Avaliou-se o efeito da eletroacupuntura (EA) sobre os aspectos macroscópicos e microscópicos em 36 coelhos que tiveram o tendão calcanear comum seccionado e que, após 30 dias, receberam enxerto de peritônio bovino conservado em solução saturada de sal. Após a cirurgia, os animais foram distribuídos em três grupos de igual número: os do grupo E10 foram estimulados pela EA do segundo ao 11º dia de pós-operatório, os do grupo E20 do 11º ao 31º dia de pós-operatório e os do grupo C não foram estimulados. Posteriormente, os animais foram redistribuídos em três subgrupos de igual número, os quais foram observados no pós-operatório por 11, 31 e 91 dias. Nos coelhos do E10 e E20, ocorreu diminuição de aderência da área enxertada aos tecidos circunvizinhos. Aos 31 dias de pós-operatório, observou-se nos do E10 tecido conjuntivo. Verificou-se intensa neovascularização no local enxertado em todos os animais que receberam estímulo pela EA. Nos coelhos do E10, aos 91 dias, verificou-se intensa quantidade de tecido conjuntivo, com fibras colágenas organizadas e paralelas ao eixo do tendão. O estímulo pela EA na fase precoce do pós-operatório estimulou a maturação cicatricial com mínima reação inflamatória.

O objetivo deste estudo foi verificar as condições periodontais e necessidades de tratamento em crianças e adolescentes, com idades entre 7 e 19 anos, matriculados em Escolas Públicas de Araraquara, SP, Brasil, em 1995, com o propósito de se obter um banco de dados-base inicial para o monitoramento das mudanças no estado periodontal. Uma amostra de 1956 crianças e adolescentes foi examinada por duas examinadoras, previamente calibradas, usando o CPITN. Os resultados mostraram que o sangramento gengival foi a condição periodontal mais frequentemente observada em todas as idades, com média de 3,0 sextantes afetados na idade de 15 anos. A presença de cálculo dental aumentou com a idade. No grupo etário de 15-19 anos, o cálculo dental e as bolsas periodontais rasas (3,5-5,5 mm) foram observados em 39,8% e 1,6% dos adolescentes, respectivamente. Com o propósito de alcançar a meta da OMS de não mais do que um sextante com escores 1 (sangramento) ou 2 (presença de cálculo dental) do CPITN na idade de 15 anos, prioridade deve ser dada aos programas de prevenção primária para que a saúde periodontal da maioria seja beneficiada.
Características morfogênicas do capim-piatã submetido à adubação com efluentes de abatedouro avícola

O objetivo do trabalho foi avaliar diferentes doses de efluente de abatedouro avícola para proporcionar melhorias nas características produtivas, morfogênicas e estruturais do capim-piatã. O experimento foi conduzido em casa de vegetação, adotando-se delineamento inteiramente casualizado, em que foram testadas cinco doses de efluentes: 324, 648, 972, 1.296 e 1620m³ ha-1 ou equivalente a 50, 100, 150, 200 e 250kg ha-1 de N. As variáveis mensuradas foram: produção de matéria seca (MS), taxa de aparecimento de folha (TApF), taxa de alongamento de folha (TAlF), filocrono, taxa de alongamento de pseudocolmo (TAlC), comprimento final de folha (CFF) e números de folhas verdes (NFV). A produção de MS seguiu um modelo linear de predição em função das doses efluente avícola, em que o tratamento com 250kg ha-1 de N foi 55% maior, quando comparado com o tratamento de 50kg ha-1 de N. Todas as características morfogênicas e estruturais avaliadas com exceção do filocrono apresentaram comportamento linear positivo. Dessa forma, o efluente de abatedouro avícola pode ser utilizado como uma alternativa para adubação do capim-piatã, pois este respondeu de maneira crescente até a dose máxima testada.

O conhecimento do genoma pode auxiliar na identificação de regiões cromossômicas e, eventualmente, de genes que controlam características quantitativas (QTLs) de importância econômica. em um experimento com 1.129 suínos resultantes do cruzamento entre machos da raça Meishan e fêmeas Large White e Landrace, foram analisadas as características gordura intramuscular (GIM), em %, e ganho dos 25 aos 90 kg de peso vivo (GP), em g/dia, em 298 animais F1 e 831 F2, e espessura de toucinho (ET), em mm, em 324 F1 e 805 F2. Os animais das gerações F1 e F2 foram tipificados com 29 marcadores microsatélites. Estudou-se a ligação entre os cromossomos 4, 6 e 7 com GIM, ET e GP. Análises de QTL utilizando-se metodologia Bayesiana foram aplicadas mediante três modelos genéticos: modelo poligênico infinitesimal (MPI); modelo poligênico finito (MPF), considerando-se três locos; e MPF combinado com MPI. O número de QTLs, suas respectivas posições nos três cromossomos e o efeito fenotípico foram estimados simultaneamente. Os sumários dos parâmetros estimados foram baseados nas distribuições marginais a posteriori, obtidas por meio do uso da Cadeia de Markov, algoritmos de Monte Carlo (MCMC). Foi possível evidenciar dois QTLs relacionados a GIM nos cromossomos 4 e 6 e dois a ET nos cromossomos 4 e 7. Somente quando se ajustou o MPI, foram observados QTLs no cromossomo 4 para ET e GIM. Não foi possível detectar QTLs para a característica GP com a aplicação dessa metodologia, o que pode ter resultado do uso de marcadores não informativos ou da ausência de QTLs segregando nos cromossomos 4, 6 e 7 desta população. Foi evidenciada a vantagem de se analisar dados experimentais ajustando diferentes modelos genéticos; essas análises ilustram a utilidade e ampla aplicabilidade do método Bayesiano.

A simulation model implemented in the programming software Delphi XE® was applied to evaluate sex selection in bovine. The hypothesis under investigation was that a dynamic model with stochastic and deterministic elements could detect the sexed semen technique to minimize pregnancy cost and to determine the adequate number of recipients required for in vivo (ET) and in vitro embryo production (IVP) in the proposed scenarios. Sex selection was compared through semen sexed using flow cytometry (C1) and density gradient centrifugation techniques (C2) in ET and IVP. Sensibility analyses were used to identify the adequate number of recipients for each scenario. This number was reinserted into the model to determine the biological and financial values that maximized ET and IVP using sexed semen (C1M and C2M). New scenarios showed that the density gradient technique minimized pregnancy cost based on the proposed scenarios. In addition, the adequate number of recipients (ET - C1M - 115 and C2M - 105)/(IVP - C1M - 145 and C2M - 140) per donor used was determined to minimize the pregnancy cost in all scenarios.

Thermal regime and stability in Jurumirim Reservoir (São Paulo, Brazil) were studied for a year. Isothermy and thermal stratification were observed from June to September and from October to March, respectively. The annual heat budget was 14.0 Kcal . cm-2 and average heat gain per day was 67.2 cal . cm-2 . day-1. Maximum stability and wind work reached 324.2 and 3,037.5 g . cm . cm-2, respectively. According to thermal structure and heat content, Jurumirim Reservoir was classified as a warm tropical monomictic lake.

The objective of this study was to examine the endothelial surface morphology and to perform morphometric analysis of the corneal endothelial cells of Yacare caiman (Caiman yacare) using scanning electron microscopy. Morphometric analysis with regard to polygonality, mean cell area, cell density and coefficient of variation of mean cell area was performed. Cell areas were measured using image analysis software. The normal corneal endothelium of Yacare caiman consisted of polygonal cells of uniform size and shape with interdigitations of the cell borders. Microvilli appeared as protrusions on the cellular surface. The average cell area was 270 +/- 24 mum(2) and the endothelial cell density was 3704 +/- 324 cells/mm(2). The coefficient of variation of cell area was 0.22. This study demonstrates that the Yacare caiman corneal endothelium is similar to those described in other vertebrates.

The objective of this work was to evaluate the total endogenous N losses, and protein and energy net requirements for maintenance in growing lambs. Thirty-four castrated lambs, 17 F1 Ideal X Ile de France wool and 17 Santa Inas hair lambs, averaging 20 +/- 0.14 kg BW, were used in the experiment. Five animals from each genotype were slaughtered at the beginning of the experiment and taken as controls. Diets (D) were composed of concentrate mix (C) and Cynodon sp. c.v. Tifton 85 hay (R), combined in three different ratios: D1 = 60C:40R; D2 = 40C:60R and D3 = 20C:80R. Animals of each group of three lambs, that showed BW of 20 kg at the beginning of the dietary regimen, were slaughtered when one of them reached 35 kg, what always happened to be the one fed with D1. Total endogenous N losses estimated for wool lambs were 250 mg N/kg BW0.75. For hair lambs, total endogenous N losses reached 324 mg N/kg BW0.75 . Hair lambs showed higher (P < 0.01) (29.9%) net requirements of protein for maintenance than wool lambs. In contrast, net energy (NE) requirement for maintenance was similar (P > 0.05) for both genotypes (74.27 kcal/kg BW0.75 per day), the average of the antilog of the two intercept values obtained from the estimated regression equations of heat production for zero metabolizable energy (ME) consumption. Further studies should be done to check if this trend is also true for metabolizable energy and protein in animals exhibiting BW gains in tropical region. (C) 2003 Elsevier B.V. B.V. The purpose of this work was to assess the effects of soil preparation methods (with or without subsoiling) on physical properties of a cerrado soil cropped to corn during the, 1986/87 season, in the Selviria county (Mato Grosso do Sul, Brazil). Also, the objective of this was to check the changes in the soil (dark red latosol-Acrustox) caused by the preparation after nine years of cultivation in comparison with natural state. The experiment consisted of six soil preparation systems: conventional, superpreparation, reversed, revolving hoe, reduced and no-tillage. Along with these systems subsoiling was also tested. The reversed system showed the best com grain yield. Subsoiling only presented positive results in productivity with the no-tillage system. All the preparation system tested degraded the physical properties in comparison with the natural soil.

The effect of temperature on the oxalic acid catalyzed sono-hydrolysis of tetramethoxysilane (TMOS) was studied by means of a heat flux calorimetric method. The activation energy of the process was measured as (24.5 +/- 0.8) kJ/mol in the temperature range between 10 and 50 degreesC. The structural characteristics of the resulting sonogels, after long period of aging in saturated conditions, were studied by means of small angle X-ray scattering. The structure can be described as formed by similar to2.7 nm mean size mass fractal-like aggregates (clusters) of primary silica particles of similar to0.3 nm mean size, all imbibed in a liquid phase. The average mass fractal dimension of the clusters was found to be 2.58. The primary particle density was estimated as 2.23 g/cm(3), in good agreement with the value frequently quoted for fused silica. The volume fraction of the clusters, in the saturated sonogels was estimated as about 28%. The moment in which the meniscus of the liquid phase penetrates into the clusters under rapid evaporation process has been detected by an inflection in the first derivative of the curve of weight loss in a simple thermogravimetric test. Adriamycin, a commonly used antineoplastic antibiotic, induces glomerular lesions in rats, resulting in persistent proteinuria and glomerulosclerosis. We studied the effects of dietary protein and of an angiotensin I converting enzyme inhibitor on the progression of this nephropathy and the evolution of the histological lesions, as well as mesangial macromolecule flow. Adriamycin nephropathy was induced by injecting a single iv dose of adriamycin (3 mg/kg body weight) into the tail vein of male Wistar rats (weight, 180-200 g). In Experiment I animals with adriamycin-induced nephropathy were fed diets containing 6% (Low-Protein Diet Group = LPDG), 20% (Normal-Protein Diet Group = NPDG) and 40% (High-protein Diet Group = HPDG) protein and were observed for 30 weeks. In Experiment II the rats with adriamycin nephropathy were divided into 2 groups: ADR, that received adriamycin alone, and ADR-ENA, that received adriamycin plus enalapril, an angiotensin I converting enzyme inhibitor. The animals were sacrificed after a 24-week observation period. Six hours before sacrifice the animals were injected with I-131-ferritin and the amount of I-131-ferritin in the glomeruli was measured. In Experiment III, renal histology was performed 4, 8 and 16 weeks after adriamycin injection. At the end of Experiment I the tubulointerstitial lesion index was 2 for LPDG, 8 for NPDG, and 7.5 for HPDG (P<0.05); the frequency of glomerulosclerosis was 19 +/- 6.1% in LPDG, 42.6 +/- 6% in NPDG, and 54 +/- 9% in HPDG (P<0.05); and proteinuria was 61.1 +/- 25 mg/24 h in LPDG, 218.7 +/- 27.5 mg/24 h in NPDG, and 324.5 +/- 64.8 mg/24 h in HPDG (P<0.05). In Experiment II, at sacrifice, 24-h proteinuria was 189 +/- 16.1 mg in ADR, and 216 +/- 26.1 mg in ADR-ENA (P>0.05); the tubulointerstitial lesion index was 5 for ADR, and 5 for ADR-ENA (P>0.05); the frequency of glomerulosclerosis was 40 +/- 5.2% in ADR and 44 +/- 6% in ADR-ENA (P>0.05); the amount of I-131-ferritin in the mesangium was 214.26 +/- 22.71 cpm/mg protein in ADR and 253.77 +/- 69.72 cpm/mg protein in ADR-ENA (P>0.05). In Experiment III, sequential histological analysis revealed an acute tubulointerstitial cellular infiltrate at week 4, which was decreased at week 8. Tubular casts and dilatation were first seen at week 8 and increased at week 16 when few glomerular lesions were found. The results suggest that the tubulointerstitial lesions may play a role in the development of glomerulosclerosis in adriamycin-induced nephropathy.

Roscovitine and flavopiridol have been shown to potently inhibit cyclin-dependent kinase 1 and 2 (CDK1 and 2). The structures of CDK2 complexed with roscovitine and deschoroflavopiridol have been reported, however no crystallographic structure is available for complexes of CDK1 with inhibitors. The present work describes two molecular models for the binary complexes CDK1:roscovitine and CDK1:flavopiridol. These structural models indicate that both inhibitors strongly bind to the ATP-binding pocket of CDKI and structural comparison of the CDK complexes correlates the structures with differences in inhibition of these CDKs by flavopiridol and roscovitine. We determined the effect of intracerebroventricular (icv) administration of losartan, an angiotensin II (ANG II) subtype 1 receptor (AT1) antagonist, on icv carbachol-induced natriuresis, kaliuresis and antidiuresis in water-loaded male Holtzman rats (250-300 g) with a cannula implanted into the lateral ventricle (LV). The rats were water loaded with 5% of their body weight by gavage twice, with the second gavage one hour after the first. Carbachol (2 nmol in 1 mu l) was injected icv immediately after the second load. When losartan (DUP-753, 50 nmol in 1 mu l) was administered icv, it was given 3 min before carbachol. Previous icv treatment with losartan significantly reduced the icv carbachol-induced natriuresis (324 +/- 17 mu Eq/120 min), kaliuresis (103 +/- 15 mu Eq/120 min) and antidiuresis (13.5 +/- 2.1 ml/120 min) compared to the effects of previous icv injection of saline (Nai excretion = 498 +/- 22 mu Eq/120 min; K+ excretion = 167 +/- 20 mu Eq/120 min; urine volume = 5.2 +/- 1.2 ml/120 min). These results, reported as means +/- SEM for 12 rats in each group, are consistent with the hypothesis that AT1 subtype receptors participate in the regulation of body electrolyte balance.

This work reports on the study about the luminescence behavior of Eu3+-O2- associates in Sr2SiO4 doped with Eu3+, or simultaneously doped with Eu3+ and Zn2+ ions, where the Zn2+ doping ion acts as a charge compensating agent. Both Sr2SiO4:Eu3+ and Sr2SiO4:Eu3+,Zn2+ emission spectra show two types D-5(0)-->F-7(0) type transitions, one type related to Eu3+ in the Sr2+ Site, at 577 and 580 nm and the other one related to Eu3+-O2- associates, at 574 nm. Excitation spectra present two CT bands at 275 and 324 nm related to each emission center, respectively. Comparing the relative intensities between the emission spectra, the 0-->0 transition at 574 nm assigned to Eu3+-O2- associates in the Sr2SiO4:Eu3+ emission spectrum is much more intense than the same transition in the Sr2SiO4:Eu3+,Zn2+ emission spectrum. Moreover, in the Sr2SiO4:Eu3+ excitation spectrum, the intensity ratio between the CT band related to the Eu3+-O2- emission center and Eu3+ F-7-->L-5(6) transition is also larger than in the Sr2SiO4:Eu3+,Zn2+ one. Therefore, the presence of Zn2+ species provides an extra charge compensating mechanism, which decreases the formation of Eu3+-O2- associates. (C) 2002 Elsevier B.V. B.V. All rights reserved.

Tendon composition changes according to differentiation, mechanical load, and aging. In this study, we attempted to identify, localize, and quantify type VI collagen in bovine tendons. Type VI collagen was identified by the electrophoretic behavior of the alpha chains and Western blotting, and by rotary shadowing. Type VI collagen was extracted from powdered tendon with three sequential 24-h extractions with 4 M guanidine-HCl. The amount of type VI collagen was determined by enzyme-linked immunosorbent assay for purely tensional areas and for the compressive fibrocartilage regions of the deep flexor tendon of the digits, for the corresponding fetal and calf tendons, and for the extensor digital tendon. The distal fibrocartilaginous region of the adult tendon was richer in type VI collagen than the tensional area, reaching as much as 3.3 mg/g (0.33%) of the wet weight. Calf tendons showed an accumulation of type VI at the fibrocartilage site. Immunocytochemistry demonstrated that type VI collagen was evenly distributed in the tensional areas of tendons but was highly concentrated around the fibrochondrocytes in the fibrocartilages. The results demonstrate that tendons are variable with regard to the presence and distribution of type VI collagen. The early accumulation of type VI collagen in the region of calf tendon that will become fibrocartilage in the adult suggests that it is a good marker of fibrocartilage differentiation. Furthermore, the distribution of type VI collagen in tendon fibrocartilage indicates that it organizes the pericellular environment and may represent a survival factor for these cells.
