Microbial enzymes are in great demand owing to their importance in several industries such as brewing, baking, leather, laundry detergent, dairy. starch processing and textiles besides pharmaceuticals. About 80% of the enzymes produced through fermentation and sold in the industrial scale are hydrolytic enzymes. Due to recognition of new and new applications, an intensive screening of different kinds of enzymes with novel properties, from various microorganisms, is being pursued all over the world. Bacillus sp are largely known to produce a-amylase, among the different groups of microoganisms, at industrial level. They are known to produce both saccharifying and liquefying a-amylases (Fukumoto 1963; walker and Campbell, 1967a). which are distinguishable by their mechanisms of starch degradation by the fact that the saccharifying asamylases produce an increase in reducing power about twice that of the liquefying enzyme (Fukumoto, 1963; Pazur and Okada, 1966). Under this circumstances, the present study was undertaken, with a View to utilise a fast growing B.coagu1ans isolated from soil, for production of thermostable and alkaline oz-amylase under different fermentation processes

the thesis entitled “Ground and Excited State Electron Transfer Reaction Between a few Anthracene Appended Tertiary Amines and Suitable Electron Acceptors” portrays our attempts to explore the solvent, concentration and temperature effect of the reaction between a few (anthracen-9- yl)methanamines with electron acceptors like DMAD, DBA and DBE. We have also studied the effect of solvent and percentage fluorescence quenching in the photoinduced electron transfer reactions of these ‘donor-spacer-acceptor’ systems. Finally we look in to the intramolecular electron transfer reactions of a few tertiary amine appended dibenzobarrelenes and bisdibenzobarrelenes

Ziel dieser Arbeit war, durch Aziridinierung homochiraler 5-Methyl-4H-1,3-dioxinen eine neue Methode zur Synthese von alpha-Aminoaldehyden und den ableitbaren Aminosäuren mit alpha-quartären Zentren zu entwickeln. Die chiralen 5-Methyl-4H-1,3-dioxine sind mit hohen Enantiomerenüberschüssen durch asymmetrische Doppelbindungsisomerisierung von 5-Methylen-1,3-dioxanen zugänglich. Die Metall-katalysierte Aziridinierung der 5-Methyl-4H-1,3-dioxine mit der Nitrenquelle (N-Tosylimino)phenyliodinan führte direkt zu N-Tosyl-geschützen 4-Methyl-1,3-oxazolidin-4-carbaldehyden. Vermutlich über ein Aziridin als nicht isolierbare Zwischenstufe werden über eine Ringöffnungs-/Ringverengungsreaktion die Oxazolidinderivate gebildet, vorzugsweise in Gegenwart von Cu(I)-Katalysatoren, während die Rhodium-katalysierte Reaktion ausschließlich zu Insertionsprodukten führt. In der Cu-katalysierten Aziridinierung ist das Verhältnis von Aziridinierung/Insertion abhängig von der Katalysatorkonzentration. Die Aziridinierung mit N-(p-Nitrobenzolsulfonyl)- und N-(Trimethylsilylethylsulfonyl)- substituierten Nitrenquellen führt zu Oxazolidinderivaten mit leichter abspaltbaren Schutzgruppen. Diese Nitrenquellen können in situ aus den korrespondierenden Sulfonamiden mit Iodosobenzol dargestellt werden. Bei dem Einsatz homochiraler 4H-1,3-Dioxine ist Erhalt der Stereoinformation abhängig vom Substituenten in 2-Position der Dioxine sowie von der Polarität des Lösungsmittels. Die höchsten Selektivitäten wurden in tert-Butylmethylether erzielt. In Falle des 2-tert-Butyl-4-methyl-3-(toluol-4-sulfonyl)-1,3-oxazolidin-4-carbaldehyds kristallisiert das Hauptdiastereomer in enantiomerenreiner Form. Die Absolutkonfiguration wurde durch Röntgenkristallstrukturanalyse ermittelt. Das Anwendungspotential dieser neuen Methode konnte durch Überführen der Serinale in Aminoalkohole und alpha-Methylserin-Derivate sowie in der Synthese der unnatürlichen Aminosäure alpha-Vinylalanin gezeigt werden.

To study the complex formation of group 5 elements (Nb, Ta, Ha, and pseudoanalog Pa) in aqueous HCI solutions of medium and high concentrations the electronic structures of anionic complexes of these elements [MCl_6]^-, [MOCl_4]^-, [M(OH)-2 Cl_4]^-, and [MOCl_5]^2- have been calculated using the relativistic Dirac-Slater Discrete-Variational Method. The charge density distribution analysis has shown that tantalum occupies a specific position in the group and has the highest tendency to form the pure halide complex, [TaCl_6-. This fact along with a high covalency of this complex explains its good extractability into aliphatic amines. Niobium has equal trends to form pure halide [NbCl_6]^- and oxyhalide [NbOCl_5]^2- species at medium and high acid concentrations. Protactinium has a slight preference for the [PaOCl_5]^2- form or for the pure halide complexes with coordination number higher than 6 under these conditions. Element 105 at high HCl concentrations will have a preference to form oxyhalide anionic complex [HaOCl_5]^2- rather than [HaCl_6]^-. For the same sort of anionic oxychloride complexes an estimate has been done of their partition between the organic and aqueous phases in the extraction by aliphatic amines, which shows the following succession of the partition coefficients: P_Nb < P_Ha < P_Pa.

Immunity to severe malaria is the first level of immunity acquired to Plasmodium falciparum. Antibodies to the variant antigen PfEMP1 (P. falciparum erythrocyte membrane protein 1) present at the surface of the parasitized red blood cell (pRBC) confer protection by blocking microvascular sequestration. Here we have generated antibodies to peptide sequences of subdomain 2 of PfEMP1-DBL1 alpha previously identified to be associated with severe or mild malaria. A set of sera generated to the amino acid sequence KLQTLTLHQVREYWWALNRKEVWKA, containing the motif ALNRKE, stained the live pRBC. 50% of parasites tested (7/14) were positive both in flow cytometry and immunofluorescence assays with live pRBCs including both laboratory strains and in vitro adapted clinical isolates. Antibodies that reacted selectively with the sequence REYWWALNRKEVWKA in a 15-mer peptide array of DBL1 alpha-domains were also found to react with the pRBC surface. By utilizing a peptide array to map the binding properties of the elicited anti-DBL1 alpha antibodies, the amino acids WxxNRx were found essential for antibody binding. Complementary experiments using 135 degenerate RDSM peptide sequences obtained from 93 Ugandan patient-isolates showed that antibody binding occurred when the amino acids WxLNRKE/D were present in the peptide. The data suggests that the ALNRKE sequence motif, associated with severe malaria, induces strain-transcending antibodies that react with the pRBC surface.

Este trabajo demuestra que los genes que codifican para los enzimas beta-galactosido alfa-2,3-sialiltransferasa 3 (ST3Gal III), y en menor medida beta-galactosido alfa-2,3-sialiltransferasa 4 (ST3Gal IV), están directamente implicados en etapas clave de la progresión tumoral como la adhesión, la migración y la formación de metástasis en las líneas de adenocarcinoma pancreático humano Capan-1 y MDAPanc-28. También, que las Especies Reactivas del Oxígeno (ROS) generadas durante los procesos de proliferación y diferenciación celular o debido a estímulos oxidantes externos, desempeñan un importante papel en el control de la síntesis de ST3Gal III y SLex, y por lo tanto en la regulación del fenotipo metastático. Además, junto al papel pro-adhesivo de la E-Selectina, este trabajo ha descrito efectos prometastáticos adicionales para esta molécula como inductora de la migración y de la secreción de VEGF a través de un mecanismo E-Selectina-SLex dependiente.

The principle theme of this thesis was the synthesis of bioactive compounds. To this end, this work was focus on two main projects. The first one, which was carried out in the Department of Chemistry of the University of Girona under the supervision of Dr Montserrat Heras, concerned the synthesis of new unnatural amino acids bearing a pyrimidine ring within their side chain for incorporation into the antimicrobial peptide BP100 following a rational design in order to improve its biological profile. On the other hand, the second chapter of this thesis was developed in collaboration with the Laboratoire de Chimie Organique (ESPCI-ParisTech, Paris, France) under the guidance of Pr Janine Cossy and Dr Arseniyadis. This chapter was centered on the total synthesis of three marine natural products with complex structures and interesting biological activities: acremolide B, (–) bitungolide F and lyngbouilloside.

The recent discovery of the contribution of alpha synuclein in the auditory system prompted further investigation of its functional role. Auditory brainstem response (ABR) and gap detection testing were completed on wild-type and transgenic M83 mice to assess the role of alpha synuclein in noise-induced hearing loss and central auditory function.

Alpha-synuclein has been implicated in the cellular mechanisms that control auditory sensitivity. In other systems it can also confer protection against cellular injury. Auditory brainstem response thresholds and immunohistochemistry were used to assess the ability of alpha-synuclein to protect against oxidative damage to the cochlea.

Alpha-synuclein is found in synaptic terminals at the base of both inner and outer hair cells, while the beta isoform is prominently localized to spiral ganglion neuron cell bodies. The present study assessed the role of beta-synuclein in auditory function, and potential interactions between isoforms.

The AMPA receptor (AMPAR) subunit GluR2, which regulates excitotoxicity and the inflammatory cytokine tumour necrosis factor alpha (TNF alpha) have both been implicated in motor neurone vulnerability in Amyotrophic Lateral Sclerosis/Motor Neurone Disease. TNF alpha has been reported to increase cell surface expression of AMPAR subunits to increase synaptic strength and enhance excitotoxicity, but whether this mechanism occurs in motor neurones is unknown. We used primary cultures of mouse motor neurones and cortical neurones to examine the interaction between TNF alpha receptor activation, GluR2 availability, AMPAR-mediated calcium entry and susceptibility to excitotoxicity. Short exposure to a physiologically relevant concentration of TNFalpha (10 ng/ml, 15 min) caused a marked redistribution of both GluR1 and GluR2 to the cell surface as determined by cell surface biotinylation and immunofluorescence. Using Fura-2 AM microfluorimetry we showed that exposure to TNFalpha caused a rapid reduction in the peak amplitude of AMPA-mediated calcium entry in a PI3-kinase and p38 kinase-dependent manner, consistent with increased insertion of GluR2-containing AMPAR into the plasma membrane. This resulted in a protection of motor neurones against kainate-induced cell death. Our data therefore, suggests that TNF alpha acts primarily as a physiological regulator of synaptic activity in motor neurones rather than a pathological drive in ALS
