Dans cet article, nous étudierons les interactions entre un MEF (maîtresses en formation) et des élèves à propos d’un objet d’enseignement/apprentissage lorsque ce dernier désire construire et mettre en scène des contenus d’enseignement innovants en JSC (jeux sportifs collectifs). Nous serons donc amenés à nous intéresser à son action en ce qui concerne le choix des contenus d’enseignement et des tâches qu’il propose aux élèves par rapport aux orientations innovantes de l’enseignement des JSC et l’étude des processus de régulation lors des interactions didactiques qu’il met en oeuvre dans la gestion des situations d’enseignement/apprentissage

El objeto de esta tesis es el análisis de la autorregulación regulada, es decir, el análisis de las normas y de los controles privados que poseen relevancia para el Derecho público. A pesar de su aparente desvinculación con el Derecho Administrativo, la autorregulación es una tendencia que se percibe cada vez con mayor nitidez; es, claramente, un fenómeno que se asocia principalmente al uso de nuevas tecnologías, pero que desborda con mucho este ámbito. La autorregulación es una noción directamente relacionada con la transformación de las formas de gobernanza impulsada desde la Unión Europea; es una nueva técnica o instrumento que las normas jurídico-públicas, a través de numerosas remisiones a la autorregulación, ponen en manos de la administración para gestionar los fines que ésta tiene encomendados. Existe, sin embargo una clara disociación entre las esperanzas puestas en la autorregulación y la falta de respuestas que ofrece el derecho positivo. Un análisis exhaustivo de la jurisprudencia y de la legislación que utilizan la voz "autorregulación" me ha llegado a concluir que este vocablo es en nuestro ordenamiento, un "término sin concepto". En esta fuentes se asimila la "autorregulación" con la capacidad de autonormación de un sujeto. Es el derecho comunitario el que la autorregulación como una alternativa o un complemento a la desreglamentación estatal y como una manifestación de un traslado de funciones y responsabilidades públicas a la sociedad. Por influencia del Derecho comunitario, el legislador pretende, a través de la autorregulación, alcanzar dos finalidades aparentemente contradictorias: (a) facilitar la función de garante que tiene atribuida el Estado, mediante una intervención más extensa i más intensa en las actividades privadas; (b) hacer efectivo los objetos propuestos con la desregulación, mediante una contención del ejercicio de la potestad reglamentaria y una disminución de los controles, preventivos o represivos, realizados directamente por la Administración. Para salvar esta contradicción, es necesario que el legislador establezca una regulación adecuada de la autorregulación. Dicha regulación es, hoy por hoy, manifiestamente insuficiente. Deberían fijarse con carácter general -y no sólo puntualmente y por sectores- las medidas adecuadas para el fomento de la autorregulación; los efectos públicos que ésta posee en cada caso y, significativamente, las garantías y controles necesarios para contrarrestar tales efectos. Esto es, los principios que rigen la actividad administrativa deberían ser aplicados también a la autorregulación en aquellos casos en los que sus diversas manifestaciones poseen efectos similares a los que son propios de los reglamentos, las inspecciones o las sanciones administrativas. Esta propuesta deja abierta la cuestión acerca de la incidencia recíproca de ambas técnicas; esto es , la incidencia de la regulación pública en la autorregulación de origen privado y, a la inversa, el impacto del desarrollo de la autorregulación regulada en el ejercicio de las potestades reglamentaria, autorizatoria, y sancionadora de la Administración. Esta cuestión sólo puede ser contestada hoy caso por caso, de modo que la respuesta es distinta si se analiza una norma técnica, un código ético, un manual de buenas prácticas, una certificación privada del cumplimiento de normas técnicas, el ejercicio de la potestad disciplinaria privada o el arbritaje. De lo que no cabe duda alguna es que ni la autorregulación es sólo una actividad de interés privado, ni la regulación pública conserva hoy sus rasgos tradicionales.

Differences in the expression of cell surface proteins between a normal prostate epithelial (1542-NP2TX) and a prostate cancer cell line (1542-CP3TX) derived from the same patient were investigated. A combination of affinity chromatographic purification of biotin-tagged surface proteins with mass spectrometry analysis identified 26 integral membrane proteins and 14 peripheral surface proteins. The findings confirm earlier reports of altered expression in prostate cancer for several cell surface proteins, including ALCAM/CD166, the Ephrin type A receptor, EGFR and the prostaglandin F2 receptor regulatory protein. In addition, several novel findings of differential expression were made, including the voltage-dependent anion selective channel proteins Porin 1 and 2, ecto-5'-nucleotidase (CD73) and Scavenger receptor B1. Cell surface protein expression changed both qualitatively and quantitatively when the cells were grown in the presence of either or both interferon INFalpha and INFgamma. Costimulation with type I and II interferons had additive or synergistic effects on the membrane density of several, mainly peripherally attached surface proteins. Concerted upregulation of surface exposed antigens may be of benefit in immuno-adjuvant-based treatment of interferon-responsive prostate cancer. In conclusion, this study demonstrates that differences in the expression of membrane proteins between normal and prostate cancer cells are reproducibly detectable following vectorial labelling with biotin, and that detailed analysis of extracellular-induced surface changes can be achieved by combining surface-specific labelling with high-resolution two-dimensional gel electrophoresis and mass spectrometry.

Ingestion of probiotics can be recommended as a preventative approach to maintaining intestinal microflora balance and thereby enhance 'well-being'. Undoubtedly, probiotic bacteria will vary in their efficacy. The literature indicates positive results in over 50 human trials with prevention/treatment of infections the most frequently reported. In theory increased levels of probiotics may induce a 'barrier' influence against common pathogens. Mechanisms of effect are likely to include the excretion of acids (lactate, acetate), competition for nutrients and gut receptor sites, immuno-modulation and the formation of specific antimicrobial agents. An alternative, or additional, approach is the prebiotic concept. This takes the view that probiotics are present indigenous to the gut and that a rational approach towards increasing their numbers would be to consume food ingredients (carbohydrates) that have a selective metabolism in the lower gut. A prebiotic is 'a nondigestible food ingredient that beneficially affects the host by selectively stimulating the growth and/or activity of one or a limited number of bacteria in the colon that can improve the host health.' In particular, the ingestion of fructo-oligosaccharides, galacto-oligosaccharides, and lactulose has shown to stimulate bifidobacteria in the lower gut.

The aim of this study was to determine the cost effectiveness of influenza vaccination for healthy people aged 65-74 years living in the UK. People without risk factors for influenza (chronic heart, lung or renal disease, diabetic, immuno-suppressed or those living in an institution) were identified from 20 general practitioner (GP) practices in Liverpool in September 1999. 729/5875 (12.4%) eligible individuals were recruited and randomised to receive either influenza vaccine or placebo (ratio 3: 1)! with all participants receiving 23-valent-pneumococcal polysaccharide vaccine unless already administered. The primary analysis was the frequency of influenza as recorded by a GP diagnosis of pneumonia or influenza like illness. In 2000, the UK vaccination policy was changed with influenza vaccine becoming available. for all people aged 65 years and over irrespective of risk. As a consequence of this policy change. the study had to be fundamentally restructured and only results obtained over a one rather than the originally planned two-year randomised controlled trial framework were used. Results from 1999/2000 demonstrated no significant difference between groups for the primary outcome (relative risk 0.8, 95%, CI 0.16-4.1). In addition. there were no deaths or hospitalisations for influenza associated respiratory illness in either group. The subsequent analysis. using both national and local sources of evidence, estimated the following cost effectiveness indicators: (1) incremental NHS cost per GP consultation avoided = pound2000; (2) incremental NHS cost per hospital admission avoided = pound61,000: (3) incremental NHS cost per death avoided = pound1.900.000 and (4) incremental NHS cost per QALY gained = pound304,000. Experiments are presented which show that Botrytis cinerea, the cause of gray mould disease, is often present in symptomless lettuce plants as a systemic, endophytic, infection which may arise from seed. The fungus was isolated on selective media from surface sterilized sections of roots, stem pieces and leaf discs from symptomless plants grown in a conventional glasshouse and in a spore-free air-flow provided by an isolation propagator. The presence of B. cinerea was confirmed by immuno-labelling the tissues with the Botrytis-specific monoclonal antibody BC-12.CA4. As plants grew, infection spread from the roots to stems and leaves. Surface sterilization of seeds reduced the number of infected symptomless plants. Artificial infection of seedlings with dry conidia increased the rate of infection in some experiments. Selected isolates were genetically finger-printed using microsatellite loci. This confirmed systemic spread of the inoculating isolates but showed that other isolates were also present and that single plants hosted multiple isolates. This shows that B. cinerea commonly grows in lettuce plants as an endophyte, as has already been shown for Primula. If true for other hosts, the endophytic phase may be as important a component of the species population as the aggressive necrotrophic phase.

A considerable amount of evidence has accumulated to support the view that the very long chain omega 3 fatty acids (eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA)) have beneficial cardiovascular and anti-inflammatory properties and that levels of their consumption are insufficient in most Western diets. More recently, attention has been given to the possibility that the precursor omega-3 PUFA, alpha linolenic acid (ALNA), may share some of the beneficial actions of EPA/DHA on human health. Further research into the metabolism and physiological actions of ALNA, and comparisons with EPA/DHA, is needed before conclusions regarding the optimal amounts and types of omega-3 PUFA for human health can be defined. Conjugated linoleic acid (CLA), which arises as a metabolic by-product of rumen hydrogenation and which is found in foods of animal origin, has been proposed to possess potent health promoting properties, but much of this research has been conducted in experimental animals. There is an urgent need for complementary studies in human volunteers, to confirm the putative anti-carcinogenic, anti-atherogenic, anti-lipogenic and immuno-suppressive properties of CLA.

There is interest in the enrichment of poultry meat with long-chain n-3 polyunsaturated fatty acids in order to increase the consumption of these fatty acids by humans. However, there is concern that high levels of n-3 polyunsaturated fatty acids may have detrimental effects on immune function in chickens. The effect of feeding increasing levels of fish oil (FO) on immune function was investigated in broiler chickens. Three-week-old broilers were fed 1 of 4 wheat-soybean basal diets that contained 0, 30, 50, or 60 g/kg of FO until slaughter. At slaughter, samples of blood, bursa of Fabricius, spleen, and thymus were collected from each bird. A range of immune parameters, including immune tissue weight, immuno-phenotyping, phagocytosis, and cell proliferation, were assessed. The pattern of fatty acid incorporation reflected the fatty acid composition of the diet. The FO did not affect the weight of the spleen, but it did increase thymus weight when fed at 50 g/kg (P < 0.001). Fish oil also lowered bursal weights when fed at 50 or 60 g/kg (P < 0.001). There was no significant effect of FO on immune cell phenotypes in the spleen, thymus, bursa, or blood. Feeding 60 g/kg of FO significantly decreased the percentage of monocytes engaged in phagocytosis, but it increased their mean fluorescence intensity relative to that of broilers fed 50 g/kg of FO. Lymphocyte proliferation was significantly decreased after feeding broiler chickens diets rich in FO when expressed as division index or proliferation index, although there was no significant effect of FO on the percentage of divided cells. In conclusion, dietary n-3 polyunsaturated fatty acids decrease phagocytosis and lymphocyte proliferation in broiler chickens, highlighting the need for the poultry industry to consider the health status of poultry when poultry meat is being enriched with FO.

Major outer membrane proteins (MOMPs) of Gram negative bacteria are one of the most intensively studied membrane proteins. MOMPs are essential for maintaining the structural integrity of bacterial outer membranes and in adaptation of parasites to their hosts. There is evidence to suggest a role for purified MOMP from Chlamydophila pneumoniae and corresponding MOMP-derived peptides in immune-modulation, leading to a reduced atherosclerotic phenotype in apoE−/− mice via a characteristic dampening of MHC class II activity. The work reported herein tests this hypothesis by employing a combination of homology modelling and docking to examine the detailed molecular interactions that may be responsible. A three-dimensional homology model of the C. pneumoniae MOMP was constructed based on the 14 transmembrane β-barrel crystal structure of the fatty acid transporter from Escherichia coli, which provides a plausible transport mechanism for MOMP. Ligand docking experiments were used to provide details of the possible molecular interactions driving the binding of MOMP-derived peptides to MHC class II alleles known to be strongly associated with inflammation. The docking experiments were corroborated by predictions from conventional immuno-informatic algorithms. This work supports further the use of MOMP in C. pneumoniae as a possible vaccine target and the role of MOMP-derived peptides as vaccine candidates for immune-therapy in chronic inflammation that can result in cardiovascular events.

Phosphorylation of the serine residues in estrogen receptor (ER) α is important in transcriptional activation. Hence, methods to detect such posttranslational modifi cation events are valuable. We describe, in detail, the analysis of the phosphorylated ERα by electrophoretic separation of proteins and subsequent immuno-blotting techniques. In particular, phosphorylation of the ERα is one possible outcome of activation of the putative membrane estrogen receptor (mER), GPR30. Hence, phosphorylation represents a cross talk event between GPR30 and ERα and may be important in estrogen-regulated physiology.

Objectives Little information is available on the molecular events that occur during graft incorporation over time. The calvarial bone (Cb) grafts have been reported to produce greater responses compared with other donor regions in maxillofacial reconstructions, but the scientific evidences for this are still lacking. The objectives of this study are (1) to study the morphological pattern of Cb onlay bone grafts and compare them with the biological events through immunohistochemical responses and (2) to establish the effects of perforations in maintaining the volume and bone density of the receptor bed. Material and methods Sixty New Zealand White rabbits were submitted to Cb onlay bone grafts on the mandible. In 30 rabbits, the receptor bed was perforated (perforated group), while for the remaining animals the bed was kept intact (non-perforated group). Six animals from each group were sacrificed at 5, 7, 10, 20 and 60 days after surgery. Histological sections from the grafted area were prepared for immunohistochemical and histological analyses. Immuno-labeling was found for proteins Osteoprotegerin (OPG), receptor activator of nuclear factor-kappa beta ligand (RANKL), alkaline phosphatase (ALP), osteopontin (OPN), vascular endothelial growth factor (VEGF), tartrate-resistant acid phosphatase (TRAP), Type I collagen (COL I) and osteocalcin (OC). The tomography examination [computerized tomography (CT) scan] was conducted just after surgery and at the sacrifice. Results The histological findings revealed that the perforations contributed to higher bone deposition during the initial stages at the graft-receptor bed interface, accelerating the graft incorporation process. The results of the CT scan showed lower resorption for the perforated group (P < 0.05), and both groups showed high bone density rates at 60 days. This set of evidences is corroborated by the immunohistochemical outcomes indicating that proteins associated with revascularization and osteogenesis (VEGF, OPN, TRAP and ALP) were found in higher levels in the perforated group. Conclusions These findings indicate that the bone volume of calvarial grafts is better maintained when the receptor bed is perforated, probably resulting from more effective graft revascularization and greater bone deposition. The process of bone resorption peaked between 20 and 60 days post-operatively in both groups although significantly less in the perforated group. Peroxiredoxins are receiving increasing attention as defenders against oxidative damage and sensors of hydrogen peroxide-mediated signaling events. In the yeast Saccharomyces cerevisiae, deletion of one or more isoforms of the peroxiredoxins is not lethal but compromises genome stability by mechanisms that remain under scrutiny. Here, we show that cytosolic peroxiredoxin-null cells (tsa1 Delta tsa2 Delta) are more resistant to hydrogen peroxide than wildtype (WT) cells and consume it faster under fermentative conditions. Also, tsa1 Delta tsa2 Delta cells produced higher yields of the 1-hydroxyethyl radical from oxidation of the glucose metabolite ethanol, as proved by spin-trapping experiments. A major role for Fenton chemistry in radical formation was excluded by comparing WT and tsa1 Delta tsa2 Delta cells with respect to their levels of total and chelatable metal ions and of radical produced in the presence of chelators. The main route for 1-hydroxyethyl radical formation was ascribed to the peroxidase activity of Cu, Zn-superoxide dismutase (Sod1), whose expression and activity increased similar to 5- and 2-fold, respectively, in tsa1 Delta tsa2 Delta compared with WT cells. Accordingly, overexpression of human Sod1 in WT yeasts led to increased 1-hydroxyethyl radical production. Relevantly, tsa1 Delta tsa2 Delta cells challenged with hydrogen peroxide contained higher levels of DNA-derived radicals and adducts as monitored by immuno-spin trapping and incorporation of (14)C from glucose into DNA, respectively. The results indicate that part of hydrogen peroxide consumption by tsa1 Delta tsa2 Delta cells is mediated by induced Sod1, which oxidizes ethanol to the 1-hydroxyethyl radical, which, in turn, leads to increased DNA damage. Overall, our studies provide a pathway to account for the hypermutability of peroxiredoxin-null strains.

O Brasil é um dos principais atores do novo ambiente econômico mundial, mas para permanecer atrativo, deve fazer progressos significativos no campo das Tecnologias de Informação e da Comunicação (TIC). Assim, um serviço de internet banda larga é primordial para o desenvolvimento de regiões distantes, geralmente de baixa renda e que não têm acesso a instalações de telecomunicações, nem aos serviços de internet banda larga. De fato, a penetração da banda larga no Brasil apresenta situação inferior à média mundial e boa parte da população ainda está excluída do mundo digital por causa das desigualdades sociais e as dimensões continentais do país. Para preencher esta lacuna, o governo federal lançou, em 2010, o Programa Nacional de Banda Larga (PNBL). No entanto, a penetração de banda larga permanece em níveis baixos, mesmo em grandes centros urbanos e com uma distribuição regional desigual, especialmente quando comparados internacionalmente. As concessionárias não estão desempenhando o seu papel de universalização dos serviços de banda larga e as pequenas e médias operadoras podem desempenhar um papel fundamental na promoção desse objetivo. Esse trabalho tem o propósito de investigar se é possível conciliar o modelo vigente de telecomunicações regido pela Lei Geral das Telecomunicações (LGT) com a universalização dos serviços de banda larga, desde que apoiado num mercado competitivo e menos concentrado, com pequenas e médias operadoras. Como verificação, foi feito um trabalho empírico com base em entrevistas semiestruturadas com profissionais de telecomunicações do setor público e privado, assim como uma análise de padrão de codificação dessas entrevistas. Os resultados mostram, porém, que o ambiente regulatório atual pode gerar incentivos à operação de qualidade nas áreas remotas e de baixa renda, mas não de forma generalizada. A universalização requer uma mudança estrutural do modelo e é a política pública federal que deve dar as diretrizes, com objetivos claros. Tampouco há modelo ideal de regulação, mas um entorno regulatório adequado pode ser motor de desenvolvimento do setor. O estabelecimento de compromissos entre os atores também é fundamental, assim como a decisão política na origem, dando suporte para o planejamento e projetos de longo prazo. E é a partir dessa decisão que se desenvolvem as medidas regulatórias de competição e fiscalização que vão garantir a execução do que foi previsto e desenhado.

Blood and bone marrow samples were taken from 112 Didelphis spp., collected between March 2005 and February 2006, from urban and peri-urban areas of Bauru, São Paulo State, Brazil, to evaluate the hypothesis that these animals might constitute a reservoir of Leishmania spp. Anti-Leishman ia ssp. antibodies were screened in the serum samples using an enzyme-linked immuno-sorbent assay (ELISA) and the polymerase chain reaction (PCR). PCR was performed on fragments of DNA samples from Leishmania spp. using primers 13A and 13B, and showed a positive outcome in 91.6% of the 112 samples tested. of the 107 samples analyzed by ELISA, 71 % were positive. Evidence of epidemiological risk factors such as a circulating parasite and freely moving vectors suggests that Didelphis spp. may participate in the transmission cycle of Leishmania spp. in Bauru.
