Os sucroésteres são empregados como surfactantes, tensoativos, substitutos de gordura e antibióticos. Este trabalho tem como objetivo sintetizar um novo sucroéster derivado do ácido picolínico (ácido 2-piridinocarboxílico), o picolinato de sacarose, e estudar sua atividade antibacteriana in vitrosobre a bactéria Gram-negativa patogênica Escherichia coli. A síntese do picolinato de sacarose foi processada a partir da transesterificação da sacarose com picolinato de metila em condição anidra à 80oC, utilizando dimetil sulfóxido (DMSO) como solvente e K2CO3 como catalisador. A separação dos isômeros formados foi realizada por HPLC no modo semipreparativo e cinco frações cromatográficas foram coletadas e aplicadas em testes de atividade antibacteriana, por disco-difusão em meio sólido, nas concentrações de 150; 300; 450; 600; 750 e 900 μg/mL. As frações 2, 4 e 6 foram ativas contra E.coli. e a fração 4 (900 μg/mL) foi a mais eficiente, sendo selecionada para testes em sinergia com EDTA nas concentrações de 250, 500 e 750 μg/mL, com melhor resultado quando empregado EDTA em 750 μg/mL. Neste caso, os discos apresentaram halos de inibição de crescimento igual ao da Tetraciclina (30 μg/mL) e superior aos produzidos pelos discos com Gentamicina (10 μg/mL). A fração 4 foi caracterizada por FTIR e espectrometria de massas (ESI-MS) e os resultados indicam que se trata sucroéster monossubstituído. Palavras-chave:Sucroquímica. Sucroéster. Sacarose. ABSTRACT Study of antimicrobial activity of sucrose picolinate against Escherichia coli Sucrose esters are generally used as surfactants, fat substitutes and antibiotics. The aim of the present study was to synthesize new sucrose esters derived from picolinic acid (2-pyridine carboxylic acid) and study in vitro antimicrobial activity on the Gramnegative pathogenic bacterium Escherichia coli. The synthesis of sucrose picolinate was performed through the transesterification of sucrose with methyl picolinate under anhydrous conditions at 80 oC using dimethyl sulfoxide (DMSO) as the solvent and K2CO3as the catalyst. The separation of the formed isomers was performed by HPLC in a semi-preparative chromatograph system. Five fractions were collected and applied to a disc-diffusion antibiogram in solidmedium tests at concentrations of 150, 300, 450, 600, 750 and 900 μg/mL. Fractions 2, 4 and 6 were active against E. coli. Fraction 4 (900 μg/mL) was the most efficient and was selected for the determination of antimicrobial activity in synergistic tests with EDTA at concentrations of 250, 500 and 750 μg/mL. The best result was obtained with 750 μg/mL of EDTA. Fraction 4 was characterized as a monosubstituted sucrose ester by FTIR and mass spectrometry (ESI-MS). Keywords: Sucrochemistry. Sucrose. Laccases (benzendiol:oxygen oxidoreductases; EC 1.10.3.2) catalyze the oxidation of a broad range of substrates, such as polyphenols, dyes and pollutants, and thus these enzymes are widely applied in industrial, biotechnological and environmental fields. In order to improve their biotechnological applications, a deep knowledge of structural factors involved in controlling their activity, in various experimental conditions and on different substrates, is required. In the present study, a laccase from the mushroom Rigidoporus lignosus was kinetically characterized. In particular, the stability, the effects of pH, ionic strength and fluoride ion concentration on the kinetic parameters were investigated, using three di-hydroxy-benzene isomers (1,2-dihydroxy-benzene, 1,3-dihydroxy-benzene and 1,4-dihydroxy-benzene) as substrates. The catalytic constant values of the laccase showed a bell-shaped pH profile, with the same optimum pH and pK(a) values for all tested substrates. This behavior appears to be due to the presence of an ionizable residue in the enzyme active site. To identify this residue, the enzyme was derivatized with diethylpyrocarbonate to modify accessible histidine residues, which, according to structural data, are present in the active site of this enzyme. The kinetic behavior of the derivatized laccase was compared with that of the native enzyme and the derivatized residues were identified by mass spectrometry. Mass spectrometry and kinetic results suggest the main role of His-457 in the control of the catalytic activity of laccase from R. lignosus. (C) 2013 Elsevier B.V. All rights reserved.

Optics consists in the study of interaction of light with physical systems. The human vision is a product of the interaction of light with the eye (a very peculiar physical system). Here we present a basic study of the relationship between the optics and human vision, including: - The fundaments and physicals properties who characterize the light and the colors and the characteristics of the incidence mediums. - The basics laws of geometrical optics, based in the rectilinear propagation of light in the form of a light ray, in the independence of light rays and in the principle of reversibility of the light beams. This principle is present in the process of image formations in lenses and mirrors and applied in the study of image formation in the human eye. - The refraction and reflection laws and types of lenses, who permits the construction of optics devices for the study of physical universe, and the appliances to correct vision diseases. - Presents the human vision process as consisting in the reception of light (electromagnetic radiation in the zone of wavelength visible to us) through the eye and the sending of information obtained by the retina to the brain where it is interpreted. The vision involves a biophysical relation between the light and the biological structure of the eye who is constituted by cornea, iris, crystalline and retina. Analyzes is made of how some parts of the eye performs a function in the reception and sending of information of the images to the brain

This study presents the results from a qualitative resource, based on research-action methodology, which examined the innovate teaching practices implementations in the Calculus I during 2011 year. The analysis of collected data from three used sources – an initial questionnaire, an exploratory-investigative classroom, and an interview with some students at the end of the second semester – reveals that the students had appropriated of the technological recourses, using it as a tool to look for the knowledge. The investigative activities with the use of information technologies made the use of multiple representations in solving mathematical tasks, making the transition of numerical, algebraic and geometrical results possible for the students when they have looking for validation of their hypotheses and conjectures during mathematical problems solving. This works helped in the insertion of new practices in the discipline, and their results validate the proposal presented by the teacher, which is the discipline of Calculus use, whose character is strongly linked to the training content of the student, as a discipline whose can contributed to the pedagogical formation of the graduation student, leading him to know the Mathematical Education possibilities, specially the Mathematical Investigation - Solving Problems, in the outlook of dialogued classes where the teacher assumes the facilitator role and where the students become actives in their pursuit of knowledge

The Bonito oil field, located on southwest of Campos Basin-RJ, has been explored since 1982. The main reservoir is composed by calcarenites of Quissamã Formation (Macaé Group) from Albian, but two other carbonate levels are present on the field, firsts is Coqueiros Formation (Aptian) and the second Siri Member (Oligo-Miocene). In this context and considering that carbonates reservoirs are a challenge for exploratory geoscientists, since the difficulty on recognize the effective reservoir distribution. This work aim to characterize the geophysical/geological facies based on seismic attributes responses, related to reservoir geometrical distribution, for the tree carbonates intervals on Bonito oil Field. A tree dimensional interpretation of the levels has been developed, based on well cross correlation and a 3D seismic interpretation, resulting on the stratigraphic and structural framework of the field, which showed a NE-SW fault trend controlling the Aptian carbonates reservoirs, and halocnetics structures showing a structural trap on Albian carbonates reservoirs. The definition of the structural/ stratigraphic framework possibly the seismic attributes calculations over the reservoir intervals. To select the best response in comparison with the reservoir distribution, obtained by seismic interpretation, the attributes response were compared with isopachs maps of each carbonate stratigraphic level. The attributes Maximum Amplitude, Maximum Magnitude and Rms Amplitude showed a good answer to reservoir distribution. The Rms Amplitude also showed a good correlation with physical rock properties, like RHOB bulk density, for the Albian and Aptian carbonates, as consequence it is possible make a characterization of reservoir distribution based on seismic attribute answer
