Cardiovascular diseases are nowadays the first cause of mortality worldwide, causing around the 30% of global deaths each year. The risk of suffering from cardiovascular illnesses is strongly related to some factors such as hypertension, high cholesterol levels, diabetes, obesity The combination of these different risk factors is known as metabolic syndrome and it is considered a pandemic due to the high prevalence worldwide. The pathology of the disorders implies a combined cardiovascular therapy with drugs which have different targets and mechanisms of action, to regulate each factor separately. The simultaneous analysis of these drugs turns interesting but it is a complex task since the determination of multiple substances with different physicochemical properties and physiological behavior is always a challenge for the analytical chemist. The complexity of the biological matrices and the difference in the expected concentrations of some analytes require the development of extremely sensitive and selective determination methods. The aim of this work is to fill the gap existing in this field of the drug analysis, developing analytical methods capable of quantifying the different drugs prescribed in combined cardiovascular therapy simultaneously. Liquid chromatography andem mass spectrometry (LCMS/MS) has been the technique of choice throughout the main part of this work, due to the high sensitivity and selectivity requirements.

The purpose of this study was to determine the actual direction of littoral transport along the north shore of Santa Rosa Island in the vicinity of Pensacola Beach, Florida. To accomplish this objective the sand tracer method was used for the study. Visual observations and instrument recordings of the environment factors were also made during the tracing operations. The investigation covered a time span from September 14, 1976 to March 12, 1977. (PDF contains 68 pages.)

[EN]Conjugated linoleic acid (CLA) has many potential healthful properties, and beef is naturally enriched with CLA. Simple and rapid methods to measure total CLA were investigated to enable sorting of beef carcasses with potential enhanced economic value. Direct alcohol extraction combined with measuring absorbance was simple, accurate and perhaps the most viable method for rapid carcass sorting compared to methods using saponification or methylation followed by extraction.

The use of growth layers in teeth as an indicator of age in odnotocetes and pinnipeds was suggested by Laws (1954) and since then the method has been used extensively in both marine and non-marine mammals. Dentinal growth layers are groups (growth layer groups) of repetitive alternating bands which in cross-section are similar to growth rings in trees. The most commonly used methods for counting growth layer groups (GLGs) are by undecalcified longitudinal thin sections (150 um) or decalcified and stained thin sections (10-30 um). In longitudinal sections viewed with light microscopy, GLGs appear as opaque and translucent cones nestled one inside another, with the oldest dentine Iying adjacent to the enamel, and the newest layer borderinq the pulp cavity.

We investigated within- and between-reader precision in estimating age for northern offshore spotted dolphins and possible effects on precision from the sex and age-class of specimens. Age was estimated from patterns of growth layer groups i n the dentine and cementum of the dolphins' teeth. Each specimen was aged at least three times by each of two persons. Two data samples were studied. The first comprised 800 of each sex from animals collected during 1973-78. The second included 45 females collected during 1981. There were significant, generally downward trends through time in the estimates from multiple readings of the 1973-78 data. These trends were slight, and age distributions from last readings and mean estimates per specimen appeared to be homogeneous. The largest factor affecting precision in the 1973-78 data set was between-reader variation. In light of the relatively high within-reader precision (trends considered), the consistent between-reader differences suggest a problem of accuracy rather than precision for this series. Within-reader coefficients of variation averaged approximately 7% and 11%. Pooling the data resulted i n an average coefficient of variation near 16%. Within- and between-reader precision were higher for the 1981 sample, and the data homogeneous over both factors. CVs averaged near 5% and 6% for the two readers. These results point to further refinements in reading the 1981 series. Properties of the 1981 sample may be partly responsible for greater precision: by chance there were proportionately fewer older dolphins included, and preparation and selection criteria were probably more stringent. One of existing strategies to engineer active antibody is to link VH and VL domains via a linker peptide. How the composition, length, and conformation of the linker affect antibody activity, however, remains poorly understood. In this study, a dual approach that coordinates molecule modeling, biological measurements, and affinity evaluation was developed to quantify the binding activity of a novel stable miniaturized anti-CD20 antibody or singlechain fragment variable (scFv) with a linker peptide. Upon computer-guided homology modeling, distance geometry analysis, and molecular superimposition and optimization, three new linker peptides PT1, PT2, and PT3 with respective 7, 10, and 15 residues were proposed and three engineered antibodies were then constructed by linking the cloned VH and VL domains and fusing to a derivative of human IgG1. The binding stability and activity of scFv-Fc chimera to CD20 antigen was quantified using a micropipette adhesion frequency assay and a Scatchard analysis. Our data indicated that the binding affinity was similar for the chimera with PT2 or PT3 and ~24-fold higher than that for the chimera with PT1, supporting theoretical predictions in molecular modeling. These results further the understanding in the impact of linker peptide on antibody structure and activity.

This thesis presents two different forms of the Born approximations for acoustic and elastic wavefields and discusses their application to the inversion of seismic data. The Born approximation is valid for small amplitude heterogeneities superimposed over a slowly varying background. The first method is related to frequency-wavenumber migration methods. It is shown to properly recover two independent acoustic parameters within the bandpass of the source time function of the experiment for contrasts of about 5 percent from data generated using an exact theory for flat interfaces. The independent determination of two parameters is shown to depend on the angle coverage of the medium. For surface data, the impedance profile is well recovered.

The second method explored is mathematically similar to iterative tomographic methods recently introduced in the geophysical literature. Its basis is an integral relation between the scattered wavefield and the medium parameters obtained after applying a far-field approximation to the first-order Born approximation. The Davidon-Fletcher-Powell algorithm is used since it converges faster than the steepest descent method. It consists essentially of successive backprojections of the recorded wavefield, with angular and propagation weighing coefficients for density and bulk modulus. After each backprojection, the forward problem is computed and the residual evaluated. Each backprojection is similar to a before-stack Kirchhoff migration and is therefore readily applicable to seismic data. Several examples of reconstruction for simple point scatterer models are performed. Recovery of the amplitudes of the anomalies are improved with successive iterations. Iterations also improve the sharpness of the images.

The elastic Born approximation, with the addition of a far-field approximation is shown to correspond physically to a sum of WKBJ-asymptotic scattered rays. Four types of scattered rays enter in the sum, corresponding to P-P, P-S, S-P and S-S pairs of incident-scattered rays. Incident rays propagate in the background medium, interacting only once with the scatterers. Scattered rays propagate as if in the background medium, with no interaction with the scatterers. An example of P-wave impedance inversion is performed on a VSP data set consisting of three offsets recorded in two wells.

The content of free formaldehyde (FA) in minced fish muscle was measured by the following procedure: A mixture of minced fish muscle and water was deproteinized by means of the Carrez reagent. The FA content of the filtrate was determined reflectometrically using the Reflectoquant test strips and the RQflex. The results agreed well with the colorimetrically (Nash test) measured FA content. Der Gehalt an freiem Formaldehyd (FA)in Fischerzeugnissen wurde mit folgender Methode bestimmt: Zerkleinertes Fischfleisch wurde mit Wasser homogenisiert und mit Carrez-Reagenz enteiweißt. Der Formaldehydgehalt des Filtrates wurde reflektometrisch unter Verwendung von Reflectoquant-Teststätbchen und des RQflex ermittelt. Die Ergebnisse stimmten gut mit kolorimetrisch (Nash Test) gemessenen FA-Gehalten überein.

A Fourier analysis method is used to accurately determine not only the absolute phase but also the temporal-pulse phase of an isolated few-cycle (chirped) laser pulse. This method is independent of the pulse shape and can fully characterize the light wave even though only a few samples per optical cycle are available. It paves the way for investigating the absolute phase-dependent extreme nonlinear optics, and the evolutions of the absolute phase and the temporal-pulse phase of few-cycle laser pulses.

Experimental stocking density of Macrobrachium rosenbergii in larval rearing was conducted in A.G. Aqua Hatchery, Chakaria, Bangladesh to study the effect of different stocking densities on growth, survival rate and diseases stress under hatchery condition. The research work was conducted using six cemented rectangular tanks having 3m3 capacity (1.5mX2mX1m) each. Stocking density were maintained in three experimental setup as 200, 150 and 100ind/L of the T1, T2 and T3 respectively with one replicate each. The larvae were fed with Artemia nauplii, Custard, Maxima and brine shrimp flakes. Water quality was maintained by exchanging 20-30% (12ppt saline water) daily. During the study period, temperature, pH, DO, salinity, nitrite-nitrogen, ammonia and alkalinity were maintained from 28.5-31.5ºC, 7.5-7.8, 5.8-5.9mg/L, 12-13ppt, 0.14-0.2 mg/L, 0.22-0.3mg/L, and 140-160mg/L respectively. The growth rates of larvae at 11th stage were recorded in terms of body length 0.115, 0.136, and 0.169 mm/day whereas body weight were observed 0.000115, 0.000180, and 0.000240g/day. The survival rate of larvae were found 21.8%, 30.4% and 51.3% in treatments T1, T2 and T3 respectively. PL was obtained as 43, 45, and 51PL/L and days required of 41, 38 and 34 days in stocking density of 200, 150, and 100ind/L respectively. It was found that the minimum of 34 days was required to attain the PL (12th stage) using the stocking density of 100 individuals/L. Cannibalism, Zoothamnium, Exuvia Entrapment Disease (EED), and Bacterial Necrosis (BN) were found to be the threat to the commercial hatchery operation that might responsible for potential larval damages which can be reduced by lowering the stocking densities in larval rearing tank that also increased the survival and growth rate.
