e Finsider ma anche progetti e impegno dei privati, quali la Fiat e la Falck) e della società civile (soprattutto quella tarantina dedicando una certa attenzione al mondo cattolico). La ricerca si è svolta su più livelli. Nel primo anno si è compiuta un'approfondito studio presso l'archivio dell'Iri (conservato presso l'Archivio Centrale dello Stato a Roma), della Dc (Istituto Sturzo, Roma), del Pci (Istituto Gramsci Roma). Il secondo ha permesso di soffermarsi sulle vicende di Taranto (Archivio dell'Arcidiocesi, Archivio del Comune, Biblioteca Comunale). Il terzo è stato focalizzato ancora su vicende politiche e finanziarie (Archivio Storico Banca d'Italia, Archivio Giulio Andreotti, Archivio storico del Senato, tutti con sede a Roma). L'esame dei verbali dei Comitati esecutivi della Finsider per gli anni 1954 -1959 rivela una chiara direzione: la Finsider studia con grande rigore la situazione del mercato italiano, delle varie aziende e dei centri produttivi del Gruppo. Un rigore finalizzato alla chiusura dei centri ritenuti antiquati e poco funzionali e al potenziamento di quelli più moderni e dalle dimensioni imponenti. Un'azione tesa ad incrementare la produzione, essere competitivi sul mercato internazionale e salvaguardare il vero faro dell'azienda in quegli anni: l'economicità. Cioè ottenere il più possibile tutelando gli equilibri finanziari del Gruppo. E' per questo che per ben due anni, i funzionari dell'azienda avanzano progetti complementari, come quello del piccolo centro di Apuania, ma il Comitato esecutivo pur considerando l'investimento indispensabile e conveniente, decide di rinviarne la realizzazione. Gli elementi più rilevanti sono emersi per l'anno 1957. E' noto come il centro siderurgico meridionale fosse messo a rischio da un altro progetto avanzato dalla Fiat, decisa a realizzare un proprio stabilimento nell'Italia settentrionale, a Vado Ligure. Il centro avrebbe dovuto rifornire di acciaio gli stabilimenti della casa automobilistica rendendola indipendente dall'Industria di Stato. Nel caso si fosse realizzato il centro di Vado (al quale la Fiat rinuncerà nel 1957) sarebbe risultato impossibile realizzarne un altro nel Meridione (a quel punto la produzione avrebbe superato i consumi). Dai dati esaminati emerge una trattativa finora inedita che vede il coinvolgimento della Finsider e di privati stranieri.

Nanomedicine is a new branch of medicine, based on the potentiality and intrinsic properties of nanomaterials. Indeed, the nanomaterials ( i.e. the materials with nano and under micron size) can be suitable to different applications in biomedicine. The nanostructures can be used by taking advantage of their properties (for example superparamagnetic nanoparticles) or functionalized to deliver the drug in a specific target, thanks the ability to cross biological barriers. The size and the shape of 1D-nanostructures (nanotubes and nanowires) have an important role on the cell fate: their morphology plays a key role on the interaction between nanostructure and the biological system. For this reason the 1D nanostructure are interesting for their ability to mime the biological system. An implantable material or device must therefore integrate with the surrounding extracellular matrix (ECM), a complex network of proteins with structural and signaling properties. Innovative techniques allow the generation of complex surface patterns that can resemble the structure of the ECM, such as 1D nanostructures. NWs based on cubic silicon carbide (3C-SiC), either bare (3C-SiC NWs) or surrounded by an amorphous shell (3C-SiC/SiO2 core/shell NWs), and silicon oxycarbide nanowires (SiOxCy NWs) can meet the chemical, mechanical and electrical requirements for tissue engineering and have a strong potential to pave the way for the development of a novel generation of implantable nano-devices. Silicon oxycarbide shows promising physical and chemical properties as elastic modulus, bending strength and hardness, chemical durability superior to conventional silicate glasses in aggressive environments and high temperature stability up to 1300 °C. Moreover, it can easily be engineered through functionalization and decoration with macro-molecules and nanoparticles. Silicon carbide has been extensively studied for applications in harsh conditions, as chemical environment, high electric field and high and low temperature, owing to its high hardness, high thermal conductivity, chemical inertness and high electron mobility. Also, its cubic polytype (3C) is highly biocompatible and hemocompatible, and some prototypes of biomedical applications and biomedical devices have been already realized starting from 3C-SiC thin films. Cubic SiC-based NWs can be used as a biomimetic biomaterial, providing a robust and novel biocompatible biological interface . We cultured in vitro A549 human lung adenocarcinoma epithelial cells and L929 murine fibroblast cells over core/shell SiC/SiO2, SiOxCy and bare 3C-SiC nanowire platforms, and analysed the cytotoxicity, by indirect and direct contact tests, the cell adhesion, and the cell proliferation. These studies showed that all the nanowires are biocompatible according to ISO 10993 standards. We evaluated the blood compatibility through the interaction of the nanowires with platelet rich plasma. The adhesion and activation of platelets on the nanowire bundles, assessed via SEM imaging and soluble P-selectin quantification, indicated that a higher platelet activation is induced by the core/shell structures compared to the bare ones. Further, platelet activation is higher with 3C-SiC/SiO2 NWs and SiOxCyNWs, which therefore appear suitable in view of possible tissue regeneration. On the contrary, bare 3C-SiC NWs show a lower platelet activation and are therefore promising in view of implantable bioelectronics devices, as cardiovascular implantable devices. The NWs properties are suitable to allow the design of a novel subretinal Micro Device (MD). This devices is based on Si NWs and PEDOT:PSS, though the well know principle of the hybrid ordered bulk heterojunction (OBHJ). The aim is to develop a device based on a well-established photovoltaic technology and to adapt this know-how to the prosthetic field. The hybrid OBHJ allows to form a radial p–n junction on a nanowire/organic structure. In addition, the nanowires increase the light absorption by means of light scattering effects: a nanowires based p-n junction increases the light absorption up to the 80%, as previously demonstrated, overcoming the Shockley-Queisser limit of 30 % of a bulk p-n junction. Another interesting employment of these NWs is to design of a SiC based epicardial-interacting patch based on teflon that include SiC nanowires. . Such contact patch can bridge the electric conduction across the cardiac infarct as nanowires can ‘sense’ the direction of the wavefront propagation on the survival cardiac tissue and transmit it to the downstream surivived regions without discontinuity. The SiC NWs are tested in terms of toxicology, biocompatibility and conductance among cardiomyocytes and myofibroblasts.

Tra il 2005 e il 2013 la disciplina italiana sulle crisi d’impresa è stata oggetto di varie riforme, che hanno cercato di rafforzare il concordato preventivo, in particolare nella sua funzione di istituto atto a favorire la continuità di aziende in crisi non irreversibile. Anche tenendo conto degli effetti della crisi economica, tali riforme hanno conseguito l’obiettivo di allargare il ricorso al concordato, in particolare a seguito dell’introduzione di quello “in bianco”, che consente di posticipare la presentazione del piano di risanamento. Le riforme hanno anche contribuito a un lieve miglioramento della continuità aziendale. Ciò nonostante, solo una quota limitata di aziende (circa il 4,5 per cento) sopravvive dopo il concordato, la cui funzione principale è rimasta quella di fornire uno strumento liquidatorio di tipo negoziale, alternativo al fallimento che vede un maggior ruolo degli organi giudiziari. Il ricorso al concordato risulta correlato, oltre che con caratteristiche strutturali dell’impresa (un maggior peso delle immobilizzazioni materiali) e delle relazioni creditizie (un minore peso dei crediti dotati di garanzie reali), anche con la durata temporale delle procedure fallimentari nel tribunale di riferimento.

O trabalho estuda a reclamação como instrumento de controle de precedentes do Supremo Tribunal Federal e do Superior Tribunal de Justiça. O estudo se inicia com a análise do desenvolvimento do instituto desde sua origem correicional, passando por sua constitucionalização até sua previsão no novo Código de Processo Civil, que generaliza seu cabimento como meio de controle da eficácia vinculante dos precedentes. Em seguida, passamos à análise do sistema brasileiro de respeito aos precedentes, fazendo uma breve comparação com países do common law, e concluímos que, no Brasil, o efeito vinculante, assim entendida a força que torna obrigatória a observância da norma extraível das decisões judiciais, só existe se houver previsão expressa na Constituição ou na lei. Também constatamos que o sistema adotou a reclamação a ser ajuizada diretamente perante o STF e o STJ como instrumento processual de controle da observância dessa força vinculante. Verificamos que, além de valorizar a segurança jurídica, a isonomia e a justiça das decisões, a adoção de um sistema de respeito a precedentes no Brasil tem como confessado objetivo a otimização do serviço judiciário e a redução dos processos pendentes nos tribunais superiores. Todavia, a utilização da reclamação como meio de controle da eficácia vinculante dos precedentes vai de encontro àqueles objetivos, pois reatomiza os litígios sem que isso signifique maior respeito à obrigatoriedade dos precedentes.
