Partindo da ideia de ʻDesign Globalʼ e tendo como pano de fundo a emergência e consolidação de novos paradigmas de interação baseados numa relação mais direta entre o corpo e os conteúdos digitais, a presente tese aborda a questão da (re)qualificação da experiência humana tecnologicamente mediada do ponto de vista do Design, designadamente do Design de Interação e do Design da Experiência. Neste contexto, a noção de ʻexperiência tecnologicamente (i)mediadaʼ representa uma ação humana que é, simultaneamente, mediada (do ponto de vista técnico) e direta ou imediada (do ponto de vista da percepção) entre o ser e a sua ação no mundo. Dado o objecto de estudo ser a experiência real que os objetos e dispositivos técnicos convocam designadamente na vida quotidiana, o nosso estudo implica toda a dinâmica holística da relação singular – racional, emocional, estética e projetiva – do ser com o seu mundo. Desse modo, defende-se a necessidade de enquadrar a raiz do pensamento e ação em Design numa hermenêutica fenomenológica e ontológica do projeto. Esta tese visa desta forma contribuir para apuramento e consolidação de uma praxis transdisciplinar para os designers e restantes intervenientes do processo do Design, cujo desígnio é a (re)qualificação da experiência que os artefactos e dispositivos técnicos convocam, assim como na apropriação e vínculo que a pessoa estabelece com estes. O resultado deste estudo sintetiza-se nos seguintes objetivos: 1. no desenvolvimento de conceitos operativos que estruturem o pensamento de uma Filosofia da Ação em Design com base numa prática que designamos por “design centrado-no-ser”; 2. na reflexão da experiência tecnológica através do conceito do estado de fluxo de Mihaly Csikszentmihalyi, enquadrando essa tipologia de experiências no contexto de uma vida, ou seja, no projeto de um eu; 3. na problematização da (re)qualificação da experiência técnica numa perspetiva de vida mais alargada que envolve o princípio de simetria entre ação ética e ação estética no pensamento e prática em Design; 4. na definição de um quadro de ações que manifesta a aplicabilidade dos conceitos desenvolvidos no ensino, investigação e prática do Design.

A engenharia de tecidos é um domínio tecnológico emergente em rápido desenvolvimento que se destina a produzir substitutos viáveis para a restauração, manutenção ou melhoria da função dos tecidos ou órgãos humanos. Uma das estratégias mais predominantes em engenharia de tecidos envolve crescimento celular sobre matrizes de suporte (scaffolds), biocompatíveis e biodegradáveis. Estas matrizes devem possuir não só elevadas propriedades mecânicas e vasculares, mas também uma elevada porosidade. Devido à incompatibilidade destes dois parâmetros, é necessário desenvolver estratégias de simulação de forma a obter estruturas optimizadas. A previsão real das propriedades mecânicas, vasculares e topológicas das matrizes de suporte, produzidas por técnicas de biofabricação, é muito importante para as diversas aplicações em engenharia de tecidos. A presente dissertação apresenta o estado da arte da engenharia de tecidos, bem como as técnicas de biofabricação envolvidas na produção de matrizes de suporte. Para o design optimizado de matrizes de suporte foi adoptada uma metodologia de design baseada tanto em métodos de elementos finitos para o cálculo do comportamento mecânico, vascular e as optimizações topológicas, como em métodos analíticos para a validação das simulações estruturais utilizando dados experimentais. Considerando que as matrizes de suporte são estruturas elementares do tipo LEGO, dois tipos de famílias foram consideradas, superfícies não periódicas e as superfícies triplas periódicas que descrevem superfícies naturais. Os objectivos principais desta dissertação são: i) avaliar as técnicas existentes de engenharia de tecidos; ii) avaliar as técnicas existentes de biofabricação para a produção de matrizes de suporte; iii) avaliar o desempenho e comportamento das matrizes de suporte; iv) implementar uma metodologia de design de matrizes de suporte em variáveis tais como a porosidade, geometria e comportamento mecânico e vascular por forma a auxiliar o processo de design; e por fim, v) validar experimentalmente a metodologia adoptada.

Composite structures incorporating piezoelectric sensors and actuators are increasingly becoming important due to the offer of potential benefits in a wide range of engineering applications such as vibration and noise supression, shape control and precisition positioning. This paper presents a finit element formulation based on classical laminated plate theory for laminated structures with integrated piezoelectric layers or patches, acting as actuators. The finite element model is a single layer triangular nonconforming plate/shell element with 18 degrees of freedom for the generalized displacements, and one electrical potential degree of freedom for each piezsoelectric elementlayer or patch, witch are surface bonded on the laminate. An optimization of the patches position is performed to maximize the piezoelectric actuators efficiency as well as, the electric potential distribuition is search to reach the specified structure transverse displacement distribuition (shape control). A gradient based algorithm is used for this purpose. The model is applied in the optimization of illustrative laminated plate cases, and the results are presented and discussed.

The design of neuro-fuzzy models is still a complex problem, as it involves not only the determination of the model parameters, but also its structure. Of special importance is the incorporation of a priori information in the design process. In this paper two known design algorithms for B-spline models will be updated to account for function and derivatives equality restrictions, which are important when the neural model is used for performing single or multi-objective optimization on-line.

All systems found in nature exhibit, with different degrees, a nonlinear behavior. To emulate this behavior, classical systems identification techniques use, typically, linear models, for mathematical simplicity. Models inspired by biological principles (artificial neural networks) and linguistically motivated (fuzzy systems), due to their universal approximation property, are becoming alternatives to classical mathematical models. In systems identification, the design of this type of models is an iterative process, requiring, among other steps, the need to identify the model structure, as well as the estimation of the model parameters. This thesis addresses the applicability of gradient-basis algorithms for the parameter estimation phase, and the use of evolutionary algorithms for model structure selection, for the design of neuro-fuzzy systems, i.e., models that offer the transparency property found in fuzzy systems, but use, for their design, algorithms introduced in the context of neural networks. A new methodology, based on the minimization of the integral of the error, and exploiting the parameter separability property typically found in neuro-fuzzy systems, is proposed for parameter estimation. A recent evolutionary technique (bacterial algorithms), based on the natural phenomenon of microbial evolution, is combined with genetic programming, and the resulting algorithm, bacterial programming, advocated for structure determination. Different versions of this evolutionary technique are combined with gradient-based algorithms, solving problems found in fuzzy and neuro-fuzzy design, namely incorporation of a-priori knowledge, gradient algorithms initialization and model complexity reduction.

Polyphase IIR structures have recently proven themselves very attractive for very high performance filters that can be designed using very few coefficients. This, combined with their low sensitivity to coefficient quantization in comparison to standard FIR and IIR structures, makes them very applicable for very fast filtering when implemented in fixed-point arithmetic. However, although the mathematical description is very simple, there exist a number of ways to implement such filters. In this paper, we take four of these different implementation structures, analyze the rounding noise originating from the limited arithmetic wordlength of the mathematical operators, and check the internal data growth within the structure. These analyses need to be done to ensure that the performance of the implementation matches the performance of the theoretical design. The theoretical approach that we present has been proven by the results of the fixed-point simulation done in Simulink and verified by an equivalent bit-true implementation in VHDL.

A novel compact tunable bandstop filter using Defected Microstrip Structure (DMS) is presented in this paper. The structure utilizes a modified T shaped DMS which helps in miniaturization of the filter. To verify the concept, one such filter was simulated, designed, fabricated and tested. Measurements on a fabricated tunable filter confirm the accuracy of the design procedure. The tuning range of 20% is achieved, ranging from 2.26 GHz to 2.747 GHz. Tuning is achieved by using NXP BB179 varactor diode. A nonlinear distortion evaluation in a tunable filter was experimentally verified. Experimental verification shows the filter is highly linear.

Angiotensin-converting enzyme (ACE) plays a critical role in rennin-angiotensin system. Recently, natural products isolated from herbal medicines revealed inhibitory effects against ACE which suggested their potential activities in regulating blood pressure. In this study, ACE inhibition (ACEI) of 21 phenylethanoid glycosides and related phenolic compounds were investigated by measuring the production of HA a rapid, sensitive, accurate and specific ultra-performance liquid chromatography-tandem quadrupole mass spectrometry (UPLC-MS/MS) method. The test compounds showed different inhibitory potencies on ACE ranging from 5.29 to 95.01% at 50 mM, and the compounds with ACEI higher than 50% were selected for further IC50 determination. The IC50 values were from 0.53 ± 0.04 to 15.035 ± 0.036 mM. The structure-inhibition relationship were then explored and the result showed that cinnamoyl groups played an essential role in ACEI of phenylethanoid glycosides. Furthermore, the sub-structures of increasing ACEI for phenylethanoid glycosides is more hydroxyls and less steric hindrance to chelate the active site Zn2+ of ACE. In summary, our results suggested that phenylethanoid glycosides are a widely available source of anti-hypertensive natural products and the information provided from structure-inhibition relationship study could aid the design of structurally modified phenylethanoid glycosides as anti-hypertensive drugs.

This paper uses some data from Igbo-English intrasentential codeswitching involving mixed nominal expressions to test the Matrix Language Frame (MLF) model. The MLF model is one of the most highly influential frameworks used successfully in the study of grammatical aspects of codeswitching. Three principles associated with it, the Matrix Language Principle, the Asymmetry Principle and the Uniform Structure Principle, were tested on data collected from informal conversations by educated adult Igbo-English bilinguals resident in Port Harcourt. The results of the analyses suggest general support for the three principles and for identifying Igbo-English as a ‘classic’ case of codeswitching.

This paper studies fractional variable structure controllers. Two cases are considered namely, the sliding reference model and the control action, that are generalized from integer into fractional orders. The test bed consists in a mechanical manipulator and the effect of the fractional approach upon the system performance is evaluated. The results show that fractional dynamics, both in the switching surface and the control law are important design algorithms in variable structure controllers.

This study was developed with the purpose to investigate the effect of polysaccharide/plasticiser concentration on the microstructure and molecular dynamics of polymeric film systems, using transmission electron microscope imaging (TEM) and nuclear magnetic resonance (NMR) techniques. Experiments were carried out in chitosan/glycerol films prepared with solutions of different composition. The films obtained after drying and equilibration were characterised in terms of composition, thickness and water activity. Results show that glycerol quantities used in film forming solutions were responsible for films composition; while polymer/total plasticiser ratio in the solution determined the thickness (and thus structure) of the films. These results were confirmed by TEM. NMR allowed understanding the films molecular rearrangement. Two different behaviours for the two components analysed, water and glycerol were observed: the first is predominantly moving free in the matrix, while glycerol is mainly bounded to the chitosan chain. Glass fibre-reinforced plastics (GFRP), nowadays commonly used in the construction, transportation and automobile sectors, have been considered inherently difficult to recycle due to both: cross-linked nature of thermoset resins, which cannot be remolded, and complex composition of the composite itself, which includes glass fibres, matrix and different types of inorganic fillers. Presently, most of the GFRP waste is landfilled leading to negative environmental impacts and supplementary added costs. With an increasing awareness of environmental matters and the subsequent desire to save resources, recycling would convert an expensive waste disposal into a profitable reusable material. There are several methods to recycle GFR thermostable materials: (a) incineration, with partial energy recovery due to the heat generated during organic part combustion; (b) thermal and/or chemical recycling, such as solvolysis, pyrolisis and similar thermal decomposition processes, with glass fibre recovering; and (c) mechanical recycling or size reduction, in which the material is subjected to a milling process in order to obtain a specific grain size that makes the material suitable as reinforcement in new formulations. This last method has important advantages over the previous ones: there is no atmospheric pollution by gas emission, a much simpler equipment is required as compared with ovens necessary for thermal recycling processes, and does not require the use of chemical solvents with subsequent environmental impacts. In this study the effect of incorporation of recycled GFRP waste materials, obtained by means of milling processes, on mechanical behavior of polyester polymer mortars was assessed. For this purpose, different contents of recycled GFRP waste materials, with distinct size gradings, were incorporated into polyester polymer mortars as sand aggregates and filler replacements. The effect of GFRP waste treatment with silane coupling agent was also assessed. Design of experiments and data treatment were accomplish by means of factorial design and analysis of variance ANOVA. The use of factorial experiment design, instead of the one factor at-a-time method is efficient at allowing the evaluation of the effects and possible interactions of the different material factors involved. Experimental results were promising toward the recyclability of GFRP waste materials as polymer mortar aggregates, without significant loss of mechanical properties with regard to non-modified polymer mortars.

The thesis is divided into two parts corresponding to structural studies on two different proteins. The first part concerns the study of two UDP-glucose dehydrogenases (UGDs) from Sphingomonas elodea ATCC 31461 and Burkholderia cepacia IST 408, both involved in exopolysaccharide production. Their relevance arises because some of these bacterial exopolysaccharides are valuable as established biotechnological products, the former case, whilst others are highly problematic, when used by pathogens in biofilm formation over biological surfaces, as the latter case, namely in the human lungs. The goal of these studies is to increase our knowledge regarding UGDs structural properties, which can potentiate either the design of activity enhancers to respond to the increased demand of useful biofilms, or the design of inhibitors of biofilm production, in order to fight invading pathogens present in several infections. The thesis reports the production and crystallisation of both proteins, the determination of initial phases by single-wavelength anomalous dispersion (SAD) in S. elodea crystals using a seleno-methionine isoform, and phasing of B. cepacia crystals by molecular replacement (MR) using the S. elodea model, as well as the refinement, structural analysis and comparison between the several UGDs structures available during this work.(...)
