O autor, depois de analisar as diversas técnicas utilizadas por diversos pesquisadores, com respeito à maior ou menor positividade obtida pela chamada técnica do bloco de parafina e técnica de Papanicoulaou, apresenta 10 casos de comprovado néo brônquico, estudados com a técnica de inclusão em parafina e subseqüente coloração epla hematoxilina-azur-eosina. Conclui afirmando que o exame citológico do escarro do lavado brônquico, deve ser, de preferência, utilizado pelo emprêgo daquela técnica, por permitir maiores detalhes nas técnicas histoquímicas e por conservaram as células as suas inter-relações.

O A. recapitula os dados já conhecidos sôbre a filtrabilidade de muitas Rickettsias e sôbre o que êle chama o ciclo evolutivo das mesmas, citando os trabalhos de S. B. Dulky e E. Gordon sôbre a Coxiella papilliae e a observação de A. Donatien e F. Lestoquard bem como os de P. Giraud. Refere-se à filtrabilidade do agente da febre "Q". Reporta-se depois aos trabalhos de Mme. Ruth Rein Gutfreund sôbre os achados de Rickettsia prowazedi em animais domésticos na Etiópia e a transmissão dos mesmos pelos carrapatos, dizendo que isto confirma suas próprias idéias, de há muito emitidas, de que não há no grupo tifo exantemático, de regra, especificidade estrita para os transmissores, não podendo êste carácter servir de base para classificações racionais. Descreve um caso clínico, mostrando a dificuldade de diagnóstico cofundido com a febre tifóide, mesmo com longa prática do exame na doença e que só a inoculação em animais sensíveis - aqui os cobaios - pode decidir a questão. Estuda depois dois surtos epidêmicos de tifo exantemático neotrópico - em 1950 em Carmópolis (Minas Gerais); outro em 1956, Mucuri (Bahia). Em ambos, a percentagem de mortes dos casos graves, não tratados, foi elevada e a terapêutica pelos antibióticos, principalmente a Cloromicetina e Terramicina foi brilhante. Nestes dois surtos epidêmicos com já se vira em 1941 (4 casos na mesma residência) e 1948 (6 casos na mesma moradia), apuraram-se 2 e 3 casos da doença na mesma casa. As reações de Fixação de Complemento (R.F.C.), Weil-Feliz (W.F.) e Widal - confirmam o diagnóstico de tifo exantemático neotrópico.

Os autores depois de apresentarem alguns métodos recentemente descritos para a coloração de protozoários intestinais em tecidos, apresentam dois métodos que pela simplicidade e rapidez que apresentam, são sem dúvida superiores a todos anteriormente utilizados.

Os autores, utilizando duas técnicas diferentes, obtiveram concentração de células tumorais do chamado tumor de Yoshida, isoladas do sangue obtido por punção cardíaca e do líquido de derrame ascítico de ratos. As técnicas empregadas foram a de SANDBERG & MOORE (fibrinogênio bovino) e a do hematócrito de Wintrobe.

A - NORTE - A floresta em tôrno de Belém parece já bastante alterada, sendo poucas as árvores de grande porte. O solo é sempre arenoso, com uma camada de húmus muito fina a qual, quando é removida, deixa ver uma areia branca. Em Utinga era de notar o grande número de árvores caídas, justamente as maiores, o que sugere um solo pobre e raso, originando um enraizamento superficial e conseqüente queda das mesmas pelos ventos. Na floresta de àgua Preta é muito maior o número de árvores finas, o que sugere um solo ainda mais pobre. Trata-se de floresta típica da planície (Quartenário antigo), onde o solo foi formado por deposição do arenitos pobres do Centro-oeste capturados pela bacia do Tocatins. B - Nordeste - 1. Caatinga: O tipo de vegetação que predomina no Nordeste é o da caatinga, vegetação de arbustiva a arbórea, espinhosa e decidual. A parte vista, de Terezina até Paulo Afonso, via Ubajara, Fortaleza, Crato e Floresta, apresenta várias nuances fisiográficas de densidade e porte. Essas são devidas, aparentemente ao menos, não só à intervenção humana, como também à grande variação na constituição física do solo. Assim é que observamos: a) Caatinga arbórea - dominada pela carnaúba: Copernicia cerifera (A. Cam.) Mart. nos lugares encharcados, pela oiticica: Licania rigida Benth. na linha demarcatória das maiores enchentes, e pelo juàzeiro: Zizyphus joazeiro Mart., fora da influência das àguas dos rios. b) Caatinga aberta - nos solos pedregosos, com dominância da imburana-de-espinho: Bursera leptophloeos Engl. e do umbu: Spondias tuberosa A. Cam. Ocorre nas enconstas dessecadas dos serrotes de quartzito. c) Caatinga arbustiva densa - existente nos largos vales dos maiores rios, provável fruto da intervenção...

The Brazilian Institute for Agrarian Reform is promoting researches on soil fertility in southern Mato Grosso, complemented by a survey of the main types of vegetation. The data obtained were also to be used to build up the legends for the map of vegetation. Photos were taken by the authors and botanical material collected in order to permit the identification of important species. Observations made led to the above table of main physiognomies.

Foram estudados os encéfalos e as supra-renais de cinco casos de raiva humana, sendo três crianças e dois adultos com óbito ocorrendo entre três e seis dias após início das manifestações clínicas. Em todos os casos o encéfalo mostrava corpúsculo de Negri nas células nervosas e infiltrado mononuclear perivascular mais evidente nos casos de evolução mais longa. O comprometimento eletivo da medular das supra-renais caracterizado por acentuado exsudato mononuclear, além de alterações dos feocromócitos ocorreu em 60% dos casos. Corpúsculos eosinófilos foram encontrados no citoplasma dos feocromócitos e, às vezes, no interstício da medular. Tais corpúsculos poderiam representar inclusões viróticas semelhantes aos corpúsculos de Negri dos neurônios. Entretanto sua verdadeira natureza precisa ser ainda esclarecida. A medulite supra-renálica aqui descrita pode ter sua gênese, pelo menos em parte, determinada por fatores que se relacionam ao "parentesco" embriológico do sistema cromafim com o tecido nervoso.

Through microsatellite analysis of 53 monoclonal populations of Trypanosoma cruzi, we found a remarkable degree of genetic polymorphism with no single multilocus genotype being observed more than once. The microsatellite profile proved to be stable during 70 generations of the CL Brener clone in culture. The microsatellite profiling presented also high diagnostic sensitivity since DNA amplifications could be achieved with less than 100 fg DNA, corresponding to half parasite total DNA content. Based on these technical attributes the microsatellite assay turns out to be an important tool for direct typing T. cruzi in biological samples. By using this approach we were able to type T. cruzi in feces of artificially infected bugs and in single cells sorted by FACS. The microsatellites have shown to be excellent markers for T. cruzi phylogenetic reconstruction. We used maximum parsimony based on the minimum number of mutational steps to build an unrooted Wagner network, which confirms previous conclusions based on the analysis of the D7 domain of the LSU rDNA gene that T. cruzi is composed by two major groups. We also obtained evidence that strains belonging to rRNA group 2 are subdivided into two genetically distant clusters, and that one of these clusters is more related to rRNA group 1/2. These results suggest different origins for these strains.

Este proyecto tiene como objetivo crear y aplicar una metodología a una aplicación llamada MATE que fue creada en en el año 2003 por Anna Sikora para su tesis doctoral. Se trata de dotar el proyecto MATE de las herramientas necesarias para garantizar su evolución. La metodología creada consta de la especificación de un entorno de trabajo y una serie de documentos que detallan los procesos relativos al desarrollo de MATE. Además se han creado algunas nuevas características que hacen de MATE una herramienta más completa y cómoda.

The prevalence, virological and epidemilogical aspects of the hepatitis C virus (HCV) and the hepatitis B virus (HBV) infections vary among hemodialysis patients in different countries. Aiming at analyzing these aspects of HCV and HBV infections in hemodialysis patients in Belo Horizonte, MG, Brazil, we studied three hemodialysis units including 434 patients. Serology was used to detect anti-HCV and HBsAg. Reverse trancriptase nested polymerase chain reaction (RT-nested-PCR) of the 5'-noncoding region was used to detect circulating HCV RNA and restriction fragment length polymorphism analysis for genotyping. Seroprevalence varied from 26.5% to 11.1% for hepatitis C and from 5.9% to 0% for hepatitis B. Risk factors observed for HBV and/or HCV infections were the number of patients per dialysis unit, duration of treatment, number of clinics attended, number of blood units transfused, and lower level scholarity. Alanine aminotransferase levels were altered with a higher frequency in HBV or HCV seropositive patients. Half of ten patients, negative for anti-HCV, had detectable viremia by RT-nested-PCR, indicating that this technique should be used to confirm infections in this group of patients. The HCV genotype 1 was the most frequently observed, followed by the genotype 2, but no correlation was detected between genotype and clinical or epidemiological data.

Effect of Aedes fluviatilis saliva on the development of Plasmodium gallinaceum experimental infection in Gallus (gallus) domesticus was studied in distinct aspects. Chickens subcutaneously infected with sporozoites in the presence of the mosquito salivary gland homogenates (SGH) showed higher levels of parasitaemia when compared to those ones that received only the sporozoites. However, the parasitaemia levels were lower among chickens previously immunized by SGH or non-infected mosquito bites compared to the controls, which did not receive saliva. High levels of anti-saliva antibodies were observed in those immunized chickens. Moreover, 53 and 102 kDa saliva proteins were recognized by sera from immunized chickens. After the sporozoite challenge, the chickens also showed significant levels of anti-sporozoite antibodies. However, the ability to generate anti-sporozoites antibodies was not correlated to the saliva immunization. Our results suggest that mosquito saliva components enhance P. gallinaceum parasite development in naive chickens. However, the prior exposure of chickens to salivary components controls the parasitemia levels in infected individuals.

In Brazil, until 2004, the immunization policy against diphtheria involved childhood vaccination with no official routine booster dose administered after 15 years of age. This study assessed functional antibody levels against diphtheria among blood donors. A total of 140 blood samples were collected, and diphtheria antitoxin levels were evaluated by Vero cell neutralization test. The mean age of the population was 34 years old (range: 18-61 years); 37.8% females and 62.2% males. Overall, 30.7% (95%, CI: 23.4-38.7) individuals presented neutralizing antitoxin antibody titers < 0.01 IU/ml; 42.1% (95%, CI: 34.1-50.4) showed values between 0.01-0.09 IU/ml and, 27.1% (95%, CI: 20.2-34.9) had ³ 0.1 IU/ml. In the subgroup of individuals with history of diphtheria immunization during childhood (85%), a number of 28.5% showed unprotective levels of circulating neutralizing antibody (< 0.01 IU/ml). Despite the continuous progress of immunization programs directed to Brazilian population, currently healthy adults remain susceptible to diphtheria.

Praziquantel (PZQ) is effective against all the evolutive phases of Schistosoma mansoni. Infected Biomphalaria glabrata snails have their cercarial shedding interrupted when exposed to PZQ. Using primary in vitro transformed sporocysts, labeled with the probe Hoechst 33258 (indicator of membrane integrity), and lectin of Glycine max (specific for carbohydrate of N-acetylgalactosamine membrane), we evaluated the presence of lysosomes at this evolutive phase of S. mansoni, as well as the influence of PZQ on these acidic organelles and on the tegument of the sporocyst. Although the sporocyst remained alive, it was observed that there was a marked contraction of its musculature, and there occurred a change in the parasite's structure. Also, the acidic vesicles found in the sporocysts showed a larger delimited area after contact of the parasites with PZQ. Damages to the tegument was also observed, as show a well-marked labeling either with Hoechst 33258 or with lectin of Glycine max after contact of sporocysts with the drug. These results could partially explain the interruption/reduction mechanism of cercarial shedding in snails exposed to PZQ.

We tested the attraction of Triatoma dimidiata and T. pallidipennis to traps baited with yeast volatiles. Two traps were simultaneously presented in opposite sides of an experimental arena. One trap presented a yeast culture in sucrose solution, while the other contained sucrose solution as control. A first experimental series was done without offering a central refuge for bugs. In a second series, one shelter where the insects could hide was offered and the traps were presented afterwards. In the first series, yeast baited traps attracted significantly more insects than control ones for both species. In the second series, T. pallidipennis was significantly attracted to yeast, whereas T. dimidiata was not attracted. The potential use of yeast baited traps for capturing these vectors of Chagas disease is discussed.

The enterococci are important nosocomial pathogens with a remarkable capacity of expressing resistance to several antimicrobial agents. Their ubiquitous nature and resistance to adverse environmental conditions take account for their ability to colonize different habitats and for their potential for easy spreading through the food chain. In the present study we evaluated the distribution of species and antimicrobial susceptibility among enterococcal isolates recovered from food obtained in retail stores in Rio de Janeiro, Brazil. The following species were identified among 167 isolates obtained from poultry meat and 127 from pasteurized milk: Enterococcus faecalis (62.6%), E. casseliflavus (17.3%), E. durans (6.5%), E. gallinarum (3.0%), E. gilvus (2.4%), E. faecium (2.0%), E. hirae (1.4%), and E. sulfureus (1.0%). The overall percentages of antimicrobial resistant isolates were: 31.2 % to tetracycline, 23.8% to erythromycin, 11.3% to streptomycin, 4.3% to chloramphenicol, 3.9% to gentamicin, 1.4% to norfloxacin, 1.1% to imipenem, 0.7% to ciprofloxacin, nitrofurantoin, and penicillin, and 0.4% to ampicillin. Intermediate resistance was detected in frequencies varying from 0.5% for linezolid to 58.2% for erythromycin. None of the isolates showed resistance to glycopeptides. High-level resistance to aminoglycosides was observed in 13.1% of the isolates. Multiresistance was observed in E. faecalis, E. casseliflavus, E. faecium, E. gallinarum, E. durans and E. gilvus.
